ABSTRACT

OBJECTIVE: To explore the experiences of clinical and non-clinical staff in an intensive care unit regarding the perceived benefits and drawbacks of using in situ simulation as a training tool. METHODS: A descriptive phenomenological qualitative study was conducted among clinical and nonclinical ICU personnel. Simulations and interviews were conducted until data saturation was achieved. The interviews were recorded, transcribed verbatim for analysis, and interpreted using the Colaizzi method. RESULTS: Ten participant interviews generated data saturation. ISS was found to be feasible and beneficial in the ICU, facilitating experiential and emotion-based learning in real-world environments. Eight result categories were identified: simulation benefits, simulation benefits in real conditions, scenario authenticity, interference with usual work, ISS sessions, high-fidelity generating affective bonding, ISS as knowledge reinforcement, and recommendations for improvement. The fundamental structure revealed that ISS is perceived as an authentic and emotionally impactful team simulation modality that promotes experiential learning, reflection, and care improvement opportunities within the complex sociotechnical system of the ICU. CONCLUSIONS: All interviewees considered ISS to be a feasible simulation tool that should be implemented in the ICU to improve knowledge and skills, thereby enhancing teamwork.

ABSTRACT

Intraspecies nucleotide sequence variation is a key to understanding the evolutionary history of a species, such as the geographic distribution and population structure. To date, numerous phylogenetic and population genetics studies have been conducted based on the sequences of a gene or an intergenic region on the mitochondrial genome (mtDNA), such as cytochrome c oxidase subunits or the D-loop. To evaluate the credibility of the usage of such 'classic' markers, we compared the phylogenetic inferences based on the analyses of the partial and entire mtDNA sequences. Importantly, the phylogenetic reconstruction based on the short marker sequences did not necessarily reproduce the tree topologies based on the analyses of the entire mtDNA. In addition, analyses on the datasets of various organisms revealed that the analyses based on the classic markers yielded phylogenetic trees with poor confidence in all tested cases compared to the results based on full-length mtDNA. These results demonstrated that phylogenetic analyses based on complete mtDNA sequences yield more insightful results compared to those based on mitochondrial genes and segments. To ameliorate the shortcomings of the classic markers, we identified a segment of mtDNA that may be used as an 'approximate marker' to closely reproduce the phylogenetic inference obtained from the entire mtDNA in the case of mammalian species, which can be utilized to design amplicon-seq-based studies. Our study demonstrates the importance of the choice of mitochondrial markers for phylogenetic analyses and proposes a novel approach to choosing appropriate markers for mammalian mtDNA that reproduces the phylogenetic inferences obtained from full-length mtDNA.

ABSTRACT

Harmful algae blooms (HABs) monitoring has been implemented worldwide, and Chile, a country famous for its fisheries and aquaculture, has intensively used microscopic and toxin analyses for decades for this purpose. Molecular biological methods, such as high-throughput DNA sequencing and bacterial assemblage-based approaches, are just beginning to be introduced in Chilean HAB monitoring, and the procedures have not yet been standardized. Here, 16S rRNA and 18S rRNA metabarcoding analyses for monitoring Chilean HABs are introduced stepwise. According to a recent hypothesis, algal-bacterial mutualistic association plays a critical synergetic or antagonistic relationship accounting for bloom initiation, maintenance, and regression. Thus, monitoring HAB from algal-bacterial perspectives may provide a broader understanding of HAB mechanisms and the basis for early warning. Metabarcoding analysis is one of the best suited molecular-based tools for this purpose because it can detect massive algal-bacterial taxonomic information in a sample. The visual procedures of sampling to metabarcoding analysis herein provide specific instructions, aiming to reduce errors and collection of reliable data.

RESUMEN La presencia de residuos mineros en la región de Coquimbo es un serio problema, que impacta al medio ambiente y a la sociedad. Se estudió el impacto que genera la presencia de un relave abandonado, ubicado en el humedal el Culebrón, que es costero y urbano, determinando la cantidad y la movilidad de algunos elementos, que se encuentran presentes en este relave. Se realizaron dos etapas de muestreos del suelo en la zona del humedal y en el relave, que fueron sometidas a digestión ácida y cuantificación, mediante espectrofotometría de absorción atómica. Los metales con mayor concentración fueron el hierro, seguido del aluminio y, en menor cantidad, zinc, arsénico y plomo. El aluminio tuvo un comportamiento distinto a los demás, ya que presentó baja concentración en el relave y concentraciones equivalentes en las distintas áreas muestreadas, dando la idea que está naturalmente en el humedal. El arsénico presentó una gran movilidad, puesto que su mayor concentración está en el límite del relave y su concentración varió en las distintas zonas del humedal. Del estudio, se concluyó que la distribución de estos elementos en la zona de estudio, se debe a dos factores importantes; el primero, por el proceso geoquímico natural de movilidad de estos componentes y el segundo y más importante es el tsunami que sufrió la comuna de Coquimbo, en 2015.


ABSTRACT The presence of mining waste in the Coquimbo Region is a serious problem, which impacts the environment and society. The impact generated by the presence of an abandoned tailings located in the El Culebrón wetland, which is coastal and urban, was studied, determining the quantity and mobility of some elements that are present in this tailings. Two stages of soil sampling were carried out in the wetland area and in the tailings, which were subjected to acid digestion and quantification by atomic absorption spectrophotometry. The metals with the highest concentration were iron, followed by aluminum, in less quantity were zinc, arsenic, and lead. Aluminum had a different behavior from the others, since it presented low concentration in the tailings, and equivalent concentrations in the different sampled areas, giving the idea that it is naturally in the wetland. Arsenic was highly mobile, since its highest concentration is at the tailings limit and its concentration varied in the different areas of the wetland. From the study, it was concluded that the distribution of these elements in the study area is due to two important factors, the first due to the natural geochemical process of mobility of these components and the second and most important is the tsunami suffered by the commune of Coquimbo in 2015.

ABSTRACT

OBJECTIVE: To describe the clinical and serological patients characteristics with Microscopic Polyangiitis (MPA) and Interstitial lung disease (ILD). METHODS: Of all the patients with AAV diagnosed between 2007-2017 at the Hospital Clinico Universidad de Chile, those with MPA and ILD were selected and studied retrospectively. RESULTS: All patients were Hispanic; median age at diagnosis 65 years (32-84). 59% were female. All were positive for p-ANCA, 16 patients for MPO. Most common manifestations were constitutional symptoms, weight loss and fever. CT-Scans patterns were Usual Interstitial Pneumonia (UIP) in 10 patients, Nonspecific Interstitial Pneumonia (NSIP) in 6 and fibrosis not UIP or NSIP pattern in 1. In 6 cases, ILD was diagnosed 0.5-14 years before MPA and concomitantly in 11. CONCLUSIONS: Although infrequent, Microscopic Polyangiitis should be suspected in patients with ILD particularly if extra-pulmonary manifestations that rise the possibility of a systemic illness are present, regardless of the time elapsed between the latter and the diagnosis of this type of lung involvement. ABSTRACT

Harmful algae blooms (HABs) cause acute effects on marine ecosystems due to their production of endogenous toxins or their enormous biomass, leading to significant impacts on local economies and public health. Although HAB monitoring has been intensively performed at spatiotemporal scales in coastal areas of the world over the last decades, procedures have not yet been standardized. HAB monitoring procedures are complicated and consist of many methodologies, including physical, chemical, and biological water sample measurements. Each monitoring program currently uses different combinations of methodologies depending on site specific purposes, and many prior programs refer to the procedures in quotations. HAB monitoring programs in Chile have adopted the traditional microscopic and toxin analyses but not molecular biology and bacterial assemblage approaches. Here we select and optimize the HAB monitoring methodologies suitable for Chilean geography, emphasizing on metabarcoding analyses accompanied by the classical tools with considerations including cost, materials and instrument availability, and easiness and efficiency of performance. We present results from a pilot study using the standardized stepwise protocols, demonstrating feasibility and plausibility for sampling and analysis for the HAB monitoring. Such specific instructions in the standardized protocol are critical obtaining quality data under various research environments involving multiple stations, different analysts, various time-points, and long HAB monitoring duration.

ABSTRACT

The current study aimed to evaluate the removal of a pesticide mixture composed of the insecticides chlorpyrifos (CP) and diazinon (DZ) from liquid medium, soil and a biobed biomixture by a Streptomyces mixed culture. Liquid medium contaminated with 100 mg L-1 CP plus DZ was inoculated with the Streptomyces mixed culture. Results indicated that microorganisms increased their biomass and that the inoculum was viable. The inoculum was able to remove the pesticide mixture with a removal rate of 0.036 and 0.015 h-1 and a half-life of 19 and 46 h-1 for CP and DZ, respectively. The sterilized soil and biobed biomixture inoculated with the mixed culture showed that Streptomyces was able to colonize the substrates, exhibiting an increase in population determined by quantitative polymerase chain reaction (q-PCR), enzymatic activity dehydrogenase (DHA) and acid phosphatase (APP). In both the soil and biomixture, limited CP removal was observed (6-14%), while DZ exhibited a removal rate of 0.024 and 0.060 day-1 and a half-life of 29 and 11 days, respectively. Removal of the organophosphorus pesticide (OP) mixture composed of CP and DZ from different environmental matrices by Streptomyces spp. is reported here for the first time. The decontamination strategy using a Streptomyces mixed culture could represent a promising alternative to eliminate CP and DZ residues from liquids as well as to eliminate DZ from soil and biobed biomixtures.

ABSTRACT

The diversity of sponge-associated fungi has been poorly investigated in remote geographical areas like Antarctica. In this study, 101 phenotypically different fungal isolates were obtained from 11 sponge samples collected in King George Island, Antarctica. The analysis of ITS sequences revealed that they belong to the phylum Ascomycota. Sixty-five isolates belong to the genera Geomyces, Penicillium, Epicoccum, Pseudeurotium, Thelebolus, Cladosporium, Aspergillus, Aureobasidium, Phoma, and Trichocladium but 36 isolates could not be identified at genus level. In order to estimate the potential of these isolates as producers of interesting bioactivities, antimicrobial, antitumoral and antioxidant activities of fungal culture extracts were assayed. Around 51% of the extracts, mainly from the genus Geomyces and non identified relatives, showed antimicrobial activity against some of the bacteria tested. On the other hand, around 42% of the extracts showed potent antitumoral activity, Geomyces sp. having the best performance. Finally, the potential of the isolated fungi as producers of antioxidant activity seems to be moderate. Our results suggest that fungi associated with Antarctic sponges, particularly Geomyces, would be valuable sources of antimicrobial and antitumoral compounds. To our knowledge, this is the first report describing the biodiversity and the metabolic potential of fungi associated with Antarctic marine sponges.
