RESUMO

Reproductive deficiency is a major outcome of pesticide exposure sequel to cellular oxidative damage to sex organs. Flavonoid possess potent antioxidant capacities to mitigate pesticide related cellular injury. The present investigation examined the mitigative effect of micronized purified fractions of diosmin and hesperidin on reproductive hormones, sperm parameters, and testicular glycogen in male Wistar rats after sub-chronic Chlorpyriphos (CPF) exposure. Twenty-five male Wistar rats (120-145 g) were randomly allocated five rats per group. Group I (DW) received distilled water (2 ml/kg), Group II (S/oil) received soya oil (2 ml/kg), Group III (DAF) received Daflon at 1000 mg/kg, Group IV (CPF) received Chlorpyriphos (7.74 mg/kg), and Group V (DAF + CPF) received Daflon (1000 mg/kg) followed by CPF (7.74 mg/kg) after 30 min of Daflon. This regimen was administered daily for 60 days. After cervical venesection under light chloroform anesthesia, blood samples were examined for levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone. Each rat's testicular tissue was quickly cut, collected, and glycogen evaluated. Sperm concentration, motility, morphology, and viability were measured in the right caudal epididymis. Results revealed that the untreated CPF group had significantly lower FSH, LH, testosterone, testicular glycogen, and sperm concentration. Additionally, CPF group sperm characteristics were abnormal compared to other groups. These reproductive hormones, testicular glycogen, and sperm parameters improved in the Daflon-treated groups. Hence, pre-treatment with flavonoid fractions of diosmin and hesperidin mitigated CPF-induced reproductive toxicity.

The study aimed to evaluate the effects of Carica papaya peel meal-based diet on spermiogram and reaction time in rabbit bucks. Ripe pawpaw fruits were harvested during the dry season. The peels were carefully removed from the pulp and sun-dried for a week. Afterward, they were ground and included in the test diets as pawpaw peel meal (PPM) at inclusion rates of 0%, 15%, and 30%. Rabbit bucks (n = 15) were randomly separated into three groups of five bucks and labeled as groups A, B, and C. Group A, the control group (0%), was fed the basal protein diet (BD), group B (PPM 15) was given a PPM-based diet (15%), while C (PPM 30) was given diet composed of PPM (30%). Semen samples were collected and evaluated fortnightly for 14 weeks. The reaction time and mean ejaculate volume were lower (P < 0.05) in the treatment groups than in the control. Sperm motility and concentration decreased significantly (P < 0.05) across the groups from week 4 to the end of the experiment. Bucks fed PPM 15%, and PPM 30% had significantly (P < 0.05) higher percentages of dead sperm cells and total spermatozoa abnormalities. The control had (86%) normal spermatozoa morphology while those of PPM 15% and PPM 30% were (61%) and (52%), respectively. PPM 30% had the highest abnormal spermatozoa (47%) compared to PPM 15% (38%) and control (13%). The findings indicate that pawpaw peels up to 15% and 30% in the diet have a negative effect on spermiogram.

Objective: The study aimed at investigating the effects of administering melatonin and a probiotic to streptozotocin-induced diabetic rats on hyperglycaemia, oxidative stress biomarkers and beta-cells. Design: Type 1 diabetes was induced in 5 months-old male Wistar rats by single intraperitoneal (i.p.) administration of freshly-prepared STZ (60 mg/kg body weight). Six groups of 10 rats were used and treated once daily for six weeks; (1) Healthy control: normal saline only; (2) Pre-treated with Melatonin (MEL); (3) Diabetic control; (4) Diabetic + Treated with MEL; (5) Diabetic + Treated with Probiotic (Prob); (6) Diabetic + Treated with MEL + Prob. Blood glucose, body weight, activities of antioxidant enzymes and malondialdehyde concentration in serum and tissues, reduced glutathione and immunohistochemical assay. Data obtained were expressed as mean ± standard error of the mean (Mean ± SEM) and subjected to ANOVA followed by Tukey's post hoc test. Results: Melatonin + Probiotic significantly decreased blood glucose concentrations in diabetic treated rats, compared to the diabetic control rats. MEL + Probiotic increased (p < 0.05) superoxide dismutase activity in serum and liver of diabetic rats. MEL + Probiotic reduced (p < 0.05) malondialdehyde concentration in the serum, liver and kidneys, respectively. MEL + Probiotic treated diabetic rats displayed islets with much greater content of insulin. Conclusion: Melatonin + Probiotic combination was more effective in mitigating hyperglycaemia, oxidative stress, and exerted cytoprotective effect on the beta-cells.

The ability of melatonin to reduce growth inadequacy induced by parental and nutritional combination administration to chlorpyriphos (Ch) and cypermethrin (Cy) was examined in male albino rats. Oral alimentation was given to gravid dams divided into six groups (n = 10; age: 12 weeks) from the first day of pregnancy to the 21st postnatal day. Distilled water (DW), Soya oil (SYO), and melatonin (MeL) groups were exposed to 2 mL/kg, 2 mL/kg, and 0.5 mg/kg, respectively; the Ch+Cy group was co-exposed to Ch (1.9 mg/kg of LD50) and Cy (7.5 mg/kg of LD50); the MChCy group was preconditioned with MeL (0.5 mg/kg), followed by co-exposure to Ch and Cy; and the ChCyM group was exposed to Ch and Cy and post treated with MeL. Male offspring rats were tested for ontogeny criteria at various points after accouchement. MeL pre- and post-administration reduced the variation in litter size and weight, number of live/dead pups, anogenital distance , crown-rump length, the timing of eye and ear openings, and testicular descent caused by fetal and nutritional co-administration to Ch+Cy in offspring male albino rats. MeL demonstrated preventive promise as a result of its apparent antioxidative capability.

The study aimed to evaluate the effects of packing (load carrying) on rectal and body surface temperatures and their circadian rhythmicity in donkeys during the hot-dry season. Twenty pack donkeys of both sexes (15 males and 5 non-pregnant females), aged 2-3 years, with average weight of 93 ± 2.7 kg and divided into two groups randomly, served as experimental subjects. Group 1 donkeys (packing + trekking) were subjected to packing in addition to trekking, while group 2 (trekking only) did not carry any load. All the donkeys were trekked, covering a distance of 20 km. The procedure was repeated three times within the week, one day apart. During the experiment, dry-bulb temperature (DBT), relative humidity (RH) and temperature-humidity index (THI), wind speed and topsoil temperature were recorded; and rectal temperature (RT) and body surface temperature (BST) were measured before and immediately after packing. Starting from 16 h after the last packing, circadian rhythms of RT and BST were recorded at 3-h intervals for 27-h period. The RT and BST were measured by digital thermometer and non-contact infrared thermometer, respectively. The DBT and RH, especially after packing (35.83 ± 0.2 °C and 20.00 ± 0.0%, respectively), were outside the thermoneutral zone for donkeys. The RT value (38.63 ± 0.1 °C) in packing + trekking donkeys recorded immediately (15 min) after packing was higher (P < 0.05) than the value obtained in trekking only donkeys (37.27 ± 0.1 °C). The overall mean RT recorded during the continuous 27-h period of measurement, starting from 16 h after the last packing procedure was higher (P < 0.05) in packing + trekking donkeys (36.93 ± 0.2 °C) compared with trekking only donkeys (36.29 ± 0.3 °C). The BSTs recorded in both groups were higher (P < 0.05) immediately after packing when compared with pre-packing values, but insignificant 16-h post-packing. The RT and BST values were generally higher in the photophase and lower during the scotophase in both groups of donkeys during the continuous recordings. The eye temperature was the closest to the RT, followed by scapular temperature, while the coronary band temperature was the farthest. The mesor of RT in packing + trekking donkeys (37.06 ± 0.2 °C) was significantly higher than in donkeys that were trekked only (36.46 ± 0.1 °C). The amplitude of RT in trekking only donkeys (1.20 ± 0.1 °C) was wider (P < 0.05) than that obtained in packing + trekking donkeys (0.80 ± 0.1 °C). The acrophase and bathyphase occurred later in packing + trekking donkeys (18:10 ± 0.3 h and 6:10 ± 0.3 h, respectively), compared to the trekking only donkeys (16:50 ± 0.2 h and 4:50 ± 0.2 h, respectively). In conclusion, exposure to thermally stressful prevailing environmental conditions during packing increased the body temperature responses, especially in packing + trekking donkeys. Packing significantly influenced the circadian rhythms of body temperatures in working donkeys, as evidenced by differences in circadian rhythm parameters in packing + trekking group, compared with trekking only donkeys during the hot-dry season.

The study aimed to evaluate the effects of L-serine on circadian variation of body temperatures in feed-restricted broiler chickens during the hot-dry season. Day-old broiler chicks of both sexes served as subjects; comprising four groups of 30 chicks each: Group A: water ad libitum + 20% feed restriction (FR); Group B: feed and water ad libitum (AL); Group C: water ad libitum + 20% feed restriction + L-serine (200 mg/kg) (FR + L-serine); Group D feed and water ad libitum + L-serine (200 mg/kg) (AL + L-serine). Feed restriction was performed on days 7-14 and L-serine was administered on days 1-14. Cloacal and body surface temperatures, recorded by digital clinical and infra-red thermometers, respectively, and temperature-humidity index were obtained over 26 h on days 21, 28 and 35. Temperature-humidity index (28.07-34.03) indicated broiler chickens were subjected to heat stress. L-serine decreased (P < 0.05) cloacal temperature in FR + L-serine (40.86 ± 0.07 °C), compared to FR (41.26 ± 0.05 °C) and AL (41.42 ± 0.08 °C) broiler chickens. Peak cloacal temperature occurred at 15:00 h in FR (41.74 ± 0.21 °C), FR + L-serine (41.30 ± 0.41 °C) and AL (41.87 ± 0.16 °C) broiler chickens. Fluctuations in thermal environmental parameters influenced circadian rhythmicity of cloacal temperature; especially the body surface temperatures, positively correlated with CT, and wing temperature recorded the closest mesor. In conclusion, L-serine and feed restriction decreased cloacal and body surface temperatures in broiler chickens during the hot-dry season.

BACKGROUND: Melatonin is a hormone with antioxidant and anti-inflammatory properties and may alleviate the effects of stress on hematologic and biochemical analytes in working donkeys that pack (load-carry). OBJECTIVES: We aimed to evaluate the effects of melatonin administration on hematologic and biochemical responses in donkeys subjected to packing during the hot-dry season. We also examined its effects on circadian rhythms. METHODS: Ten male Nubian pack donkeys with (Packing + Melatonin; P + M) and without melatonin (Packing-Melatonin; P-M) administration were included. Blood samples were collected before, immediately after (15 min), and from 16 h to over 27-h postpacking at 3-h intervals for hematologic and total protein analyses. Serum from blood samples collected before and immediately after packing was analyzed for muscle enzyme activities and electrolyte concentrations. Student's t-test, one-way ANOVA, and online cosinor analysis were used for statistical analyses. RESULTS: Packed cell volumes (PCVs) of the P + M donkeys did not differ before and after packing. However, for the P-M donkeys, values before packing (30.20 ± 0.8%) were higher than those (26.87 ± 0.5%) after packing. The hematologic values recorded in the P + M donkeys were higher than those in the P-M donkeys, especially during the scotophase (P Ë‚ 0.05). The mesor PCV, hemoglobin (Hb), and red blood cell (RBC) values for the P + M donkeys were significantly (P Ë‚ 0.05) higher than those in the P-M donkeys. Lactate dehydrogenase (LDH) activity was not different before (289.40 ± 60.8 U/L) or after packing (323.20 ± 36.0 U/L) in P + M donkeys, but the activity was significantly (P Ë‚ 0.05) higher postpacking than prepacking in the P-M donkeys (294.20 ± 66.9 U/L and 513.40 ± 68.5 U/L, respectively). The relationships between the meteorologic parameters and PCV, Hb, and RBC concentrations in the P + M donkeys were significantly negative (P Ë‚ 0.05). CONCLUSIONS: Melatonin prevented a decrease in PCV and increase in LDH activity immediately after packing compared with non-treated donkeys. There was a marked effect during the scotophase on PCV, Hb, and RBC concentrations. The circadian parameters indicated a significant effect of melatonin on the rhythmicity of the PCV, Hb, and RBC concentration in the treated donkeys. Melatonin administration may reduce the risk of adverse effects of environmental stress and stress due to packing in donkeys during the hot-dry season.

BACKGROUND: Exercise-induced oxidative stress is a challenge in equine sports. This study aims at determining the effects of ergothioneine on heat shock protein-70 (HSP-70) following the stress of an endurance exercise of 30 km by Arabian stallions. Molecular docking was also done to investigate the interaction between the ligand ergothioneine and heat shock protein-70 using sulfogalactosylceramide and sulfogalactoglycerolipid as standards. The study involved a total of 18 clinically healthy stallions, with an average age of 6.7 ± 2.4 years and an average weight of 411.54 ± 12.46 kg. Only clinically healthy stallions were selected as subjects. The stallions were divided into two groups of nine stallions each. Group I (ERGX) was administered ergothioneine at a dose of 0.02 mg/kg once daily orally for four weeks while group II (ERGN) was not administered ergothioneine. The activities of the antioxidant enzymes superoxide dismutase, catalase, and glutathione peroxidase were determined in the two groups before and post-exercise. The concentrations of malondialdehyde and HSP-70 were also determined. RESULTS: The results obtained showed that the activities of the antioxidant enzymes and concentration of HSP-70 were higher (P < 0.05) in the ERGX group compared to the ERGN group. The concentration of malondialdehyde was however lower in the ERGX group. Following molecular docking, ergothioneine and the selected standards have common amino acids at the site of interaction with the target protein (HSP-70) suggesting that ergothioneine may have a modulatory effect on the synthesis of HSP-70. CONCLUSION: The results obtained indicated that ergothioneine modulated the synthesis of HSP-70 and the biomarkers of oxidative stress. It was therefore concluded that ergothioneine may be beneficial to horses subjected to endurance exercise.

The study aimed to evaluate effects of packing (load-carrying) and trekking on hematological and biochemical parameters in donkeys during the hot-dry season. Ten pack donkeys, aged 2-3 years, weighing 93 ± 2.7 kg were enrolled in the study and randomly divided into two groups of five animals each. Group 1 donkeys were subjected to packing + trekking while group 2 trekked only. All the donkeys trekked 20 km thrice, 1-day apart. Blood was collected before and after packing or trekking for hematological evaluation; and serum analyzed for electrolyte concentrations, and aspartate aminotransferase, creatine kinase and lactate dehydrogenase activities. Temperature-humidity index pre and postpacking was 70.00 ± 0.1 and 79.67 ± 0.3, respectively. The packed cell volume (30.93 ± 1.1%) was higher (P < .05) before packing than postpacking value (27.83 ± 0.7%) in group 1 donkeys. Hemoglobin concentration (9.01 ± 0.3 g/dL) decreased (P < .05) after trekking (7.81 ± 0.1 g/dL) in group 2 donkeys. Neutrophil count increased (P < .05) in group 1 donkeys after packing (from 8.63 ± 0.9 × 109/L to 10.59 ± 1.2 × 109/L). Lymphocyte count (8.58 ± 0.8 × 109/L) decreased (P < .05), compared to postpacking (5.23 ± 0.5 × 109/L) in group 1 donkeys. Postpacking neutrophil/lymphocyte ratio in group 1 donkeys (2.33 ± 0.4) was higher (P < .05) than in group 2 donkeys (1.31 ± 0.3). Electrolyte concentrations and enzyme activities did not change significantly. In conclusion, packing + trekking was more stressful to the donkeys than trekking only, evidenced by postpacking increased neutrophil count, neutrophil/lymphocyte ratio and phosphate ion concentration, but decreased lymphocyte count. If this stress is not mitigated, the performance of pack donkeys during hot-dry season may not be optimum.

The study aimed to evaluate the effects of melatonin administration on rectal and body surface temperature (RT and BST, respectively) responses and their circadian rhythmicity in donkeys subjected to packing (load carrying) during the hot-dry season. Twenty donkeys were divided into two equal groups randomly: Groups 1 (packing + melatonin) and 2 (packing - melatonin), subjected to packing and both covered 20 km. The RT, BST, and thermal environmental parameters were measured before and after packing. The procedure was carried out three times within the week, one day apart. This was followed 16-h after the last (third) packing procedure by 27-h recording period of all the parameters at 3-h intervals. The RT (37.77 ± 0.1℃) recorded in packing + melatonin donkeys was lower (P < 0.05) than in (packing - melatonin) (38.29 ± 0.1℃) post-packing, while the BSTs in packing + melatonin donkeys were lower than in (packing - melatonin) donkeys, especially the neck (33.07 ± 0.6℃ vs 35.4 ± 0.7℃, respectively) and coronary band (30.58 ± 0.5℃ vs 33.38 ± 0.7℃, respectively) temperatures. In both groups 16-h post-packing, overall mean RT and BST values were not different, except the coronary band temperature (26.61 ± 0.8℃) that was lower (P < 0.05) in packing + melatonin donkeys than (packing - melatonin) donkeys (28.78 ± 1.4℃). Melatonin-exerted biphasic effects on circadian rhythms of RT and BSTs by reducing body temperatures during the photophase and increasing the values during the scotophase in pack donkeys. In conclusion, melatonin may enhance packing output in donkeys during the hot-dry season.

In mammals a central circadian clock is located in the suprachiasmatic nuclei of the hypothalamus, which regulates the innate physiological rhythms to the ambient 24-h light-dark cycle. Melatonin is an essential component of circadian rhythm. The study aimed to evaluate the effects of melatonin administration on the respiratory rate (RR) and heart rate (HR) and their circadian rhythmicity in donkeys subjected to packing (load carrying) during the hot-dry season. Twenty healthy pack donkeys, aged 2-3 years with average weight of 93 ± 2.7 kg were divided into two groups randomly for the experiment. Group 1 donkeys (packing with melatonin) were administered melatonin for seven days during the study and subjected to packing, while group 2 were packed without melatonin administration. Both groups of donkeys were packed three times within the week, one day apart, covering a total distance of 20 km on each experimental day. Meteorological parameters were recorded during the study period. RR and HR were measured pre- and immediately (15 min) post-packing. Continuous measurement of the parameters started 16 h later, after the last packing procedures for a period of 27 h at intervals of 3 h. Temperature-humidity index (THI) pre-packing (73.67 ± 0.7) was lower (P < 0.05) than that obtained post-packing (80.33 ± 1.2). The THI recorded during the continuous measurement was at its peak at 15:00 h (86), indicating that the afternoon hours were thermally stressful to the donkeys. The THI was strongly correlated with HR recorded in packing (without melatonin) compared to packing (with melatonin) donkeys. The RR and HR values in both groups of donkeys were significantly (P < 0.05) higher immediately after packing. However, the post-packing values of the parameters were not different (P > 0.05) between the two groups of donkeys. The mesor (adjusted arithmetic mean) and amplitude of RR and HR in packing (without melatonin) donkeys were significantly (P < 0.05) higher than that recorded in packing (with melatonin) donkeys. In conclusion, melatonin reduced negative influence on the circadian rhythmicity (mesor and amplitude) of RR and HR in donkeys by exerting its anti-stress and antioxidant effects. The study has demonstrated the beneficial effect of melatonin and its administration may mitigate excessive respiratory and cardiac activities that may reduce the work output of donkeys during the hot-dry season.

The study evaluated effects of L-serine on lipid profile, performance, carcass weight and small intestinal parameters in heat-stressed broiler chickens subjected to feed restriction. Broiler chickens were divided into four groups, comprising 30 each. Group 1, feed restriction (FR); Group 2, feed restriction + L-serine (200 mg/kg) (FR + L-serine); Group 3, ad libitum (AL); Group 4, ad libitum + L-serine (200 mg/kg) (AL + L-serine). L-serine was administered orally from days 1 to 14, and feed restriction was performed on days 7-14. Serum harvested from blood samples on days 21, 28 and 35 was evaluated for lipid profile. Feed and water intake, live weight gain, organ and carcass weight were measured. At 35 days old, broiler chickens (n = 7) per group were sacrificed to evaluate small intestinal morphology. Temperature-humidity index in the pen (30.88 ± 0.81) was above thermoneutral zone, indicating that chickens were subjected to heat stress. Concentrations of low-density lipoprotein, total cholesterol and total triglycerides were lower (p < 0.05), while higher concentration of high-density lipoprotein was recorded in L-serine groups than in the controls. Feed intake and live weight gain on day 35 in L-serine groups were higher (p < 0.05) than in controls. In L-serine groups, liver, spleen, pancreas and heart weight were higher, but abdominal fat was lower than in FR and AL groups. Villus height:crypt height ratio and area of villus surface were highest in L-serine groups than any other group. In conclusion, L-serine decreased low-density lipoprotein, increased feed intake, live weight, organ and carcass weight, villus height:crypt height ratio and villus surface area.

RESUMO

The aim of the study was to evaluate the effects of lycopene and vitamin E on cloacal temperature (CT), thyroid hormones and performance indices in laying hens (Gallus domesticus) during the hot-dry season. The dry-bulb temperature and temperature-humidity index in the pen and CT were measured in all hens twice weekly and thyroid hormones for five consecutive weeks. Ovarian and follicular activities were assessed at the end of the study after slaughter. The CT values in control hens at 09:00 h, 12:00 h and 15:00 h (41.20 ± 0.07 °C, 41.84 ± 1.8 °C and 42.1 ± 1.1 °C, respectively) were higher (P < 0.05), compared to the corresponding values recorded in lycopene (41.50 ± 0.07 °C, 41.50 ± 0.07 °C and 41.73 ± 0.08 °C, respectively), and lycopene + vitamin E (41.31 ± 0.07 °C, 41.40 ± 0.05 °C and 41.63 ± 0.09 °C, respectively). In lycopene + vitamin E laying hens, plasma thyroxine concentration (15.22 ± 1.74 nmol/L) was greater (P < 0.05) than in lycopene (7.64 ± 0.8 nmol/L), vitamin E hens (6.80 ± 1.3 nmol/L) and controls (6.5 ± 0.9 °C nmol/L). Plasma triiodothyronine concentration was highest (P < 0.05) in lycopene + vitamin E (4.80 ± 0.37 nmol/L), compared to lycopene (3.42 ± 0.4 nmol/L), vitamin E (1.96 ± 0.2 nmol/L) and control (1.2 ± 0.1 nmol/L) laying hens. Lycopene + vitamin E hens recorded higher (P < 0.05) count of preovulatory follicles (6.0 ± 0.2) than the controls (4.5 ± 0.3). Countable white follicles were higher (P < 0.05) in lycopene + vitamin E and lycopene hens (58.0 ± 1.4 and 48.5 ± 0.5, respectively) than controls (33.0 ± 2.5). In conclusion, lycopene and vitamin E, especially their combination, modulated the heat stress-induced responses in the laying hens by decreasing CT values, and increasing thyroid hormone concentrations, the count of hierarchical preovulatory and white ovarian follicles during the hot-dry season.

Overworking and overloading of donkeys may cause fatigue. The experiment aims to determine the effects of epicatechin-(4ß-8)-catechin (EEC) on some fatigue biomarkers in packed donkeys during the dry season. 10 jacks, aged between four and five years were used for the study. Five were given EEC in feed seven days before packing (test), while another five were given feed only (control). Blood samples collected for biochemical analyses were analyzed for superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), total antioxidant capacity (TAC), creatine kinase (CK) and tumor necrosis factor-alpha (TNF-α). The SOD activity increased significantly (P < 0.05) in the test group during the cold-dry season after packing. The values of TAC during the packing period were significantly (P < 0.05) higher in test group during the hot-dry season. The TNF-α concentration was higher (P > 0.05) in the control group especially one-hour on route. The CK activity was significantly (P < 0.05) higher in the control donkeys during the hot-dry season. In conclusion, packing donkeys during the dry season resulted in fatigue, but EEC served as an anti-fatigue agent.

The aim of the study was to evaluate oxidative stress and behavioural responses of donkeys during the cold-dry and hot-dry seasons. Thirty donkeys divided into three groups of ten donkeys each according to their age served as experimental subjects. During each season, behavioural and blood parameters of the donkeys were evaluated three times with an interval of one week at 06:00 h, 12:00 h and 18:00 h (GMT+1). During the hot-dry season, the peak dry-bulb temperature (DBT) value of 36.00 ± 1.00°C was recorded at 12:00 h, indicating that the season induced heat stress. The time spent feeding demonstrated diurnal fluctuations in adult, yearling and foal donkeys, with the highest time occurring at 12:00 h in both seasons. The malondialdehyde (MDA) concentrations in adults (2.34 ± 0.14 nMol/L), yearlings (2.28 ± 0.19 nMol/L) and foals (3.48 ± 0.17 nMol/L) during the hot-dry season were significantly (P < 0.05) higher, when compared to the corresponding values (0.96 ± 0.09, 1.18 ± 0.10 and 1.82 ± 0.10 nMol/L, respectively) recorded during the cold-dry season. At 0.3% NaCl-concentration, the percentage haemolysis obtained during the hot-dry season in adult (90.33 ± 0.49%), yearling (91.24 ± 0.88%) and foal (97.44 ± 0.24%) donkeys were significantly (P < 0.05) higher than the corresponding values (78.53 ± 0.98, 78.67 ± 0.58 and 88.23 ± 0.47%, respectively), recorded during the cold-dry season. In conclusion, the hot-dry season induced heat stress in the donkeys, resulting in a decrease in time spent on feeding and oxidative stress, as evidenced by high erythrocyte MDA concentration and percentage haemolysis.

This study aimed to evaluate the effects of packing (load carrying) on the diurnal rhythms of respiratory and heart rates in donkeys during the hot-dry season. Twenty 2- to 3-years-old donkeys weighing 93 ± 2.7 kg were divided into two groups to serve as subjects: Group 1 was subjected to packing + trekking, while group 2 was subjected to trekking only. All donkeys trekked 20 km on each experimental day. The procedure was repeated three times at one-day intervals. Thermal environmental parameters were recorded. Respiratory rate (RR) and heart rate (HR) were measured before and immediately (15 min) after the packing and/or trekking period. Continuous recordings of parameters were performed for 27 hr at 3-hr intervals beginning 16 hr after the last packing and/or trekking period. The RR rose significantly (P<0.05) immediately after packing + trekking and trekking only, from 30.15 ± 0.5 and 27.52 ± 0.5 cycles/min before packing + trekking and trekking only, respectively, to 43.78 ± 3.0 and 46.30 ± 1.8 cycles/min after them, respectively. The HR (76.63 ± 4.5 beats/min) in the packing + trekking donkeys was higher (P<0.05) than that of the trekking-only donkeys (62.33 ± 2.7 beats/min). The acrophase and bathyphase of the HR in both groups were recorded during the photophase and scotophase, respectively, with higher values (P<0.05) in the packing + trekking donkeys than in the trekking-only donkeys. The circadian rhythms of RR and HR did not differ between the groups of donkeys. In conclusion, packing + trekking was more stressful to the donkeys, significantly increasing the HR of donkeys subjected to packing + trekking, compared with trekking only.

The aim of the study was to investigate the effects of L-carnosine administration to prepartum ewes on oxidative status, lipid profile and thermoregulatory responses in the tropics during the hot-dry season. Twenty pregnant ewes were allotted each into two groups of treated and untreated ewes. The untreated ewes received orally 10 mL of water, while treated ewes were administered orally with L-carnosine (100 mg/kg) daily during the last 3 weeks of gestation. Blood samples were obtained during pre-gestation on day 0, gestation days 136 and 143 and after lambing. Dry-bulb temperatures (DBTs) of the pen were measured once per week at 06:00, 14:00 and 18:00 h as well as the thermoregulatory responses of the ewes. Birth weights of the lambs were determined 2-3 h postpartum. The DBT (37.33 ± 1.20 °C) and temperature-humidity index (85.24 ± 1.91) were above the thermoneutral zone. The results revealed no significant effect of L-carnosine on thermoregulatory responses of ewes and on the live-birth weight of their lambs. Malondialdehyde, 8-isoprostane and triglyceride concentrations were lower (P < 0.05) in treated ewes than in untreated ewes, but superoxide dismutase, catalase and glutathione peroxidase activities were higher (P < 0.05) in treated ewes compared to untreated ewes. In conclusion, L-carnosine reduced lipo-peroxidative products and augmented antioxidant enzyme activities in prepartum ewes under hot-dry conditions.

Chlorpyrifos (CPF), a chlorinated organophosphate insecticide that is widely used in agriculture and public health, has neurotoxic effects in animals. In addition to acetylcholinesterase inhibition, CPF has been shown to induce alterations such as oxidative stress and lipid peroxidation. Fisetin is a dietary flavonol that protects the brain tissue against oxidative stress by modulating the activity of antioxidant enzymes. This study was designed to investigate the protective role of fisetin against brain oxidative damages and neurobehavioral parameters induced by subchronic oral exposure to CPF in albino mice. Adult albino mice (males, n = 32, weighing 20 ~ 25 g) were assigned randomly into 4 groups and treated accordingly for 7 weeks as follows: Group 1(S/OIL): served as the control group and were given 2 ml/kg of soya oil; Group 2 (CPF): received CPF (6.6 mg/kg; 1/5th of the LD50); Group 3 (FIS): fisetin (15 mg/kg) and Group 4 (FIS + CPF): received fisetin at 15 mg/kg, followed by CPF (6.6 mg/kg) 30 min later. Co-treatment with FIS + CPF mitigated the increase in brain malondialdehyde concentration (0.28 ± 0.02 nmol/mg) and orchestrated the increase in the activities of catalase (81.35 ± 7.26 µ/mg), superoxide dismutase (93.03 ± 6.63 IU/mL), glutathione peroxidase (68.76 ± 3.554 nmol/mL) and acetylcholinesterase (11.59 ± 0.72 nmol/min/mL) when compared to the CPF group. The result showed that deficits in motor strength and excitability scores induced by subchronic CPF were mitigated by fisetin administration. It was concluded that fisetin has a protective potential in mitigating against oxidative stress and damages in the brain tissues, induced by subchronic exposure to CPF in adult male albino mice.

The study aimed at comparing variations in body temperature values recorded using rectal digital, infrared, and mercury-in-glass thermometers in donkeys during the hot-dry season, prevailing under tropical savannah conditions. Thirty donkeys that served as subjects were divided into three groups of adults, yearlings, and foals. Values of the body temperature of each donkey were recorded bihourly, starting from 06:00 h till 18:00 h, by digital (5-cm depth of insertion), mercury-in-glass (3 cm depth), and infrared thermometers. The values obtained by each type of the thermometer were compared with those recorded using a 15-cm digital probe (Model HI935007, Hanna Instruments, range -50.0 to 150.0°C; accuracy ± 0.2°C) which served as the gold standard. Dry-bulb temperature (34.00 ± 0.50°C), temperature-humidity index (79.65 ± 0.15), and wet-bulb globe temperature (28.00 ± 0.50) index peaked at 14:00 h. The mean body temperatures for rectal probe, digital, mercury-in-glass, and infrared thermometers were 38.35 ± 0.11°C, 37.24 ± 0.04°C, 36.76 ± 0.06°C, and 36.92 ± 0.07°C, respectively. In comparison to the rectal probe, the mean bias for digital (-1.11 ± 0.05°C), mercury-in-glass (-1.59 ± 0.07°C), and infrared thermometers (-1.38 ± 0.07°C) was large. The Passing-Bablok regression plot demonstrated significant deviation from linearity (p < 0.01) when digital, infrared, and mercury-in-glass thermometers were compared to the rectal probe. The area under the curve (AUC) for digital (AUC: 0.7005 ± 0.01 [95%: 0.6853 - 0.7310], infrared (AUC: 0.6711 ± 0.01 [95%: 0.6322 - 0.7100], and mercury-in-glass (AUC: 0.6321 ± 0.01 [95%: 0.6001 - 0.7873] thermometers showed poor accuracy with low sensitivity. In conclusion, the use of digital, mercury-in-glass, and infrared thermometers in recording body temperature in donkeys during the hot-dry season underestimated the values. Their use in measuring body temperature may result in wrong diagnosis, and compromise the control of hyperthermia and diseases associated with thermoregulatory impairments in donkeys.

The objective of the present study was to evaluate the effect of the depth of insertion and environmental parameters on the rectal temperature (RT) in donkeys during the hot-dry season in a tropical savannah zone of Nigeria. The experimental subjects were comprised of thirty donkeys divided into three groups based on age: group I, 10 foals (40.67 ± 2.20 kg; 1.50 ± 0.02 months); group II, 10 yearlings (91.53 ± 0.54 kg; 1.51 ± 0.01 years); and group III, 10 adults (140 ± 0.71 kg; 8.03 ± 0.06 years). Each group was divided into 5 male and 5 female donkeys. Measurements of the RT were recorded with a digital thermometer probe (model HI935007, Hanna Instruments), which was inserted into the rectum at varying depths of 3.5, 7, 10.5, and 14 cm in the same animal in each group. There was a gradual increase in the RT in donkeys as the depth of insertion was increased from 3.5 cm (36.60°C) to 14 cm (38.40°C). Data obtained from the study were subjected to repeated-measures analysis of variance, followed by Tukey's post-hoc test to compare mean values between different depths of RT measurements. Overall, there was a variation in the RT by the depth of insertion with the shallow depth of 3.5 cm having a lower RT than the depths of 7, 10.5, and 14 cm. The variation of the RT observed in donkeys showed that there is need to standardize the probe-insertion depth in veterinary clinical practice for accurate measurement of the RT in donkeys in the Northern Guinea savannah zone of Nigeria.
