Considering the differences in molecular structure and function, the effects of ß-1,3-glucans from Euglena gracilis and ß-1,3/1,6-glucans from Saccharomyces cerevisiae on immune and inflammatory activities in dogs were compared. Four diets were compared: control without ß-glucans (CON), 0.15 mg/kg BW/day of ß-1,3/1,6-glucans (Β-Y15), 0.15 mg/kg BW/day of ß-1,3-glucans (Β-S15), and 0.30 mg/kg BW/day of ß-1,3-glucans (Β-S30). Thirty-two healthy dogs (eight per diet) were organized in a block design. All animals were fed CON for a 42-day washout period and then sorted into one of four diets for 42 days. Blood and faeces were collected at the beginning and end of the food intake period and analysed for serum and faecal cytokines, ex vivo production of hydrogen peroxide (H2O2) and nitric oxide (NO), phagocytic activity of neutrophils and monocytes, C-reactive protein (CRP), ex vivo production of IgG, and faecal concentrations of IgA and calprotectin. Data were evaluated using analysis of covariance and compared using Tukey's test (P<0.05). Dogs fed Β-Y15 showed higher serum IL-2 than dogs fed Β-S30 (P<0.05). A higher phagocytic index of monocytes was observed in dogs fed the B-S15 diet than in those fed the other diets, and a higher neutrophil phagocytic index was observed for B-S15 and B-Y15 than in dogs fed the CON diet (P<0.05). Monocytes from dogs fed B-S15 and B-S30 produced more NO and less H2O2 than those from the CON and B-Y15 groups (P<0.05). Despite in the reference value, CRP levels were higher in dogs fed B-S15 and B-S30 diets (P<0.05). ß-1,3/1,6-glucan showed cell-mediated activation of the immune system, with increased serum IL-2 and neutrophil phagocytic index, whereas ß-1,3-glucan acted on the immune system by increasing the ex vivo production of NO by monocytes, neutrophil phagocytic index, and serum CRP. Calprotectin and CRP levels did not support inflammation or other health issues related to ß-glucan intake. In conclusion, both ß-glucan sources modulated some immune and inflammatory parameters in dogs, however, different pathways have been suggested for the recognition and action of these molecules, reinforcing the necessity for further mechanistic studies, especially for E. gracilis ß-1,3-glucan.

High-protein low-carbohydrate diets have been widely used for adult maintenance dogs, as well as in specific weight loss diets and maintenance programs. However, increasing dietary protein may increase undigested protein in the hindgut, modifying intestinal fermentation and fecal metabolite concentrations. The aim of this study was to evaluate the effects of protein source and concentration on apparent total tract digestibility (ATTD) of nutrients, metabolizable energy (ME), fecal and urinary characteristics, and fecal metabolites of dogs. Twelve healthy adult dogs were distributed into six treatments (n = 6 per diet) in a balanced incomplete Latin square design consisting of three periods of 30 days each. Six diets, varying in protein source [poultry byproduct meal (PBPM) and hydrolyzed chicken liver powder (HCLP)] and concentration [24, 32, and 40% crude protein (CP) on dry matter (DM) basis] were tested: PBPM24 (PBPM based diet with 24% CP); PBPM32 (PBPM with 32% CP); PBPM40 (PBPM with 40% CP); HCLP24 (HCLP based diet with 24% CP); HCLP32 (HCLP with 32% CP); HCLP40 (HCLP with 40% CP). The ATTD of CP was greater in dogs-fed HCLP and higher protein concentrations diets (P < 0.05). However, dogs-fed HCLP diets had lower ATTD of fat and carbohydrates, and ME (P < 0.05). Similarly, high-protein diets reduced the ATTD of DM, OM, fat, carbohydrates, and energy (P < 0.05). High-protein diets increased the daily fecal output and moisture (P = 0.004 and P < 0.05, respectively), as well as the fecal score (P < 0.0001), verified as soft, moist stools, but still within the ideal range. Fecal valerate concentration was greater in dogs-fed PBPM at 32% CP (P = 0.007). Fecal isobutyrate tended to increase in dogs-fed PBPM and high-protein diets (P < 0.10). Also, dogs-fed PBPM and high-protein diets had greater fecal concentrations of isovalerate, branched-chain fatty acids (BCFA), and ammonia (P < 0.05). Finally, the fecal lactate concentration increased in dogs-fed HCLP and high-protein diets (P < 0.05). The HCLP increased the ATTD of CP, being a highly digestible protein. Although the inclusion of HCLP slightly increased fecal score and moisture, it decreased the amount of fecal metabolites of protein fermentation ammonia and BCFA, both of which are associated with proteolytic fermentation in the colon.


Feeding companion animals with high-protein diets has been a demand of the market and pet owners. However, the protein quality and quantity consumed can interfere with the amount of undigested protein that reaches the hindgut and be fermented. Intestinal fermentation can be desired when well controlled. This study tested two protein sources (hydrolyzed chicken liver and poultry byproduct meal) combined at three dietary protein concentrations (24, 32, and 40% crude protein on dry matter basis) and their effects on the apparent total tract digestibility (ATTD), fecal and urinary characteristics, and fecal metabolites of healthy adult dogs. In summary, diets containing higher inclusion of hydrolyzed chicken liver had improved protein ATTD. However, the same diets impaired the ATTD of fat and carbohydrates and decreased metabolizable energy. High-protein diets retained more water in the feces and increased the fecal output. Fecal consistency was affected, scored as soft and moist stools, but remained within an acceptable score. Dogs-fed poultry byproduct meal diets had greater concentrations of fecal protein fermentation metabolites, such as ammonia and branched-chain fatty acids, possibly related to a greater amount of undigested protein that reached the hindgut and was fermented.

Biogenic amines are synthesized through the bacterial decarboxylation of amino acids, commonly found in high levels in animal by-product meals due to spoilage. Furthermore, biogenic amines and other metabolites can be produced by the fermentation of proteins in the hindgut according to the protein source and concentration of crude protein (CP) in the diet. Thus, this study aimed to evaluate two protein sources (poultry by-product meal (PBPM) and hydrolyzed chicken liver powder (HCLP)) and three CP concentrations (24, 32, and 40%) and their effects on the consumption and fecal excretion of biogenic amines, plasma monoamine oxidase (MAO) and diamine oxidase (DAO) activities, and total antioxidant capacity (TAC) of healthy adult dogs after 30 days of feeding the experimental diets. Twelve dogs were randomly distributed into six treatments (n = 6/treatment): PBPM24 (PBPM with 24% CP); PBPM32 (PBPM with 32% CP); PBPM40 (PBPM with 40% CP); HCLP24 (HCLP with 24% CP); HCLP32 (HCLP with 32% CP); HCLP40 (HCLP with 40% CP). The PBPM and PBPM-based diets had higher concentrations of putrescine, cadaverine, tyramine, histamine, agmatine, and total biogenic amines. In contrast, HCLP and HCLP-based diets contained higher concentrations of spermidine, phenylethylamine, and spermine. The PBPM and PBPM-diets had higher biogenic amine index (BAI) indicating lower quality due to the high content of putrescine, cadaverine and tyramine. Dogs fed diets with PBPM and higher protein concentrations consumed more putrescine, cadaverine, tyramine, agmatine, and total amines (p < 0.0001), while dogs fed with HCLP consumed more spermidine, phenylethylamine, and spermine (p < 0.0001). Fecal excretion of phenylethylamine was greater in dogs fed HCLP32 and HCLP40 diets (p = 0.045). Dogs fed with HCLP tended to excrete more spermidine and tryptamine via feces, while higher protein concentrations tended to increase fecal excretion of cadaverine (p < 0.10). Plasma MAO activity was higher in dogs fed HCLP24 and PBPM32 diets (p = 0.024). The plasma activities of DAO and TAC were not different between diets (p > 0.05). Although we did not evaluate the intestinal activities of MAO and DAO, our results suggest that healthy adult dogs have an efficient deamination process on the gut epithelium.

The objective was to evaluate through three experiments the effects of a fine cassava fibre (CA: 106 µm) on kibble characteristics, coefficients of total tract apparent digestibility (CTTAD) of macronutrients, diet palatability and faecal metabolites and microbiota of dogs. Dietary treatments consisted of a control diet (CO), without an added fibre source and with 4.3% total dietary fibre (TDF), and a diet with 9.6% CA (106 µm), with 8.4% TDF. Experiment I evaluated the physical characteristics of the kibbles. The palatability test was evaluated in experiment II, which compared the diets CO versus CA. In experiment III, 12 adult dogs were randomly assigned to one of the two dietary treatments for 15 days, totalling six replicates/treatment, to assess the CTTAD of macronutrients; faecal characteristics, faecal metabolites and microbiota. The expansion index, kibble size and friability of diets with CA were higher than the CO (p < 0.05). Additionally, the CA diet presented higher palatability than the CO (p < 0.05) but did not affect CTTAD except for those of fibre (p > 0.05). Moreover, a greater faecal concentration of acetate, butyrate and total short-chain fatty acids (SCFA) and a lower faecal concentration of phenol, indole and isobutyrate were observed in dogs fed the CA diet (p < 0.05). Dogs fed with the CA diet presented a greater bacterial diversity and richness and a greater abundance of genera considered to be beneficial for gut health, such as Blautia, Faecalibacterium and Fusobacterium when compared to the CO group (p < 0.05). The inclusion of 9.6% of a fine CA improves the expansion of kibbles and diet palatability without affecting most of the CTTAD of nutrients. Besides, it improves the production of some SCFA and modulates the faecal microbiota of dogs.

Dogs with food allergies and enteropathies may require hydrolyzed diets to prevent or reduce clinical signs, therefore the protein sources used in these diets must be previously characterized and evaluated in healthy dogs. This study aimed to investigate the effects of a hydrolyzed chicken liver powder-based diet (HCLP) versus a poultry by-product meal and bovine meat and bone meal-based diet (Control), on complete blood count (CBC), cytokine, immunoglobulins responses (assessed on days 0, 15, 30 and 45), and fecal microbiota (assessed on day 45) in healthy adult dogs. The CBC did not differ between diets (P>0.05), remaining within reference range. Total plasma IL-4 concentrations were decreased over time independent of the dietary treatment (P<0.001). Total plasma IgA decreased on day 30 compared to days 0 and 45 in dogs fed the control diet (P<0.001). Total plasma IgE concentrations were reduced on days 30 and 45 in dogs fed the control diet, and on days 15 vs 30 and 15 vs 45 in dogs fed HCLP diet (P = 0.001). The 16S rRNA gene sequencing showed similar species richness and abundances of phyla and genera between diets (P>0.05). ß-diversity principal coordinate analysis plots demonstrated that HCLP group had a higher similarity than control. Based on our results, healthy adult dogs fed a HCLP based diet maintained normal values for hematological and immunological characteristics, and fecal microbiota after 45 days of feeding.

A glicerina é um coproduto originado da produção de biodiesel, que apresenta possibilidade de uso como fonte energética na nutrição animal. Objetivou-se avaliar os coeficientes de digestibilidade aparente (CDA) e a energia metabolizável (EM) de dietas contendo glicerina, bem como a EM da glicerina em cães. Foram avaliadas quatro dietas contendo crescentes níveis de glicerina: 0%, 3%, 6% e 9%. As dietas foram fornecidas a oito cães adultos, os quais foram distribuídos em delineamento Quadrado Latino duplo (4x4). A adição de glicerina aumentou os CDA da matéria seca, energia bruta e a EM, sem afetar os CDA da proteína bruta e dos extrativos não nitrogenados da dieta. Entretanto, resultou em redução no CDA do extrato etéreo da dieta. Embora a adição de glicerina tenha reduzido o teor de matéria seca fecal, não houve efeito sobre o escore das fezes. Dentro dos níveis avaliados, a glicerina pode ser utilizada na dieta como fonte energética altamente disponível para cães.


Glycerin is a by-product originated from biodiesel production and it is an energetic ingredient of increasing possibility of use in diets of different species. This study aimed to evaluate the digestibility in dogs fed diets containing increasing levels of glycerol (0%, 3%, 6% and 9%). Diets were fed to eight adult dogs, which were distributed in Latin square design (4x4). The glycerol addition in the diet increased the digestibility of dry matter, gross energy and metabolizable energy, without affecting the digestibility of crude protein and nitrogen free extract. However, resulted in lower digestibility of ether extract. Although the addition of glycerin has reduced the amount of fecal dry matter, had no effect on fecal score. Within the levels evaluated, the glycerin can be used as dietary energy source highly available for dogs.

As características físicas da dieta, dentre elas a granulometria, exercem importante papel no aproveitamento da dieta pelos animais. Desse modo, o presente estudo teve como objetivo avaliar os efeitos da granulometria da dieta sobre a digestibilidade e a metabolizabilidade da energia de rações secas extrusadas para cães. Oito cães adultos da raça Beagle foram distribuídos em delineamento quadrado latino duplo (4 x 4), totalizando oito repetições no tempo. Os ingredientes foram moídos em peneiras de 0,8; 1,0; 1,2 e 1,5mm, resultando em rações com diâmetro geométrico médio (DGM) de 468, 476, 499 e 588µm, respectivamente. Cada período experimental foi composto por cinco dias para adaptação às dietas e cinco dias de coleta total de fezes. As análises de correlação e regressão linear demonstraram relação negativa entre o DGM e os coeficientes de digestibilidade aparente (CDA) da proteína bruta (y=94,82-42,05x), o extrato etéreo em hidrólise ácida (y=86,57-35,02x), os extrativos não nitrogenados (y=102,48-48,42x) e a energia metabolizável (y=3597,56-1697,00x). Não foi observado efeito do DGM sobre o consumo de matéria seca e o CDA da matéria seca. Houve piora na qualidade das fezes à medida que o DGM das dietas aumentou. Portanto, a menor granulometria da ração está relacionada ao melhor aproveitamento dos nutrientes e à energia da dieta e à produção de fezes de melhor qualidade pelos cães.


The physical characteristics of the diet, including particle size, exert important role in animals diet usage. So, the current study aimed to evaluate the effect of diet granulometry on nutrients digestibility and metabolizability of energy in dogs. Eight adult Beagle dogs were distributed in a double latin square design (4 x 4), totaling eight replicates in time. The ingredients of diets were ground in sieves of: 0.8, 1.0, 1.2 and 1.5mm, and the diets particle size were expressed as geometric mean diameter (GMD): 468, 476, 499 and 588µm, respectively. Each experimental period was composed by five days for diet adaptation and five days of total feces collection. The correlation and regression analyses showed a negative relationship between GMD and coefficient of apparent digestibility (CAD) of crude protein (y=94.82-42.05x), acid ether extract (y=86.57-35.02x), nitrogen free-extract (y=102.48-48.42x) and metabolizable energy (y=3597.56-1697.00x). It was not observed effect of GMD on dry matter intake and dry matter CAD. There was lower fecal score with increasing GMD of foods. Therefore, the lowest particle size of food resulted in better use of nutrients and diet energy and production of drier feces by dogs.
