Background: It was previously shown that a bodyweight reduction among patients with nonalcoholic fatty liver (NAFLD) was connected to the lower concentration of arachidonic and linoleic acid derivatives in their blood. We hypothesized that the concentration of these lipids was correlated with the extent of their body mass reduction and, thus, liver steatosis. Methods: We analyzed 68 individuals who completed the dietary intervention. Patients were divided into two groups depending on their body mass reduction (more or less than 7%). Before and after the dietary intervention, all patients had the following measurements recorded: body mass, waist circumference, stage of steatosis, fatty liver index, liver enzymes, lipid parameters, insulin and glucose. Concentrations of lipoxins A4 (LTX A4), hydroxyeicosatetraenoic fatty acids (5(S)-HETE, 12(S)-HETE and 16(S)-HETE), hydroxyoctadecaenoic acids (9(S)-HODE and 13(S)-HODE) and 5-oxo-eicosatetraenoic acid (5-oxo-ETE) were measured in serum samples collected before and after the dietetic intervention using high-performance liquid chromatography (HPLC). Results: Patients who reduced their body mass by more than 7% revealed a significant improvement in their steatosis stage, waist circumference, fatty liver index, triglycerides and cholesterol. Conclusion: A reduction in body mass by more than 7% but not by less than 7% revealed a significant improvement in steatosis stage; waist circumference; fatty liver index; and levels of triglycerides, cholesterol, 5-oxo-ETE and LTXA-4.

OBJECTIVE: The aim of the study was to compare the impact of 6 weeks of reducing daily caloric intake by 20% of total daily energy expenditure (TDEE)-CRI vs. reducing daily caloric intake by 30% of TDEE-CRII on body mass reduction and insulin metabolism in former athletes. METHODS: 94 males aged 35.7 ± 5.3 years, height 180.5 ± 4.1 cm, and body mass 96.82 ± 6.2 kg were randomly assigned to the CRI (n = 49) or CRII (n = 45) group. Thirty-one participants (18 subjects from CRI and 13 from CRII) resigned from the study. The effects of both diets on the body composition variables (body mass-BM; body fat-BF; fat free mass-FFM; muscle mass-MM; total body water-TBW), lipid profile (total lipids-TL; total cholesterol-TCh; HDL cholesterol-HDL; LDL cholesterol-LDL; triglycerides-TG), and glucose control variables (glucose-GL, insulin-I, HOMA-IR, insulin-like growth factor-1-IGF-1, leptin and adiponectin) were measured. RESULTS: After adhering to the CR I diet, significant differences were observed in FFM, MM and TG. After adhering to the CR II diet, significant differences were registered in tCh, TL and LDL. Both diets had a significant influence on leptin and adiponectin concentrations. Significant differences in FFM, MM, and tCh were observed between the CR I and CR II groups. At the end of the dietary intervention, significant differences in BF, FFM, MM and TBW were observed between the CR I and CR II groups. CONCLUSION: The 6 weeks of CR II diet appeared to be more effective in reducing BF and lipid profile and proved to be especially suitable for subjects with high body fat content and an elevated level of lipoproteins and cholesterol. Both reductive diets were effective in improving the levels of leptin and adiponectin in obese former athletes.

This study is aimed at determining the content of water soluble vitamins in amateur sportsmen before and after the use of reduction diet. Twenty adult male amateur sportsmen aged between 20 and 43 qualified for this study. The participants adhered to individually adjusted reduction diets for 3 months. Diet supplements were excluded from the diet during that time. Vitamins were analyzed before and after the reduction diet using HPLC. The following vitamins were analyzed: B1, B2, B3, PP, B5, B6, B7, B9, B12 and vitamin C. Statistically significant increase of riboflavin in blood plasma (p=0.0254) and statistically significant (p=0.048) decrease in the concentration of niacin was found when compared to the period before the diet. Appropriate level of riboflavin can be obtained by the consumption of proper amount of diary in the diet. Supplementation with PP is advised and, in case of improper diet, the supplementation of B1, B7 and B12 may also be considered.

Background: Nonalcoholic fatty liver disease (NAFLD) is closely related to the metabolism disorders of fatty acids. The pathogenesis of the disease includes an increased concentration of FFA in blood, an increase in the biosynthesis of fatty acids, and disorders in the process of ß-oxidation. Objective: The aim of the study was to analyze the fatty acids in erythrocyte membranes among 55 patients with NAFLD who were subjected to a 6-month dietary intervention in order to reduce fatty liver. Materials and Methods: Basic anthropometric and biochemical measurements were performed. The profile of fatty acids was measured in the membranes of erythrocytes and analyzed by gas chromatography. The dietary compliance was evaluated using 72-diary questionnaires, anthropometric measurements. Results: With the reduction of fatty liver (p<0.01), the patients' biochemical and anthropometric parameters were significantly improved. A significant decrease in the concentration of alanine aminotransferase (p<0.01) and asparagine aminotransferase (p<0.01) was observed, along with a decrease in the amount of insulin (p<0.05) and insulin resistance (p<0.05). Significant changes in terms of the fatty acid profile were observed among patients who followed the dietary intervention. There was a noticeable tendency in terms of the reduction palmitic acid (p<0.055) and a significant reduction of stearic acid (p<0.05). Significant changes in the profile of fatty acids were also associated with the reductionof palmitoleic (p<0.05) and oleic acids (p<0.05). Another statistically significant change observed was the increase in polyunsaturated fatty acids. In particular (p<0.01) the rise of eicosapentaenoic (p<0.055) and docosahexaenoic acids (p<0.55) was noted. Conclusion: The profile of fatty acids turned out to be a potential biomarker of the liver changes during NAFLD regression. Further research is needed to fully elucidate the usefulness and applicability of our findings in the management of NAFLD.

INTRODUCTION: The proper development of a child is linked with proper nutrition, including nutritional habits which are formed from childhood. THE AIM OF THE STUDY: The aim of the study was to establish a list of the most popular food products among children and to develop a register of potentially dangerous substances on a Facebook website. MATERIALS AND METHODS: A website was created on Facebook. The participants provided lists of favorite dishes or products. RESULTS: The study involved 264 participants. An inverse correlation was observed with reference to the age of the subjects and the occurrence of sugar syrup in their diet (R=-0.20; p<0.001), glucose-fructose (R= -0.18; p< 0.004), and glucose (R=-0.13; p< 0.039) syrups. The most common potential food allergens are: gluten (R=0.28; p<0.001), eggs (R=0.28; p<0.001), and wheat (R=0.25; p<0.001). The main substances added to food that are present in a child's diet that increase proportionally with reference to the child's age are: salicylates (R=0.37; p<0.001), iron and ammonium sulfates (R=0.21; p<0.001). CONCLUSION: The choices of favorite products are related to age and sex. Products containing gluten, the consumption of which increases with age, carry a risk of undiagnosed celiac disease and non-celiac gluten sensitivity in people with a genetic predisposition. Facebook has fulfilled its role as an effective tool for gathering information about the food preferences of children and adolescents.

Solute carrier (SLC) transporters are a diverse group of membrane transporter proteins that regulate the cellular flux and distribution of endogenous and xenobiotic compounds. Post-translational modifications (PTMs), such as ubiquitination, have recently emerged as one of the major regulatory mechanisms in protein function and localization. Previously, we showed that SLC amino acid transporters were on average 6-fold de-ubiquitinated and increased amino acid levels were detected in ρ° cells (lacking mitochondrial DNA, mtDNA) compared to parental cells. Here, we elucidated the altered functionality of SLC transporters and their dynamic ubiquitination status by measuring the uptake of several isotopically labeled amino acids in both human osteosarcoma 143B.TK- and ρ° cells. Our pulse chase analysis indicated that de-ubiquitinated amino acid transporters in ρ° cells were accompanied by an increased transport rate, which leads to higher levels of amino acids in the cell. Finding SLC transport enhancers is an aim of the pharmaceutical industry in order to compensate for loss of function mutations in these genes. Thus, the ubiquitination status of SLC transporters could be an indicator for their functionality, but evidence for a direct connection between de-ubiquitination and transporter activity has to be further elucidated.

The aim of the study was to investigate the efficacy of 6 week Mediterranean diet or 30% calorie restriction on the fatty acid profile and eicosanoids (hydroxyoctadecadienoi acids and hydroxyeicosatetraenoic acids) concentration. Furthermore, basic biochemical variables such as insulin, glucose, HOMA-IR, and a lipid profile were estimated. The study enrolled 94 Caucasian former athletes aged 20-42, with body height of 179 ± 16.00 cm and body mass of 89.26 ± 13.25 kg who had not been active for at least 5 years. The subjects were randomly assigned to one of the three intervention groups: CR group - the 30% calorie restriction (n = 32), MD group - the Mediterranean diet (n = 34), and C group - a control group (n = 28). The pattern of nutrition was analysed before and after the experiment using the 72 h food diaries. In order to evaluate the effect of diet intervention, the following variables were measured: anthropometrics, basic biochemical variables (insulin, fasting glucose, HOMA-IR, lipid profile), fatty acids and their blood derivatives profiles. The CR group showed significantly lower levels of several biochemical variables, i.e., BMI, total cholesterol LDL, TG, total lipids, insulin and HOMA - IR (p < 0.05). Subjects consuming the MD diet significantly decreased their BMI and reduced the level of total lipids (p < 0.05). We did not find any significant changes in the C group. The analysis of the fatty acid profile revealed that the CR group had a significantly decreased EPA level (p < 0.05). The MD group showed a significantly increased level of the DHA (p < 0.05) and improvement in the omega - 3 index (p < 0.05). Subjects following the MD also showed significantly lower concentrations of 15 - hydroxyicosatetraenoic acid (15-HETE). We did not observe any significant differences between the CR and C groups. Within short time, calorie restriction helps to improve lipid variables and insulin resistance. The MD diet seems to be more advantageous in the decrease of inflammation, but does not improve basic biochemical variables. We can conclude that calorie restriction can be a good choice for former athletes, although EPA and DHA supplementation is needed.

Nonalcoholic fatty liver disease (NAFLD) is one of the most common liver diseases in the world. Nowadays, the number of people with type II diabetes and hypertension is growing. These symptoms belong to metabolic syndrome and the liver manifestation of this disease is NAFLD. Most people suffering from NAFLD are obese but this disease can also affect the lean individuals. This disease in extreme cases can lead to liver fibrosis and cirrhosis. Although NAFLD remains largely asymptomatic for the most of the time it can progress to a severe form of disease called nonalcoholic steatohepatitis (NASH) or even hepatocellular carcinoma. In this review we describe a role of factors that influence NAFLD pathogenesis such as: insulin resistance, epigenetic factors, adipokine dysfunction, intestinal flora disorder, oxidative stress and markers of inflammatory states in the form of fatty acid derivatives.

Nutrients play a fundamental role as regulators of the activity of enzymes involved in liver metabolism. In the general population, the action of nutrients may be affected by gene polymorphisms. Therefore, individualization of a diet for individuals with fatty liver seems to be a fundamental step in nutritional strategies. In this study, we tested the nutrient-induced insulin output ratio (NIOR), which is used to identify the correlation between the variants of genes and insulin resistance. We enrolled 171 patients, Caucasian men (n = 104) and women (n = 67), diagnosed with non-alcoholic fatty liver disease (NAFLD). From the pool of genes sensitive to nutrient content, we selected genes characterized by a strong response to the NIOR. The polymorphisms included Adrenergic receptor (b3AR), Tumor necrosis factor (TNFα), Apolipoprotein C (Apo C III). Uncoupling Protein type I (UCP-1), Peroxisome proliferator activated receptor γ2 (PPAR-2) and Apolipoprotein E (APOEs). We performed three dietary interventions: a diet consistent with the results of genotyping (NIOR (+)); typical dietary recommendations for NAFLD (Cust (+)), and a diet opposite to the genotyping results (NIOR (-) and Cust (-)). We administered the diet for six months. The most beneficial changes were observed among fat-sensitive patients who were treated with the NIOR (+) diet. These changes included improvements in body mass and insulin sensitivity and normalization of blood lipids. In people sensitive to fat, the NIOR seems to be a useful tool for determining specific strategies for the treatment of NAFLD.

Introduction: The aim of this study is to assess the nutritional habits of participants in the research project "CRON -diet as an alternative method for athletes who have completed their sporting career". Materials and methods: 94 former athletes from Poland participated in this study. A standardized Food Frequency Questionnaire was used for interview. Respondents were interviewed once, during the first visit after inclusion in the study. Results: Most former athletes had 3 meals per day during the last 12 months (39.4%), not respecting specified consumption hours (44.7%). The most commonly consumed foods are: whole grain bread, fruits and vegetables. Respondents reported the consumption of these foods several times a week. The study revealed a correlation between the consumption of fruit in general and a preference for apples and pears (r = 0.7340; p < 0.05). Conclusions: The study showed that former athletes' diets are quite diversified. It should be mentioned that former athletes' diets consists of a low number of meals, which are not consumed at fixed times. Respondents were aware that their diet provided too much sugar and fat; some of them declared that they try to reduce the consumption of these products in their daily diet.

Introduction: Vegetables and fruit are an essential part of a healthy and balanced diet. It has been shown that regular consumption of fruit and vegetables can reduce the risk of chronic diseases. However, in the past 8 years fruit and vegetable consumption among Polish youth has significantly decreased. Materials and methods: Patients involved in the project were participants of the Woodstock Festival Poland 2014. During three days of the festival, 615 young adults (18­35 years) were enrolled in the study. The study participants had to fill in a questionnaire that included questions about the frequency of fruit and vegetable consumption, portion sizes, and knowledge of the recommended daily intake. We also measured basic anthropometric parameters. Results: The data from our study showed that 97.6% of respondents consumed fewer than 4 servings of vegetables and fruit. In addition 39% of them ate 2 or fewer vegetable servings per day. We found that men ate significantly less fruit and fewer vegetables than women (p < 0.05). Additionally, almost 75% of men responded that proper consumption of fruit and vegetables should be at the level of 2­3 servings, and only 22.3% of them said that the proper level is 4­5 servings. Conclusions: We need to keep in mind that only clear information about vegetables and fruit, as well as highlighting the importance of the principles of nutrition, can give effective results in the future. It also appears that nutritional education should be conducted not only at the early stages of development but also in adolescents.

BACKGROUND: Nonalcoholic fatty liver disease (NAFLD) is a spectrum of liver conditions related to fat infiltration. The role of liver triacylglycerol accumulation in NAFLD is not fully understood. METHODS: Twenty-four patients, 12 in the first and 12 in the second stage of NAFLD, were prospectively enrolled in this study. Biochemical parameters and eicosanoids (HETE and HODE) were compared between the first and the second stage of hepatic steatosis and the effect of a 6-month dietary intervention on these parameters was evaluated. Eicosanoid profiles were extracted from 0.5 ml of plasma using solid-phase extraction RP-18 SPE columns. The HPLC separations were performed on a 1260 liquid chromatograph. RESULTS: Patients with stage I NAFLD had a significantly higher level of HDL cholesterol and a lower level of 5-HETE. Patients with grade II steatosis had higher concentrations of 9-HODE. Following the six-month dietary intervention, hepatic steatosis resolved completely in all patients. This resulted in a significant decrease in the concentrations of all eicosanoids (LX4, 16-HETE, 13-HODE, 9-HODE, 15-HETE, 12-HETE, 5-oxoETE, 5-HETE) and key biochemical parameters (BMI, insulin, HOMA-IR, liver enzymes). CONCLUSION: A significant reduction in the analyzed eicosanoids and a parallel reduction in fatty liver confirmed the usefulness of HETE and HODE in the assessment of NAFLD.

AIM: To investigate whether liver steatosis reduction due to a six-month dietary intervention results in significant changes in the concentrations of fatty acids. METHODS: A group of 35 Caucasian individuals diagnosed with different levels of steatosis were prospectively enrolled in the present study. Analysis of the fatty acid profiles was performed according to changes in liver steatosis (liver steatosis reduction by one or two degrees) after a six-month dietary intervention. The diet helped reduce body mass in obese and overweight patients, and stabilize both glycemia and dyslipidemia. Fatty acids were extracted according to the Folch method and analyzed by gas chromatography. RESULTS: This study showed significant changes in fatty acid profiles in patients who had reduced liver steatosis by one as well as two degrees. A reduction in liver steatosis by one degree caused a significant increase in the level of the n-3 family: eicosapentaenoic acid (P < 0.055), docosapentaenoic acid-C 22:5 (P < 0.05) and docosahexaenoic acid (P < 0.05). A reduction in liver steatosis by two degrees caused a significant decrease in serum palmitoleic acid-C 16:1 (P < 0.05). CONCLUSION: Liver steatosis reduction is associated with changes in fatty acid profiles, and these changes may reflect an alteration in fatty acid synthesis and metabolism. These findings may help better understand regression of nonalcoholic fatty liver disease.

INTRODUCTION: Caloric restriction is the only well-documented nutritional intervention prolonging the life of mammals. This method modifies the lipid levels in blood, controlling obesity and delaying the onset of many medical conditions associated with metabolic disorders. The aim of the study was to carry out a comparative analysis of lipid profile in patients on Mediterranean or CRON (Caloric Restriction with Optimal Nutrition) diets, before and after six weeks of dieting. MATERIAL AND METHODS: The following parameters were compared: total cholesterol, total lipids, triglycerides, LDL cholesterol, HDL cholesterol, and BMI. Additionally, we measured the levels of insulin, HOMA score, and anthropometric parameters. The comparative analysis demonstrated a statistically significant correlation between mild caloric restriction and blood lipid profile. Results from studies on patients who underwent six-week dietetic intervention indicated statistically significant changes in biochemical parameters due to caloric restrictions. Such changes were not found in subjects following the Mediterranean diet. The greatest decrease in the blood level of triglycerides was found in subjects on the CRON diet, LDL cholesterol, total cholesterol and total lipid levels also decreased. No significant changes if biochemical parameters were found in patients on the Mediterranean diet. CONCLUSIONS: A comparative analysis of all parameters demonstrated that the use of mild caloric restrictions with ensured supply of all necessary nutrients seems to be the most effective solution for reducing fatty tissue.
