RESUMO

Aqueous zinc-ion batteries (AZIBs) based on vanadium oxides or sulfides are promising candidates for large-scale rechargeable energy storage due to their ease of fabrication, low cost, and high safety. However, the commercial application of vanadium-based electrode materials has been hindered by challenging problems such as poor cyclability and low-rate performance. To this regard, sophisticated nanostructure engineering technology is used to adeptly incorporate VS2 nanosheets into the MXene interlayers to create a stable 2D heterogeneous layered structure. The MXene nanosheets exhibit stable interactions with VS2 nanosheets, while intercalation between nanosheets effectively increases the interlayer spacing, further enhancing their stability in AZIBs. Benefiting from the heterogeneous layered structure with high conductivity, excellent electron/ion transport, and abundant reactive sites, the free-standing VS2/Ti3C2Tz composite film can be used as both the cathode and the anode of AZIBs. Specifically, the VS2/Ti3C2Tz cathode presents a high specific capacity of 285 mAh g-1 at 0.2 A g-1. Furthermore, the flexible Zn-metal free in-plane VS2/Ti3C2Tz//MnO2/CNT AZIBs deliver high operation voltage (2.0 V) and impressive long-term cycling stability (with a capacity retention of 97% after 5000 cycles) which outperforms almost all reported Vanadium-based electrodes for AZIBs. The effective modulation of the material structure through nanocomposite engineering effectively enhances the stability of VS2, which shows great potential in Zn2+ storage. This work will hasten and stimulate further development of such composite material in the direction of energy storage.

RESUMO

Solar-driven interfacial evaporation and purification is a promising solar energy conversion technology to produce clean water or solve water scarcity. Although wood-based photothermal materials have attracted particular interest in solar water purification and desalination due to their rapid water supply and great heat localization, challenges exist given their complicated processing methods and relatively poor stability. Herein, we propose a facile approach for fabricating a bilayered wood-poly(3,4-ethylenedioxythiophene):polystyrene sulfonate (wood-PEDOT:PSS) hydrogel interfacial evaporator by direct drop-casting and dry-annealing. Benefiting from the unique combined merits of the wood-PEDOT:PSS hydrogel evaporator, i.e., excellent light absorption (~99.9%) and efficient photothermal conversion of nanofibrous PEDOT:PSS and the strong hydrophilicity and fast water transport from wood, the as-fabricated bilayered wood-PEDOT:PSS hydrogel evaporator demonstrates a remarkably high evaporation rate (~1.47 kg m-2 h-1) and high energy efficiency (~75.76%) at 1 kW m-2. We further demonstrate the practical applications of such an evaporator for sewage purification and desalination, showing outstanding performance stability and partial salt barrier capability against a continuous 10-day test in simulated seawater and an ultrahigh ion removal rate of 99.9% for metal ion-containing sewage. The design and fabrication of such novel, efficient wood-based interfacial evaporators pave the way for large-scale applications in solar water purification.

RESUMO

As one of major food crops, soybean is grown over a broad ecological region in China with considerable variations in environmental conditions, and the seed compositions of soybeans are diverse among different regions. To clarify the spatial patterns of soybean seed compositions, crude oil, protein, and 11 categories of functional components were quantified in 1792 soybean samples collected from a vast range of soybean planting regions across China spanning from 2010 to 2017. The Kriging interpolation maps presented a clear north-to-south (high latitude to low latitude) increasing trend in contents of crude protein and dietary fiber and decreasing trend in contents of crude oil, phospholipids, saponins, and carotenoids. Soybeans with high-level of total oligosaccharide were concentrated in the central region. Based on the geographical distribution of soybean nutritional components, weather conditions, and cultivation systems, the soybean production areas in China were divided into three regions and 10 subregions. This study highlights the geographic distribution of soybean nutritional compositions and provides scientific evidence for guiding the construction of high-quality edible soybean production bases in China.

RESUMO

Objective: A deep learning algorithm was developed for automatic detection and localization of intracranial aneurysms in DSA, and its clinical characteristics were analyzed, and targeted nursing measures were formulated. Methods: Using a retrospective multicenter study method based on radiology reports, DSA images of aneurysms were randomly divided into 75 cases in the training set, 20 cases in the internal test set, and 35 cases in the external test set. Using a computer-aided detection method based on the three-dimensional U-Net (3D U-Net), after preprocessing DSA images, automatic segmentation of intracranial blood vessels is performed to obtain regions of interest, and based on the segmentation results, physicians' annotations are introduced. The 3D U-Net network model is trained and adjusted, and the obtained model is used to automatically detect the cerebral aneurysm area. Results: Fivefold cross-validation was used for the training set and the internal test set, and a sensitivity of (94.4 ± 1.1)% was obtained. Automatic detection of aneurysms was performed on the external test set, and the average false positive rate was 0.86 FPs/case (false positives/case). The resulting sensitivity was 82.9%. The classification comparison of external test sets showed that the sensitivity of the method for detecting aneurysms with sizes of 5.00~<10.00 mm and ≥10.00 mm (88.2% and 100.0%) was higher than that for aneurysms with sizes of <3.00 mm and 3.00~<5.00 mm (50.0% and 72.7%). The sensitivity of patients aged 50-60 years and >60 years (90.0% and 87.5%) was higher than that of patients aged <50 years (66.7%), and there was little difference between different genders (84.6% in males and 81.8% in females). Conclusion: The deep learning algorithm has high diagnostic performance in detecting intracranial aneurysms, which is verified by external datasets.

RESUMO

Tussilagone is a sesquiterpenoid extracted from Tussilago farfara and is used as an oriental medicine for asthma and bronchitis. Although previous studies have shown that tussilagone has an inhibitory effect on platelet aggregation, no studies have been performed to investigate its precise effect on platelets, and the underlying mechanism remains unclear. In the present study, we showed that tussilagone inhibited platelet aggregation induced by collagen, thrombin and ADP, as well as platelet release induced by collagen and thrombin, in mice. Tussilagone decreased P-selectin expression and αIIbß3 activation (JON/A binding) in activated platelets, which indicated that tussilagone inhibited platelet activation. Moreover, tussilagone suppressed platelet spreading on fibrinogen and clot retraction. The levels of phosphorylated Syk, PLCγ2, Akt, GSK3ß, and MAPK (ERK1/2 and P38) and molecules associated with GPVI downstream signaling were downregulated in the presence of tussilagone. In addition, tussilagone prolonged the occlusion time in a mouse model of FeCl3-induced carotid artery thrombosis and had no effect on mouse tail bleeding time. These results indicate that tussilagone inhibits platelet function in vitro and in vivo and that the underlying mechanism involves the Syk/PLCγ2-PKC/MAPK and PI3K-Akt-GSK3ß signaling pathways downstream of GPVI. This research suggests that tussilagone is a potential candidate antiplatelet drug for the prevention of thrombosis.

RESUMO

An efficient ultrasonic disruption extraction (UDE) of polysaccharides from Armillariamellea (AM) were optimized by response surface methodology (RSM). Under optimum conditions: ultrasonic power 915.00 W, temperature 69.27 °C and time 39.13 min, the crude polysaccharides (AMPs) yield was 19.5%. Two purified fractions AMPs-1-1 and AMPs-2-1 were obtained through anion-exchange and gel chromatography. AMPs-1-1 was a heteropolysaccharide with average molecular weights of 1.23 × 105 Da, and composed of Glc, Gal and GlcA with mole percentages of 89.06%, 9.59% and 1.34%, respectively, owning a backbone structure of (1→)-ß-d-Glcp, (1 â†’ 3,6)-α-d-Glcp and (1 â†’ 3)-ß-d-Glcp residues. AMPs-2-1 was a heteropolysaccharide with average molecular weights of 6.76 × 104 Da, and composed of Glc, Gal, GlcA and Man with mole percentages of 65.28%, 22.87%, 2.87% and 8.98%, containing a main backbone chain of (1 â†’ 3,6)-α-d-Glcp and (1 â†’ 6)-ß-d-Glcp residues. AMPs-2-1 possessed obviously antioxidant activities in terms of stronger scavenging activity against DPPH· and ABTS+⋅, higher FRAP and ORAC value than AMPs-1-1. AMPs-2-1 could promote splenocyte lymphocytes and RAW264.7 macrophages proliferation and enhanced the phagocytosis of macrophages, exhibited significant immunomodulatory activities. These results suggested that UDE is an effective extract technology, and AMPs-2-1 could be explored as potential natural antioxidants and immunomodulatory agents.

RESUMO

Neferine has long been recognized as a medicinal herbal ingredient with various physiological and pharmacological activities. Although previous studies have reported its antithrombotic effect, the underlying mechanisms have not been thoroughly investigated. Since platelets play a key role in thrombosis, we investigated the effects of neferine on human platelet function and the potential mechanisms. Platelet aggregation, adhesion and spreading were performed to investigate the effect of neferine on inhibition of platelet function. Flow cytometry was used to determine platelet alpha granule secretion and integrin IIb/IIIa activation, as detected by CD62P (P-selectin) expression, PAC-1 and fibrinogen binding. Western blotting was utilized to investigate the effect of neferine on intracellular signaling of activated platelet. We found that neferine significantly suppressed platelet aggregation and remarkably promoted the dissociation of platelet aggregates induced by collagen, thrombin, U46619, ADP and adrenaline in a dose-dependent manner. Flow cytometry analysis showed that neferine inhibited thrombin-induced platelet P-selectin expression, PAC-1 and fibrinogen binding. In addition, neferine reduced the adhesion of human platelets on coated collagen under both static and shearing condition at an arterial shear rate of 40 dyne/cm2. Neferine also inhibited the spreading of human platelets on immobilized fibrinogen. Western blot analysis showed that neferine inhibited PI3K activation, and decreased the levels of phosphorylation of Akt, GSK3ß and p38 MAPK in platelets. In summary, neferine has the potential to be an antiplatelet and antithrombotic agent by inhibiting the PI3K-Akt-GSK3ß/p38 MAPK signaling pathway.

RESUMO

Salvia miltiorrhiza Bunge contains various active constituents, some of which have been developed as commercially available medicine. Moreover, some other ingredients in Salvia miltiorrhiza play roles in anti-platelet activity. The aim of the present study was to investigate the effects and the underlying mechanism of miltirone, a lipophilic compound of Salvia miltiorrhiza Bunge. The ability of miltirone to modulate platelet function was investigated by a variety of in vitro and in vivo experiments. Platelet aggregation and dense granule secretion induced by various agonists were measured with platelet aggregometer. Clot retraction and spreading were imaged by digital camera and fluorescence microscope. Ferric chloride-induced carotid injury model and pulmonary thromboembolism model were used to check miltirone antithrombotic effect in vivo. To elucidate the mechanisms of anti-platelet activity of miltirone, flow cytometry and western blotting were performed. Miltirone (2, 4, 8 µM) was shown to suppress platelet aggregation, dense granule, and α granule secretion in a dose-dependent manner. Meanwhile, miltirone inhibited the clot retraction and spreading of washed platelets. It reduced the phosphorylation of PLCγ2, PKC, Akt, GSK3ß and ERK1/2 in the downstream signal pathway of collagen receptor. It also reduced the phosphorylation of Src and FAK in the integrin αIIbß3-mediated "outside-in" signaling, while it did not suppress the phosphorylation of ß3. In addition, miltirone prolonged the occlusion time and reduced collagen/epinephrine-induced pulmonary thrombi. Miltirone suppresses platelet "inside-out" and "outside-in" signaling by affecting PLCγ2/PKC/ERK1/2, PI3K/Akt, and Src/FAK signaling. Therefore, miltirone might represent a potential anti-platelet candidate for the prevention of thrombotic disorders.

RESUMO

Platelets in the primary tumor microenvironment play crucial roles in regulating tumor growth, metastasis, and angiogenesis, but the underlying mechanisms are unclear. Here, we show that platelet releasates exhibited a proliferative effect on HeLa cells, and this effect correlated with a reduction of KLF6 expression. After incubation with either washed human platelets or collagen-related peptide (CRP) activated platelet releasates, expression of KLF6 in the HeLa cervical tumor cell line was markedly reduced. However, no significant difference was observed between control HeLa cells and HeLa cells incubated with resuspended activated platelet pellet. Moreover, the platelets' promoting effect on HeLa cell growth was significantly abolished in KLF6 silenced HeLa cells. In addition, blocking TGF-ß signaling with SB431542, a TGF-ß receptor inhibitor, also counteracted the effect of platelets on proliferation and KLF6 expression in HeLa cells. From these findings, we conclude that platelet derived TGF-ß promotes proliferation of HeLa cells by decreasing the expression of KLF6. The discovery that KLF6 is a key target of platelet-derived TGF-ß signaling in HeLa cells identifies a potential new therapeutic target for the prevention and treatment of cervical carcinoma.

RESUMO

Water shortage and low phosphorus (P) availability limit yields in soybean. Roots play important roles in water-limited and P-deficient environment, but the underlying mechanisms are largely unknown. In this study we determined the responses of four soybean [Glycine max (L.) Merr.] genotypes [Huandsedadou (HD), Bailudou (BLD), Jindou 21 (J21), and Zhonghuang 30 (ZH)] to three P levels [applied 0 (P0), 60 (P60), and 120 (P120) mg P kg-1 dry soil to the upper 0.4 m of the soil profile] and two water treatment [well-watered (WW) and water-stressed (WS)] with special reference to root morphology and architecture, we compared yield and its components, root morphology and root architecture to find out which variety and/or what kind of root architecture had high grain yield under P and drought stress. The results showed that water stress and low P, respectively, significantly reduced grain yield by 60 and 40%, daily water use by 66 and 31%, P accumulation by 40 and 80%, and N accumulation by 39 and 65%. The cultivar ZH with the lowest daily water use had the highest grain yield at P60 and P120 under drought. Increased root length was positively associated with N and P accumulation in both the WW and WS treatments, but not with grain yield under water and P deficits. However, in the WS treatment, high adventitious and lateral root densities were associated with high N and P uptake per unit root length which in turn was significantly and positively associated with grain yield. Our results suggest that (1) genetic variation of grain yield, daily water use, P and N accumulation, and root morphology and architecture were observed among the soybean cultivars and ZH had the best yield performance under P and water limited conditions; (2) water has a major influence on nutrient uptake and grain yield, while additional P supply can modestly increase yields under drought in some soybean genotypes; (3) while conserved water use plays an important role in grain yield under drought, root traits also contribute to high nutrient uptake efficiency and benefit yield under drought.

RESUMO

From 2010 to 2013, 763 soybean samples were collected from an extensive area of China. The correlations between seed compositions and climate data were analyzed. The contents of crude protein and water-soluble protein, total amount of protein plus oil, and most of the amino acids were positively correlated with an accumulated temperature ≥15 °C (AT15) and the mean daily temperature (MDT) but were negatively correlated with hours of sunshine (HS) and diurnal temperature range (DTR). The correlations of crude oil and most fatty acids with climate factors were opposite to those of crude protein. Crude oil content had a quadratic regression relationship with MDT, and a positive correlation between oil content and MDT was found when the daily temperature was <19.7 °C. A path analysis indicated that DTR was the main factor that directly affected soybean protein and oil contents. The study illustrated the effects of climate factors on soybean protein and oil contents and proposed agronomic practices for improving soybean quality in different regions of China. The results provide a foundation for the regionalization of high-quality soybean production in China and similar regions in the world.

RESUMO

The potato/soybean intercropping trials using three soybean varieties including Zhonghuang 30 (early-maturing variety) , Jidou 17 (mid maturing variety) and Qihuang 34 (late maturing variety) with the sole cropping potato as control were carried out to determine the dynamic changes of leaf area index (LAI) of soybean, accumulation of dry matter, photosynthetic characteristics, yield and yield components. The results showed that the LAI, dry matter accumulation, net photosynthesis (Pn), transpiration rate (Tr) and stomatal conduction (g(s)) of soybean in all intercropping systems were lower than in monoculture because of the influence of intensified lower light during earlier growing stage, and the duration from planting to flowering was extended. When the potato was harvested, the LAI, dry matter accumulation, Pn, Tr and g(s) of soybean in all intercropping systems increased, especially for mid-maturing and late-maturing varieties, which became much closer to those in the monoculture. Compared with sole cropping, the pods per plant, seeds per plant and seeds per pod in intercropping system significantly decreased by 22.0%, 36.0% and 17.6% for early-maturing soybean, 5.1%, 13.1% and 8.9% for mid-maturing soybean, 5.7%, 7.6% and 2.1% for late-maturing soybean, respectively. The yields of mid-maturing and late-maturing varieties in intercropping systems were higher than that of the early-maturing, which increased by 92.4% and 163.4%, with the land equivalent ratio (LER) of 1.81 and 1.84, respectively. This suggested that mid-maturing and late-maturing soybean varieties were suitable for intercropping with the potato to improve photosynthetic efficiency, dry matter accumulation and yield of intercropping soybean.
