In this context, the aim of this study was to verify the applicability and effectiveness of transcranial stimulation (tACS / tDCS / rTMS) in sleep disorders. A systematic search was performed according to PRISMA guidelines in the Web of Science, PubMed, LILACS, and SciELO databases. Initially, 448 articles were found, according to the eligibility criteria. The physiotherapy evidence database (PEDro) was used to assess the methodological quality of the 11 final articles. In general, the results indicate that there are both favorable and unfavorable reports on the effectiveness of the therapeutic use of transcranial stimulation techniques in sleep disorders and, therefore, it is still an open question, depending on multiple methodological and conceptual factors.

RESUMO

The chronotype refers to the individual differences related to the preference to perform activities or to rest during the wake or in the preferences for a certain period of the day. In this study, we evaluated how the chronotype can be considered a variable of interest for individual personality differences. Still, it was verified how the positive and negative effects and self-esteem interact with the quality of sleep and the circadian personality according to the Big Five personality factors. This study included 150 volunteers of both sexes (41 men and 109 women) aged between 16 and 44 years old (M=22.08; SD=3.8 in age). The analysis of variance showed significant differences for Horne and Ostberg [F(2.148) = 401.69; η2=0.85] usual sleep efficiency [F(2.148) = 4.83; η2=0.6] and the sleep quality index [F(2.148) = 3.25; η2=13.0]. Morning subjects had better behavioral indexes of sleep quality when compared to evening subjects. Regarding positive affects [F(2.147) = 3.54; η2=0.53], morning subjects had a higher score than afternoon subjects (p=0.34) and consequently had higher scores in kindness traits [F(2,148) = 6.81; η2=0.95] and emotional stability [F(2.188) = 6.58; η2=0.91]. The chronotype is associated with personality factors and sleep behavior (efficiency and sleep latency as basic requirements for good quality of sleep) and variables such as sleep efficiency and quality of sleep can be moderators of this behavior.

RESUMO

Limiting CD4+ T cell responses is important to prevent solid organ transplant rejection. In a mouse model of costimulation blockade-dependent cardiac allograft tolerance, we previously reported that alloreactive CD4+ conventional T cells (Tconvs) develop dysfunction, losing proliferative capacity. In parallel, induction of transplantation tolerance is dependent on the presence of regulatory T cells (Tregs). Whether susceptibility of CD4+ Tconvs to Treg suppression is modulated during tolerance induction is unknown. We found that alloreactive Tconvs from transplant tolerant mice had augmented sensitivity to Treg suppression when compared with memory T cells from rejector mice and expressed a transcriptional profile distinct from these memory T cells, including down-regulated expression of the transcription factor Special AT-rich sequence-binding protein 1 (Satb1). Mechanistically, Satb1 deficiency in CD4+ T cells limited their expression of CD25 and IL-2, and addition of Tregs, which express higher levels of CD25 than Satb1-deficient Tconvs and successfully competed for IL-2, resulted in greater suppression of Satb1-deficient than wild-type Tconvs in vitro. In vivo, Satb1-deficient Tconvs were more susceptible to Treg suppression, resulting in significantly prolonged skin allograft survival. Overall, our study reveals that transplantation tolerance is associated with Tconvs' susceptibility to Treg suppression, via modulated expression of Tconv-intrinsic Satb1. Targeting Satb1 in the context of Treg-sparing immunosuppressive therapies might be exploited to improve transplant outcomes.

RESUMO

Neural mechanisms are traditionally examined from different theoretical angles. Similarly, social neuroscience emphasizes the important role that the social environment and neural systems play in the development of social behavior. Thus, this study aims to describe how social neuroscience uses the brain mapping theory and hypothesis testing approach to explain the functioning of social cognition. This approach points out that brain areas, such as the ventromedial prefrontal cortex (vmPFC), lateral temporal cortex, and dorsal anterior cingulate cortex, are specialized in processing social information. These areas are also linked to the regulation of interpersonal relationships, social cooperation, values, empathy, and moral behavior. However, how does social neuroscience explain the phenomena of social behavior? What is the explanation of the social self? Social neuroscience has theoretical and methodological features that bring it closer to a paradigm of social research and neuroscience; the form of mechanistic analysis of biological and cognitive structures broadly describes a neurocognitive mechanism for explaining human social behavior.

RESUMO

Dominant infectious tolerance explains how brief tolerance-inducing therapies result in lifelong tolerance to donor antigens and "linked" third-party antigens, while recipient sensitization and ensuing immunological memory prevent the successful induction of transplant tolerance. In this study, we juxtapose these 2 concepts to test whether mechanisms of dominant infectious tolerance can control a limited repertoire of memory T and B cells. We show that sensitization to a single donor antigen is sufficient to prevent stable transplant tolerance, rendering it unstable. Mechanistic studies revealed that recall antibody responses and memory CD8+ T cell expansion were initially controlled, but memory CD4+Foxp3- T cell (Tconv) responses were not. Remarkably, naive donor-specific Tconvs at tolerance induction also acquired a resistance to tolerance, proliferating and acquiring a phenotype similar to memory Tconvs. This phenomenon of "linked sensitization" underscores the challenges of reprogramming a primed immune response toward tolerance and identifies a potential therapeutic checkpoint for synergizing with costimulation blockade to achieve transplant tolerance in the clinic.

RESUMO

INTRODUCTION: Obesity and diabetes are characterized by chronic inflammatory responses. Roux-en-Y gastric bypass (RYGB) is increasingly regarded as an effective approach for the improvement of glucose homeostasis. In this study, we examined the effects of RYGB on the regulation of early inflammatory responses in the liver and adipose tissue in high-fat diet (HFD)-induced obese (DIO) mice. MATERIALS AND METHODS: RYGB was performed in DIO mice followed by analyses of adiposity, insulin sensitivity, plasma and tissue cytokines and adipokines, tissue NF-κB and JNK/c-Jun activation, and tissue macrophage and T-cell subsets. RESULTS: We found that RYGB resulted in sustained improvement of adiposity and insulin sensitivity. Plasma insulin and leptin levels were increased in untreated DIO mice and reduced in RYGB mice. RYGB maintained plasma adiponectin levels and inhibited monocyte chemoattractant protein-1 and interleukin 6 in white adipose tissue (WAT) and liver. RYGB inhibited NF-κB activation in WAT and muscle, but not in the liver. However, RYGB attenuated the JNK/c-Jun signaling pathway in the liver and WAT at 1 wk after surgery, suggesting that RYGB regulates the tissue-specific inflammatory pathway. RYGB reduced M1-like (F4/80+/CD11c+) differentiation and enhanced M2-like population (F4/80+/CD206c+). RYGB also regulated CD4+ and CD8+ T-cell infiltration and increased Treg cells in the liver and WAT at the same time point. CONCLUSIONS: Our findings demonstrate that RYGB improves obesity and insulin resistance, which are associated with the regulation of early inflammatory reactions in the liver and WAT.

RESUMO

ABSTRACT BACKGROUND: This article systematically updates the literature on changes in visual functions during the phases of the normal menstrual cycle in women. OBJECTIVES: To update Guttridge's 1994 review of visual structures and functions associated with the menstrual cycle and broaden the search through psychophysical, neuroimaging and neurobehavioral measurements covering 1994-2020. DESIGN AND SETTING: Narrative review conducted in a neurosciences and behavior laboratory in Brazil. METHODS: The PubMed, Cochrane Clinical Answers and Google Scholar databases were searched. After screening and applying the eligibility criteria, 32 articles were examined. Through this analysis, the following information was extracted: (1) geographical distribution of the study; (2) sample size (according to age and phase of the menstrual cycle); (3) type of measurements according to psychophysical, neuroimaging and neurobehavioral instruments; (4) vision testing model; (5) visual subcategory evaluated; (6) categories of processed visual stimuli; and (7) main findings. RESULTS: The menstrual phases give rise to significant changes in visual functions, including in relation to orientation and spatial attention, visual campimetry and visual sensitivity. These relate specifically to the follicular and luteal phases. CONCLUSIONS: These findings theoretically expand the effects of menstrual cycles on visual functions found by Guttridge (1994). Despite some inconsistencies in the studies analyzed, it was found that visual processing during the follicular and luteal phases of the normal menstrual cycle of healthy women can explain physiological, cognitive, behavioral and social modulations.

RESUMO

Obese subjects have increase probabilities of developing type 2 diabetes (T2D). In this study, we sought to determine whether gastric bypass prevents the progression of prediabetes to overt diabetes in genetically modified mice and chemically induced diabetic mice. Roux-en-Y gastric bypass (RYGB) was performed in C57BL/KsJ-db/db null (BKS-db/db,) mice, high-fat diet (HFD)-fed NONcNZO10/LtJ (NZO) mice, C57BL/6 db/db null (B6-db/db) mice and streptozotocin (STZ)-induced diabetic mice. Food consumption, body weight, fat mass, fast blood glucose level, circulating insulin and adiponectin and glucose tolerance test were analyzed. The liver and pancreatic tissues were subjected to H&E and immunohistochemistry staining and islet cells to flow cytometry for apoptotic analysis. RYGB resulted in sustained normoglycemia and improved glucose tolerance in young prediabetic BKS-db/db mice (at the age of 6 weeks with hyperglycemia and normal insulinemia) and HFD-fed NZO and B6-db/db mice. Remarkably, RYGB improved liver steatosis, preserved the pancreatic ß-cells and reduced ß-cell apoptosis with increases in circulating insulin and adiponectin in young prediabetic BKS-db/db mice. However, RYGB neither reversed hyperglycemia in adult diabetic BKS-db/db mice (12 weeks old) nor attenuated hyperglycemia in STZ-induced diabetic mice. These results demonstrate that gastric bypass improves hyperglycemia in genetically modified prediabetic mice; however, it should be performed prior to ß-cells exhaustion.

RESUMO

The predominance of visual content in dreams raises a fundamental issue in the formation of images and for the construction of ideas based on the activity of the visual cortex in people with visual impairments. The central question for students of the visual system and for dream connoisseurs is: to what extent the absence or loss of vision will affect the sensory sensitivity for dream construction. Sensory modalities other than vision (tactile and auditory) can influence the functional development of the occipitotemporal visual system in the absence of visual stimulation early in life. What I mean is that blind individuals have significantly less visual capacity, but they also have an increase in the number of auditory, tactile, olfactory, and gustatory sound impressions.

RESUMO

Immunological tolerance to semiallogeneic fetuses is necessary to achieving successful first pregnancy and permitting subsequent pregnancies with the same father. Paradoxically, pregnancy is an important cause of sensitization, resulting in the accelerated rejection of offspring-matched allografts. The underlying basis for divergent outcomes following reencounter of the same alloantigens on transplanted organs versus fetuses in postpartum females is incompletely understood. Using a mouse model that allows concurrent tracking of endogenous fetus-specific T and B cell responses in a single recipient, we show that semiallogeneic pregnancies simultaneously induce fetus-specific T cell tolerance and humoral sensitization. Pregnancy-induced antibodies, but not B cells, impeded transplantation tolerance elicited by costimulation blockade to offspring-matched cardiac grafts. Remarkably, in B cell-deficient mice, allogeneic pregnancy enabled the spontaneous acceptance of fetus-matched allografts. The presence of pregnancy-sensitized B cells that cannot secrete antibodies at the time of heart transplantation was sufficient to precipitate rejection and override pregnancy-established T cell tolerance. Thus, while induction of memory B cells and alloantibodies by pregnancies establishes formidable barriers to transplant success for multigravid women, our observations raise the possibility that humoral desensitization will not only improve transplantation outcomes, but also reveal an unexpected propensity of multiparous recipients to achieve tolerance to offspring-matched allografts.

RESUMO

This article aims to identify the importance of markers at the neurophysiological, physiological, and behavioral levels of mental states linked to the performance of workers as an influence in the field of neuroscience and psychology at work. The author discusses the emergence of neuroergonomy as an area of neuroscience principles and cognitive psychology related to human factors for fundamentals of analysis in brain dynamics, mainly through the use of noninvasive tools, such as the use of electroencephalography for physiological aspects of mental exhaustion of the worker. Neuroergonomics is seen as fundamental for the investigation of complex relationships of organizations, especially when interacting with technology. Future research should involve more complex team-building scenarios and enrich the different neuroergonomic solutions.

RESUMO

Studies reported that tobacco addiction was related to visual impairments, but one unresolved issue is whether the impairments are related to the many compounds existing in the cigarettes or to the effects of nicotine. On the other hand, nicotine gum can be used as replacement therapy or as a neuroprotective agent for some diseases. The main purpose of this controlled trial is to investigate the effects of nicotine gum on vision. The ENIGMA-Vis trial aims to compare two dosages of nicotine gum (2 and 4 mg) and a placebo gum in a randomized, double-blind, placebo-controlled trial of 100 participants to be allocated into a single group assignment of repeated measures (two studies; N = 50 for each one). Eligibility criteria are healthy non-smokers not diagnosed with substance abuse and without an acute or chronic medical condition. Intervention will last three sessions for each participant with a window frame of 1 week per session. Study outcomes are (1) short-term effects of nicotine gum on contrast sensitivity; (2) short-term effects of nicotine gum on chromatic contrast discrimination; and (3) whether demographics, body mass index, or serum cotinine predicts response of visual processing. This study addresses an important gap in the effects of nicotine on vision. One of the main takeaways of this study is to understand the effects of nicotine on contrast sensitivity and chromatic contrast discrimination. This information will provide a further understanding of how nicotine interacts with early visual processes and help determine how the different components present during smoking can affect vision. Clinical Trial Registration Number: RBR-46tjy3.

Abstract The purpose of this research was to adapt the Structured Interview for Assessing Perceptual Anomalies (SIAPA) for use in Brazil as a self-report tool. A non-probabilistic sample of 854 undergraduate students was used, mostly men (51.4%) and aged between 16 and 65 years (M = 23.7, SD = 6.8). After the translation and semantic validation of SIAPA, the validity and precision parameters were tested by means of factor analysis, Cronbach's alpha coefficients, and Item Response Theory (IRT) analysis. The results confirmed the psychometric adequacy of the measure, pointing to a one-dimensional solution with high loads on the general factor and high accuracy (α = 0.88; Ω = 0.88). It is believed that the proposed objective was satisfied, so that at the end of the investigation there was a valid and reliable measure that can be used, both for screening and research, to understand and diagnose perceptual anomalies. RESUMO

The current paradigm that subunit vaccines require adjuvants to optimally activate innate immunity implies that increased vaccine reactogenicity will invariably be linked to improved immunogenicity. Countering this paradigm, nanoparticulate vaccines have been reported to act as delivery systems for vaccine antigens and induce immunity without the need for exogenous adjuvants or local inflammation; however, the mechanisms underlying the immunogenicity of nanoparticle vaccines are incompletely identified. Here, we show that antigens displayed on self-assembling nanofiber scaffolds and delivered intranasally are presented by CD103+ and CD11b+ lung dendritic cells that up-regulate CD80 and migrate into the draining lymph node (LN). This was accompanied by a nearly exclusive priming and accumulation of antigen-specific TH17 cells occurring independently in both LN and lung. Thus, self-assembling peptide nanofiber vaccines may represent a novel, needle- and adjuvant-free means of eliciting protective immunity against fungal and bacterial infections at skin and mucosal barrier surfaces.

RESUMO

The aim of this study was to analyze the contrast sensitivity of spatial luminance and the color discrimination thresholds of the protan, deutan, and tritan axes of people with leprosy. This study included 8 subjects with leprosy (M = 4, W = 4, M = 33.38 ± 8.7) and 8 healthy subjects (M = 4, W = 4, M = 30.89 ± 5.8). The contrast sensitivity was evaluated by the Metropsis software version 11.0 with vertical sinusoidal grids of frequencies of 0.2, 0.5, 1, 5, 10, and 16 cycles per degree of visual angle (cpd) and color vision by the desaturated Lanthony D15 tests and the trivector and ellipse protocols of the Cambridge Color Test. The results showed significant differences between the groups in the processing of spatial frequencies of 0.2 (U = 14; p = .018); 5.0 (U = 45.0; p = .001); 10.0 (U = 45.0; p = .001), and 16.0 (U = 45.0; p = .001) cpd. The difference in color recognition through D15d (U = 4.0; p = .002). Ellipse 2 (U = 10.0; p = .012) and ellipse 3 (U = 9.0; p = .009) were discriminated against. Overall, the results indicate that leprosy changes the visual processing of low, medium, and high spatial frequencies, as well as the sensitivity of the short wavelength (tritan line of confusion) and long (protan line of confusion) cones.

The goal of this research was to obtain normative data of set of faces from Karolinska Directed Emotional Faces (KDEF) in a Brazilian sample. It was used a non-probabilistic sample (convenience sample) of 100 participants from the city of João Pessoa-PB. Age ranged from 18 to 62 years (M=21.6, SD=6.2), in which the majority was female (76%). Results showed that participants achieved a mean hit rate of 76.2%, and expressions of joy (94.7%) and surprise (90.3%) were the easiest emotions to identify and fear (40.65%) the most difficult. Regarding the measures of intensity and valence, disgust and surprise obtained more intense classifications, and joy was the only emotion with high positive valence. These findings were quite similar to those reported in previous research, providing subjective norms of classification suitable to the characteristics of the Brazilian population.

The circadian typology is a behavioral dimension associated with preferences of daily habits. Thus, this study aimed to evaluate the quality of sleep and excessive sleepiness of undergraduates in psychology. Fifty-five students aged between 17 and 34 years participated in this quasi-experimental study. The Pittsburgh Sleep Quality Index, the Morningness-Eveningness Questionnaire, the Karolinska Sleepiness Scale, and the Brief Social Rhythm Scale were used as instruments. After assessing the PSQI, all participants answered the instruments for a period of 14 consecutive days. MANOVA showed significant difference for daily sleepiness during the working days [λλ=.81; F(4.47)=451.53; p<.05], specifically for business hours of 21h [F(2;5)=3.03; p<.05]. It is concluded that students have poor sleep quality and generally try to compensate for the lack of nights of sleep during the week on weekends.

RESUMO

This study evaluated visual sensitivity to luminance contrast during a daily period. Twenty-eight young male adults (M = 24.85; SD = 2.4) with normal color vision and 20/20 visual acuity participated in this study. The circadian pattern was assessed using the Karolinska Sleepiness Scale (KSS), the Pittsburgh Sleep Quality Index (PSQI), and a sleep diary. To measure the luminance contrast, we used version 11.0 of the Metropsis software with sine-element frequency stimuli for spatial frequencies of 0.2, 0.6, 1, 3.1, 6.1, 8.8, 13.2, and 15.6 cycles per degree of visual angle (cpd). The stimuli were presented on a 19-inch color cathode ray tube (CRT) video monitor with a resolution of 1024 × 786 pixels, an update rate of 100 Hz, and a photopic luminance of 39.6 cd/m2. There was a significant difference in KSS on the weekdays [χ2(2) = 20.27; p = .001] and in the luminance contrast for frequencies of 13.2 cpd [χ2(2) = 8.27; p = .001] and 15.6 cpd [χ2(2) = 13.72; p = .041]. The results showed greater stability of the measurement during the afternoon and a reduction in the visual sensitivity in the high spatial frequencies during the night.
