ABSTRACT

This study aimed to evaluate arsenic (As), cadmium (Cd), chromium (Cr), and lead (Pb) levels in dried chili peppers from nine districts in Guizhou Province. These heavy metals, widely dispersed and capable of transferring to crops, pose potential health risks to humans. The assessment included modeling daily intake (EDI), target hazard quotient (THQ), total target hazard index (TTHQ), and target carcinogenic risk (TCR) to assess health risks across different population groups. Results showed chromium (0.9540 ± 0.301 mg/kg) and lead (0.8949 ± 0.266 mg/kg) had the highest concentrations, followed by arsenic (0.3287 ± 0.093 mg/kg) and cadmium (0.0627 ± 0.017 mg/kg). Children exhibited higher EDI values than adults, indicating greater health risks from dried chili pepper consumption at equivalent levels. THQ and TTHQ values were below 1 across all regions, indicating no significant health risks associated with dried chili pepper consumption. Similarly, TCR values were below 10-4 for all nine regions, indicating an acceptable level of carcinogenic risk. Overall, consuming dried chili peppers in Guizhou Province poses an acceptable health risk, but caution is advised, especially for children, to limit heavy metal exposure.

ABSTRACT

China, is characterized by its remarkable ethnical diversity, which necessitates whole genome variation data from multiple populations as crucial tools for advancing population genetics and precision medical research. However, there has been a scarcity of research concentrating on the whole genome of ethnic minority groups. To fill this gap, we developed the Guizhou Multi-ethnic Genome Database (GMGD). It comprises whole genome sequencing data from 476 healthy unrelated individuals spanning 11 ethnic minorities groups in Guizhou Province, Southwest China, including Bouyei, Dong, Miao, Yi, Bai, Gelo, Zhuang, Tujia, Yao, Hui, and Sui. The GMGD database comprises more than 16.33 million variants in GRCh38 and 16.20 million variants in GRCh37. Among these, approximately 11.9% (1,956,322) of the variants in GRCh38 and 18.5% (3,009,431) of the variants in GRCh37 are entirely new and do not exist in the dbSNP database. These novel variants shed light on the genetic diversity landscape across these populations, providing valuable insights with an average coverage of 5.5 ×. This makes GMGD the largest genome-wide database encompassing the most diverse ethnic groups to date. The GMGD interactive interface facilitates researchers with multi-dimensional mutation search methods and displays population frequency differences among global populations. Furthermore, GMGD is equipped with a genotype-imputation function, enabling enhanced capabilities for low-depth genomic research or targeted region capture studies. ABSTRACT

Guizhou Province ranks first in terms of Hg reserves and production in the country, and rice is its largest grain crop. In order to study the characteristics and pollution causes of soil-rice Hg content at the provincial level in Guizhou and to carry out safe planting zoning, 1 564 pairs of soil-rice samples, 470 natural soil samples, and 203 individual paddy soil samples were collected to test their Hg content and basic physical and chemical properties of the soil. The results showed that:① Paddy soil was mainly neutral and acidic, the paddy soil ω (Hg) range was 0.005-93.06 mg·kg-1, and the geometric mean was 0.864 mg·kg-1. The Hg content of paddy soil in Guizhou Province was significantly higher than that in natural soil (0.16 mg·kg-1,P < 0.05). Compared with the filtered value and control value, the soil samples exceeded the standard by 63.25% and 14.71%, respectively. Among them, the soil Hg pollution in Danzhai County of Qiandongnan Prefecture, Wuchuan County of Zunyi City, Zhenfeng County of Qianxinan Prefecture, and Wanshan District of Tongren City was more prominent. ② Rice ω(Hg) ranged from 0.000 5 to 0.52 mg·kg-1, and the geometric mean was 0.010 mg·kg-1, the percentage of rice Hg content exceeding the standard was 25.87%, and the exceeding points were mainly distributed in Suiyang County of Zunyi City, Zhenfeng County of Qianxinan Prefecture, Xixiu District of Anshun City, Bijiang District of Tongren City, and other industrial and mining activity-intensive areas. ③ The majority of the study area was in the priority protection category (74.75%); the safe use category accounted for (24.62%); and the strictly controlled category (0.93%) was scattered in Danzhai County at the border between Qiannan Prefecture and Qiandongnan Prefecture, Zhenfeng County in Qianxinan Prefecture, and Wanshan District in Tongren. It is not recommended to plant rice, which can be used as feed for reproduction.

ABSTRACT

Introduction: Salmonella is a major foodborne pathogen worldwide that causes severe morbidity and mortality. It is mainly caused by consuming contaminated food, with retail food considered the primary source. Methods: In Guizhou, China, 102 Salmonella strains isolated from 2016 to 2021 underwent phenotypic antimicrobial resistance testing and whole-genome sequencing (WGS) to understand Salmonella diversity, including serotypes, sequencing types (STs), antimicrobial genes, virulence genes, plasmid types, multi-locus sequence types (MLST), and core genome MLST (cgMLST). Results and discussion: S.Typhimurium was the dominant serotype, and O:4(B) was the leading serogroup. The most prevalent genotype was ST40. Phenotypic antimicrobial resistance identified 66.7% of the sampled isolates as multi-drug resistant (MDR). S.Enteritidis (n = 7), S.Typhimurium (n = 1), S.Indiana (n = 1), S.Kentucky (n = 1), S.Uganda (n = 1), all of which were MDR, were resistant to Colistin. Resistance rates varied significantly across different strains and food types, particularly meat products exhibiting higher resistance. Notably, significant increases in resistance were observed from 2016 to 2021 for the following: ≥ 1 resistant (P = 0.001), MDR (P = 0.001), ampicillin (P = 0.001), tetracycline (P < 0.001), chloramphenicol (P = 0.030), and trimethoprim/sulfamethoxazole (P = 0.003). The marked escalation in drug resistance over the recent years, coupled with the varying resistance rates among food sources, underscores the growing public health concern. Our findings highlight the need for a coordinated approach to effectively monitor and respond to Salmonella infections in Guizhou, China.

To explore the distribution characteristics of paddy soil and rice AS content, as well as the health risks of rice consumption, and to evaluate the safe planting ability of rice, 209 paddy soil samples and 1 567 groups of paddy soil-rice samples were collected, their As content and basic soil physical and chemical properties were determined, and the single-factor pollution index method was used to evaluate the pollution degree of the samples. The results showed that:① the soil of paddy fields in Guizhou Province was mainly neutral, and its fertilizer retention capacity and organic matter content were above the medium level, and the soil was relatively fertile. The range of ω(As) in paddy soil was 0.042-91.75 mg·kg-1, the geometric mean was 10.03 mg·kg-1, and the cumulative effect of paddy soil As was lower than that of natural soil As (P<0.05) by independent sample T. Compared with the screening value (0.2 mg·kg-1) of the Soil Pollution Risk Management and Control Standard for Agricultural Land (GB 15618-2018), the excess rate of soil samples was 15.37%. ② The ω (As) range of rice grain samples was 0.001-0.937 mg·kg-1, the geometric average value was 0.108 mg·kg-1, 10.21% of the rice grain samples exceeded the limit value of "Limit of Contaminants in Food (trial)" (GB 2762-2022), and the locations where the exceedances are mainly found are in the central and northern parts of Qiannan Prefecture, as well as around industrial and mining activity zones in the southern counties and districts of Zunyi. ③ As ingested through rice posed non-carcinogenic risk and carcinogenic risk for adults and children, and the impact on children was greater than that of adults. There is no strict control area for safe rice planting in Guizhou Province, and rice can be safely planted.

ABSTRACT

Expressways are essential for intercounty trips of passenger travel and freight mobility, which are also an important source of vehicular CO2 emissions in transportation sector. This study takes the expressway system of Guizhou Province as the research objective, and establishes the multi-year expressway vehicular CO2 emission inventories at the county level from 2011 to 2019. We employ the extended STIRPAT model incorporating ridge regression to identify driving factors from six different aspects, and then utilize the affinity propagation cluster method to conduct the differentiation research by dividing Guizhou's counties into four clusters. Based upon clustering analysis, localized and targeted policies are formulated for each cluster to reduce expressway vehicular CO2 emissions. The results indicate that generally: (1) Guizhou's expressway vehicular CO2 emissions manifest a continuously upward trend during 2011-2019. Small-duty passenger vehicle (SDV), light-duty truck (LDT), and heavy-duty truck (HDT) contribute to the largest CO2 emissions in eight vehicle types. (2) GDP and population are the foremost two positive driving factors, followed by urbanization rate and expressway length. The proportion of secondary industry is also a positive driver, but that of tertiary industry exhibits an opposite effect. (3) Regional disparity exists in four county clusters of Guizhou Province. Efficient policies are proposed, such as improving the layout and infrastructure of transportation hubs, promoting multimodal integration, and implementing industrial upgrading as per regional advantages. Sustainable expressway vehicular CO2 emission reduction is realized from both the source of industry and low-carbon modes of transport.

ABSTRACT

TheWudu River is a typical mining-type watershed in the karst mountainous area of western Guizhou Province. Based on the collection of the main stream, tributaries, spring water, and mine water samples in Wudu River Basin, the hydrochemical characteristics and control factors of Wudu River Basin were studied using Gibbs diagram, Piper diagram, and mathematical statistics analysis, and the solute contribution rate of different sources was calculated. The results revealed that the pH value of the water in the Wudu River Basin ranged from 7.87 to 8.52, with an average of 8.14. The TDS values ranged from 135 to 243 mg·L-1, with an average of 191.7 mg·L-1. The major cations in natural river and spring water were Ca2+ and Mg2+, the major anion was HCO3-, and the hydrochemical type was HCO3-Ca. However, owing to the influence of mining activities, the major cations in some tributaries were Ca2+ and Na+, and the hydrochemical types transitioned to HCO3·SO4-Ca and HCO3·SO4-Ca·Na. The ion components of river water in Wudu River Basin were affected by mine water discharge and cation exchange, carbonate rock weathering, silicate rock weathering, and agricultural fertilization. The high concentration of SO42- and Na+in mine water was the primary source of SO42- and Na+in the tributaries of the Wudu River. The method for calculating chemical material balance showed that the contribution rate of carbonate rock weathering ranged from 44.12% to 86.92%, with an average of 74.32%. The contribution rate of mining activities ranged from 3.28% to 37.07%, with an average of 11.61%. Carbonate rock weathering was the main controlling factor of hydrochemical components in the Wudu River Basin; meanwhile, mining activities also had a certain impact on river water chemistry but they showed spatial heterogeneity. The average contribution rates of atmospheric precipitation, silicate rock weathering, agricultural activities, and domestic sewage were 3.75%, 4.67%, 2.85%, and 2.81%, respectively, which had a limited impact on the hydrochemical components of the basin.

BACKGROUND: Mosquitoes carry a variety of viruses that can cause disease in humans, animals and livestock. Surveys for viruses carried by wild mosquitoes can significantly contribute to surveillance efforts and early detection systems. In addition to mosquito-borne viruses, mosquitoes harbor many insect-specific viruses (ISVs). Quang Binh virus (QBV) is one such example, categorized as an ISV within the Flavivirus genus (family Flaviviridae). QBV has been specifically documented in Vietnam and China, with reports limited to several mosquito species. METHODS: The homogenate obtained from female mosquitoes was cultured on C6/36 (Aedes albopictus) and BHK-21 (baby hamster kidney) cell lines. Positive cultures were identified by reverse transcription-polymerase chain reaction (RT‒PCR) with taxon- or species-specific primers. Next-generation sequencing was employed to sequence the complete genomes of the identified positive samples. Subsequently, phylogenetic, gene homology, molecular evolutionary and genetic variation analyses were conducted. RESULT: In 2021, a total of 32,177 adult female mosquitoes were collected from 15 counties in Guizhou Province, China. The predominant mosquito species identified were Culex tritaeniorhynchus, Armigeres subalbatus and Anopheles sinensis. Among the collected mosquitoes, three positive cultures were obtained from Cx. tritaeniorhynchus pools, revealing the presence of Quang Binh virus (QBV) RNA sequences. Phylogenetic analysis indicated that the three Guizhou isolates, along with the prototype isolate from Vietnam, formed distinct branches. These branches were primarily closely related to other QBV isolates reported in China. Comparative analysis revealed a high degree of nucleotide and amino acid homology between the Guizhou isolates and both Vietnamese and other indigenous Chinese isolates. Additionally, nonsynonymous single-nucleotide variants (SNVs) were observed in these strains compared to the QBV prototype strain. CONCLUSION: This study represents the first report of QBV presences in Cx. tritaeniorhynchus mosquitoes in Guizhou Province, China. Phylogenetic tree analysis showed that the three Guizhou isolates were most closely related to the QBV genes found in China. In addition, the study of the genetic characteristics and variation of this virus provided a deeper understanding of QBV and enriched the baseline data of these insect-specific flaviviruses (ISFVs).

Introduction: Brucellosis was made statutorily notifiable in 1955, in China, while in Guizhou Province, the pathogen of human brucellosis was isolated for the first time in 2011. However, currently, the brucellosis epidemic is becoming more and more severe in Guizhou Province. The type distribution and genetic characteristics of Brucella in Guizhou Province, as well as its evolutionary relationship with domestic and foreign strains, are still unclear. Methods: MLST, MLVA, and rpoB typing techniques were used for the molecular epidemiological study of the 83 Brucella isolates in Guizhou province. Results: Among the 83 Brucella strains, MLST identified three ST genotypes, of which ST39 is a newly reported type in China. MLVA-16 generated 49 genotypes, and MLVA-11 generated 5 known genotypes and 2 unreported genotypes. Six genotypes were identified by rpoB technology. Discussion: MLVA has a high resolution, but differences at the Bruce 04 and 16 loci cannot exclude associations between epidemics, and combining MLST and rpoB typing methods for epidemiologic tracing can avoid erroneous judgments. Moreover, through the combined analysis of the three typing techniques, the possible origin of the new Brucella can be reasonably inferred, which is also conducive to promoting the subsequent research of the novel Brucella.

The purpose of this study was to investigate tick species around Mount Fanjing and analyze bacterial communities in two species - Rhipicephalus microplus and Haemaphysalis longicornis - parasitizing cattle in Tongren, Guizhou province, Southwest China, using high-throughput sequencing methods. In April 2019, ticks were collected from five sites in Jiangkou County, Yinjiang County, and Songtao County. In total, 296 ticks were collected, comprising two genera and three species: H. longicornis, Haemaphysalis flava, and R. microplus. Rhipicephalus microplus was the most representative species (57.4%) within the collected group, being the dominant species in Tongren City, followed by H. longicornis (39.5%) and H. flava (3.0%). Beta-diversity analysis revealed differences in bacterial community composition among the tick species. The bacterial community structure of R. microplus collected in the three counties was highly similar. Chlorella and Bacillus were highly abundant in H. longicornis. Rickettsia was detected at high relative abundance in R. microplus but in low relative abundance in H. longicornis, suggesting that Rickettsia is more associated with R. microplus than with H. longicornis. More in-depth investigations are needed to determine the pathogenic risk of Rickettsia and its relationship with the host. This is the first survey on tick-borne bacterial communities in this area, which is of great significance for the prevention and control of tick-borne diseases locally.

ABSTRACT

With the objective to ascertain the emissions of biomass combustion in Guizhou, the activity levels were measured through data collection and field surveys, and the emission factors were acquired using actual monitoring data and data cited from previous literature. A 3 km×3 km-gridded emission inventory of nine air pollutants from biomass combustion sources in Guizhou Province in 2019 was developed in combination with GIS technology. The results showed that the total emissions of CO, NOx, SO2, NH3, VOCs, PM2.5, PM10, BC, and OC in Guizhou were estimated to be 293505.53, 14781.19, 4146.11, 8501.07, 45025.70, 39463.58, 41879.31, 6832.33, and 15134.74 t, respectively. The distribution of atmospheric pollutants emitted by biomass combustion sources in different cities was noticeably uneven, being mainly concentrated in Qiandongnan Miao and Dong Autonomous Prefecture. The analysis of variation characteristics of emissions indicated that the monthly emissions were concentrated in February, March, April, and December, and the hourly emissions peaked daily from 14:00 to 15:00. Some uncertainty remained in the emission inventory. It is necessary to perform in-depth analyses of the accuracy of obtaining activity-level data, localize the emission factors through more combustion experiments in subsequent research for improving the emission inventory of air pollutants from biomass combustion in Guizhou Province, and provide a basis for the cooperative governance of the atmospheric environment.

ABSTRACT

China's rapid urbanization has greatly boosted the quality of life of its traditionally impoverished regions. Research into the spatial distribution characteristics, evolution and spatial correlation of the quality of life in impoverished regions can help illuminate the experience of successful development and construct a knowledge base for authorities to devise development strategies. This study focuses its attention on the historically impoverished districts and counties (which are designated as parallel administrative units in China) of Guizhou Province in southwestern China. Extensively citing official statistics on districts and counties released by China's National Bureau of Statistics and local governments, it assesses the quality of life of those places in the 3 years of 2000, 2010, and 2020 from the four dimensions of economy, society, culture, and environment. The aim is to illustrate the distribution characteristics and the evolution of quality of life in Guizhou's historically impoverished districts and counties. In order to understand the characteristics of spatial clustering as well as the patterns of evolution of the quality of life of Guizhou's impoverished districts and counties, the study incorporates spatial autocorrelation analysis into a spatio-temporal analysis of local quality of life. It could presumably help enrich the knowledge base that local authorities draw on to formulate development strategies that are scientific and adapted to local conditions. The study found that while the overall quality of life in all the impoverished districts and counties of Guizhou Province has improved, large gaps in quality of life between eastern and western regions of the province persisted. In addition, the driving force behind the evolution in the overall quality of life of those places changed with time, as did the characteristics of the spatial aggregation in quality of life.

Tuochangjiang River is a typical mining-type watershed in the karst mountainous area of western Guizhou province. The study of its hydrochemical evolution characteristics and driving factors is of great significance to the local economic development and the scientific management of water resources. The samples of river water, spring water, and mine water in the Tuochangjiang River Basin were collected, and the sources of solutes and their contribution to the chemical components of river water were discussed by means of hydrochemical diagrams, mathematical statistics, and the absolute principal component scores-multivariate linear regression model (APCS-MLR). The results showed that the pH of the river water ranged between 7.30 to 8.31, and the TDS value ranged between 40 to 520 mg·L-1, which was mainly contributed by Ca2+, Na+, HCO3-, and SO42-. The dominant cations in river water were Ca2+ and Na+, the dominant anions were HCO3- and SO42-, and the main water chemistry transitioned from HCO3-Ca to HCO3-Ca·Na and HCO3·SO4-Ca·Na type, whereas that of the mine water was mainly the HCO3-Na and HCO3·SO4-Na types. The chemical composition of river water was affected by rock weathering, exchange adsorption of anions, mineral dissolution and precipitation, and human activities, in which Ca2+, Mg2+, and HCO3- were mainly derived from the weathering and dissolution of carbonate and silicate rocks, Na+and SO42- were mainly from the discharge of mining wastewater, and Cl- and NO3- were affected by domestic sewage and agricultural activities, respectively. APCS-MLR analysis further showed that the river water solutes mainly included five sources:discharge of mining wastewater, dissolution of soil minerals, geological background, agricultural activities, and unknown sources, and their contribution rates to river water were 23.49%, 35.04%, 13.87%, 7.96%, and 20.63%, respectively. Mining factors and soil factors were the most important sources of solutes in the river water, and they were the main driving factors for the hydrochemical evolution of the Tuochangjiang River Basin.

Rapid urbanization and irrational human activities have induced in numerous environmental problems, seriously threatening regional ecological security. The establishment and optimization of ecological security patterns (ESPs) were considered as a nature-based solution and an effective way for sustainable development. In this study, the Guizhou Province, a representative karst mountainous region in the southwest of China, was used as the study region. The ecological sources were identified and optimized through integrating ecosystem services and landscape connectivity, and the ecological resistance surface was corrected by representative features of karst areas. The circuit theory was adopted to extract the ecological corridors and barriers. We found that the three ecosystem services (i.e., water conservation, biodiversity maintenance, and soil conservation) had remarkable spatial heterogeneity. The area of optimized ecological sources was enlarged 4752.14 km2. The number of corridors was reduced from 73 to 47 after optimization, with a total length decreased by 1251.97 km. The optimized ecological network structure considerably enhanced ecological connectivity, among the γ index increased by 0.0014, the ß index reduced by 0.0833, while the α index did not change significantly. We concluded that quantitatively exploring the impacts of ecological source optimization are significant for enhancing ecological connectivity. The approach of our study proposes a novel idea into the ESP construction that can provide a meaningful reference for ecological protection and restoration.

ABSTRACT

In order to understand the distribution characteristics of As content in dryland soils and maize seeds in Guizhou province and to evaluate the safety of maize cultivation, 468 natural soil samples, 1260 dryland surface soil samples, and 980 corresponding maize seed samples were systematically collected to determine their As content and basic physicochemical properties. The degree of contamination of the samples was evaluated by using the single-factor contamination index method. The results showed that: ① the range of ω(As) in dryland soil was 0.35-758.53 mg·kg-1, with a geometric mean of 23.28 mg·kg-1. The independent sample T-test showed that the As content of dryland soil in Guizhou province was significantly higher than that of natural soil ω(As) 21.29 mg·kg-1 (P<0.01), indicating the existence of arsenic accumulation effects in dryland soil; compared with the screening value of the "Agricultural Land Soil Pollution Risk Management and Control Standard" (GB 15618-2018), the excess rate of soil samples was 33.81%. ② Corn seed ω(As) ranged from 0.001 to 0.868 mg·kg-1, with a mean value of 0.064 mg·kg-1, and 0.61% of the corn seed samples exceeded the limit value of "Limits of Contaminants in Foods" (GB 2762-2017), with the exceeding points distributed in Bijie City, Qianxinan Prefecture, and Tongren City. ③ When corn seeds were used as feed and grain, maize could be safely grown in dryland soils in Guizhou province. This study showed that the soil As pollution in the dryland of Guizhou province was relatively serious, and overall safe maize cultivation can be achieved; however, the cultivation of maize around the As-related non-ferrous metal mining areas is still of concern.

ABSTRACT

To assess the effect of zinc smelting on environment and human health, the concentration, distribution, sources and health risk of eight heavy metals (Pb, Cd, Cu, Zn, Cr, Ni, Mn, and As) were investigated from agricultural soils in a long-term Zn smelting area in Guizhou, China. Different health risk assessment methods and models were used to access health risks. The results show that the concentrations of all the eight heavy metals were significantly higher than their corresponding background values (p < 0.05). Pb, Cd, and As were the most contaminated elements, with 93.6%, 90.3% and 48.4% of agricultural soil samples higher than the risk screening values, respectively. Statistical analysis indicated that Pb, Cd, Cu, Zn, Mn, and As could be mainly accounted for Zn smelting activities, while Cr and Ni may be generally more controlled by natural sources. The geo-accumulation index (Igeo) suggested that the most seriously contaminating heavy metals were Pb, Cd, and Zn, with 96.8%, 90.3%, and 96.8%, respectively, of the soil samples classified as moderately to extremely contaminated. The non-carcinogenic health risk associated with Pb, Cd and As were observed for children, meanwhile, the carcinogenic risk due to As was found for both adults and children. Regardless of cancer risk or non-cancer risk, local children are at greater risk than adults. Therefore, Pb, Cd and As play the leading role to cause potential health risks in the study area, which need to be paid more attention and also effective control measures should be taken.

ABSTRACT

Rickettsiales (Rickettsia spp., Ehrlichia spp., and Anaplasma spp., etc.) are generally recognized as potentially emerging tick-borne pathogens. However, some bacteria and areas in China remain uninvestigated. In this study, we collected 113 ticks from mammals in Guizhou Province, Southwest China, and screened for the Rickettsiales bacteria. Subsequently, two spotted fever group Rickettsia species and one Candidatus Lariskella sp. were detected and characterized. "Candidatus Rickettsia jingxinensis" was detected in Rhipicephalus microplus (1/1), Haemaphysalis flava (1/3, 33.33%), Haemaphysalis kitaokai (1/3), and Ixodes sinensis (4/101, 3.96%), whereas Rickettsia monacensis was positive in H. flava (1/3), H. kitaokai (2/3), and I. sinensis ticks (74/101, 73.27%). At least two variants/sub-genotypes were identified in the R. monacensis isolates, and the strikingly high prevalence of R. monacensis may suggest a risk of human infection. Unexpectedly, a Candidatus Lariskella sp. belonging to the family Candidatus Midichloriaceae was detected from Ixodes ovatus (1/4) and I. sinensis (10/101, 9.90%). The gltA and groEL gene sequences were successfully obtained, and they show the highest (74.63-74.89% and 73.31%) similarities to "Candidatus Midichloria mitochondrii", respectively. Herein, we name the species "Candidatus Lariskella guizhouensis". These may be the first recovered gltA and groEL sequences of the genus Candidatus Lariskella.

ABSTRACT

Rhipicephalus microplus ticks are vectors for multiple pathogens infecting animals and humans. Although the medical importance of R. microplus has been well-recognized and studied in most areas of China, the occurrence of tick-borne Rickettsiales has seldom been investigated in Guizhou Province, Southwest China. In this study, we collected 276 R. microplus ticks from cattle (209 ticks) and goats (67 ticks) in three locations of Guizhou Province. The Rickettsia, Anaplasma, and Ehrlichia were detected by targeting the 16S rRNA gene and were further characterized by amplifying the key genes. One Rickettsia (Ca. Rickettsia jingxinensis), three Ehrlichia (E. canis, E. minasensis, Ehrlichia sp.), and four Anaplasma (A. capra, A. ovis, A. marginale, Ca. Anaplasma boleense) species were detected, and their gltA and groEL genes were recovered. Candidatus Rickettsia jingxinensis, a spotted fever group of Rickettsia, was detected in a high proportion of the tested ticks (88.89%, 100%, and 100% in ticks from the three locations, respectively), suggesting the possibility that animals may be exposed to this type of Rickettsia. All the 16S, gltA, groEL, and ompA sequences of these strains are 100% identical to strains reported in Ngawa, Sichuan Province. E. minasensis, A. marginale, and Candidatus Anaplasma boleense are known to infect livestock such as cattle. The potential effects on local husbandry should be considered. Notably, E. canis, A. ovis, and A. capra have been reported to infect humans. The relatively high positive rates in Qianxinan (20.99%, 9.88%, and 4.94%, respectively) may indicate the potential risk to local populations. Furthermore, the genetic analysis indicated that the E. minasensis strains in this study may represent a variant or recombinant. Our results indicated the extensive diversity of Rickettsiales in R. microplus ticks from Guizhou Province. The possible occurrence of rickettsiosis, ehrlichiosis, and anaplasmosis in humans and domestic animals in this area should be further considered and investigated.

ABSTRACT

The potato cyst nematodes (PCN) Globodera rostochiensis and Globodera pallida are the very important quarantine nematode pests of potato [Stone 1973]. Both species cause serious potato tuber yield losses. These species are subjected to strict quarantine regulations in many countries (EPPO 2017). G. rostochiensis was detected in Sichuan and Yunnan province, China in 2022 (Jiang et al. 2022). A survey for cyst nematodes in potato fields was conducted in Guizhou Province from 2018 to 2020. A total of 200 samples, including roots and soil, were collected from 40 potato fields in Hezhang (N27 06.145, E104 39.153) and Weining (N26 50.541, E104 09.885) counties in Guizhou Province, China. The Cobb decanting and sieving method was used to isolate cysts and J2s from the soil samples (Southey 1986). The potato roots were stained with acid fusion to observe cyst development. Morphological and molecular analyses indicated that 27 (13.5%) of the samples contained G. rostochiensis. The cyst density ranged between 1-85 cysts per 100 cm3 of soil and a mean density was 15 cysts per 100 cm3 soil. The smoothly rounded cysts were in brown and golden color, and the terminal cone was absent and circumfenestrate. The key morphometrics of cysts (n=20) were 695 ± 26 (685-757) µm in length excluding neck and 690 ± 30 (668 to769) µm in width; the number of cuticular ridges between anus and vulval fenestra was 16.3 ±2.1 (14 to 18); fenestral length was 15.1 ± 2.1 µm (13.18 to 19.27); distance from anus to the edge of fenestra was 61.12 ± 8.9 (49.22 to76.27) µm; and Granek's ratio was 4.54 ± 0.8 (3.97-5.26). The key morphometrics of J2 (n = 20): 468.0 ± 20.1 (427 to - 521) µm in body length, 20.58 ± 0.7 (20.2 to 21.8) µm in stylet length, 43.9 ± 5.6 (40.3 to 53.9) µm in tail length, and 23.1 ± 1.8 (21.77 to 25.32) µm in hyaline region length. The cyst and J2 morphologies were consistent with those of G. rostochiensis (Subbotin et al. 2010, EPPO 2017). Genomic DNA was isolated from cysts (n=20). DNA extraction was performed in a volume of 20 µl containing 3 µl 10× PCR buffer, 3 µl Proteinase K (600 µg µl-1), 14 µl distilled water and a single cyst was added and ground in an ice bath as described by Ou et al. (2008). The internal transcribed spacer (ITS) regions were amplified using the universal primers: rDNA1 (5'-TTGATTACGTCCCTGCCCTTT-3') and rDNA2 (5'-TTTCACTCGCCGTTACTAAGG-3') (Fleming 1998 ), and the 28S rDNA-D2/D3 regions were amplified using the primers: D2A (5´-ACAAGTACCGTGAGGGAAAGTTG-3´) and D3B (5´-TCGGAAGGAACCAGCTACTA-3´) (Subbotin et al. 2006). After the brackets at the beginning and end of the sequences were closed-up, the ITS rDNA sequences (GenBank Accession No. MZ042367 and MZ042368) showed 99.66% - 99.92% identity to G. rostochiensis sequences available in GenBank (FJ212166.1, GQ294513, FJ212164.1 and KJ409617.1). Sequences from the 28S region (GenBank Accession No. MZ057597 and MZ057598) were 99.23% - 99.74% similar to those of G. rostochiensis isolate from Slovakia (KJ409625.1), Italy (KJ409631.1) and United Kingdom (KJ409633.1). We used species specific primers ITS5(5'-GGAAGTAAAAGTCGTAACAAGG-3')and PITSr3 (5'AGCGCAGACATGCCGCAA-3') to amplify the product (Bulman & Marshall 1997; EPPO 2017). A single 434bp fragment was obtained from Hezhang and Weining populations. A host test for the Hezhang and Weining populations were performed by inoculating 1,000 eggs per plant of varieties Qingshu 9, Huize 2 and Hezuo 88 grown in the pots containing 800 cm3 of sterilized soil (soil: sand ratio was 3:1), and four replications were tested in greenhouse under 16 h light, 22°C in the day and 8 h dark in the night. At 90 days post inoculation, 32.6 ± 7, 31.2 ± 8, and 29.5 ± 8 females and cysts were extracted from the infected roots and soils of the varieties Qingshu 9, Huize 2 and Hezuo 88, respectively. No females and cysts were observed on the control plants. The trial indicated that potato cultivars Qingshu 9, Huize 2 and Hezuo 88 are hosts for the Hezhang and Weining populations of Globodera rostochiensis. To the best of our knowledge, this is the first detection of potato cyst nematode Globodera rostochiensis in Guizhou Province, China.

ABSTRACT

Background: Sweltsa is a genus of green stoneflies in the family Chloroperlidae and is distributed throughout the Nearctic and East Palaearctic Regions. As they are sensitive to pollutants, they are often used as an indicator species for determining the quality of water bodies. There are around 57 species of this genus worldwide and 11 of those have been identified from China. New information: A new species of the Alloperlini genus Sweltsa Ricker, 1943, Sweltsaligula Rehman, Huo & Du sp. n. is described from Kuankuoshui National Natural Reserve, Suiyang County, Guizhou Province, southwest China. This is the first report of the family Chloroperlidae from Guizhou Province. Diagnosis, description of male, female and nymph, illustration of terminalia and similarities with closely-related species are provided and discussed.
