RESUMO

Changes in keratin gene expression and spatiotemporal regulation determine the compositional content and cellular localization of wool keratin, thereby affecting wool traits. Therefore, keratin gene family member 32 (KRT32) was selected for a study using RT-qPCR, immunofluorescence, and penta-primer amplification refractory mutation system (PARMS) techniques. The results showed that KRT32 mRNA was highly expressed in the skin and localized to the inner root sheath (IRS), outer root sheath (ORS) and dermal papilla (DP). Sequencing results identified eight SNPs in KRT32, and association analyses revealed that the variations were significantly associated with multiple traits in wool (p < 0.05), including MFD, CF and MFC. The constructed haplotype combination H2H3 has higher CF and smaller MFD than other haplotype combination (p < 0.05). In conclusion, KRT32 can be used as a candidate gene for molecular genetic improvement of wool in Gansu Alpine Fine-wool sheep.

Two strains (NYNU 218101 and NYNU 218104) of an asexual yeast species were isolated from insect frass collected in insect tunnels of red leaf plum trees in the Henan Province, central China. Molecular phylogenetic analysis of the D1/D2 domain of the large subunit rRNA gene and the internal transcribed spacer (ITS) region showed that these two strains belonged to the genus Danielozyma, with Danielozyma litseae as the closest known species. They differed from the type strain of D. litseae by 0.6 % substitutions (three substitutions and one gap) in the D1/D2 domain and by 5.1 % substitutions (19 substitutions and six gaps) in the ITS region, respectively. When compared with the partial ACT1, TEF1 and RPB1 gene sequences, they differed by 3 % (26 substitutions), 2.7 % (25 substitutions) and 9 %(54 substitutions) from D. litseae NRRL YB-3246T in these regions. Physiologically, they also differed from its closest known species D. litseae based on the ability to assimilate inulin and galactitol, as well as to grow in 0.1 % cycloheximide and its inability to ferment maltose and raffinose. In order to classify the two new isolates based on morphological and molecular evidence, we proposed the description of a novel species Danielozyma pruni sp. nov. with strain JCM 35735T as holotype (Mycobank MB 849101).

RESUMO

With the emergence of challenges in the environmental degradation and resource scarcity fields, the research of biobased self-healing polyurethane (BSPU) has become a prevailing trend in the technology of the polyurethane industry and a promising direction for developing biomass resources. Here, the production of BSPU from lignocellulose, vegetable oil, chitosan, collagen, and coumarin is classified, and the principles of designing polyurethane based on compelling examples using the latest methods and current research are summarized. Moreover, the impact of biomass materials on self-healing and mechanical properties, as well as the tailored performance method, are presented in detail. Finally, the applications of BSPU in biomedicine, sensors, coatings, etc. are also summarized, and the possible challenges and development prospects are explored to helpfully make progress in the development of BSPU. These findings demonstrate valuable references and practical significance for future BSPU research.

Cellulose nanocrystal (CNC)-based chiral nematic structure is widely used in stimulus response and sensing. A popular area of research is enhancing the mechanical characteristics and environmental adaptability of chiral nematic materials. In this paper, a flexible photonic film with self-healing ability (FPFS) was prepared by combining waterborne polyurethane containing dynamic covalent disulfide bonds (SSWPU) with CNC. The results found that the FPFS showed excellent toughness under the action of stretching, bending, twisting, and folding. The FPFS exhibited an amazing self-healing efficiency, which can be self-healed within 2 h at room temperature. Moreover, the FPFS could respond immediately and produce reversible color change when it was soaked in typical solvents. In addition, when ethanol was used as ink to paint on the FPFS, a visible pattern only under polarized light was formed. This study offers fresh perspectives in the areas of self-healing, biological anticounterfeiting, solvent response, and flexible photonic materials.

To meet more application requirements, improving mechanical properties and self-healing efficiency has become the focus of current research on self-healing PU. The competitive relationship between self-healing ability and mechanical properties cannot be avoided by a single self-healing method. To address this problem, a growing number of studies have combined dynamic covalent bonding with other self-healing methods to construct the PU structure. This review summarizes recent studies on PU materials that combine typical dynamic covalent bonds with other self-healing methods. It mainly includes four parts: hydrogen bonding, metal coordination bonding, nanofillers combined with dynamic covalent bonding and multiple dynamic covalent bond bonding. The advantages and disadvantages of different self-healing methods and their significant role in improving self-healing ability and mechanical properties in PU networks are analyzed. At the same time, the possible challenges and research directions of self-healing PU materials in the future are discussed.

Objective: The detection of Helicobacter pylori mutations that result in antimicrobial resistance can serve as a guideline of antimicrobial therapeutics and probably prevent the failure of clinical treatments. Evaluating the potential of Sanger sequencing to identify genetically resistant determinants in Helicobacter pylori clinical isolates will be important. Methods: 180 cultured strains have been tested using agar dilution for antibiotic susceptibility. NCBI BLAST was used to perform genotypic analysis on the sequencing data. Sanger sequencing was evaluated as an alternative method to detect resistant genotypes and susceptibility. Results: By the conventional E-test, resistance to levofloxacin, amoxicillin, metronidazole, and clarithromycin was 67.3%, 15.1%, 96.4%, and 25.5%, respectively. In contrast, tetracycline had no resistance. Resistance to multiple drugs was observed in 8.12% of the strains. The genetic determinants of resistance to CLA was 23s rRNA, the determinants of resistance to amoxicillin was Pbp1, the determinants of resistance to metronidazole was rdxA, and the determinants of resistance to levofloxacin were GyrA and GyrB. However, there was no association of resistance in tetracycline. Conclusion: We found increased rates of metronidazole antibiotic resistance, highlighting the necessity for alternative therapies and periodic evaluation. Sanger sequencing has proved to be highly effective and holds the potential to be implemented in policies catering to local treatments.

Three thousand and ninety Unigenes were obtained from 10 778 Hevea brasiliensis ESTs. Four hundred and thirty SSRs were distributed in 353 Unigenes, which accounts for 11.42% of the total number of Unigenes. The frequency of SSRs was 1/3.93 kb. Dinucleotide and trinucleotide repeats were the dominant types among the obtained unigenes, accounting for 63.49% and 32.09%, respectively. TC/AG, CT/GA and CTT/GAA, AAG/TTC, and AGA/TCT were the most abundant motifs for dinucleotide and trinucleotide motifs. One hundred and forty-eight primer pairs were designed by PRIMER5.0 and 21 primer pairs were synthesized. Among them, 15 primer pairs can produce clear and stable bands, and the PCR products were screened in denaturing polyacrylamide gel following silver staining. Genetic diversity of 44 rubber clones were investigated with these primer pairs, and a dendrogram of 44 rubber clones was built. The results indicated that it is an effective and feasible way to develop EST-SSR markers from H. brasiliensis EST sequences, and the primers designed in this study can be used in genetic study of rubber tree.

OBJECTIVE: To observe the changes of thromboxane B(2) (TXB(2)), 6-keto-prostaglandin F1alpha (6-K-PGF1alpha) and anticardiolipin antibody (ACA) in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS) before and after institution of nasal continuous positive airway pressure (nCPAP). METHODS: Sixty cases of OSAHS confirmed by polysomnography (PSG) were selected as the trial group, and 20 normal donors without OSAHS were recruited as the control group. Nineteen patients with severe OSAHS were treated by nCPAP. Plasma levels of TXB(2), 6-K-PGF1alpha were detected by enzyme-linked immunosorbent assay (ELISA). RESULTS: Plasma (serum) level of TXB(2) (ACA) was significantly higher in patients with moderate to severe OSAHS than that in control group (P < 0.01), and nCPAP therapy decreased its level significantly (P < 0.01). Plasma level of 6-K-PGF1alpha was significantly lower than that in the control group (P < 0.01), and nCPAP therapy increased its level significantly (P < 0.01). TXB(2) and ACA were correlated positively with AHI, and negatively with minimal oxygen saturation (P < 0.01). 6-K-PGF1alpha was correlated negatively with AHI, and positively with minimal oxygen saturation (P < 0.01). CONCLUSIONS: The results indicate that patients with OSAHS are susceptible to thromboembolism disease. TXB(2), 6-K-PGF1alpha, ACA may be associated with the high prevalence of thromboembolism in patients with OSAHS. nCPAP therapy is effective in correcting TXB(2), 6-K-PGF1alpha, ACA.
