ABSTRACT

BACKGROUND: Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) variants demonstrate predilection for different regions of the respiratory tract. While saliva-based reverse transcription-polymerase chain reaction (RT-PCR) testing is a convenient, cost-effective alternative to nasopharyngeal swabs (NPS), few studies to date have investigated whether saliva sensitivity differs across variants of concern. METHODS: SARS-CoV-2 RT-PCR was performed on paired NPS and saliva specimens collected from individuals with acute coronavirus disease 2019 (COVID-19) symptoms or exposure to a COVID-19 household contact. Viral genome sequencing of NPS specimens and Los Angeles County surveillance data were used to determine the variant of infection. Saliva sensitivity was calculated using NPS-positive RT-PCR as the reference standard. Factors contributing to the likelihood of saliva SARS-CoV-2 RT-PCR positivity were evaluated with univariate and multivariable analyses. RESULTS: Between June 2020 and December 2022, 548 saliva samples paired with SARS-CoV-2 positive NPS samples were tested by RT-PCR. Overall, saliva sensitivity for SARS-CoV-2 detection was 61.7% (95% CI, 57.6%-65.7%). Sensitivity was highest with Delta infection (79.6%) compared to pre-Delta (58.5%) and Omicron (61.5%) (P = 0.003 and 0.01, respectively). Saliva sensitivity was higher in symptomatic individuals across all variants compared to asymptomatic cases [pre-Delta 80.6% vs 48.3% (P < 0.001), Delta 100% vs 72.5% (P = 0.03), Omicron 78.7% vs 51.2% (P < 0.001)]. Infection with Delta, symptoms, and high NPS viral load were independently associated with 2.99-, 3.45-, and 4.0-fold higher odds of SARS-CoV-2 detection by saliva-based RT-PCR (P = 0.004, <0.001, and <0.001), respectively. CONCLUSIONS: As new variants emerge, evaluating saliva-based testing approaches may be crucial to ensure effective virus detection.

ABSTRACT

Mucosal immunity may contribute to clearing SARS-CoV-2 infection prior to systemic infection, thereby allowing hosts to remain seronegative. We describe the meaningful detection of SARS-CoV-2-specific nasal mucosal antibodies in a group of exposed-household individuals that evaded systemic infection. Between June 2020 and February 2023, nasopharyngeal swab (NPS) and acute and convalescent blood were collected from individuals exposed to a SARS-CoV-2-confirmed household member. Nasal secretory IgA (SIgA) antibodies targeting the SARS-CoV-2 spike protein were measured using a modified ELISA. Of the 36 exposed individuals without SARS-CoV-2 detected by the RT-PCR of NPS specimens and seronegative for SARS-CoV-2-specific IgG at enrollment and convalescence, 13 (36.1%) had positive SARS-CoV-2-specific SIgA levels detected in the nasal mucosa at enrollment. These individuals had significantly higher nasal SIgA (median 0.52 AU/mL) compared with never-exposed, never-infected controls (0.001 AU/mL) and infected-family participants (0.0002 AU/mL) during the acute visit, respectively (both p < 0.001). The nasal SARS-CoV-2-specific SIgA decreased rapidly over two weeks in the exposed seronegative individuals compared to a rise in SIgA in infected-family members. The nasal SARS-CoV-2-specific SIgA may have a protective role in preventing systemic infection.

ABSTRACT

Twist-bend nematic liquid crystals (N_{TB} LCs), although consisting of achiral molecules, possess a spontaneous conic helix. They have been intensively studied and utilized in many applications in recent years. Herein we add chiral molecules to N_{TB} LCs and study their effects on the structure of the conic helix. We observe that the system is in the regular chiral nematic phase at high temperature and is still in the twist-bend nematic phase at low temperature. The addition of the chiral molecules does not induce a twist of the conic helical axis. The main effect of the chiral molecules is increasing the cone angle of the conic helix. We show that the structural chirality parameters in the chiral nematic phase and the twist-bend chiral nematic phase can be calculated from the same intrinsic chirality parameter, which only depends on the molecular structure and concentrations of the chiral molecule. We also observe a pretransitional phenomenon that the helical pitch of the chiral nematic phase increases dramatically when temperature is decreased toward the chiral nematic to twist-bend nematic phase transition temperature.

ABSTRACT

The accurate modeling of the complex dynamic stiffness of inflated rubber diaphragms in pneumatic springs is necessary for an efficient design of vibration isolation tables for precision instruments, such as optical devices and nano-scale equipment. In addition to pressurized air, rubber diaphragms, essentially employed for the prevention of air leakage, make a significant contribution to the total complex stiffness. To reflect the effect of the dynamic stiffness of the inflated rubber diaphragm on the total complex stiffness during the initial design or design improvement stage, it is desirable to predict the complex stiffness of the inflated rubber diaphragm beforehand. In this paper, an estimation method for the complex stiffness of inflated rubber diaphragms using the commercial finite element method (e.g., ABAQUS) is proposed. The proposed method reflects their dynamic characteristics under the large static deformation by the Mooney-Rivlin and Morman's constitutive equations. The results of comparison with experimental results indicate that the predictions obtained by the proposed method are congruent with the experimental values of the diaphragm.

ABSTRACT

A vibration isolator embedded in precision equipment, such as a scanning electron microscope (SEM), wafer inspection equipment, and nanoimprint lithography equipment, play a critical role in achieving the maximum performance of the equipment during the fabrication of nano/micro-electro-mechanical systems. In this study, the factors that degrade the performance of SEM equipment with isolation devices are classified and discussed, and improvement measures are proposed from the viewpoints of the measured image patterns and vibrations in comparison with the relevant vibration criteria. In particular, this study quantifies the image patterns measured using SEMs, and the results are discussed along with the measured vibration. A guide for the selection of mounting equipment is presented by performing vibration analysis on the lower mount of the dual elastic mount configuration applied to the SEM, as well as the image patterns analyzed with that configuration. In addition, design modifications for the mount and its arrangement are suggested based on impact tests and numerical simulations.

ABSTRACT

Chirality is of great importance in physical and biological systems. It helps differentiate chemical reactions and physical processes. It was reported that the diffusion of mesogenic chiral guests in chiral nematic liquid crystal hosts profoundly depends on the chirality of the guest and host molecules. When the guest and host molecules have the same handedness, the diffusion is much faster than that when the guest and host molecules have the opposite handedness. In this paper, we report the discovery that chiral differentiation also exists in the diffusion of limonene, which is non-mesogenic in chiral nematic liquid crystals. The diffusion of l-limonene (right-handed) in chiral nematic liquid crystals with a right handed helical structure is faster than that of d-limonene (left-handed). This result might be important in understanding the effects of chirality on physical processes that take place in biological systems. In addition, this effect could be utilized for enantiomer separation in the pharmaceutical industry.

ABSTRACT

We report a novel polymer stabilized liquid crystal based light waveguide display whose performance is significantly improved by using patterned photo-polymerization or an electrode. The waveguide display is edge-lit and operates on the light scattering of the polymer stabilized liquid crystal. When no voltage is applied, the liquid crystal is uniformly aligned and is transparent. The incident light is waveguided through the display by total internal reflection and no light comes out of the viewing side of the display. When a voltage is applied, the liquid crystal is switched to a micron-sized poly-domain structure and becomes scattering. The incident light is scattered out of the viewing side of the display. We demonstrated that by using patterned photo-polymerization or an ITO electrode, the scattering efficiency of the liquid crystal in the voltage-on state is significantly enhanced. We have achieved high brightness, low driving voltage, sub-millisecond switching time and high contrast ratio. This new display is suitable for transparent and augmented display applications.

ABSTRACT

The air-balloon can effectively neutralize hull excitations induced by the propeller cavitation. For the design, it is essential to derive the destructive frequency of an oblate spheroidal air-bubble, which is elaborated on in this paper. Beginning with the exact modal-series solution proposed by Yeh [Ann. Phys. 468, 53-61 (1964)], an approximated form of the scattered pressure is set up by assuming that the acoustic wavelength is much larger than the size of the balloon in the low frequency ranges. An algebraic formula for the destructive frequency can then be written as a function of the resonance frequency and a spatial variable. It is well known that the resonance frequency of a deformed bubble is higher than that of an ideal spherical one with the same volume. In addition to this, the current investigation puts an emphasis on the fact that asphericity induces a more severe shift of the destructive frequency than the resonance frequency, and that its effect needs to be reflected in the balloon design.

ABSTRACT

A colonic muco-submucosal elongated polyp (CMSEP) was identified at colonoscopy in a 53-year-old male patient with lower gastrointestinal bleeding. Non-polypoid depressed type of early cancer was noted at the tip of the colonic polyp. The CMSEP is very rare and incidentally found in most cases. Moreover, its association with colonic neoplasia is extremely rare. To our knowledge, this is the second case report of CMSEP associated with a cancerous transformation.
