On-farm hatching is a relatively new method in the broiler industry, in which fertilized broiler eggs are transported to the farms at the stage of 17-19 days of incubation. Once hatched, the broiler chicks have direct access to feed and water. Previous studies have shown on-farm hatching to increase animal welfare and intestinal development. However, no studies have yet aimed to quantify and compare the antimicrobial use in on-farm hatched flocks with that of traditionally hatched flocks. In this study, information on antimicrobial use (AMU) was collected from 211 Belgian conventional broiler farms, including data from 2244 traditionally hatched flocks and 227 on-farm (NestBorn) hatched flocks. On-farm hatched flocks had significantly (p < 0.001) more antimicrobial-free flocks (n = 109, 48.01%) compared to traditional flocks (n = 271, 12.08%) and a 44% lower (p < 0.01) treatment incidence (TI) at flock level (TI 8.40 vs. TI 15.13). Overall, the farms using traditional hatching had 5.6 times (95% CI 3.6-8.7) higher odds to use antimicrobials than the farms using on-farm hatching. Treated on-farm hatched flocks received three times less lincomycin-spectinomycin (linco-spectin) and less (routine) treatments at the start of the production round. However, both traditional and on-farm flocks experienced outbreaks later in the production round. These results show that on-farm hatching can contribute to the reduction in antimicrobial use in conventional broiler production.

Reduced and responsible antimicrobial use leads to a lower risk of developing antimicrobial resistance. Raised Without Antibiotics (RWA) is a certification label that is recognized in only a few countries, and it is often unclear what the specific criteria and characteristics of RWA farms are. The objectives of this study were to describe the criteria for a Belgian RWA program; to coach farms towards reduced antimicrobial usage (AMU); to assess if it was possible to obtain and maintain the RWA status; and to determine differences between RWA and conventional pig farms. Pig farms (n = 28) were visited three times for the following reasons: (1) data collection, (2) farm-specific coaching (2 months later), and (3) evaluation (7 months later). AMU was followed from before the start of the study up to one year after the last visit. AMU, biosecurity (Biocheck.UGentTM), and farm characteristics of (non-)RWA farms were compared. RWA was defined as no antibiotics from birth until slaughter. Pigs requiring individual treatment received a special ear tag and were excluded from the program. The status of the farms varied over time, and the distribution of RWA vs. non-RWA was 10-18, 13-15, and 12-16, before intervention, after coaching, and after one year, respectively. For the non-RWA farms, there was a reduction in AMU of 61%, 38%, and 23%, for the suckling piglets, fattening pigs, and sows, respectively, indicating that they were moving toward the RWA status. There were no significant differences in biosecurity status between RWA and non-RWA farms, but biosecurity improved in all farms throughout the study. RWA farms were smaller (median 200 sows) compared to non-RWA farms (median 350 sows). The 4-week system was used more in non-RWA farms, while the 3- and 5-week systems were used most often in RWA farms. This study showed that farmers could achieve and maintain the RWA status through farm-specific coaching related to prudent AMU and improved biosecurity.

This study presents a case of clubbed down syndrome in conventional broilers. During the first week of life, severe growth retardation was observed in approximately 25% of the flock. The growth-retarded chicks weighed only 45 g and showed a typical feather disorder which was most apparent on their abdomen and was defined in literature as typical for clubbed down syndrome. Necropsies, histology, biochemical analysis of blood and liver samples, serology and different PCR tests were performed in broilers to assess the aetiology of the clinical signs that were present in the affected broiler farm. Because of the suspicion of a possible link with the broiler-breeder farms, different investigations including serology, PCR and feed analysis were also performed on these farms. The results suggest that an accidentally excessive amount of calcium and iron in the feed of broiler-breeders, 3 weeks prior to first clinical signs in broilers, led to the development of clubbed down in the offspring, because of a relative Zn-deficiency in broiler-breeders and an absolute Zn-deficiency in the hatching eggs that were produced during this period. This appeared to be a reversible process as no clinical signs were observed in younger offspring of these broiler-breeders after they had consumed more of the new batch of feed. A potential involvement of Astrovirus could not be completely ruled out. This study demonstrates the importance of correct mineral concentrations in broiler-breeder feed and the impact it can have on the development of the offspring.

A multifactorial approach using environmental, performance, health and welfare parameters was used to investigate the numerous associations of ventilation throughout three consecutive fattening batches (08/2015 to 12/2016) in a farrow-to-finish commercial pig farm in Belgium. Two fattening pig units were used, unit A (1256 pigs) with mechanical ventilation and unit B (1264 pigs) with natural ventilation. Animal genetics, nutrition, stocking density and health management were the same for both units. Key environmental indicators were monitored in real-time (temperature, humidity, CO2 and NH3) and the daily prevalence of respiratory disease cases was recorded to monitor the temporal expression of disease over time within a farm environment. The welfare status of the animals was assessed twice per production round (batch) with a simplified version of the Welfare quality® protocol. Serological tests for the most prevalent respiratory infectious agents (Mycoplasma hyopneumonniae, swine influenza virus (subtypes H1N1, H1N2 and H3N2), Actinobacillus pleuropneumoniae serotypes 1-2-9-11 and porcine circovirus type 2) were performed during the first, second and final third of each fattening period. Finally, key performance parameters were calculated (average daily growth, feed conversion ratio) and upon slaughter lungs from pigs from unit A (n: 782) and from unit B (n: 544) were assessed for the presence of lung lesions, pleurisy and fissures. To identify the associations of ventilation on the aforementioned parameters, statistical models were run that also included other factors (when applicable) namely production batch, season, age and sampling. Overall, the use of natural ventilation was associated with a less optimal environment with regards to thermal comfort (p < 0.001), CO2 (p < 0.001) and NH3 (p < 0.001). A higher daily prevalence of respiratory disease cases was seen in the naturally-ventilated unit (p < 0.001). Concerning the sero-prevalence of the infectious agents tested, the odds to have a positive H1N1 sample were 3.17 higher in the naturally-ventilated unit (p = 0.003). From the visual assessment of the lungs no statistically significant associations were seen between ventilation type and the presence of lesions, fissures or pleuritis. Yet, the lung lesion score was expected to be lower in the naturally-ventilated unit (p = 0.010). Regarding performance parameters, feed conversion ratio and average daily gain were overall better in the mechanically-ventilated unit (descriptive results). Finally, a better welfare score was seen in the mechanically-ventilated unit in all three production batches (descriptive results). In conclusion, the mechanically-ventilated farm was associated with better environmental conditions for the fattening pigs. Yet, further research is needed to reach definite causal claims.

BACKGROUND: Conventional broilers are currently one of the most efficient protein converters. Although decades of progress in genetic selection and feed formulation have lead to high standards of efficient broiler production, still a lot of variability is found between farms and between successive flocks. The aim of this study was to investigate risk- and/or protective factors for poor health and performance in conventional broiler-farms in Europe by developing eight multivariable linear mixed models. Three different models were used to investigate mortality (overall, first week, after first week), three models for performance variables (growth, feed conversion, European production index) and two models were related to slaughterhouse data (i.e. dead on arrival and condemnation rate). RESULTS: Several factors related to management and housing were significantly associated with health and performance of broilers. The following factors were associated with increased mortality: floor quality, neonatal septicemia, ventilation type and other professional activities of the farmer. The factors associated with performance were chick sex, coccidiosis infections, necrotic enteritis, dysbacteriosis, light intensity adaptations, ventilation type, comparing daily flock results with previous flock results by farmer, daily check of feed and water system and type of feed. For dead on arrival three risk factors were identified i.e. daily growth, type of light adaptation and type of drinkers system. For condemnation rate seven risk factors were found, i.e. type of drinking system, daily growth, feed withdrawal time, type of ventilation, house size, septicemia after seven days and type of feed. CONCLUSIONS: These results imply that a multifactorial approach is required with adaptations involving both improvements in management, housing, health programs and an increasing level of professionalism of the farmer in order to improve broiler performance and health.

Necrotic enteritis (NE) caused by Clostridium perfringens is commonly reported in broilers. Recently, increased NE prevalence in layer breeds was reported in the Indian subcontinent. NE is also frequently observed by veterinary practitioners in Europe, mainly during the pullet rearing phase. In this study, data from layer pullet flocks in Belgium over a 5-year period (2013-2017) were used to assess the incidence of NE and identify potential risk factors for NE in layer pullets. NE was observed in 26% of the layer pullet flocks receiving veterinary intervention. This accounts for an overall estimated NE incidence of 12.3% in Belgian layer pullet flocks. Occurrence of NE was significantly associated with coccidiosis, with flocks being diagnosed with coccidiosis being two-fold more likely to develop NE. Additionally, birds kept in aviary houses were less prone to NE than flocks reared in floor systems or enriched cages. At necropsy, necrotic lesions in the small intestine were comparable to NE in broilers. A single strain of C. perfringens was isolated from the necrotic lesions of three different birds from the same flock; however, no NetB could be detected.

Coccidiosis in broiler chickens, caused by infection with Eimeria spp. remains one of the most economically important production diseases. Development of a genetic biomarker panel of sub-clinical infection would be an important biological tool for the management of broiler flocks. We analysed expression of MicroRNAs (miRNAs) to determine the potential for these in diagnosing coccidiosis in broiler flocks. miRNA expression, in the ilea of Ross 308 broilers, was compared between chickens naturally clinically or sub-clinically infected with Eimeria maxima and Eimeria acervulina using NextSeq 500 sequencing. 50 miRNAs with greatest coefficient of variance were determined and principal component analysis showed that these miRNAs clustered within the clinical and sub-clinical groups much more closely than uninfected controls. Following false detection rate analysis and quantitative PCR we validated 3 miRNAs; Gallus gallus (gga)-miR-122-5p, gga-miR-205b and gga-miR-144-3p, which may be used to diagnose sub-clinical coccidiosis.

We analysed intestinal tissues from groups of fast growing (Ross 308) broilers with natural or experimental coccidiosis, by genomic microarray. We identified genes that were differentially expressed (DE) in all groups and analysed expression of a panel of these, by qPCR, in Ross 308 and slow growing (Ranger classic) broilers, infected with 2500 or 7000 oocysts of Eimeria maxima for 6 or 13 days post-infection (dpi). Four genes (ADD3, MLLT10, NAV2 and PLXNA2) were upregulated (P <0.05) in Ross 308 but were not DE in Ranger Classic at 6 dpi with 2500 oocysts. Six genes (PTPRF, NCOR1, CSF3, SGK1, CROR and CD1B) were upregulated (P <0.05) in both Ross 308 and Ranger Classic infected with 2500 oocysts at 6 dpi but were not DE at 6 dpi with 7000 oocysts. At 13 dpi with 7000 oocysts, NAV2 and NCOR1 were upregulated in Ross 308 (P <0.05) and PTPRF was upregulated in both genotypes (P <0.05). DE of immune genes within the biomarker panel also occurred, with CSF3 upregulated in both genotypes infected with 2500 oocysts at 6 dpi and in Ranger Classic infected with 7000 oocysts, at 6 and 13 dpi (P <0.05). IL-22 was down-regulated in Ranger Classic infected with 2500 or 7000 oocysts at 6 dpi (P <0.05) but upregulated in both genotypes at 13 dpi (P <0.05). CD72 was down-regulated in Ranger Classic infected with 2500 oocysts at 6 dpi and with 7000 oocysts at 6 and 13 dpi (P <0.05). CD72 was upregulated in Ross 308 infected with 2500 oocysts at 6 dpi but was down-regulated following infection with 7000 oocysts at 13 dpi (P <0.05). In conclusion, differential gene expression occurs in fast and slow growing broiler genotypes with coccidiosis. In addition, we highlight a potential genetic biomarker panel for early diagnosis of coccidiosis.

Production diseases are often of multi-factorial origin in which environment (housing, nutrition and management) health and reproductive challenges show complex interactions. The aim of this study was to identify specific environment-related factors and to discuss their associations with health, welfare and reproductive performance in sows and piglets, in diverse systems using data from 130 farms from five EU countries. Two sets of data were used: a) a questionnaire was developed for sows and piglets covering farm management, interventions and housing and b) farm production data covering various performance parameters. Eight parameters were further selected, four of which were related to sow reproductive performance (litter index, replacement rate, repeat breeding (i.e. failure to breed after one mating), weaning to first mating interval) and the remaining four to litter / piglet health performance (piglets born alive per litter, piglets born dead per litter, preweaning mortality rate and weaned piglets per litter). Univariable and multivariable linear models were employed to identify risk factors. Associations were considered significant if P ≤ 0.007 (a criterion of p ≤ 0.05 corrected for the number of parameters tested). Various risk and protective factors were identified for each tested outcome variable. Country effects were included in all models as a fixed factor. Adjusted R-squared values for the multivariable models varied between 9.6% (preweaning mortality) and 66% (litter index). Litter index (litters/sow/year) was negatively associated with a higher weaning age of the piglets. Housing recently weaned sows to be inseminated in a separate unit from the gestation unit had a positive association with litter index. Repeat breeding was negatively associated with PRRS-free farms, farms that bred (raised) all gilts on the farm and farms that perform farrowing induction of sows. PRRS-free farms were also associated with a higher replacement rate. Farms that bred gilts on the farm and PRRS-free farms were negatively associated with preweaning mortality. Natural ventilation in the gestation unit was associated with fewer piglets born alive and with fewer weaned piglets. Closed type of farms was associated with less piglets born dead. The use of open box housing system for pregnant sows (provision of individual resting areas) was associated with more weaned piglets. In conclusion, several factors related to applying good farm and health management, and optimal housing conditions showed positive association with various sow and piglet performance parameters. Further studies will help to assess causal links for these factors.

This study aimed to investigate a possible involvement of mycotoxins in neonatal tail necrosis in piglets. Ten affected and 10 non-affected farms were selected. Sow feed samples were analysed for the presence of 23 mycotoxins by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Blood plasma samples of sows and their piglets were analysed for the presence of deoxynivalenol (DON), de-epoxydeoxynivalenol, T-2 and HT-2 toxin, zearalenone, alfa-zearalenol, and beta-zearalenol, using LC-MS/MS. Additionally, high-resolution mass spectrometry (HRMS) was performed to detect DON-glucuronide (DON-Glca). There was a significant difference between case herds and control herds for mean DON concentrations in feed and sow plasma. For piglet samples, concentrations of DON were above the limit of quantification of 0.1 ng/ml in only 12 samples. Positive correlations were found between DON concentrations in sow feed and plasma of sows; DON concentration in sow feed and DON-Glca concentration in plasma of sows; and between DON and DON-Glca concentration in sow-plasma. In conclusion, high prevalence of DON in feed samples was found, with significantly higher concentrations in case herds, as well as the presence of DON and DON-Glca in sow plasma. Additional research is needed to identify risk factors, including within-herd factors, associated with neonatal tail necrosis in piglets.

BACKGROUND: Infections with encephalomyocarditis virus may cause myocarditis and sudden death in young pigs and reproduction disorders in sows. The presence of encephalomyocarditis virus infected rodents is considered a major risk factor for transmission of the virus to pigs. There is currently no effective treatment. Tightening up biosecurity, applying effective rodent control and reducing stress are the main control measures. CASE PRESENTATION: Two farrow-to-finish herds suffering from problems with sudden death are presented. In herd A, suckling piglets from 3 to 12 days old were dying acutely whereas in herd B, piglets at the end of the nursery period (8-10 weeks) were showing identical problems. A presumptive diagnosis of encephalomyocarditis virus infection was made because typical lesions were observed in some of the affected pigs. These lesions were not always present in pigs dying acutely or in some cases the lesions were very subtle. Therefore other causes had to be ruled out based upon clinical history, clinical signs and diagnostic tests. A conclusive diagnosis was finally established by showing encephalomyocarditis virus in heart tissue using conventional gel-based polymerase chain reaction tests. The real-time PCR test that gave initially negative result was further optimized to avoid false negative results. CONCLUSIONS: Typical lesions are not always present in piglets infected with encephalomyocarditis virus, indicating the importance of examining multiple animals. Problems in suckling piglets may occur in affected herds without reproductive problems in sows. Transmission routes of EMCV in swine are not fully understood. A stand-empty period following thorough cleaning and disinfection is recommended for controlling EMC virus infections.
