RESUMO

PURPOSE: Canine ehrlichiosis is an important tick-borne disease of dogs worldwide. In the present study, we aimed to determine the serum total antioxidant capacity (TAC), malondialdehyde (MDA), nitric oxide (NO), superoxide dismutase, (SOD), glutathione peroxidase (GSH-Px), adenosine deaminase (ADA) activity and serum protein profiles in dogs affected with naturally acquired ehrlichiosis. METHODS: The animal materials had been consisted of ten dogs naturally infected with Ehrlichia canis, and ten controls negative for Ehrlichia canis. TAC, MDA, NO, SOD, GSH-Px, ADA activity and TP, ALB, GLOB levels were measured in sera of the animals. The serum protein concentrations were measured by autoanalyzer. The electrophoretic profiles of serum total protein were determined by native polyacrylamide gel electrophoresis (Native-PAGE). RESULTS: In dogs with ehrlichiosis, decreased TAC (P < 0.05) and GSH-Px (P > 0.05) levels were determined. However, NO (P > 0.05), SOD (P < 0.05), ADA (P > 0.05), MDA (P > 0.05), TP (P < 0.05) and GLO (P < 0.05) levels were found as increased in the Ehrlichia positive dogs. ALB levels were decreased without a statistical significance (P > 0.05). ALB, α1 and ß2 globulin strip densities were found as decreased in native-PAGE, while ß1 and γ globulin strip densities were significantly increased in the E. canis positive group when compared to the control. CONCLUSION: It was determined that the oxidative stress decreased high antioxidant activity in dogs naturally infected with E. canis, and consequently, pro-oxidant and antioxidant defense and serum protein profiles were affected. It was thought that antioxidant supplementation could be beneficial to the treatment of the disease.

RESUMO

Objective: The aim of the present study was to determine tick species found on humans who suffered from tick bite in the Southwestern Anatolia Region, Turkey. Methods: Between January and October 2007, ticks were collected from people admitted to the city and/or town hospitals with complaints of tick bites in nine different provinces of Turkey. Genus and/or species of the ticks in adult, larva and nymph stages were identified microscopically. Identification was done using related taxonomic keys. Results: A total of 2.610 ticks were collected from humans who were admitted to the hospitals with complaints of tick bites in the Southwestern Anatolia Region in the present study. Of these, 1.858 samples were collected from the Aegean Region and the remaining 752 from the Mediterranean Region of the country. The ticks were identified as Hyalomma spp. (78.58%), Rhipicehalus spp. (18.89%), Ixodes spp. (0.88%), Dermacentor spp. (0.77%), Haemaphysalis spp. (0.61%), Argas spp. (0.23%), and Ornithodoros spp. (0.04%). Results indicated that the majority of the ticks were nymphs of Hyalomma spp. (n=1.582). Nymphal stage was most commonly encountered from the Aegean Region and the Mediterranean Region with a prevalence of 46.13% (n=1.204) and 14.48% (n=378) respectively. Within the collected adult ticks (n=913), the majority of the samples were identified as H. marginatum (n=233, 26.09%). Conclusion: The results indicate the high diversity of tick species infesting humans in the Southwestern Anatolia Region, Turkey. So, it is crucial to publish information on tick bite prevention, which would play an important role in reducing the incidence of tick-borne diseases.

RESUMO

The purpose of the present study was to investigate the prevalence and molecular characterization of Giardia duodenalis among dogs in Aydin, Turkey. A total of 473 faecal samples from dogs were collected. The overall prevalence of G. duodenalis was 18.8%. Higher infection rates were observed in dogs younger than three months old, from shelters, and with diarrhoea. Faecal samples of 89 dogs, diagnosed Giardia-positive by microscopy, were found positive by nested PCR. The ß-giardin nested PCR assay revealed assemblage B in all samples (100%), whereas 38 of the samples were mixed with assemblage A (42%). Sequence analysis of isolates indicated sub-genotypes A3 and B4 which have been previously detected in human isolates from Turkey. The results of the present study indicated the relatively high prevalence of giardiasis and the presence of the zoonotic sub-genotypes suggesting the important role of dogs as potential reservoirs of human infections.

RESUMO

ABSTRACT Objective. The aim of the present study was to determine the efficacy of Chloroquine (Cq), an antimalarial medicine, administered at a dose of 2.5 mg/kg, orally, during 5 consecutive days, in Sakiz and Merino lambs naturally infected with Giardia duodenalis. Materials and methods. To this extent weaned 10 weeks of aged lambs were enrolled and randomly assigned into two groups based on treatment (group C, n=18 lambs treated with Cq) and placebo (group P, n=8 untreated control animals). Diagnosis was based on detection of trophozoit and/or cysts on fecal flotation. Cyst count per gram feces (days 0, 3, 7 and 10) was analyzed among groups. Results. During the trial, regarding the efficacy of Cq on days 3., 7., and 10. There was significant (p<0.01) reduction in cyst excretion; whereas evaluation of mean geometric cyst excretion revealed 100% reduction. Conclusions. There was a very high (100%) reduction in cyst excretion in the Cq treatment group compared to the positive untreated control group on days 3, 7 and 10, resulting in a significant (p<0.001) reduction, making Cq, safety, and thus highly effective treatment option of lambs with naturally occuring giardiasis.

Materiales y metodos. RESUMO

Oxidative stress was defined as corruption of balance between oxidant-antioxidant states in favor of oxidants. In this study, it was aimed to determine oxidative stress in naturally infected dogs with Babesia vogeli. The 8-hydroxy-2'-deoxyguanosine (8-OHdG), nitric oxide (NO) and total antioxidant capacity (TAC) levels in the sera were analyzed by ELISA. 8-OHdG levels increased in B. vogeli infected dogs compared to control group (P<0.05). Also, NO levels increased while TOC levels decreased in B. vogeli infected dogs but these variations were not found as statistically important (P>0.05).

RESUMO

Objetive. The purposes of this study were to determine whether secnidazole administered at a single dose of 10 mg/kg, orally, lessens or eliminates Giardia cyst shedding, and to validate the benefit of secnidazole treatment on clinical signs and faecal consistency in lambs naturally infected with Giardia duodenalis. Materials and methods. To this extent weaned 12 weeks of age lambs were selected and randomly assigned into two groups based on placebo (group C, n=7 untreated control group) or treatment (group S, n=10 lambs treated with a single dose of secnidazole at 10 mg/kg). Results. On days 0 and 10, before and after treatment, respectively, reduction in cyst excretion was determined. The faecal consistency and general health were recorded at each sampling day. Conclusions. During the study there was a high (99.98%) reduction in cyst excretion in the secnidazol treatment group compared to the positive control group on day 10, resulting in a significant (p<0.001) reduction, making secnidazole highly effective treatment option.

OBJECTIVE: In this study, it was aimed to determine the prevalence of E. granulosus in the Aydin district. METHODS: Clean and fresh faeces samples were collected off the ground from owned dogs (n=100; 25 female, 75 male) with the range of 6-16 months old. These faeces samples were obtained in 13 villages from five different locations (Centre, Söke, Çine, Bozdogan and Kuyucak) of the Aydin district. The prevalence of E. granulosus was investigated with PCR and prior to applications of molecular techniques, faeces samples were investigated with macroscopic and floatation methods RESULTS: E. granulosus was determined in only one dog (1%; 1/100) and Toxocara canis (11%), Ancylostoma caninum (%3), Taenia spp. (2%) and Capillaria spp. (1%) eggs were also identified. CONCLUSION: Current prevalence (1%) of E. granulosus in this study is lower than the other studies conducted in different parts of Turkey. Determination of E. granulosus with PCR in faeces without determining any parasites with either macroscopic or microscopic examinations could indicate the potential risk of dog faeces for animal owners, veterinarians and parasitology laboratory technicians.

RESUMO

The tick-borne apicomplexan parasite Theileria annulata is endemic in many sub-tropical countries and causes the bovine disease tropical theileriosis. Although the parasite is known to be highly diverse, detailed information is lacking on the genetic structure of natural populations and levels of multiplicity of infection in the cattle host. With the widespread deployment of live attenuated vaccines and the emergence of drug-resistant parasites in the field, it is vital to appreciate the factors which shape genetic diversity of the parasite both within individual hosts and in the wider population. This study addresses these issues and represents an extensive genetic analysis of T. annulata populations in two endemic countries utilising a high-throughput adaptation of a micro- and mini-satellite genotyping system. Parasite material was collected from infected cattle in defined regions of Turkey and Tunisia to allow a variety of analyses to be conducted. All animals (n=305) were found to harbour multiple parasite genotypes and only two isolates shared an identical predominant multi-locus profile. A modelling approach was used to demonstrate that host age, location and vaccination status play a measurable role in determining multiplicity of infection in an individual animal. Age was shown to positively correlate with multiplicity of infection and while positive vaccination status exerted a similar effect, it was shown to be due not simply to the presence of the immunising genotype. Importantly, no direct evidence was found for the immunising genotype spreading or recombining within the local parasite community. Genetic analysis confirmed the tentative conclusion of a previous study that the parasite population appears to be, in general, panmictic. Nevertheless, evidence supporting linkage disequilibrium and a departure from panmixia was uncovered in some localities and a number of explanations for these findings are advanced.

RESUMO

The present study was carried out to find out the prevalence of tropical theileriosis in the Aydin region and to determine immune status of cattle vaccinated with Theileria annulata schizont vaccine using the indirect fluorescence antibody test (IFAT) during and after the disease season. A total of 236 out of 466 cattle found to be seronegative with IFAT were vaccinated in the Aydin region (Cine, Incirliova, Nazilli, and Centrum) before the disease season (March). The remaining cattle (230/466) served as controls. Blood samples were collected from all cattle following the vaccination once in each month of April, June, September, and December (a total of 4 times) to determine the incidence of the disease using microscopic and serologic evaluations. Results indicated that the incidence of the disease varied among the regions. Clinical theileriosis was found in a total of 22 cattle (10 vaccinated, 12 unvaccinated) during the disease season.
