RESUMO

Uric acid is the end product of purine metabolism in humans due to inactivation of the uricase determined by the mutated uricase gene. Uricase catalyzes the conversion of uric acid into water-soluble allantoin that is easily excreted by the kidneys. Hyperuricemia occurs when the serum concentration of uric acid exceeds its solubility (7 mg/dL). However, modifications to improve the uricase activity is under development for treating the hyperuricemia. Here we designed 7 types of human-porcine chimeric uricase by multiple sequence comparisons and targeted mutagenesis. An optimal human-porcine chimeric uricase mutant (uricase-10) with both high activity (6.33 U/mg) and high homology (91.45 %) was determined by enzyme activity measurement. The engineering uricase was further modified with PEGylation to improve the stability of recombinant protein drugs and reduce immunogenicity, uricase-10 could be more suitable for the treatment of gout and hyperuricemia theoretically.

Based on the reported research, hydroxyl radicals can be rapidly transformed into carbonate radicals in the carbonate-bicarbonate buffering system in vivo. Many of the processes considered to be initiated by hydroxyl radicals may be caused by carbonate radicals, which indicates that lipid peroxidation initiated by hydroxyl radicals can also be caused by carbonate radicals. To date, theoretical research on reactions of hydrogen abstraction from and radical addition to polyunsaturated fatty acids (PUFAs) of carbonate radicals has not been carried out systematically. This paper employs (3Z,6Z)-nona-3,6-diene (NDE) as a model for polyunsaturated fatty acids (PUFAs). Density functional theory (DFT) with the CAM-B3LYP method at the 6-311+g(d,p) level was used to calculate the differences in reactivity of carbonate radicals abstracting hydrogen from different positions of NDE and their addition to the double bonds of NDE under lipid solvent conditions with a dielectric constant of 4.0 (CPCM model). Grimme's empirical dispersion correction was taken into account through the D3 scheme. The energy barrier, reaction rate constants, internal energy, enthalpy and Gibbs free energy changes in these reactions were calculated With zero-point vibrational energy (ZPVE) corrections. The results indicated that carbonate radicals initiate lipid peroxidation primarily through hydrogen abstraction from diallyl carbon atoms. The reaction of hydrogen abstraction from diallyl carbon atoms exhibits the highest reaction rate, with a reaction rate constant approximately 43-fold greater than the second-ranked hydrogen abstraction from allyl carbon atoms. This process has the lowest energy barrier, internal energy, enthalpy, and Gibbs free energy changes, indicating that it is also the most spontaneous process.

In this study, pyrolysis and hydrothermal methods were used for Enteromorpha biochar that was co-modified with l-cysteine and barium titanate (LBCBa). It has great environmental tolerance and can remove 93.0 % of atrazine (ATZ, 10 mg·L-1) within 60 mins of ultrasonic treatment. The enhanced hydrophilicity, electron-donating capability, and piezoelectricity of LBCBa are considered to induce excellent performance. The apparent reaction rate of the LBCBa-2/PMS/ATZ system with ultrasonic was 2.87 times that without ultrasonic. The density functional theory points out that, introducing l-cysteine to carbon edges improves the adsorption of ATZ and peroxymonosulfate (PMS), making PMS easier to activate. This work offered unique insights for fabricating effective catalysts and demonstrated the combination of hydrophilic functional groups and piezoelectricity in improving catalytic performance and stability.

The distinct characteristics of active species produced during the photocatalytic reaction can result in alterations in the degradation routes of organic pollutants with diverse chemical structures. The relationship between the active species and degradation pathways of organic pollutants lacks a direct experimental or characterization method, so in-depth research is still needed to understand the details of their interactions. In this study, sulfur-doped bulk carbon nitride (SBCN) was prepared based on bulk carbon nitride (BCN), and the process of S-doping enhancing the production of O21 was revealed. Through the degradation experiment, the degradation rate of CIP by SBCN reached 91 %, which was higher than that of BCN (66 %). The increase of degradation rate was mainly attributed to the increase of O21. Through the density functional theory (DFT) calculation of CIP and its degradation intermediate, due to the preferential oxidation of CIP by O21, O21 changes the initial degradation direction of CIP, releasing more attack sites for ˙O2-, thereby achieving more efficient degradation of CIP through the synergy of O21 and ˙O2-. In this study, the attack preferences of the active species and their synergistic promotion provide important insights for the efficient photocatalytic degradation of organic pollutants.

Over the past few years, high-valent iron oxo species (Fe(IV)) have shown considerable promise. However, an improved solution is needed for the bottleneck of unsatisfactory electron transfer efficiency in Fe-based catalyst/PMS systems. In this study, Enteromorpha-derived biochar was pyrolyzed with iron and barium titanate (FeBCBa). Under ultrasonic treatment, it removes 94.5% of atrazine (10 mg/L) within 60 min, and is environmentally friendly. BaTiO3's piezoelectricity enhances Fe(IV) production in FeBCBa, resulting in superior performance. In the ultrasonic condition, the apparent reaction rate was 1.42 times higher than in the non-ultrasonic condition. Using density functional theory calculations, it can be shown that due to the Fe dopant, electrons in ATZ's LUMO are more easily transferred to the catalyst's HOMO, which is beneficial for ATZ removal. The results of this study provide new guidance for constructing stable and efficient catalysts for environmental remediation.

Fibroblast growth factor 2 (FGF-2) is not only an angiogenic factor, but also a mitogen for epidermal keratinocytes. FGF-2 has been shown to be positively immunoreactive in the basal layer of psoriatic lesions. In previous work, we used the Escherichia coli (E. coli) expression system to biosynthesize a biologically active anti-FGF-2 nanobody (Nb) screened by phage display technology, but the low yield limited its clinical application. In this study, we aimed to increase the yield of anti-FGF-2 Nb, and evaluate its therapeutic potential for psoriasis by inhibiting FGF-2-mediated mitogenic signaling in psoriatic epidermal keratinocytes. We demonstrated a 16-fold improvement in the yield of anti-FGF-2 Nb produced in the Pichia pastoris (P. pastoris) compared to the  E. coli expression system. In vitro, the FGF-2-induced HaCaT cell model (FHCM) was established to mimic the key feature of keratinocyte overproliferation in psoriasis. Anti-FGF-2 Nb was able to effectively inhibit the proliferation and migration of FHCM. In vivo, anti-FGF-2 Nb attenuated the severity of imiquimod (IMQ)-induced psoriatic lesions in mice, and also improved the inflammatory microenvironment by inhibiting the secretion of inflammatory cytokines (IL-1ß, IL-6, IL-23, and TNF-α), chemokines (CXCL1 and CCL20), and neutrophil infiltration in skin lesions. These were mainly related to the suppression of FGF-2-mediated mitogenic signaling in psoriatic keratinocytes. In conclusion, we have improved the production of anti-FGF-2 Nb and demonstrated the modality of attenuating the abnormal proliferative behavior of psoriatic keratinocytes by inhibiting FGF-2-mediated mitogenic signaling, which offers the possibility of treating psoriasis.

Erythromycin could be used to treat various bacterial infection, but it was harmful to the colonic microflora. Therefore, it is highly desirable to develop a fluorescence probe that could selectively and sensitively detect Erythromycin in pure water. In this work, a fluorescent probe named EHMC, which exhibited aggregation-induced emission (AIE) characteristic in solid state and water/EtOH binary solvent was developed for "turn on" sensing Erythromycin in pure water with high selectivity and sensitivity (detection limit: 1.78 × 10-8 M). Also, there are fewer interference from other antibiotics in the detection process of probe EHMC for Erythromycin. Moreover, probe EHMC could as a portable test strips for highly selective detection of Erythromycin and identification of different concentrations of Erythromycin. In addition, living cells imaging experiments displayed that probe EHMC could detect Erythromycin in A549 cells and BEAS-2B cells successfully. Combined with the theoretical calculation results The sensing mechanisms that the CO in Erythromycin and OH in EHMC formed intermolecular hydrogen bond and further formed new aggregates were confirmed by job' plot, 1H NMR, FT-IR, ESI-MS, DLS and TEM and DFT calculation.

The internal electric field induced by the lattice interfaces in a heterojunction can facilitate charge transfer, thereby improving the photocatalytic performance. However, the details of the relationship between the lattice interfaces and the charge transfer mechanism in heterojunctions remain unclear. In this study, a Bi2WO6/Bi2O2CO3/C3N4 heterojunction (BBC) with an interlaced lattice was prepared, and the role of the interlaced lattice in charge transfer was revealed. Compared to pristine Bi2O2CO3, Bi2WO6, and C3N4, BBC exhibited an increased ciprofloxacin degradation rate constant (0.0573 min-1). A series of experiments were performed to reveal the role of the interlaced lattice interface in the enhanced photocatalytic performance. The results show that the driving force provided by the interlaced lattice interface changes the charge transfer mechanism from a dual Ⅱ-scheme to a dual S-scheme. This work provides profound insights into the effects of lattice interfaces in heterojunctions and the design of efficient photocatalysts.

PURPOSE: Human endothelial growth factor receptor-2 (HER2) is a leucine kinase receptor that is closely related to cell growth and differentiation. It is very weakly expressed in a few epithelial cells in normal tissue. Abnormal expression of HER2 usually leads to sustained activation of downstream signaling pathways, enabling epithelial cell growth, proliferation, and differentiation; this disturbs normal physiological processes and causes tumor formation. Overexpression of HER2 is related to the occurrence and development of breast cancer. HER2 has become a well-established immunotherapy target for breast cancer. We chose to construct a second-generation CAR targeting HER 2 to test whether it kills breast cancer. METHODS: We constructed a second-generation CAR molecule targeting HER2, and we generated cells expressing this second-generation CAR through lentivirus infection of T lymphocytes. LDH assay and flow cytometry were perform to detect the effect of cells and animal models. RESULTS: The result indicated that the CARHER2 T cells could selectively kill cells with high Her2 expression. The PBMC-activated/CARHer2 cells had stronger in vivo tumor suppressive activity than PBMC-activated cells, and administration of PBMC-activated/CARHer2 cells significantly improved the survival of tumor-bearing mice, and induced the production of more Th1 cytokines in tumor-bearing NSG mice. CONCLUSIONS: We prove that the generated T cells carrying the second-generation CARHer2 molecule could effectively guide immune effector cells to identify and kill HER2-positive tumor cells and inhibit tumors in model mice.

BACKGROUND: Improved markers for predicting recurrence are needed to stratify patients with localised (stage I-III) renal cell carcinoma after surgery for selection of adjuvant therapy. We developed a novel assay integrating three modalities-clinical, genomic, and histopathological-to improve the predictive accuracy for localised renal cell carcinoma recurrence. METHODS: In this retrospective analysis and validation study, we developed a histopathological whole-slide image (WSI)-based score using deep learning allied to digital scanning of conventional haematoxylin and eosin-stained tumour tissue sections, to predict tumour recurrence in a development dataset of 651 patients with distinctly good or poor disease outcome. The six single nucleotide polymorphism-based score, which was detected in paraffin-embedded tumour tissue samples, and the Leibovich score, which was established using clinicopathological risk factors, were combined with the WSI-based score to construct a multimodal recurrence score in the training dataset of 1125 patients. The multimodal recurrence score was validated in 1625 patients from the independent validation dataset and 418 patients from The Cancer Genome Atlas set. The primary outcome measured was the recurrence-free interval (RFI). FINDINGS: The multimodal recurrence score had significantly higher predictive accuracy than the three single-modal scores and clinicopathological risk factors, and it precisely predicted the RFI of patients in the training and two validation datasets (areas under the curve at 5 years: 0·825-0·876 vs 0·608-0·793; p<0·05). The RFI of patients with low stage or grade is usually better than that of patients with high stage or grade; however, the RFI in the multimodal recurrence score-defined high-risk stage I and II group was shorter than in the low-risk stage III group (hazard ratio [HR] 4·57, 95% CI 2·49-8·40; p<0·0001), and the RFI of the high-risk grade 1 and 2 group was shorter than in the low-risk grade 3 and 4 group (HR 4·58, 3·19-6·59; p<0·0001). INTERPRETATION: Our multimodal recurrence score is a practical and reliable predictor that can add value to the current staging system for predicting localised renal cell carcinoma recurrence after surgery, and this combined approach more precisely informs treatment decisions about adjuvant therapy. OBJECTIVES: To establish a rapid and nondestructive identification method for human body fluid stains and non-biological stains using three-dimensional fluorescence spectroscopy. METHODS: The collected three-dimensional fluorescence spectrum data of human saliva, 3% blood, coffee and Fanta® stains were processed with dimensionality reduction. After wavelet transform, spectral denoising and feature extraction, the classification formula was established. The Fisher discriminant was used for spectrum matching and recognition to establish the analysis method to distinguish stain types. RESULTS: According to the results of data training and comparison, all the recognition accuracies of Fanta®, coffee, saliva and blood were more than 91.39%. Among them, saliva reached 100% recognition accuracy. CONCLUSIONS: Three-dimensional fluorescence spectroscopy is a potential method for rapid and nondestructive identification of biological and non-biological stains.

Herein, a twisty C-TiO2 /PCN (CNT) Step-scheme (S-scheme) heterojunction is fabricated and applied to degrade ciprofloxacin (CIP) with the assistance of ultrasonic vibration and visible light irradiation. The nitrogen-rich twisty polymeric carbon nitride (PCN) can not only induce a non-centrosymmetric structure with enhanced polarity for a better piezoelectric effect but also provide abundant lone pair electrons to promote n→π* transition during photocatalysis. Its hybridization with C-TiO2 particles can construct S-scheme heterojunction in CNT. During the piezo-photocatalysis, the strain-induced polarization electric field in the heterojunction can regulate the electron migration between the two components, resulting in a more effective CIP degradation. With the synergistic effect of ultrasonic vibration and visible light irradiation, the reaction rate constant of CIP degradation by CNT increases to 0.0517 min-1 , which is 1.86 times that of photocatalysis and 6.46 times that of ultrasound. This system exhibits a stable CIP decomposition efficiency under the interference of various environmental factors. In addition, the in-depth investigation found that three pathways and 12 major intermediates with reduced toxicity are produced after the reaction. Hopefully, the construction of this twisty CNT S-scheme heterojunction with enhanced piezo-photocatalytic effect offers inspiration for the design of environmentally functional materials.

Herein, PDI-g-C3N4/g-C3N4 homojunction has been fabricated via the multiscale modification strategy to enhance photocatalytic atrazine degradation. The morphological scale modification was realized by multistep thermal condensation, where the released gas could act as templates to form the porous structure. The molecular scale modification was achieved by the pyromellitic diimide (PDI) decoration, which could distort the planar nanosheet to induce a porous structure and provide more chromophores for better light absorption. The electronic scale modification was realized by the built-in electric field between the PDI-g-C3N4/g-C3N4 homojunction interfaces, which suppressed the recombination of photocarriers. The PDI-g-C3N4/g-C3N4 strengthened photocatalytic atrazine degradation was well-adapted to different environmental influence interference. The optimum atrazine degradation rate within one hour reached 90% in a strong acidic condition (pH = 3.09). It was found that the highly pH-dependent ATZ removal is related to the H2O2 generation during photocatalysis. Within one hour, the PDI-g-C3N4/g-C3N4 could generate 147.38 µM H2O2. In addition, 1O2, ·O2-, h+, and ·OH were found to have contributed to the ATZ decomposition. During photocatalysis, 14 intermediates and three pathways for atrazine degradation have been found. Hopefully, this study could pave a way for the development of the multiscale modification for the photocatalyst.

Low back pain is common after lumbar spine surgery and the injury from extensive detachment of paraspinal muscles during the surgery may play a vital role. Previously, we prepared a bovine acellular tendon fiber (ATF) material through lyophilization and proved that it could retain its original fibrillar structure and mechanical properties. The objective of this study is to evaluate the effectiveness of this new fiber material used for attachment structure reconstruction of paraspinal muscle. Defect of spinous process, interspinous and supraspinous ligament was established on lumbar spine in rabbit and rat and ATF linear material was implanted to reconstruct the attachment structure. Ultrasound showed the cross-sectional area of the paraspinal muscle in ATF group was larger than that of control group in rats. MRI showed the irregular shape and high signal changes in control group, but regular shape and uniform signal in the ATF group in rabbit. For Electromyogram, the frequency of evoked potential in control group was lower than ATF group and normal rats. HE and Masson staining showed good tissue healing, and immunohistochemical results showed the immune rejection of ATF is significantly lower than that of suture. Reconstruction of the attachment structure of paraspinous muscles with ATF linear material could maintain the morphology, volume and function of paraspinal muscle. ATF material has the potential to be used to manufacture personalized ligaments and other tissue engineering scaffolds. Graphical abstract.

Chronic refractory wounds are one of the complications of diabetes mellitus that require effective therapy. The dermal-wound-healing property of IL-33 in diabetics is little understood. Therefore, this study aimed to express recombinant humanized mature IL-33 (rhmatIL-33) in Escherichia coli BL21 (DE3) and demonstrate its efficacy on dermal wounds in streptozotocin (STZ)-induced diabetic and nondiabetic mice by the dorsal incisional skin wound model. Results revealed that the rhmatIL-33 accelerated the scratch-healing of keratinocytes and fibroblasts at the cellular level. The wounds of diabetic mice (DM) showed more severe ulceration and inflammation than wild-type mice (WT), and the exogenous administration of rhmatIL-33 increased wound healing in both diabetic and wild-type mice. Compared with the up-regulation of endogenous IL-33 mRNA after injury in WT mice, the IL-33 mRNA decreased after injury in DM mice. Exogenous IL-33 administration increased the endogenous IL-33 mRNA in the DM group but decreased the IL-33 mRNA expression level of the WT group, indicating that IL-33 plays a balancing role in wound healing. IL-33 administration also elevated ILC2 cells in the wounds of diabetic and non-diabetic mice and improve the transcript levels of YM1, a marker of M2 macrophages. In conclusion, Hyperglycemia in diabetic mice inhibited the expression of IL-33 in the dermal wound. Exogenous addition of recombinant IL-33 promoted wound healing in diabetic mice by effectively increasing the level of IL-33 in wound tissue, increasing ILC2 cells, and accelerating the transformation of macrophage M1 to M2 phenotype.

Detergent treatment is the most commonly used method for the decellularization of ligaments and tendon grafts. However, it is well recognized that detergent treatment can also adversely affect the extracellular matrix. This study found that discission into the aponeurosis layer of the patellar tendon (PT) before decellularization is conducive to extracting cells from the PT using a low quantity of detergent in a short time period. The acellular aponeurosis discission ligament (AADL) retains its native collagen fibril structure and mechanical properties. Moreover, the PT retained cell and tissue compatibility in vitro and in vivo. After implantation into a defective allogeneic PT, we found that the AADL healed well in the host, and its collagen structure exhibited gradual improvement 12 months after implantation with satisfactory reconstruction. IMPACT: The aponeurosis of tendons/ligaments is the main barrier to achieving complete decellularization, and it thus prevents complete recellularization for applications in tissue engineering. Aponeurosis can obstruct the removal of cell components. We found that excising the aponeurosis before decellularization allows for the removal of cellular components with a reduced amount of detergent, thus improving the biological properties of the acellular ligament. To the best of our knowledge, no similar studies have been performed. Graphical abstract.

BACKGROUND: Small extracellular vesicles (sEVs) are nanometer-sized membranous particles shed by many types of cells and can transfer a multitude of cargos between cells. Recent studies of sEVs have been focusing on their potential to be novel drug carriers due to natural composition and other promising characteristics. However, there are challenges in sEVs-based drug delivery, one of which is the inefficient loading of drugs into sEVs, especially for large biomolecules. RESULTS: In this study, we proposed a membrane-associated protein, milk fat globule-epidermal growth factor 8 protein (MFG-E8), to produce αvß3-targeted sEVs with high delivery efficiency of interested protein. MFG-E8 is a secreted protein with NH2-terminal epidermal growth factor (EGF)-like domains, containing an Arg-Gly-Asp(RGD) sequence that binds αvß3 and αvß5 integrins, and COOH terminal domains C1 and C2, which can bind to lipid membrane with strong affinity. Firstly, we transiently expressed MFG-E8 in HEK293F cells and found that this protein could be secreted and adhere to the cell membrane. The recombinant MFG-E8 is also found to locate at the outer membrane of sEVs. Then we generated engineered sEVs by expressing high levels of the EGFP fused to MFG-E8 in HEK293F cells and showed that MFG-E8 could increase the delivery efficiency of EGFP into sEVs. Further delivery of Gaussia luciferase (GL) by fusion expression with MFG-E8 in donor cells demonstrated that target proteins fused with MFG-E8 still kept their activity. Finally, we identified the sEVs' target to integrin αvß3 by comparing the transfection efficiency with MFG-E8 loaded sEVs (MFG-E8-sEVs) in αvß3 positive cells and αvß3 negative cells. Analysis showed higher target protein could transfect into αvß3 positive cells with MFG-E8-sEVs than with EGFP loaded sEVs (EGFP-sEVs), meaning the engineered sEVs with MFG-E8 not only could increase the delivery of target protein into sEVs, but also could target the αvß3 positive cells. CONCLUSION: This study suggests that recombinant MFG-E8 is an ideal protein to increasingly deliver the drug into sEVs and give sEVs the ability to target the αvß3 positive cells.

Psoriasis is a complex chronic skin inflammatory disease characterized by abnormal proliferation, differentiation of keratinocytes and infiltration of lymphocytes and neutrophils. The tripeptide KdPT, structurally derived from the C-terminal amino acid of alpha-melanocyte-stimulating hormone, has shown a significant anti-inflammatory effect on mild-to-moderate active ulcerative colitis in previous reports. In this research, we investigated whether KdPT could consistently ameliorate disease in a mouse model of imiquimod (IMQ)-induced psoriasis by inhibiting proliferation and inflammation response. We demonstrated that KdPT in vitro significantly inhibited the proliferation of human keratinocytes and endothelial cells, and also downgraded the expression of inflammatory factors in LPS-induced RAW264.7, including IL-6, TNF-α and NO. In vivo, KdPT attenuates the severity of IMQ-induced psoriasis-like phenotype in mice. Such an effect was achieved by downregulating the expression of the inflammatory cytokines interleukin (IL)-6, TNF-α, and the proliferation marker Ki67. These results suggested that KdPT might be useful in the treatment for psoriasis.

In this study, an investigation is conducted into the degradation of nitenpyram (NTP) using highly efficient APMMO/PDS/Vis system. As photocatalysts, silver phosphate (AP) and calcined Zn-Al layered double hydroxides (MMO) exhibit high efficiency in achieving charge separation. Besides, the injection of electrons into peroxydisulfate (PDS) from the APMMO can contribute to obtaining the species in the active state with higher efficiency. Based on the APMMO/PDS/Vis system, 50 mg/L of nitenpyram (NTP, 50 mL) can be completely removed in 60 min using 0.8 g/L photocatalyst and 0.2 g/L PDS under the optimum condition and visible light (780 nm > λ > 420 nm). Meanwhile, as demonstrated under visible light within 30 min, an ultrahigh degradation efficiency can be achieved by NTP based on APMMO1/PDS/Vis system. Besides, the electron paramagnetic resonance (EPR) technique and radical quenching experiments suggested 1O2, h+, SO4-•, •O2-, and •OH are all contributory to the removal of pollutants. Given the outcomes achieved by LC/MS system and mass spectrometry, the primary degradation intermediates of NTP end up being converted into photodegradation products (such as 2-Chloropyridine, 6-Chloropurine Riboside and dl-Leucine). Additionally, there are three potential photodegradation pathways to NTP degradation have been deployed. Moreover, the NTP light degradation occurring in APMMO1/PDS/Vis system is competent under the three types of real water sample. Accordingly, the high-efficiency APMMO1/PDS/Vis system is fit for use in water pollution control for agricultural productions.

The severe pollution caused by antibiotics has raised serious concerns in recent decades. In this study, graphene-like Enteromorpha biochar modified g-C3N4 (BC/CN) was synthesized and applied to degrade tetracycline by activating PMS under visible light, obtaining around 90% removal rate within 1 h. The Enteromorpha biochar can provide electron-withdrawing groups to adjust the electronic structure of g-C3N4, and induces more π-π interaction to decline the recombination of photocarriers. The environmental adaptability of the BC/CN/PMS/vis system was confirmed by the TC degradation in different initial pH, coexisting ions, and natural organic materials. In most cases, the system maintained over 78% degradation rate. The kinetics and mechanism of the system indicating that ∙O2-, 1O2 contributed more to the TC photocatalytic degradation than ∙OH, SO4∙-, and h+. During the process, TC underwent serials hydroxylation, demethylation, and ring-opening processes, and produced more than 40 intermediates in three pathways. Moreover, the BC/CN/PMS/vis system was proved to have at least a 50% degradation rate for more tetracyclines and quinolone antibiotics with the same condition.
