BACKGROUND: Inappropriate use of antibiotics is a major cause of antimicrobial resistance (AMR). Inadequate knowledge about AMR among healthcare students could affect their practice of antimicrobial stewardship as future healthcare professionals. This study aims to assess the use of antibiotics and knowledge of AMR among future healthcare professionals of a Nigerian University. METHODS: Respondents' knowledge of antimicrobial resistance, use of antibiotics, and source of antibiotics in the past 12 months was explored using a self-administered questionnaire. Multivariate analyses were used to evaluate the relationship between specific variables and respondents' knowledge. RESULTS: Of the 939 questionnaires administered to the students, 866 were filled given a response rate of 92.2 %. A total of (765; 88.3 %) of the respondents were aware that antimicrobial resistance makes it harder to eliminate the infection from the body as existing drugs become less effective. In all 824, (95.2 %) of the respondents had use antibiotics in the past 12 months. The use of antibiotics to treat malaria was self-reported by (175; 21.2 %). About half (432; 52.4 %) purchased the antibiotics from community pharmacies, while others obtained their antibiotics from the hospitals (192; 23.3 %), patent medicine stores (150; 18.2 %), and friends and family (50; 6.1 %) in the last 12 months. In all 506, (58.4 %) had good knowledge of antimicrobial resistance. Logistic regression shows that students in 3rd to 6th year 9.29 [AOR = 9.29, 95 % CI: (3.7-22.96)], had greater knowledge of antimicrobial resistance. CONCLUSIONS: The healthcare students demonstrated a moderate knowledge of AMR. This underscores the need to adopt several educational tactics to introduce the concepts of AMR to the students and ensure there are strict policies to regulate the flow of antibiotics.

BACKGROUND: Globally, self-medication is a common practice, and an increasingly perceived necessity to relieve burdens on health services. However, inappropriate self-medication may result to reduced health outcomes, increased antimicrobial resistance and economic waste. Healthcare students are the future health professionals who will be consistently responsible for educating the public on rational use of medication. This study therefore aimed to assess the prevalence, knowledge and perception of self-medication practices among healthcare students. METHODS: A cross-sectional study was carried out among 866 healthcare students in a Nigerian University, comprising medical, nursing and pharmacy students. Information was garnered from respondents using a self-administered questionnaire. Data were summarized with descriptive statistics, while Chi-square and logistic regression tests were used for categorical variables at p < 0.05. RESULTS: Mean age was 21 ± 2.8 years, and female respondents were 447 (51.6%). Prevalence of self-medication among respondents was 473 (54.6%). A total of 288 (55.3%) demonstrated good knowledge of self-medication practices, comprising 250 (52.2%) among those who have previously self-medicated and 229 (47.8%) among those who had not. Reasons for engaging in self-medication practices were mentioned to include treatment of minor ailments (357; 32.4%), while 248 (22.5%) believed they had the medical knowledge of what to use. Analgesic (353; 30.1%), antimalarial (352; 30.0%), and antibiotics (182; 15.5%) were the commonest classes of medication used for self-medication. Headache (363; 18.4%), malaria (334; 16.9%), and cough (184; 9.3%) were the most frequently treated conditions. More than half (281; 59.4%) of the respondents' purchased their self-medicated drugs from the community pharmacy. Gender and respondents' disciplines were found to be the independent predictors for good knowledge of self-medication practice. CONCLUSION: Prevalence of self-medication among the studied healthcare students is moderately high, while approximately half demonstrates good knowledge and perception of self-medication practices. Stimulation for self-medication practice largely arise from the perception of treating minor ailments. This underscores a need for advocacy on responsible self-medication practice during the formal training of these future health professionals, in order to avert its imminent/widespread negative consequences.

Yeast display has become an important tool for modern biotechnology with many advantages for eukaryotic protein engineering. Antibody-based peptide interactions are often used to quantify yeast surface expression (e.g., by fusing a target protein to a FLAG, Myc, polyhistidine, or other peptide tag). However, antibody-antigen interactions require high stability for accurate quantification, and conventional tag systems based on such interactions may not be compatible with a low pH environment. In this study, a SNAP tag was introduced to a yeast display platform to circumvent disadvantages of conventional antibody display tags at low pH. SNAP forms a covalent bond with its small-molecule substrate, enabling precise and pH-independent protein display tagging. We compared the SNAP tag to conventional antibody-based peptide fusion and to direct fluorescent domain fusion using antibody fragment crystallizable (Fc) gene libraries as a case study in low pH protein engineering. Our results demonstrated that covalent SNAP tags can effectively quantify protein-surface expression at low pH, enabling the enrichment of Fc variants with increased affinity at pH 6.0 to the neonatal Fc receptor (FcRn). Incorporation of a covalent SNAP tag thus overcomes disadvantages of conventional antibody-based expression tags and enables protein-engineering applications outside of physiological pH.

Emulsion-based techniques have dramatically advanced our understanding of single-cell biology and complex single-cell features over the past two decades. Most approaches for precise single cell isolation rely on microfluidics, which has proven highly effective but requires substantial investment in equipment and expertise that can be difficult to access for researchers that specialize in other areas of bioengineering and molecular biotechnology. Inspired by the robust droplet generation technologies in modern flow cytometry instrumentation, here we established a new platform for high-throughput isolation of single cells within droplets of tunable sizes by combining flow focusing with ultrasonic vibration for rapid and effective droplet formation. Application of ultrasonic pressure waves to the flowing jet provided enhanced control of emulsion droplet size, permitting capture of 25,000 to 50,000 single cells per minute. As an example application, we applied this new droplet generation platform to sequence the antibody variable region heavy and light chain pairings (VH:VL) from large repertoires of single B cells. We demonstrated the recovery of > 40,000 paired CDRH3:CDRL3 antibody clusters from a single individual, validating that these droplet systems can enable the genetic analysis of very large single-cell populations. These accessible new technologies will allow rapid, large-scale, and precise single-cell analyses for a broad range of bioengineering and molecular biotechnology applications.

Predictive ability of genomic EBV when using single-step genomic BLUP (ssGBLUP) in Angus cattle was investigated. Over 6 million records were available on birth weight (BiW) and weaning weight (WW), almost 3.4 million on postweaning gain (PWG), and over 1.3 million on calving ease (CE). Genomic information was available on, at most, 51,883 animals, which included high and low EBV accuracy animals. Traditional EBV was computed by BLUP and genomic EBV by ssGBLUP and indirect prediction based on SNP effects was derived from ssGBLUP; SNP effects were calculated based on the following reference populations: ref_2k (contains top bulls and top cows that had an EBV accuracy for BiW ≥0.85), ref_8k (contains all parents that were genotyped), and ref_33k (contains all genotyped animals born up to 2012). Indirect prediction was obtained as direct genomic value (DGV) or as an index of DGV and parent average (PA). Additionally, runs with ssGBLUP used the inverse of the genomic relationship matrix calculated by an algorithm for proven and young animals (APY) that uses recursions on a small subset of reference animals. An extra reference subset included 3,872 genotyped parents of genotyped animals (ref_4k). Cross-validation was used to assess predictive ability on a validation population of 18,721 animals born in 2013. Computations for growth traits used multiple-trait linear model and, for CE, a bivariate CE-BiW threshold-linear model. With BLUP, predictivities were 0.29, 0.34, 0.23, and 0.12 for BiW, WW, PWG, and CE, respectively. With ssGBLUP and ref_2k, predictivities were 0.34, 0.35, 0.27, and 0.13 for BiW, WW, PWG, and CE, respectively, and with ssGBLUP and ref_33k, predictivities were 0.39, 0.38, 0.29, and 0.13 for BiW, WW, PWG, and CE, respectively. Low predictivity for CE was due to low incidence rate of difficult calving. Indirect predictions with ref_33k were as accurate as with full ssGBLUP. Using the APY and recursions on ref_4k gave 88% gains of full ssGBLUP and using the APY and recursions on ref_8k gave 97% gains of full ssGBLUP. Genomic evaluation in beef cattle with ssGBLUP is feasible while keeping the models (maternal, multiple trait, and threshold) already used in regular BLUP. Gains in predictivity are dependent on the composition of the reference population. Indirect predictions via SNP effects derived from ssGBLUP allow for accurate genomic predictions on young animals, with no advantage of including PA in the index if the reference population is large. With the APY conditioning on about 10,000 reference animals, ssGBLUP is potentially applicable to a large number of genotyped animals without compromising predictive ability.

Structural and functional characteristics of the yeast red pigment (product of polymerization of N1-(beta-D-ribofuranosyl)-5-aminoimadazole), isolated from adel 1 mutant cells of Saccharomyces cerevisiae, its deribosylated derivatives (obtained by acid hydrolysis) and its synthetic pigment analogue (product of polymerization of N1-methyl-5-aminoimadazole in vitro) has been obtained. Products of in vitro polymerization were identified using mass spectrometry. The ability of these pigments to inhibit amyloid formation using insulin fibrils was compared. The entire compounds studied were able to interact with amyloids and inhibit their growth. Electron and atomic force microscopy revealed a common feature inherent in the insulin fibrils formed in presence of these compounds--they were merged into conglomerates that were more stable and resistant to the effects of ultrasound in comparison with insulin aggregates grown without pigments. We speculate that all these compounds can cause coalescence of fibrils, partially block their loose ends and, thereby, inhibit the attachment of new monomers to growing fibrils.

RESUMO

We have developed a technique for discharging fused silica test masses in a gravitational-wave interferometer with nitrogen ionized by an electron beam. The electrons are produced from a heated filament by thermionic emission in a low-pressure region to avoid contamination and burnout. Some electrons then pass through a small aperture and ionize nitrogen in a higher-pressure region, and this ionized gas is pumped across the test mass surface, neutralizing both polarities of charge. The discharge rate varies exponentially with charge density and filament current, quadratically with filament potential, and has an optimal working pressure of ∼8 mT. Adapting the technique to larger test mass chambers is also discussed.

Angus (A) and Bos indicus (B; Brahman or Nellore) reciprocal backcross, embryo transfer calves, belonging to 28 full-sib families, were evaluated for differences in birth weight, gestation length, and weaning weight. Two methods were investigated; method I made no distinction between how the F(1) parents were produced, whereas method II distinguished between the 2 types of F(1) parents (AB vs. BA corresponding to A x B vs. B x A, respectively). Bos indicus backcross calves had a 4.3 d longer (P < 0.05) gestation length but did not differ in their average birth weight from A backcrosses. Among B backcrosses, B x F(1) calves had a 5.2 d longer (P = 0.01) gestation length than F(1) x B calves (290.5 vs. 285.3, respectively). Under method II analysis, there was a consistent trend for gestation length, in which BA F(1) parents produced calves that ranked greater than calves from AB F(1) parents, as sires and dams. Crosses with a greater proportion of B in the sire in relation to the amount in the dam had a heavier (P < 0.05) birth weight (F(1) x A and B x F(1); 38.1 and 38.4 kg, respectively) than their respective reciprocal crosses (A x F(1) and F(1) x B; 34.3 and 33.5 kg, respectively). The F(1) x A and B x F(1) crosses showed a large difference in birth weight between males and females (5.3 and 4.1 kg, respectively), whereas A x F(1) and F(1) x B crosses showed a small difference (P > 0.10) in birth weight between males and females (1.5 and 1.1 kg, respectively). Further examination within each sex showed a difference between male reciprocals that was generally much larger than that between female reciprocals. Calves with a greater percentage of B in the sire compared with the proportion in the dam ranked heavier for weaning weight as for birth weight, though these differences were not significant. In breeding systems involving B x Bos taurus crosses, even when using embryo transfer, not only does the breed composition of the calves affect their preweaning performance, but the particular cross that produces the calves also should be considered in making breeding decisions.

Angus (A) x Bos indicus (B; Brahman or Nellore) reciprocal backcross, embryo transfer calves belonging to 28 full-sib families were evaluated for differences in feedyard initial BW, feedyard final BW, carcass weight, LM area, adjusted fat thickness, intramuscular fat, and Warner-Bratzler shear force. Two methods of analysis were investigated; method I made no distinction between how the F(1) parents were produced, whereas method II distinguished the 2 types of F(1) parents (AB vs. BA, corresponding to A x B vs. B x A, respectively). No significant reciprocal differences for these weight and carcass traits were detected under method I analyses, although the same trend existed for subsequent BW rankings as for birth weight and weaning weight. For each weight phase, the cross that involved a larger proportion of B in the sire in relation to the amount in the dam (F(1) x A and B x F(1)) ranked heavier than the respective reciprocal cross (A x F(1) and F(1) x B). As a whole, A backcross calves had larger (P < 0.001) LM area, more (P < 0.001) marbling, and lower (P < 0.001) Warner-Bratzler shear force than B back-cross calves, but no consistent trends were detected between reciprocal crosses for any of these traits, in contrast with the trends observed for the weight traits. Furthermore, males were heavier than females entering (P < 0.001) and leaving (P < 0.001) the feedyard, produced a heavier carcass (P < 0.001), and had larger LM area (P < 0.05) with less adjusted fat (P < 0.001). No difference existed between the sexes for Warner-Bratzler shear force or marbling. No interactions involving sex, sire type, and dam type were observed for any of these traits. The results were similar under methods I and II analyses, with the exception that a significant sire type x dam type interaction was observed for initial feedyard BW. Results from this study suggest that for weight-related traits, both the breed constitution of the embryo transfer calf and the cross that produces the calf play an important role in its ultimate performance for B crossbred calves. For body composition and meat-related traits, it appears that the breed makeup of the embryo transfer calf itself is more important to animal performance than the specific cross used to produce the calf.
