RESUMO

Increasing cancer cell sensitivity to chemotherapy by amending aberrant metabolism using plant extracts represents a promising strategy to lower chemotherapy doses while retaining the same therapeutic outcome. Here, we incubated HepG2 cells with four plant extracts that were selected based on an earlier assessment of their cytotoxicity, viz asparagus, green tea, rue, and avocado, separately, before treatment with doxorubicin. MTT assays elucidated a significant decrease in doxorubicin-IC50 following HepG2 incubation with each extract, albeit to a variable extent. The investigated extract's ultra-performance liquid chromatography and gas chromatography coupled with mass spectrometry (UPLC/MS and GC/MS) revealed several constituents with anticancer activity. Biochemical investigation displayed several favorable effects, including the inhibition of hypoxia-inducible factor1α (HIF1α), c-Myc, pyruvate kinase-M2 (PKM2), lactate dehydrogenase-A (LDH-A), glucose-6-phosphate dehydrogenase (G6PD), and glutaminase by asparagus and rue extracts. To less extent, HIF1α, c-Myc, PKM2, and LDH-A were partially inhibited by green tea extract, and HIF1α and glutaminase activity was inhibited by avocado oil. Undesirably, green tea extract increased glutaminase; avocado oil rose c-Myc, and both increased G6PD. In conclusion, our study confirms the potential cytotoxic effects of these plant extracts. It highlights a strong association between the ability of asparagus, green tea, rue, and avocado to sensitize HepG2 cells to doxorubicin and their power to amend cell metabolism, suggesting their use as add-on agents that might aid in clinically lowering the doxorubicin dose.

RESUMO

Flavonoids are compounds abundantly found in nature and known as a polyphenolic group of compounds having flavancore and show the utmost abundant collection of complexes and are found in fiber-rich root vegetables, fruits, and their eatable items. Due to the presence of hydroxyl groups, flavonoids show various therapeutic activities like antioxidant, antibacterial, antiviral, and antiinflammatory. Substituent groups are responsible for the onset of biochemical actions of flavonoids, which affect the metabolism. The major example of flavonol is rutin, which is constituted of rutinose and quercetin. It is a glycosidic type of flavonoid, similarly identified as purple quercitrin and Vitamin P with over 70 plant species and plant-derived foods, exclusively grapefruit, buckwheat seeds, cherries, apricots, grapes, onions, plums, and oranges. Another name forrutin is a citrus flavonoid known as rutoside, and sophorin. Rutin is a polyphenolic compound having a low molecular weight.

RESUMO

The spectral-based photochemical reflectance index (PRI) and leaf surface temperature (Tleaf ) derived from thermal imaging are two indicative metrics of plant functioning. The relationship of PRI with radiation-use efficiency (RUE) and Tleaf with leaf transpiration could be leveraged to monitor crop photosynthesis and water use from space. Yet, it is unclear how such relationships will change under future high carbon dioxide concentrations ([CO2 ]) and drought. Here we established an [CO2 ] enrichment experiment in which three wheat genotypes were grown at ambient (400 ppm) and elevated (550 ppm) [CO2 ] and exposed to well-watered and drought conditions in two glasshouse rooms in two replicates. Leaf transpiration (Tr ) and latent heat flux (LE) were derived to assess evaporative cooling, and RUE was calculated from assimilation and radiation measurements on several dates along the season. Simultaneous hyperspectral and thermal images were taken at ~ $\unicode{x0007E}$ 1.5 m from the plants to derive PRI and the temperature difference between the leaf and its surrounding air ( ∆ $\unicode{x02206}$ Tleaf-air ). We found significant PRI and RUE and ∆ $\unicode{x02206}$ Tleaf-air and Tr correlations, with no significant differences among the genotypes. A PRI-RUE decoupling was observed under drought at ambient [CO2 ] but not at elevated [CO2 ], likely due to changes in photorespiration. For a LE range of 350 W m-2 , the ΔTleaf-air range was ~ $\unicode{x0007E}$ 10°C at ambient [CO2 ] and only ~ $\unicode{x0007E}$ 4°C at elevated [CO2 ]. Thicker leaves in plants grown at elevated [CO2 ] suggest higher leaf water content and consequently more efficient thermoregulation at high [CO2 ] conditions. In general, Tleaf was maintained closer to the ambient temperature at elevated [CO2 ], even under drought. PRI, RUE, ΔTleaf -air , and Tr decreased linearly with canopy depth, displaying a single PRI-RUE and ΔTleaf -air  Tr model through the canopy layers. Our study shows the utility of these sensing metrics in detecting wheat responses to future environmental changes.

RESUMO

In order to explore the physiological mechanism of different yield of summer maize (Zea mays L.) hybrids with different growth duration, a field experiment was conducted to study the growth stage, leaf photosynthetic characteristics, dry matter accumulation (DMA), transport and distribution characteristics and yield of the early maturity hybrid Denghai 518 (DH518) and the mid-late maturity hybrid Denghai 605 (DH605) from 2017 to 2021. The results showed that the yield of DH605 was significantly higher than that of DH518. The growth period of DH518 was 7-10 days shorter and the days of the growth stage of the sowing-silking stage (R1) were 5-6 days shorter compared to that of DH605. The contribution to grain dry matter by leaf and stalk dry matter remobilization (DMRC) of DH518 was significantly higher than that of DH605. There was a significant negative correlation between pre-silking growth days and harvest index (HI). The 13C distribution to grains of DH518 was significantly higher than that of DH605, and the HI and the corresponding contribution of HI to yield was also higher than that of DH605. The light and temperature resource use efficiency from silking to physiological maturity stage of DH605 was significantly higher than that of DH518. The yield per GDD of DH605 increased by 7.25% than that of DH518. At post-silking, the duration of higher leaf area index (DLAI) (>56 days) and active photosynthesis duration (APD) (>50 days) of DH605 were longer compared with that of DH518, and the average plant growth rate was 7.15% higher than that of DH518, which significantly increased the DMA of DH605. Therefore, the significant reduction of DH518 yield compared with DH605 was not due to the shortening of the growth stage of sowing-R1, but the lower light and temperature resource use efficiency from silking to physiological maturity stage.

RESUMO

Canopy photosynthesis integrates leaf functional and structural traits in space and time and correlates positively with yield formation. Many models with different levels of architectural details ranging from zero-dimensional (0D) to three-dimensional (3D) have been developed to simulate canopy light interception and photosynthesis. Based on these models, a crop growth model can be used to assess crop yield in response to genetic improvement, optimized practices, and environmental change. However, to what extent do architectural details influence light interception, photosynthetic production, and grain yield remains unknown. Here, we show that a crop growth model with high-resolution upscaling approach in space reduces the departure of predicted yield from actual yield and refines the simulation of canopy photosynthetic production. We found crop yield predictions decreased by 12.0-48.5% with increasing the resolution of light simulation, suggesting that a crop growth model without architectural details may result in a considerable departure from the actual photosynthetic production. A dramatic difference in light interception and photosynthetic production of canopy between cultivars was captured by the proposed 3D model rather than the 0D, 1D, and 2D models. Furthermore, we found that the overestimation of crop yield by the 0D model is caused by the overestimation of canopy photosynthetically active radiation (PAR) interception and the RUE and that by the 1D and 2D model is caused by the overestimated canopy photosynthesis rate that is possibly related to higher predicted PAR and fraction of sunlit leaves. Overall, this study confirms the necessity of taking detailed architecture traits into consideration when evaluating the strategies of genetic improvement and canopy configuration in improving crop yield by crop modeling.

RESUMO

Genetic progress in seed yield in lentils (Lens culinaris Medik) has increased by 1.1% per year in Australia over the past 27 years. Knowing which plant traits have changed through breeding during this time can give important insights as to how lentil yield has increased. This study aims to identify morphological and physiological traits that were directly or indirectly selected between 1993 and 2020 in the Australian lentil breeding program using 2 years of experimental data. Major changes occurred in plant architecture during this period. Divergent selection has seen the release of varieties that have sprawling to very upright types of canopies. Despite this genetic diversity in recently released varieties, there is an overall tendency of recently released varieties having increased plant height and leaf size with reduced number of branches. Increased light interception was positively correlated with year of release (YOR) and yield, and likely results from indirect selection of yield and taller plant types. There is an indication that recently released varieties have lower CO2 assimilation rate, stomatal conductance and canopy temperature depression (CTD) at high ambient temperatures (~30°C). Understanding lentil physiology will assist in identifying traits to increase yield in a changing climate with extreme weather events.

RESUMO

The present report describes two novel cases of suspected intoxication with Galega officinalis in 6- and 21-year-old Arabian mares displaying acute respiratory signs. Both animals showed signs of pulmonary edema at physical examination, with the oldest of the two also manifesting severe dyspnea and foamy nasal discharge. The mares were grazing on the same meadow with hay available ad libitum. Botanical analysis of the latter showed traces of the toxic plant Galega officinalis (L.), which has been daily ingested at a dose of around 14 g of dry matter for three days. Based on the respiratory signs and the presence of goat's rue in the mares' feed, a presumptive diagnosis of plant poisoning was assumed. Dietary change and treatment allowed the 6-year-old mare to fully recover in 3 days while a longer period of about 2 weeks was necessary for the older horse. Horses avoid eating fresh goat's rue as its palatability is low, yet poisoning may still happen in these species when the plant is found in dried and processed feed material.

RESUMO

The iron nanoparticles with different physic-chemical properties induce inconsistent effects on various studied plant species. Thus, the effect of ferric oxide (Fe2O3) nanoparticles was compared with Fe2O3 microparticles and FeSO4complexes of EDTA for major physiological and gene expression in Rue (Ruta graveolens). Iron root content increased as Fe-MPs + EDTA ˂˂ Fe-NPs + EDTA˂ FeSO4 + EDTA. The shoot's iron remained unchanged or slightly increased under most of FeSO4 and Fe-MPs + EDTA treatments. Under Fe-NPs + EDTA treatment, 50 and 250 µM concentration decreased on shoot iron by 23.2% and 19.4% compared to control, respectively. But the shoot iron at 500 µM NPs was 28.2% higher than that of the control. A 46-58 fold lower Fe translocation was observed under Fe-NPs + EDTA than Fe-MPs + EDTA. The effect of Fe-NPs + EDTA was more significant on plant fresh and dry mass than the control. All treatments showed an increase in anthocyanin by 19-84% in leaves compared to the control. The Fe-NPs + EDTA and MPs + EDTA induced similar effects on enhanced growth parameters, total chlorophyll, catalase enzyme activity, gene, and reduced chlorophyll a/b and oxidants. Catalase enzyme activity in FeSO4 and MPs + EDTA was similar, and in Fe-NPs + EDTA treatments were influenced by coarse and fine regulation mechanisms, respectively. Iron MPs + EDTA had a more negative effect on IRT1 relative gene expression in roots as compared to other iron forms. The IRT1 relative gene expression in shoots was positively affected by 31-81% under all treatment types (except control and 250 µM Fe-NPs + EDTA, and 250 µM MPs + EDTA). These results could reveal the potential mechanism of plant response to nanoparticles.

RESUMO

Thalictrum aquilegiifolium (Ranunculaceae) is widely distributed in the Eurasian Continent and Japan and comprises some intraspecific taxa. We report here the complete chloroplast genome of T. aquilegiifolium var. sibiricum. The plastome of T. aquilegiifolium var. sibiricum is 156,074 bp in length, containing large (85,457 bp) and small (17,642 bp) single-copy regions which are separated by a pair of inverted repeats (26,487 bp each). The genome consists of 119 genes, including 88 protein-coding, four ribosomal RNA genes, and 27 transfer RNA genes. Our phylogenetic analysis revealed that Thalictrum species formed a highly supported clade, indicating that these species are monophyletic.

RESUMO

Accurate crop growth and radiation use efficiency (RUE) measurements under optimal growth conditions are required to predict grain yield near the genetic growth potential and estimate climate change effects on crop production. Research was conducted to quantify RUE of maize during the whole growth period, vegetative period (VP, phenological development stage DVS < 1.0), and reproductive period (RP, DVS ≥ 1.0) under the optimal growth condition in Northeast China. Maize was grown in three population density treatments (9.0, 7.5, and 6.0 plants m-2, referred to HD, MD, and LD respectively) over three growing seasons (2017-2019). The maximum Leaf Area Index (LAI) was observed after the anthesis period (DVS = 1.2), ranged from to 4.9 to 6.9, and then gradually decreases. Photosynthetically active radiation (PAR) intercepted by the canopy of maize was approximate 10% during the early stage of the vegetative period (DVS < 0.3). Then greatest differences occurred during the late vegetative period (DVS = 0.3 to 0.8). When DVS > 1.0, there is no significant difference between the three population density conditions. Estimates of RUE were obtained based on the intercepted PAR (IPAR) and absorbed PAR (APAR). RUE value was 3.97 g MJ-1 APAR or 3.73 g MJ-1 IPAR in the whole growth period, and did not vary among density treatments. Moreover, maximum RUE occurred during VP (4.11 g MJ-1 APAR) and declined during RP (3.95 g MJ-1 APAR). Therefore, we recommend that maize models that rely on RUE for aboveground biomass accumulation should be using a value of 3.97 g MJ-1 APAR for predicting optimum maize grain yield in Northeast China, especially for the potential yield.

RESUMO

To achieve food security, it is necessary to increase crop radiation use efficiency (RUE) and yield through the enhancement of canopy photosynthesis to increase the availability of assimilates for the grain, but its study in the field is constrained by low throughput and the lack of integrative measurements at canopy level. In this study, partial least squares regression (PLSR) was used with high-throughput phenotyping (HTP) data in spring wheat to build predictive models of photosynthetic, biophysical, and biochemical traits for the top, middle, and bottom layers of wheat canopies. The combined layer model predictions performed better than individual layer predictions with a significance as follows for photosynthesis R 2 = 0.48, RMSE = 5.24 µmol m-2 s-1 and stomatal conductance: R 2 = 0.36, RMSE = 0.14 mol m-2 s-1. The predictions of these traits from PLSR models upscaled to canopy level compared to field observations were statistically significant at initiation of booting (R 2 = 0.3, p < 0.05; R 2 = 0.29, p < 0.05) and at 7 days after anthesis (R 2 = 0.15, p < 0.05; R 2 = 0.65, p < 0.001). Using HTP allowed us to increase phenotyping capacity 30-fold compared to conventional phenotyping methods. This approach can be adapted to screen breeding progeny and genetic resources for RUE and to improve our understanding of wheat physiology by adding different layers of the canopy to physiological modeling.

RESUMO

Despite progress in understanding of the genetic basis of gout, the precise factors affecting differences in gout susceptibility among different gout subtypes remain unclear. Using clinically diagnosed gout patients, we conducted a genome-wide meta-analysis of two distinct gout subtypes: the renal overload type and the renal underexcretion type. We provide genetic evidence at a genome-wide level of significance that supports a positive association between ABCG2 dysfunction and acquisition of the renal overload type.

RESUMO

The maximizing of water use efficiency (WUE) and radiation use efficiency (RUE) is vital to improving crop production in dryland farming systems. However, the fundamental question as to the association of WUE with RUE and its underlying mechanism under limited-water availability remains contentious. Here, a two-year field trial for maize designed with five progressive soil drying regimes applied at two different growth stages (three-leaf stage and seven-leaf stage) was conducted during the 2013-2014 growing seasons. Both environmental variables and maize growth traits at the leaf and canopy levels were measured during the soil drying process. The results showed that leaf WUE increased with irrigation reduction at the early stage, while it decreased with irrigation reduction at the later stage. Leaf RUE thoroughly decreased with irrigation reduction during the progressive soil drying process. Aboveground biomass (AGB), leaf area index (LAI), a fraction of absorbed photosynthetically active radiation (fAPAR), and light extinction coefficient (k) of the maize canopy were significantly decreased by water deficits regardless of the growth stages when soil drying applied. The interrelationships between WUE and RUE were linear across the leaf and canopy scales under different soil drying patterns. Specifically, a positive linear relationship between WUE and RUE are unexpectedly found when soil drying was applied at the three-leaf stage, while it turned out to be negative when soil drying was applied at the seven-leaf stage. Moreover, the interaction between canopy WUE and RUE was more regulated by fAPAR than LAI under soil drying. Our findings suggest that more attention must be paid to fAPAR in evaluating the effect of drought on crops and may bring new insights into the interrelationships of water and radiation use processes in dryland agricultural ecosystems.

The possible relationship between apneas during early childhood and Sudden Infant Death Syndrome (SIDS) has never been demonstrated, and there is evidence that the two conditions may not be related. The American Academy of Pediatrics (AAP) defines ALTE (Acute Life Threatening Event), as an abrupt and unexpected event that includes manifestations of apnea along with changes in skin color and muscle tone, where the observer believes that the child has died. The AAP has recently proposed replacing the term ALTE with Brief Resolved Unexplained Events (BRUE). The new concept makes it possible to categorize brief, resolved and unexplained events, to better optimize the health resource, through objectifying the event and delivering management strategies by categorizing the risk. Objective: To describe the clinical characteristics and lethality of patients younger than 12 months who consult for BRUE in a referral hospital. Materials and methods: Descriptive cross-sectional study with revision of the file of 46 patients from the Infant and Nutrition Unit of the Dr. Luis Calvo Mackenna Hospital, with a diagnosis of BRUE, between January and December 2017. Results: Of the total number of patients with BRUE, 45% were men and 55% women. The average age was 1.37 + 0.51 months. An etiology was demonstrated in 70%, of these 31% with gastroesophageal reflux disease (GERD), this being the most frequent cause, followed by 19% with acute respiratory infections (ARI) and 9% with neurological causes. In 30% they were idiopathic causes. Conclusion: In our study, the most frequent causes of BRUE were GERD and respiratory infections. During the study period, no patient studied died, so we found no relationship between apnea in the infant and sudden death syndrome.

RESUMO

Crop growth largely depends on radiation. Radiation is the main impetus for photosynthesis and movement of photosynthates from source to sink. Therefore, identification of the optimum sowing windows and suitable cultivars for efficient utilization of radiation is of prime importance. A field study was conducted in red clay soil during 2014 and 2015 Kharif season and the treatments consisted of three genotypes and three sowing windows by using randomized complete block design with three replications. The effect of genotypes and sowing windows was found significant with respect to number of trifoliate leaves, leaf area ratio, dry matter production, grain numbers, pod length, test weight, grain yield, and stover yield of guar during 2014 as compared to 2015 sown crop. Statistically significant plant height, number of trifoliate leaves, number of branches, leaf area ratio, absolute growth rate, leaf area index, dry matter, grain number, pod length, grain yield, stover yield and a higher cumulative radiation interception were recorded with 15th August sown crop as compared to other sowing windows. The plant height, number of trifoliate leaves, number of branches, leaf area ratio, absolute growth rate, leaf area index, dry matter, grain number, pod length, grain yield, stover yield and maximum cumulative interception of radiation were significant with RGC-1003 as compared to RGC-936 and HG-365. It is observed that the incident PAR to dry matter accumulation conversion efficiency was varied with cultivars and different sowing windows which ranges from 0.74 g MJ-1 to 0.79 g MJ-1.

RESUMO

Wheat yields are stagnating or declining in many regions, requiring efforts to improve the light conversion efficiency, known as radiation use efficiency (RUE). RUE is a key trait in plant physiology because it links light capture and primary metabolism with biomass accumulation and yield, but its measurement is time consuming and this has limited its use in fundamental research and large-scale physiological breeding. In this study, high-throughput plant phenotyping (HTPP) approaches were used among a population of field-grown wheat with variation in RUE and photosynthetic traits to build predictive models of RUE, biomass, and intercepted photosynthetically active radiation (IPAR). Three approaches were used: best combination of sensors; canopy vegetation indices; and partial least squares regression. The use of remote sensing models predicted RUE with up to 70% accuracy compared with ground truth data. Water indices and canopy greenness indices [normalized difference vegetation index (NDVI), enhanced vegetation index (EVI)] are the better option to predict RUE, biomass, and IPAR, and indices related to gas exchange, non-photochemical quenching [photochemical reflectance index (PRI)] and senescence [structural-insensitive pigment index (SIPI)] are better predictors for these traits at the vegetative and grain-filling stages, respectively. These models will be instrumental to explain canopy processes, improve crop growth and yield modelling, and potentially be used to predict RUE in different crops or ecosystems.

RESUMO

BACKGROUND: Brainstem and spinal cord hemangioblastomas are a common manifestation of von Hippel-Lindau (VHL) disease. Cysts and associated syringes are the most common cause of significant morbidity in these patients. Surgical treatment of symptomatic hemangioblastomas are often complicated by the presence of multiple potential lesions, leading to cyst and syrinx formation. OBSERVATIONS: The authors present a case of a patient with multiple VHL-related hemangioblastomas who presented with syringobulbia and holocord syrinx. Resection of two cyst wall hemangioblastomas and one cervical hemangioblastoma only transiently improved syringobulbia. Eventual resolution of syringobulbia and collapse of the holocord syrinx only occurred following removal of a large lower thoracic hemangioblastoma. LESSONS: Surgical management of hemangioblastomas and associated cysts in patients with VHL should only target lesions most likely contributing to neurological deficits as excess surgical intervention risks treatment-related morbidity. The authors illustrate how anatomical and pathophysiological considerations as well as patient symptoms are key to identifying target lesions for resection and developing deliberate treatment plans.

RESUMO

The use of drugs is not solely determined by its therapeutic properties but refers to diversions of these properties. OBJECTIVE: To assess the level of knowledge of the diversion of drugs for abortifacient purposes among hairdressers and seamstresses of the suburb of Togblékopé. MATERIAL AND METHOD: We conducted a cross-sectional study among hairdressers and seamstresses living in Togblékopé, a suburb on the outskirts of the city of Lomé. RESULTS: Of the 114 respondents, 74.6% knew that drugs were diverted for abortion, mostly among older young people, singles and apprentices. In addition, 70.6% reported having at least once attended a drug abortion by someone. Thus, drugs such as paracetamol, chloroquine and acetylsalicylic acid have been reported for abortifacient purposes' diversion, most often purchased on the street and responsible for serious side effects. CONCLUSION: This study has highlighted the knowledge of drugs' use for abortifacient purposes among hairdressers and seamstresses. It is necessary to carry out actions that will aim at teaching the risks of drug intoxication and reduce the risk of pregnancy through family planning.

Ruta chalepensis L., most commonly known as 'fringed rue,' is an excellent and valuable bioactive plant that produces a range of complex flavonoids, of which rutin is the major compound present in this plant of great pharmaceutical and medicinal significance. The present study is a pioneering attempt to examine the changes in the transcriptomic landscape of leaf, stem, and root tissues and correlate this with rutin quantity in each tissue in order to identify the candidate genes responsible for rutin biosynthesis and to increase genomic resources in fringed rue. Comparative transcriptome sequencing of leaves, stems and roots were performed using the NovaSeq 6000 platform. The de novo transcriptome assembly generated 254,685 transcripts representing 154,018 genes with GC content of 42.60 % and N50 of 2280 bp. Searching assembled transcripts against UniRef90 and SwissProt databases annotated 79.7 % of them as protein coding. The leaf tissues had the highest rutin content followed by stems and roots. Several differentially expressed genes and transcripts relating to rutin biosynthesis were identified in leaves comparing with roots or stems comparing with roots. All the genes known to be involved in rutin biosynthesis showed up-regulation in leaves as compared with roots. These results were confirmed by gene ontology (GO) and pathway enrichment analyses. Up-regulated genes in leaves as compared with roots enriched GO terms with relation to rutin biosynthesis e.g. action of flavonol synthase, biosynthetic mechanism of malonyl-CoA, and action of monooxygenase. Phylogenetic analysis of the rhamnosyltransferase (RT) gene showed that it was highly homologues with RT sequence from Citrus species and all were located in the same clade. This transcriptomic dataset will serve as an important public resource for future genomics and transcriptomic studies in R. chalepensis and will act as a benchmark for the identification and genetic modification of genes involved in the biosynthesis of secondary metabolites.
