RESUMO

Particle image velocimetry is an important optical flow diagnostic tool due to its capacity for investigating a whole flow field without introducing disturbances. However, a significant drawback of PIV methods is their requirement for optical access, making capturing data in closed cavities and confined spaces extremely challenging. A potential approach to overcome this difficulty is miniaturising the system and placing the optical components inside the model. Conventional cross-correlation PIV methods do not allow this due to the size of current PIV cameras. In this study, a miniaturised autocorrelation-based stereo PIV system, which is volumetrically 1.2% of the conventional PIV cameras, was developed and tested. The miniature system is compared with a conventional stereo PIV in wind tunnel experiments up to 16 m/s free stream velocity and a 1.6% velocity difference is observed in the boundary layer flow. Despite a comparatively slow measurement rate of 4.5 Hz, the miniature PIV system demonstrates the ability to measure inside confined spaces and cavities and the ability to be mounted on board models and vehicles. However, limitations remain around conducting measurements with large velocity ranges and with regions of reversed flow due to the challenge of resolving a velocity of 0 m/s.

BACKGROUND: Acidic phytohormones are small molecules controlling many physiological functions in plants. A comprehensive picture of their profiles including the active forms, precursors and metabolites provides an important insight into ongoing physiological processes and is essential for many biological studies performed on plants. RESULTS: A high-throughput sample preparation method for liquid chromatography-tandem mass spectrometry determination of 25 acidic phytohormones classed as auxins, jasmonates, abscisates and salicylic acid was optimised. The method uses a small amount of plant tissue (less than 10 mg fresh weight) and acidic extraction in 1 mol/L formic acid in 10% aqueous methanol followed by miniaturised purification on reverse phase sorbent accommodated in pipette tips organised in a 3D printed 96-place interface, capable of processing 192 samples in one run. The method was evaluated in terms of process efficiency, recovery and matrix effects as well as establishing validation parameters such as accuracy and precision. The applicability of the method in relation to the amounts of sample collected from distantly related plant species was evaluated and the results for phytohormone profiles are discussed in the context of literature reports. CONCLUSION: The method developed enables high-throughput profiling of acidic phytohormones with minute amounts of plant material, and it is suitable for large scale interspecies studies.

Sample preparation is an essential step focused on eliminating interfering compounds while pre-concentrating the analytes. However, its multiple steps are laborious, time-consuming, and a source of errors. Currently, automated approaches represent a promising alternative to overcome these drawbacks. Similarly, miniaturisation has been considered an ideal strategy for creating greener analytical workflows. The combination of these concepts is currently highly desired by analytical chemists. However, most automated and miniaturised sample preparation techniques are primarily concerned with liquid samples, while solids are frequently overlooked. We present an approach based on a cartridge packed with solids (soil samples) coupled with a capillary LC-MS, combining sample preparation and analytical steps into a unique platform. As a proof-of-concept, nine pesticides used in sugarcane crops were extracted and analysed by our proposed method. For optimisation, a fractional factorial design (25-1) was performed with the following variables: aqueous dilution of the sample (V1), extraction strength (V2), matrix washing time (V3), extraction flow (V4), and analytical flow (V5). After, the most influential ones (V1, V2, and V3) were taken into a central composite design (23) to select their best values. Under optimised conditions, the method reported linear ranges between 10 and 125 ng g-1 with R2 > 0.985. Accuracy and precision were in accordance with the values established by the International Council for Harmonisation (Q2(R1)). Therefore, the proposed approach was effective in extracting and analysing selected pesticides in soil samples. Also, we carried out initial qualitative tests for pesticides in honeybees to see if there is the possibility to apply our method in other solids.

Pattern hair loss (PHL) is a chronic regressive condition of the scalp, where follicular miniaturisation and decreased scalp hair coverage occurs in affected areas. In all PHL cases, there is a measurable progressive shortening of the terminal hair growth duration, along with reduced linear growth rates. In both genders, PHL initially shows an increase in short telogen hairs ≤30 mm in length, reflecting a cycle completion of under 6 months in affected terminal hair follicles. To understand the miniaturisation process, we re-examine the dynamics of miniaturisation and ask the question, "why do miniaturised hair follicles resist treatment?" In the light of recent developments in relation to hair regeneration, we looked back in the older literature for helpful clues "lost to time" and reprise a 1978 Hermann Pinkus observation of an array of elastin deposits beneath the dermal papilla following subsequent anagen/telogen transitions in male balding, originally described by Arao and Perkins who concluded that these changes provide a "morphologic marker of the entire biologic process in the balding scalp." Thus, we have reviewed the role of the elastin-like bodies in hair pathology and we propose that alterations in elastin architecture may contribute to the failure of vellus-like hair reverting back to their terminal status and may indicate a new area for therapeutic intervention.

Molecular heterogeneity exists at different spatial scales in biological samples and is an important parameter in the development of pathologies and resistances to therapies. When aiming to reach molecular heterogeneity of cells at extremely low spatial scales, single-cell analysis can be the ultimate choice. Proteomics performed in bulk population of cells (macroproteomics) is prone to mask molecular heterogeneity. Mass spectrometry-based single cell proteomics (SCP-MS) is the right solution to overcome this issue. Three main problems can be identified using SCP-MS: (i) analytical loss during sample preparation, (ii) inefficient microinjection/delivery of proteins/peptides from samples to MS and (iii) low analytical throughput. Technologies for automation of SCP have recently gained attention to improve methods accuracy, sensitivity, throughput and in-depth and low-biased proteome analysis. In this minireview, we therefore overview the state-of-the-art of automation of SCP-MS sample preparation approaches.

This study presents the doping of higher alkanes, namely, pentadecane (C15) and hexadecane (C16), with ZnS:Mn nanoparticles to create new types of in-line optical fiber sensors with unique optical properties. In this research, the phenomenon of light beam leakage out of the taper and its interaction with the surrounding materials is described. The fabricated new materials are used as cladding in a tapered optical fiber to make it possible to control the optical light beam. The manufactured sensor shows high sensitivity and fast response to the change in the applied materials. Results are presented for a wide optical range of 1200-1700 nm with the use of a supercontinuum source and an optical spectrum analyzer, as well as for a single wavelength of 800 nm, corresponding to the highest transmitted power. The results present a change in the optical property dependence on the temperature in the cooling and heating process. For all materials, the measurements in a climatic chamber are provided between 0 and 40 °C, corresponding to the phase change of the alkanes from solid to liquid. The addition of nanoparticles to the volume of alkanes is equal to 1 wt%. To avoid a conglomeration of nanoparticles, the anti-agglomeration material, Brij 78 P, is used.

Saccharomyces cerevisiae cells grown in a small volume of chemically defined media neither reach the desired cell density nor grow at a fast enough rate to scale down the volume and increase the sample number of classical biochemical assays, as the detection limit of the readout often requires a high number of cells as an input. To ameliorate this problem, we developed and optimised a new high cell density (HCD) medium for S. cerevisiae. Starting from a widely used synthetic medium composition, we systematically varied the concentrations of all components without the addition of other compounds. We used response surface methodology to develop and optimise the five components of the medium: glucose, yeast nitrogen base, amino acids, monosodium glutamate, and inositol. We monitored growth, cell number, and cell size to ensure that the optimisation was towards a greater density of cells rather than just towards an increase in biomass (i.e., larger cells). Cells grown in the final medium, HCD, exhibit growth more similar to the complex medium yeast extract peptone dextrose (YPD) than to the synthetic defined (SD) medium. Whereas the final cell density of HCD prior to the diauxic shift is increased compared with YPD and SD about threefold and tenfold, respectively. We found normal cell-cycle behaviour throughout the growth phases by monitoring DNA content and protein expression using fluorescent reporters. We also ensured that HCD media could be used with a variety of strains and that they allow selection for all common yeast auxotrophic markers.

A robust, portable and miniature battery powered gradient capillary liquid chromatograph (total weight ∼2.7 kg, without battery ∼2.0 kg), with integrated microfluidic injection, column heating and high sensitivity low-UV absorbance detection is presented. The portable capillary chromatograph, was applied with a packed reversed-phase capillary column (100 mm × 300 µm I.D., 5 µm ODS), housed within an integrated capillary column heater controlled by a proportional-integral-derivative (PID) chip module. The system delivered retention time and peak area relative standard deviation in isocratic mode of <0.7% (n = 10) and <3.3% (n = 10), respectively, and <0.1% (n = 10) and <2.3% (n = 10) respectively, for gradient elution mode. Detection was based upon a 255 nm light-emitting diode (LED) using one of two commercial capillary flow-cell options, namely a high sensitivity 12 nL Agilent capillary z-cell (HSDC) and a 45 nL Thermo Fisher Scientific UZ-View™ flow cell (UZFC). The HSDC, housed within a 3D printed detector arrangement, gave an effective pathlength of 1.01 mm (84% of nominal pathlength) and stray light of only 0.2%. Limits of detection for four test small molecule pharmaceuticals ranged from 65 to 101 µg L-1 based upon a 316 nL injection volume, with separation efficiencies of between 18,000 and 29,700 N m-1, with sub-4 min run times. The portable capillary LC system was successfully coupled to a small footprint portable mass spectrometer (Microsaic 4500 MiD) to demonstrate compatibility and 'point-of-need' miniaturised LC-MS capability.

This paper proposes an ultra-miniaturised, flexible 5.8 GHz antenna to give better performance for an ingestible wireless capsule endoscopy system. The use of an artificial magnetic conductor (AMC) unit cell in this design helps in enhancing the gain, improving the efficiency and reducing the size at resonance. This novel AMC-based layer is modelled as a radiator, and is excited by a complementary strip feed. The proposed design is printed on a flexible polyimide material of size 4.6 × 7.6 × 0.15 mm, with dielectric constant 3.5 and loss tangent 0.008. The antenna has an omni-directional radiation pattern with gain 1.64 dBi. The size is reduced drastically by bending the structure to fit inside a standard-sized ingestible capsule of 26 × 11 mm. The performance metrics of the antenna after placing inside the wireless ingestible capsule are presented with the help of an analysis of the reflection coefficient curves, radiation pattern, specific absorption rate (SAR) and field distributions. In addition, different positions and orientations of the antenna and different bending conditions are used, and these cases are solved using our AMC-based unit cell radiator. We carry out simulations using High Frequency Structure Simulator (HFSS) and measurements are made by immersing the fabricated antenna in minced meat. The results of this novel structure are compared with those of other ingestible antennas proposed in the literature. The proposed design has a SAR value of 0.82505 W/kg at around 5.8 GHz over 1 g of tissue, thereby redirecting the radiation away from the human body. This conformal AMC structure achieved better miniaturisation while maintaining the antenna resonance and SAR within permissible limits.

We explored the macroevolutionary dynamics of miniaturisation in New World anchovies by integrating a time-calibrated phylogeny, geometric morphometrics and phylogenetic comparative methods. We found that the paedomorphic species Amazonsprattus scintilla occupies a novel region of shape space, while the dwarf species Anchoviella manamensis has an overall shape consistent with other anchovies. We found that miniaturisation did not increase overall clade disparity in size or shape beyond the expectations of Brownian motion, nor were there differences in rates of size or shape evolution among clades. Overall, our study shows that while the mode of miniaturisation influences shape evolution, the phenotypic novelty produced by the evolution of miniaturisation did not seem to alter macroevolutionary dynamics.

BACKGROUND: In this work, a small-scale ceramic microbial fuel cell (MFC) with a novel type of metal-carbon-derived electrocatalyst containing iron and nicarbazin (Fe-NCB) was developed, to enhance electricity generation from neat human urine. Substrate oxidation at the anode provides energy for the separation of ions and recovery from urine without any chemical or external power additions. RESULTS: The catalyst was shown to be effective in clear electrolyte synthesis of high pH, compared with a range of carbon-based metal-free materials. Polarisation curves of tested MFCs showed up to 53% improvement (44.8 W m-3) in performance with the use of Fe-NCB catalyst.Catholyte production rate and pH directly increased with power performance while the conductivity decreased showing visually clear extracted liquid in the best-performing MFCs. CONCLUSIONS: Iron based catalyst Fe-NCB was shown to be a suitable electrocatalyst for the air-breathing cathode, improving power production from urine-fed MFCs. The results suggest electrochemical treatment through electro-osmotic drag while the electricity is produced and not consumed. Electro-osmotic production of clear catholyte is shown to extract water from urine against osmotic pressure. Recovering valuable resources from urine would help to transform energy intensive treatments to resource production, and will create opportunities for new technology development. © 2018 The Authors. A new genus and two new species of miniature clingfishes are described based on specimens collected from dense stands of macroalgae in intertidal and shallow subtidal areas along the coast of southern Australia. The new genus, Barryichthys, is distinguished from other genera of the Gobiesocidae by unique features of the adhesive disc, including elongate papillae in adhesive disc regions A and B, the reduction and/or loss of several elements of the cephalic lateral line canals, the lower gill arch skeleton, and the neurocranium, and by having two distinct types of pectoral-fin rays. Barryichthyshutchinsi is described based on 19 specimens (12.4-18.7 mm SL) from Western Australia and South Australia. Barryichthysalgicola is described based on 22 specimens (9.0-21.0 mm SL) from Victoria, New South Wales and Tasmania. The new species are distinguished from each other by characters of body and head shape, vertebral counts, and aspects of live colour pattern. The new genus shares several characters in common with Parvicrepis, another genus of miniature gobiesocids from southern Australia that also inhabits macroalgae habitats. The many reductions and novel characters of Barryichthys are discussed within the context of miniaturisation.

Artemisinin, the main antimalarial compound of Artemisia annua L., is currently attracting increasing interest for its antiproliferative properties, but its content is highly variable, depending on several genetic, environmental and processing conditions. Aim of the present study is to analyse the artemisinin content in different plant extracts, to test their in vitro activity on cell proliferation and then to correlate these data to the active principle concentration. For this purpose, an innovative miniaturised sample pretreatment strategy based on microextraction by packed sorbent (MEPS) was developed and coupled to an original advanced method based on liquid chromatography with diode array detection and tandem mass spectrometry (LC-DAD-MS/MS). The method was fully validated, granting consistent data. Good linearity was found over a suitable concentration range, i.e. 5-1000ng/mL. Extraction yields (>85%), precision (RSD < 3.5%) and accuracy (recovery 88-93%) were all within acceptable levels of confidence. After validation, the method was successfully applied to the determination of artemisinin in A. annua extracts. Analyte content was widely variable (up to twenty-fold) according to the starting material and the extraction procedure, ranging between 5.9µg/g and 109µg/mL. The cytotoxic activity of all analysed extracts was also tested on human leukemic cells by viable cell count and cell cycle analysis. Artemisinin concentrations and biological activity were carefully evaluated and the observed antiproliferative effects varied according to artemisinin content in each extract type. This highlights the need to quantitatively analyse the main active constituent of plant extracts and the obtained data have shown to be promising for the choice of the related herbal product dosage.

RESUMO

Volumetric absorptive microsampling (VAMS) is a recent microsampling technique used to obtain dried specimens of blood and other biological matrices for application to a plethora of bioanalytical purposes. As such, it can be likened to dried blood spot (DBS) technique that has been in wide use for the last 40 years. However, VAMS promises to bring some significant advantages over DBS, related to sampling volume accuracy, haematocrit (HCT) dependence, pre-treatment and automation. Although some aspects still need to be investigated in depth, VAMS is increasingly recognised as a viable alternative to DBS and other dried microsampling techniques. In this tutorial, different aspects of VAMS approach are described and discussed, presenting the procedures adopted and the results obtained by those authors who have developed this kind of analytical workflow in the last few years. Hopefully, this will help other scientists to find new solutions to old and recent problems related to microsampling and to produce new, sound and interesting science in this field.

The performance of three portable NIR spectrometers was compared by analysing the total antioxidant capacity (TAC) of different species of gluten-free grains. TAC is often used to evaluate the quality of foods and was determined using Folin-Ciocalteu measurements and used as reference data for establishing PLS-R models with NIR data. NIRS enables fast and non-invasive measurements. The microPhazir RX and the MicroNIR 2200 are broadly used in chemical and pharmaceutical industries, whereas SCiO is a pocket-sized, consumer-oriented spectrometer. The devices work in different regions of the NIR spectrum and their performances was compared using statistical parameters. 77 samples were measured and analysed using the software The Unscrambler X, as well as SCiO-Lab. All models established were cross- and test set validated. The multivariate data processing using The Unscrambler X yielded similar results as SCiO-Lab. The best model was established for non-milled samples measured with the MicroNIR 2200 and analysed using The Unscrambler X.

Moreover, medicine must be personalised, from screening and throughout the care pathway of the patient with chronic kidney failure. Thereby, the risk of morbidity can be reduced and the quality of life improved, with the help of connected tools. The role of the patients themselves and all healthcare professionals is essential.

Investigations of brain complex integrative actions should consider beside neural networks, glial, extracellular molecular, and fluid channels networks. The present paper proposes that all these networks are assembled into the brain hyper-network that has as fundamental components, the tetra-partite synapses, formed by neural, glial, and extracellular molecular networks. Furthermore, peri-synaptic astrocytic processes by modulating the perviousness of extracellular fluid channels control the signals impinging on the tetra-partite synapses. It has also been surmised that global signalling via astrocytes networks and highly pervasive signals, such as electromagnetic fields (EMFs), allow the appropriate integration of the various networks especially at crucial nodes level, the tetra-partite synapses. As a matter of fact, it has been shown that astrocytes can form gap-junction-coupled syncytia allowing intercellular communication characterised by a rapid and possibly long-distance transfer of signals. As far as the EMFs are concerned, the concept of broadcasted neuroconnectomics (BNC) has been introduced to describe highly pervasive signals involved in resetting the information handling of brain networks at various miniaturisation levels. In other words, BNC creates, thanks to the EMFs, generated especially by neurons, different assemblages among the various networks forming the brain hyper-network. Thus, it is surmised that neuronal networks are the "core components" of the brain hyper-network that has as special "nodes" the multi-facet tetra-partite synapses. Furthermore, it is suggested that investigations on the functional plasticity of multi-partite synapses in response to BNC can be the background for a new understanding and perhaps a new modelling of brain morpho-functional organisation and integrative actions.

Conventional PCNL leads to a high stone-free rate but is associated with a significant treatment morbidity. To decrease the morbidity of PCNL but to achieve an equal stone-free rate, in 2001 a miniaturised version of a conventional PCNL (Mini-PCNL) was introduced. Meanwhile numerous modifications of the Mini-PCNL were reported. The ongoing experience with the Mini-PCNL technique showed that Mini-PCNL is not only a miniaturisation but also a different method to remove the stones, as the stones come out of the calyceal system only by means of the irrigation flow without any further need of forceps or baskets. Due to a limited number of papers in the literature, the guidelines so far do not distinguish between conventional and Mini-PCNL, although the Mini-PCNL already became a frequent treatment modality in daily routine worldwide. From the current point of view, Mini-PCNL can be recommended to treat all kinds of upper urinary tract stones exceeding a diameter of 10 mm. In this context, Mini-PCNL is a treatment alternative to flexible ureterorenoscopy, SWL and conventional PCNL. The available literature shows that the morbidity of Mini-PCNL is less and the stone-free rate is equal to that of conventional PCNL. It seems that there is no limitation to Mini-PCNL in the treatment of upper urinary tract stones. The future will show what kind of further miniaturisation of the Mini-PCNL procedure will be appropriate.

Mutualisms between ants and sap-sucking insects generally involve clear benefits for both partners: the ants provide protection in exchange for honeydew. However, a single ant genus associates with armoured scale insects (Diaspididae) that do not excrete honeydew. We studied three colonies of Melissotarsus emeryi ants from two localities in Mozambique. Vast numbers of the diaspidid Morganella conspicua occupied galleries dug by the ants under the bark of living trees. Unlike free-living M. conspicua and other diaspidids, M. conspicua living with ants are known to lack shields, likely because they gain protection against enemies and desiccation. Nevertheless, we documented the occurrence of rare individuals with shields inside ant galleries, indicating that their glands continue to secrete wax and proteins as building material. This is likely to constitute a significant portion of the ants' diet, in addition to diaspidid exuviae and excretions from the Malpighian tubules. Indeed, Melissotarsus workers cannot walk outside the galleries due to modified middle legs, forcing them to obtain all nourishment within the tree. Melissotarsus founding queens, however, must locate a suitable host tree while flying, and acquire diaspidid crawlers. This mutualism involves ants that are highly specialised to chew through living wood, and diaspidids that can also live freely outside the bark. It is extremely widespread in Africa and Madagascar, recorded from 20 tree families, and harmful effects on plant hosts require rapid study.

OBJECTIVE: To present our novel miniaturised endoscopic system and describe a step-by-step guide for successful implementation of the super-mini percutaneous nephrolithotomy (SMP). PATIENTS AND METHODS: The new-generation SMP endoscopic system consists of (i) a 40 000-pixel super-mini nephroscope with an 8.0-F outer diameter and 7.5-F inner diameter dismountable sheath, and (ii) a newly designed irrigation-suction sheath available in either 12 F or 14 F. The irrigation-suction sheath is a two-layered metal structure. The key feature of the irrigation-suction sheath is to allow irrigation and suction respectively (the inflow through the space between the two layers of the sheath, the outflow through the central lumen of the sheath). This property improves irrigation and stone clearance despite reduced instrument dimension. In all, 59 patients with renal stones underwent new-generation SMP between April 2016 and December 2016. The percutaneous tract dilatation was carried out to 14 F. Lithotripsy was performed using either holmium laser or a pneumatic lithotripter. Stone fragments were sucked out by vacuum suctioning through the sheath. A nephrostomy tube or JJ stent was placed only if clinically indicated. Low-dose computed tomography was performed to assess the stone-free status on the morning after the procedure. RESULTS: The mean stone burden was 2.4 cm. Of the 59 patients, nine had diabetes and five had hypertension. SMP was completed successfully in all patients with a mean operation duration of 32.9 min and a mean haemoglobin decrease of 13 g/L. The stone-free rate was 91.5%. Complications occurred in 5.1% of the patients, all of them were Clavien-Dindo Grade I (minor fever managed by antipyretic therapy), and no transfusions were needed. CONCLUSION: The new-generation SMP system is safe, feasible, and effective for managing renal calculi of <3 cm, with the advantages of a small percutaneous tract, less blood loss, high efficacy in stone clearance, improved visual field, short operation duration, and ease of operating.
