RESUMO

In early 2024, explosive outbreaks of Oropouche virus (OROV) linked to a novel lineage were documented in the Amazon Region of Brazil. We report the introduction of this lineage into Colombia and its co-circulation with another OROV lineage. Continued surveillance is needed to prevent further spread of OROV in the Americas.

Tick-borne febrile illnesses caused by pathogens like Anaplasma spp., Bartonella spp., Borrelia spp., Ehrlichia spp., Coxiella burnetii, Francisella tularensis, and Rickettsia spp., are significant health concerns in Africa. The epidemiological occurrence of these pathogens is closely linked to the habitats of their vectors, prevalent in rural and semi-urban areas where humans and livestock coexist. The overlapping clinical presentations, non-specific symptoms, and limited access to commercially available in vitro diagnostics in resource-limited settings exacerbate the complexity of accurate diagnoses. This review aimed to systematically extract and analyze existing literature on tick-borne febrile illnesses in Africa, highlighting the diagnostic challenges and presenting an up-to-date overview of the most relevant pathogens affecting human populations. A comprehensive literature search from January 1990 to June 2024 using databases like PubMed, Cochrane Library, Science Direct, EMBASE, and Google Scholar yielded 13,420 articles, of which 70 met the inclusion criteria. Anaplasma spp. were reported in Morocco, Egypt, and South Africa; Francisella spp. in Kenya and Ethiopia; Ehrlichia spp. in Cameroon; Bartonella spp. in Senegal, Namibia, South Africa, and Ethiopia; Borrelia spp. in Senegal, Gabon, Tanzania, and Ethiopia; Coxiella burnetii in 10 countries including Senegal, Mali, and South Africa; and Rickettsia spp. in 14 countries including Senegal, Algeria, and Uganda. Data were analyzed using a fixed-effect model in R version 4.0.1 and visualized on an African map using Tableau version 2022.2. This review highlights the urgent need for improved diagnostics to better manage and control tick-borne febrile illnesses in Africa.

BACKGROUND AND OBJECTIVES: The diagnostic challenges associated with T. solium continue to hamper control efforts of the world's most significant foodborne parasite and leading cause of epilepsy in low and middle-income countries. This study aimed to validate two conventional PCRs for taeniasis and estimate the diagnostic performance of microscopic and molecular tools. METHODS: Formalin and ethanol-fixed samples were tested by formalin-ethyl acetate concentration technique (FECT), Malachite smear, McMaster2 method, rrnS PCR and cox1 PCR. Initial validation of PCR methods was completed on 45 microscopy positive individuals. After validation, the performance of microscopic methods and the rrnS were estimated using samples from 1,156 individuals in Laos. Bayesian latent class models (BLCMs) and a composite reference standard were used to estimate diagnostic sensitivity and specificity. RESULTS: On preliminary validation the rrnS was able to detect 27/45 (60.00%) infections whereas the cox1 detected 21/45 (46.67%). As a result, the cox1 was excluded from further performance analysis. Microscopy methods and the rrnS were highly specific with estimates above 99.02% regardless of analytical method. The rrnS was the most sensitive test by informed BCLM (91.45%, CrI: 73.41-99.52%) followed by the FECT (71.20%, CrI: 50.53-85.48%), McMaster2 (51.31%, CrI: 32.00-71.29%) and Malachite smear (32.23%, CrI: 15.40-54.47%). DISCUSSION: The inability to validate the cox1 PCR suggests that it may not be suitable in its current form for routine characterisation of Taenia spp. detected by microscopy. The rrnS presents a suitable alternative to the cox1, however, requires its products to be sequenced. Given the low prevalence of taeniasis in most populations, this should be a feasible approach that may be able to be integrated with existing soil-transmitted helminth surveys that often use FECT for microscopic diagnosis.

The public's health and the financial sustainability of international societies remain threatened by bacterial zoonoses, with limited reliable diagnostic and therapeutic options available for bacterial diseases. Bacterial infections influence mammalian miRNA expression in host-pathogen interactions. In order to counteract bacterial infections, miRNAs participate in gene-specific expression and play important regulatory roles that rely on translational inhibition and target gene degradation by binding to the 3' non-coding region of target genes. Intriguingly, according to current studies, that exogenous miRNAs derived from plants could potentially serve as effective medicinal components sourced from traditional Chinese medicine plants. These exogenous miRNAs exhibit stable functionality in mammals and mimic the regulatory roles of endogenous miRNAs, illuminating the molecular processes behind the therapeutic effects of plants. This review details the immune defense mechanisms of inflammation, apoptosis, autophagy and cell cycle disturbance caused by some typical bacterial infections, summarizes the role of some mammalian miRNAs in regulating these mechanisms, and introduces the cGAS-STING signaling pathway in detail. Evidence suggests that this newly discovered immune defense mechanism in mammalian cells can also be affected by miRNAs. Meanwhile, some examples of transboundary regulation of mammalian mRNA and even bacterial diseases by exogenous miRNAs from plants are also summarized. This viewpoint provides fresh understanding of microbial tactics and host mechanisms in the management of bacterial illnesses.

We conducted a self-controlled case series study to investigate the association between COVID-19 vaccination and facial palsy (FP) in South Korea. We used a large immunization registry linked with the national health information database. We included 44,564,345 patients >18 years of age who received >1 dose of COVID-19 vaccine (BNT162b2, mRNA-1273, ChAdOx1 nCoV-19, or Ad.26.COV2.S) and had an FP diagnosis and corticosteroid prescription within 240 days postvaccination. We compared FP incidence in a risk window (days 1-28) with a control window (the remainder of the 240-day observation period, excluding any risk windows). We found 5,211 patients experienced FP within the risk window and 10,531 experienced FP within the control window. FP risk increased within 28 days postvaccination, primarily after first and second doses and was observed for both mRNA and viral vaccines. Clinicians should carefully assess the FP risk-benefit profile associated with the COVID-19 vaccines and monitor neurologic signs after vaccination.

BACKGROUND: Yersiniosis is one of the most significant intestinal disorders caused by Yersinia enterocolitica and affects both humans and animals. This study aimed to investigate the prevalence of Y. enterocolitica in New Valley Governorate, Egypt in animals, humans, fresh milk and dried milk. Additionally, this study analyzed the presence of virulence genes, including ail and Yst in tested isolates and conducted a phylogenetic analysis to determine the genetic similarity between human, and animal Y. enterocolitica isolates. Finally, the antimicrobial resistance patterns of the isolates were examined. RESULTS: Among the 982 samples examined, the prevalence of Y. enterocolitica based on ISO10273-2017 was 11.7% in animal samples including 12.8% of animal faeces, and 10.4% in milk samples. Moreover, the prevalence of Y. enterocolitica was 13.2% in human stool, and 9.5% in dried milk samples. The molecular characterization of the six randomly selected isolates showed that the 16S rRNA, ail and Yst genes were found in 50, 33.3 and 100% of the examined Y. enterocolitica isolates, respectively. Phylogenetic analysis of animal and human isolates based on the 16S rRNA gene revealed a high degree of similarity between the isolates. All the tested animal and human Y. enterocolitica isolates (100%) were resistant to ampicillin and cefotaxime, but highly sensitive to norfloxacin. CONCLUSIONS: The high prevalence of Y. enterocolitica in animal and human samples with high degrees of genetic similarity poses a threat to public and animal health. Animal faeces, milk and milk powder represent the main sources of Y. enterocolitica infection in humans. Additionally, high levels of antibiotic resistance of Y. enterocolitica can cause public health hazards by leading to the failure of disease prevention and treatment programs in humans and animals.

The ongoing increase in wild boar populations across Europe has fostered human-wildlife conflicts, including the transmission of emerging pathogens with zoonotic importance. Blastocystis is a ubiquitous, faecal-oral transmitted protist that can cause gastrointestinal illnesses and is observed in humans and animals worldwide. The role of wildlife in the epidemiology of Blastocystis is insufficiently understood. Thus, we investigated the occurrence and subtype diversity of Blastocystis in free-ranging wild boars from the Iberian Peninsula using conventional PCR and next-generation amplicon sequencing of a fragment of the ssu RNA gene. A total of 459 wild boar faecal samples were collected across Spain (n = 360) and Portugal (n = 99) between 2014 and 2021. Blastocystis was present in 15.3% (70/459; 95% CI 12.1-18.9) of the wild boars analysed, and its occurrence was significantly higher in Portugal (34.3%, 34/99; 95% CI 25.1-44.6) than in Spain (10.0%, 36/360; 95% CI 7.1-13.6). Seven Blastocystis subtypes (ST5, ST10b, ST13-ST15, ST24b, and ST43) were detected among the surveyed wild boar populations, with greater variability detected in Portuguese samples. ST5 was identified in all the Blastocystis-positive animals, whereas 14.3% of them harboured ST mixed colonisations. Our results demonstrate that Blastocystis ST5 is particularly adapted to infect wild boars. The additional identification of zoonotic STs reinforces the role of wild boars as spreaders of zoonotic infections with public health significance.

Leptospirosis is a bacterial zoonosis that poses an increasing global public health risk. Pacific Island communities are highly vulnerable to leptospirosis outbreaks, yet the local drivers of infection remain poorly understood. We conducted a systematic review to identify the drivers of human Leptospira infection in the Pacific Islands. There were 42 included studies from which findings were synthesized descriptively. In tropical Pacific Islands, infections were a product of sociodemographic factors such as male gender/sex, age 20 to 60 years, Indigenous ethnicity, and poverty; lifestyle factors such as swimming, gardening, and open skin wounds; and environmental factors, including seasonality, heavy rainfall, and exposure to rodents, cattle, and pigs. Possible mitigation strategies in these islands include strengthening disease reporting standards at a regional level; improving water security, rodent control, and piggery management at a community level; and information campaigns to target individual-level drivers of infection. By contrast, in New Zealand, exposures were predominantly occupational, with infections occurring in meat and farm workers. Accordingly, interventions could include adjustments to occupational practices and promoting the uptake of animal vaccinations. Given the complexity of disease transmission and future challenges posed by climate change, further action is required for leptospirosis control in the Pacific Islands.

RESUMO

Highly pathogenic avian influenza A(H5N1) detected in dairy cows raises concerns about milk safety. The effects of pasteurization-like temperatures on influenza viruses in retail and unpasteurized milk revealed virus resilience under certain conditions. Although pasteurization contributes to viral inactivation, influenza A virus, regardless of strain, displayed remarkable stability in pasteurized milk.

BACKGROUND: Non-human primate (NHPs) conservation sites could be sites of exchange of pathogens involved in infectious diseases. It is important to assess the potential risks associated with this type of structure. The objective of this study was to carry out a risk assessment of the Primatology Centre housed in the Interdisciplinary Centre for Medical Research of Franceville (CIRMF). METHODS: A questionnaire was administered to the centre's staff to assess the risk associated with each workstation, followed by a review of the various pathogens identified in NHPs. The data were analysed using two diagrams: the Kiviat diagram and the Pareto diagram. RESULTS: Based on our results, a variety of pathogens such as viruses, bacteria and parasites, potentially transmissible to humans, were described in the NHPs at the Primatology Centre of CIRMF. The position most exposed to zoonotic risks was that of animal handlers. CONCLUSION: The Primatology Centre of CIRMF is a potential transfer site for the transfer of zoonotic agents. To avoid the risk of parasite exchange between staff and animals, the implementation of biosecurity measures is essential.

The orf virus is one of the parapoxviruses. It occurs worldwide and its natural reservoir are sheep and goats. Infections in humans can occur through direct or indirect contact with affected animals. The infectious ulcerations usually heal spontaneously after a few weeks in immunocompetent patients. An infection must be reported to the responsible professional association if the exposure occurs in the occupational environment, for example in farmers.

Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that is responsible for COVID-19 pandemic, is a zoonotic RNA virus that has been reported in animals, including domestic animals. Due to the growing concern of health threat that could arise from active transmission of SARS-CoV-2 between pet owners and their pets, there is need to monitoring the emergence of a highly pathogenic strain of SARS-CoV-2 that is capable of transboundary infection, or a serious outbreak among human populations. Methods: We carried out a search in English, on PubMed and NCBI (National Center for Biotechnology Information) SARS-CoV-2 resources for relevant journals and nucleotide sequence data, that were published between 2019 and 2023. The CoVsurver mutations application on GISAID webpage was used to analyse mutation, nucleotide sequence alignment was carried out using MAFFT (Multiple Alignment using Fast Fourier Transform) version 7 and maximum likelihood tree was constructed by bootstrapping with 1000 replicates on MEGA 11 software. Results: A total of 47 mutations at the Spike gene region were identified, and mutation D614 was the most observed mutation. Nucleotide sequences of isolates from domestic animals had high sequence identity with Wuhan-Hu-1 reference sequence and the representative sequences of previously circulating VOCs from humans. Conclusion: This reveals that there is spill over of previously circulating variants of concern (VOC) to household pets from their infected owners. Hence, there is an urgent need for more intense surveillance to be carried out globally to monitor evolution of SARS-CoV-2 coronaviruses as a result of human - pet association.

PURPOSE: This study aimed to investigate the occurrence of leishmaniasis in domestic rabbits (Oryctolagus cuniculus) in the state of Sergipe, Brazil, and to evaluate the associated clinical signs. METHODS: A total of 31 rabbits from urban and rural areas were clinically examined using cytological, immunological, and serological tests. Blood and cytological samples were collected and analysed for the presence of Leishmania parasites and antibodies. Immunochromatographic tests were used to screen for anti-Leishmania antibodies, and cytological analysis of skin lesions was performed to detect the presence of Leishmania amastigotes. RESULTS: Of the rabbits tested, 19.35% were reactive in the anti-Leishmania antibody screening, and 3.33% tested positive for Leishmania amastigotes in skin lesion cytology. Clinical signs included cachexia, lymphadenomegaly, dehydration, apathy, dermatitis, ophthalmopathy, and alopecia. Cytological analysis revealed pyogranulomatous inflammation with Leishmania amastigotes present. The findings suggest that leishmaniasis is present among domestic rabbits in this region CONCLUSION: This study demonstrates the presence of leishmaniasis in domestic rabbits in the Northeast of Brazil. The findings underline the importance of early diagnosis and intervention in preventing the spread of the disease, and highlight the need for further research into the role of rabbits as potential reservoirs of Leishmania.

Background: Cystic echinococcosis (CE) is a widespread neglected zoonotic disease caused by Echinococcus granulosus sensu lato (EG) with a global burden of control in the billions of dollars. E. granulosus' life cycle involves definitive, intermediate, and humans as dead-end hosts. Echinococcosis control programs use strategies that focus on any of these hosts. We aimed to provide a comprehensive and up-to-date overview of the EG control interventions worldwide. Methods: We conducted a scoping review by mapping all studies on interventions for EG control following the Arksey and O'Malley Framework. We screened identified articles, and charted and coded selected papers. We classified the data based on target host, type of study, and control mechanism. We described the efficacy or safety outcomes, and the associated barriers/facilitators for the intervention. Critical appraisal was conducted. Results: From 7,853 screened studies, we analyzed 45: seven centered on human interventions, 21 on animals, and 17 on both. Studies on humans focused on educational strategies and human CE monitoring. The studies on animals were field trials and most were based on Praziquantel (PZQ) for dogs. Studies focused on both animals and humans had, in general, more participants, lasted longer, and covered larger geographical areas. Overall, the quality of studies was moderate to low. Conclusions: Available evidence suggests that long-term interventions aimed at both animals and humans can achieve significant reduction in EG transmission, particularly when PZQ treatment for dogs is included. Higher quality evidence, standardization of methodologies, and better reporting on post-intervention outcomes are necessary for drawing stronger conclusions. Further evidence is needed to assess the sustainability and scalability of control measures. Nonetheless, an integrative One Health approach is essential for overcoming the multiple challenges associated with sustaining long-term control efforts for Echinococcosis.

INTRODUCTION: Anthrax is a World Organisation for Animal Health (WOAH)-listed disease that must be reported upon confirmation based on the Terrestrial Animal Health Code. Anthrax poses a serious health issue for unvaccinated livestock, is a threat to humans through interaction with contaminated livestock and animal products and is endemic in many areas throughout the world, including the Transcaucasian Region. Despite several control and eradication efforts that have been implemented by the government of the Republic of Armenia (RA), sporadic cases of anthrax are still reported. We sought to understand the epidemic situation of anthrax in RA during the last 10 years (2012-2023) based on analysis of outbreaks and reported cases in cattle and humans. METHODS: We collected and evaluated officially reported data from human and animal cases, such as time, location, animal species, disease intensity and spread radius. The data and various parameters were mapped using ArcGIS to prepare a viable risk assessment. RESULTS: Based on the officially available data and reports, there have been 80 human cases and 55 animal cases of anthrax confirmed in RA from 2012 to 2023. We also identified the presence of anthrax spores in soil and environmental samples near animal burial sites in RA in 2015-2017 through polymerase chain reaction (PCR) testing. Upon comparing the human and animal cases by frequency and intensity, the human cases are directly proportional to the animal husbandry practices performed in RA. CONCLUSION: The detection of the anthrax pathogen at the burial sites highlights the continued threat in these areas. Thus, it is imperative to secure and monitor any areas that have been used for anthrax burial and limit the movement of animals in these areas. In the future, legislation should be updated to prioritise incineration of anthrax-infected carcasses instead of burial to limit further exposure to animals and humans.

PURPOSE: HEV is an emerging pathogen in Europe and was previously shown to be hyperendemic in areas of Abruzzo and Lazio, Central Italy. No systematic analysis of the HEV strains responsible for human infections over several years in Central Italy has previously been reported. Aim of the study was the molecular characterization of HEV from autochthonous hepatitis E cases occurred in Abruzzo and Lazio between 2015 and 2023. METHODS: Samples from 118 cases collected as part of virological surveillance in Abruzzo and Lazio from 2015 to 2023 were subjected to HEV sequencing and phylogenetic analysis. RESULTS: The main observed subtype was 3f, followed by 3c and 3e. The annual subtype distribution was quite stable over the observation period, but 3f cases tended to concentrate in winter/early spring whereas 3e cases in summer. Phylogenetic clusters of highly related sequences (a) highlighted unrecognized "point source outbreaks", (b) provided molecular support to temporally and/or geographically linked cases and (c) provided evidence for transmission of identical/highly related strains up to months/years following their first detection. CONCLUSIONS: The data provide an overview of the HEV strains responsible for human infections over eight years in Central Italy. The observed subtype distribution appears to agree better with the subtype distribution reported in Italy in pigs rather than in geographically matched wild boars, suggesting pig and its derivate food was a more frequent source of infection than wild boar in Abruzzo and Lazio. Molecular characterization is essential to recognize "point source outbreaks" and to monitor HEV circulation.

Zoonoses are responsible for many of all emerging infectious diseases as well as for those already established. Rocahepevirus ratti is a rat-originated virus related to the hepatitis E virus (Paslahepevirus balayani) but highly divergent genetically from this, with a high cross-species infection potential and zoonotic transmission. It can infect humans, leading to acute hepatitis, and is primarily transmitted through the consumption of contaminated water. Rocahepevirus ratti was first discovered in Germany in 2010. The first human case was described in 2017 in Hong Kong in an immune-compromised patient. The first case of chronic infection with Rocahepevirus ratti was described in 2023. A meta-analysis based on 38 studies published between 2000 and 2023 identified 21 cases in humans described up to this date and 489 infections in different animals. Raising awareness regarding this virus is essential, as there are probably many cases that remain undiagnosed, and the virus even has the ability to produce chronic infections in selected patients.

OBJECTIVE: Monkeypox is a viral zoonotic disease endemic to West and Central Africa; it has been reported in more countries during the last decade than in the previous 40 years. In 2022 a multinational outbreak occurred. This change in the epidemiology of the virus may represent an evolutionary adaptation. The purpose of this study is to analyze the molecular aspects of Monkeypox virus (MPXV) disease that may explain the latter's change in epidemiology during the 2022 outbreak. METHODS: From July 2022 through December 2022, the period of the outbreak, a narrative review was conducted on the available literature, with a total of 271 articles published in the MEDLINE/PubMed and LILACS databases being examined. The chosen articles were organized using the search and reference manager Mendeley Desktop 1.19.4. Duplicates and articles that did not meet the study's objective were eliminated, resulting in the selection of 49 articles for the present review. DISCUSSION: MPXV resurgence poses challenges due to waning immunity and changing epidemiological patterns. Recent outbreaks show different transmission routes, affecting new demographics. Genomic evolution, vaccination history, and potential new animal reservoirs complicate containment efforts. Continued surveillance and vaccination are crucial for control. CONCLUSIONS: It seems possible that MPXV has (re-)emerged to occupy the ecological niche left by the smallpox virus. Mutations of the apolipoprotein B mRNA editing enzyme, catalytic subunit 3G motif, in MPXV clade IIb since 2017 may explain the epidemiological change that has occurred in recent years. This pattern could be due to sustained transmission in a new host or a new route of infection.

Reptiles are among the most diverse groups of animals, inhabiting nearly all continents and environments. Understanding their parasite biodiversity has garnered significant interest, particularly from a One Health perspective. Although the highly diverse reptile fauna of Iran, comprising 272 species i.e. 89 snakes (Serpentes), 171 lizards (Sauria), 8 turtles, 2 tortoises (Testudines), 1 crocodile (Crocodilia), and 1 worm-lizard (Amphisbaenia), there is a shortage of information about parasites. The present review is a compilation of 62 studies published from 1922 to August 2024. We present information on 56 species of reptiles from five groups (amphisbaenians, crocodiles, testudines, snakes, and lizards) and 98 parasitic taxa belonging to different protozoa and metazoa i.e. nematodes, cestodes, trematodes, acanthocephala, leeches, ticks, mites, and myiasis -producing flies. Although 63 taxa were diagnosed at the species level, 35 parasite taxa were only reported at the genus or family levels. Reviewing the literature, we found a paucity of information about endemic reptiles several of which are vulnerable species. Considering that some of the detected parasites e.g. Cryptosporidium and amoebae have serious clinical and/or public health threats molecular diagnostic techniques are needed for precise identification and understanding of the epidemiology and the potential zoonotic implications associated with parasites of reptiles. There is also a need to understand the exact distribution and host-parasite associations in different reptilian species present in Iran including the role of the reptiles as intermediate and reservoir hosts.

Methicillin-resistant Staphylococcus (MRS) bacteria, including methicillin-resistant S. aureus and methicillin-resistant S. pseudintermedius (MRSP), pose a significant threat in veterinary medicine, given their potential for zoonotic transmission and their implications for companion animals and humans' health. This study aimed to assess the prevalence of MRS and anti-microbial resistance patterns at a university clinical hospital in Madrid, Spain. Samples were collected from both the environment and hospital staff at Veterinary Clinical Hospital of Alfonso X el Sabio University. Anti-microbial susceptibility assays, molecular detection of mecA gene and genetic relationships among the identified bacterial strains were performed. The study revealed an MRS prevalence of 1.50% in environmental samples, with MRSP accounting for 0.75% of the cases. Genetically related MRSP strains were found in different hospital areas. Among hospital staff, there was a MRS prevalence of 14.03%, including S. pseudintermedius and S. epidermidis strains. Antibiogram tests revealed multi-drug resistance among MRSP strains. Additionally, methicillin-resistant coagulase-negative staphylococci were isolated, suggesting potential cross-species transmission. This study underscores the presence of MRS in a veterinary clinical hospital, highlighting the significance of infection control through the implementation of protective measures, stringent hygiene practices among personnel and in the environment and responsible use of antibiotics. Further research is necessary to assess MRS incidence in animal patients and explore geographical variations, enhancing our understanding of MRS in veterinary medicine and addressing its challenges.
