During the 20th century, there has been an ongoing agricultural expansion and global warming, two of the main determinants influencing biodiversity changes in Argentina. The red hocicudo mouse (Oxymycterus rufus) inhabits subtropical grasslands and riparian habitats and has increased its abundance in recent years in central Argentina agroecosystems. This paper describes the long-term temporal changes in O. rufus abundance in Exaltación de la Cruz department, Buenos Aires province, Argentina, in relation to weather fluctuations and landscape features, as well as analyzes the spatio-temporal structure of captures of animals. We used generalized liner models, semivariograms, the Mantel test, and autocorrelation functions for the analysis of rodent data obtained from trappings conducted between 1984 and 2014. O. rufus showed an increase in abundance across the years of study, with its distribution depending on landscape features, such as habitat types and the distance to floodplains. Capture rates showed a spatio-temporal aggregation, suggesting expansion from previously occupied sites. O. rufus was more abundant at lower minimum temperatures in summer, higher precipitation in spring and summer, and lower precipitations in winter. Weather conditions affected O. rufus abundance, but there was local variation that differed from global patterns of climate change.

This study aimed to analyze the role of Mus musculus as a host of Leptospira spp., lymphocytic choriomeningitis virus (LCMV) and Toxoplasma gondii, in poultry farms of Buenos Aires province, Argentina, and to assess the potential risk of transmission to humans and domestic or breeding animals. Samplings were performed between 2009 and 2011 (S1) and during 2016 (S2). In S1, we studied the prevalence of infection for Leptospira spp. and LCMV, whereas, in S2, we studied the prevalence of infection for Leptospira spp. and T. gondii. In S1, we found an overall Leptospira spp. prevalence in M. musculus of 18% (14/79) and no positive serum samples for LCMV (0/166). In S2, we detected no positive individuals for Leptospira spp. (0/56) and an overall T. gondii seroprevalence of 3.6% (2/56). The probability of Leptospira spp. infection in M. musculus was higher in reproductively active individuals and in samplings subsequent to months with high accumulated precipitation. Our results suggest that, in the poultry farms studied, the presence of M. musculus may be a risk factor in the transmission of Leptospira spp. and T. gondii to humans and domestic animals. The management of farms should include biosecurity measures for farm workers and more effective rodent control.

Mus musculus and Rattus sp. are considered pest species because they reach high densities in urban areas, crop fields and food storage and productive systems such as breeding farms and orchards. Their control relies mainly on rodenticide application, but the effectiveness of this application is reduced due to behavioural responses and resistance. Novel methods are based on the use of chemical signals as odours that may be attractants, repellents or may reduce the reproductive success of pest species. The aim of this paper is to study the aversive effect of TMT, cat urine and cat body odour on predator-inexperienced Mus musculus and Rattus norvegicus under laboratory conditions. The experimental apparatus comprised three boxes connected by PVC pipes in a linear arrangement. In lateral boxes, odour sources or distilled water were introduced, while animals were placed in the central box at the beginning of the experiment. Rats showed freezing behaviour, reduced visits in the presence of TMT and cat fur. Mice reduced their visits with cat body and cat urine. This study provides evidence of the usefulness of using fear responses as a way to control rodent pests, which must be adapted to the environment and species to be applied.

Rodents are reservoirs of various types of hantavirus, some of which are agents of hantavirus pulmonary syndrome in humans. Each hantavirus is associated with a single rodent host species but successive spill-over events may eventually lead to host-switching and new species' becoming host of a given pathogen. This study aims to gain an understanding of the spatial ecology of two hantavirus-host species, Akodon azarae, and Oligoryzomys flavescens, by identifying factors modulating their home range sizes and stability, and by evaluating intra- and interspecific spatial aggregation for these species and a third one-Oxymycterus rufus-living in sympatry. For this, eleven capture-mark-recapture surveys were carried out, spanning 22 months. We found that A. azarae males have larger and more mobile home ranges than females, independently of the season. Consequently, males could likely have a more relevant role in the transmission of hantavirus because of their greater exposure both to a higher number of contacts between individuals and viral contamination of the environment. Contrasting, O. flavescens individuals showed negligible displacements of their home range through time, which could limit the range of hantavirus spread in host populations. Since O. flavescens is host to Lechiguanas hantavirus (pathogenic to humans) this result encompasses epidemiological relevance, for it may imply the existence of local foci of infection. Additionally, individuals of both species performed excursions outside their home ranges. These events could enable hantavirus spread over distances beyond the normal range of movements and lead to new hantavirus outbreaks in formerly non-infected rodent populations, favoring the persistence of the virus in nature.

Glutamate secretion at excitatory synapses is tightly regulated to allow for the precise tuning of synaptic strength. Vesicular Glutamate Transporters (VGLUT) accumulate glutamate into synaptic vesicles (SV) and thereby regulate quantal size. Further, the number of release sites and the release probability of SVs maybe regulated by the organization of active-zone proteins and SV clusters. In the present work, we uncover a mechanism mediating an increased SV clustering through the interaction of VGLUT1 second proline-rich domain, endophilinA1 and intersectin1. This strengthening of SV clusters results in a combined reduction of axonal SV super-pool size and miniature excitatory events frequency. Our findings support a model in which clustered vesicles are held together through multiple weak interactions between Src homology three and proline-rich domains of synaptic proteins. In mammals, VGLUT1 gained a proline-rich sequence that recruits endophilinA1 and turns the transporter into a regulator of SV organization and spontaneous release.

BACKGROUND: The house mouse (Mus musculus) is a cosmopolitan rodent that has become adapted to living in close association with humans and is considered a serious pest because it poses a risk to human health, and causes economic losses due to food and crop consumption and damage to buildings. Its control in livestock farms is achieved mainly through the application of anticoagulant rodenticides, but the effect of these compounds is limited due to the presence of resistant individuals and aversive behaviours. A potential alternative method is the use of chemical signals to reduce rodent reproductive success. In this study, we assessed the effects of odours from an unfamiliar male, 17ß-oestradiol, overcrowding, cat urine and 2,5-dihydro-2,4,5-trimethylthiazoline (TMT) on the reproductive success of laboratory Mus musculus females. RESULTS: According to the generalized linear mixed models, cat urine odour increased the proportion of abortions per female, unfamiliar male odour decreased the mean number of offspring born per female, and TMT had an overall negative effect on mean offspring production at birth and at weaning. The other odours had no significant effects on reproductive success. CONCLUSIONS: TMT seems to be the best candidate for population control because it caused a decrease in the mean number of offspring born and the mean number of live offspring at weaning. TMT also has the advantage of being available in commercial forms. To be useful for rodent management in field conditions, these results should be confirmed using wild house mice females. © 2019 Society of Chemical Industry.

To contribute to the knowledge of the role of small mammals in the transmission cycle of tegumentary leishmaniasis caused by Leishmania braziliensis, we studied the small mammal community and its temporal and spatial association with phlebotominae, as well as small mammal infection by Leishmania spp. by PCR-RFLP analyses in an endemic area of northeastern Argentina. Ten small mammal samplings were conducted (2007-2009, 7506 Sherman trap nights and 422 cage trap nights). In two of these samplings, 16 capture stations each one consisting of a CDC light trap to capture phlebotominae, two to four Sherman traps and two cage traps were placed. We found co-occurrence of phlebotominae and small mammal captures in four stations, which were all the stations with small mammal captures and yielded 97% (2295 specimens, including 21 gravid females) of the total phlebotominae captures, suggesting that small mammals may provide a potential source of blood for phlebotominae females. One Didelphis albiventris and two Rattus rattus were associated with high captures of Nyssomyia whitmani, vector of L. braziliensis in the study area. The PCR-RFLP analyses confirm the presence of L. braziliensis in two sigmodontine small mammals (Akodon sp. and Euryoryzomys russatus) for the first time in Argentina, to our knowledge.

BACKGROUND: In multiple sclerosis (MS), neurological disability results from incomplete remission of relapses and from relapse-independent progression. Intravenous high dose methylprednisolone (IVMP) is the established standard treatment to accelerate clinical relapse remission, although some patients do not respond. Most studies of relapse treatment have been performed when few patients received disease-modifying treatment and may no longer apply today. METHODS: We prospectively assessed, over one year, the course of patients who presented with a clinically isolated syndrome (CIS) or MS relapse, documenting demographic, clinical, treatment and outcome data. A standardized follow-up examination was performed 10-14 days after end of relapse treatment. RESULTS: We documented 119 relapses in 108 patients (31 CIS, 77 MS). 114 relapses were treated with IVMP resulting in full remission (29.2%), partial remission (38.7%), no change (18.2%) or worsening (4.4%). In 27 relapses (22.7%), escalating relapse treatment was indicated, and performed in 24, using double-dose IVMP (n = 18), plasmapheresis (n = 2) or immunoadsorption (n = 4). CONCLUSIONS: Standardised follow-up visits and outcome documentation in treated relapses led to escalating relapse treatment in every fifth relapse. We recommend incorporating scheduled follow-up visits into routine relapse management. Our data facilitate the design of prospective trials addressing methods and timelines of relapse treatment.

INTRODUCTION: In this study we evaluated freehand 3D ultrasound (3DUS) of the median nerve in comparison to 2D ultrasound (2DUS) and assessed the influence of tilting the transversal plane on cross-sectional area (CSA) measurement. METHODS: Two examiners investigated the median nerves of 22 healthy subjects over a distance of 20 cm using 3DUS. Image quality and CSA were assessed at random points within the virtual 3D volume and compared with 2DUS. RESULTS: Image quality within the virtual 3D volume was good/sufficient/poor in 53.0%/40.2%/6.8% (examiner 1) and 21.6%/69.6%/8.8% (examiner 2), respectively. CSA measurements with 3DUS were smaller than with 2DUS (-12% and -17%; Wilcoxon test, P < 0.001). Interrater agreement for 3DUS and intermethod agreement between 2DUS and 3DUS were moderate. Stepwise tilting of the transversal plane increased CSA significantly. CONCLUSION: Freehand 3DUS of the median nerve over 20 cm is feasible and may help overcome some of the limitations and pitfalls of 2DUS. Muscle Nerve 55: 206-212, 2017.

Introduction: The HTR2C gene is an important candidate in pharmacogenetic studies of antipsychotic-induced weight gain (AIWG). However, inconsistent results have been obtained. The present study investigated the association between -759C>T, functional polymorphism of the HTR2C receptor, and AIWG. Methods: A prospective cohort of 48 female inpatients with schizophrenia and related illness treated according to normal clinical practice with second generation antipsychotics (SGAs) risperidone, clozapine, quetiapine, and olanzapine were evaluated. Patients were weighted at admission and again at 6 weeks of hospitalization. Weight gain was defined as an increase≥7% of baseline weight. The association between polymorphisms HTR2C and weight gain was evaluated. Multiple logistic regression was run to determine potential confounders. Results: Patients with the T allele at position -759 (TT or CT) gained less weight as compared to patients who did not have the allele. This association was not affected by possible confounding factors such as age, baseline BMI, and prior psychopharmacological treatment. Discussion: The T allele at position -759 protects against AIWG in female patients with schizophrenia.

BACKGROUND: Mus musculus is a pest in urban and rural habitats where it consumes and contaminates food and may transmit diseases to human and domestic animals. Its control by anticoagulants is partially effective because of aversive behaviours and resistance. In this context, we wanted to assess the potential of the use of predator odours as repellents in experimental feeding trials using urine and faeces of domestic cats and faeces of geoffroyi cat, a wild small felid that is one of the main rodent predators in the study area. We also assessed the effect of previous experience and moonlight on foraging activity. RESULTS: We did not find an aversive response to cat odours in Mus musculus individuals. There was a trend to consume food in the same feeding stations over time, and the visit rate was lower in periods with high moonlight than in periods with low moonlight. CONCLUSIONS: Predator odours did not seem to be useful as rodent repellents, but maintaining illumination may lower rodent foraging activity. As rodents maintain their feeding sites over time, toxic baits may be more efficiently placed at sites previously known to be used by rodents.

The main goal of the paper was to determine the habitat distribution of the house mouse (Mus musculus) within a rural landscape of Buenos Aires province, Argentina. We also studied the seasonal variation in abundance and reproductive activity. The habitats studied were poultry farms, human houses in a small village, cropfields, pastures, cropfield and pasture edges, riparian habitats (streams), railway embankments and woodlots. We captured 817 M. musculus and 690 individuals of 5 native rodent species. M. musculus was captured in poultry farms, houses, riparian habitats, cropfield and borders, but it showed a significantly higher abundance in poultry farms compared to the other habitats. Its presence outside poultry farms was significantly related to the distance to streams and poultry farms. The mean trapping success index of M. musculus did not show significant variations between periods, but the proportion of active males was significantly higher in the spring-summer period than in the autumn-winter period. All captures of M. musculus in cropfields, borders and riparian habitats occurred in the spring-summer period. The capture of M. musculus in many types of habitats suggests that it can disperse outside poultry farms, and streams may be used as corridors.

BACKGROUND: Rodents are major pests in many agricultural systems, where they can cause significant economic losses and involve a sanitary risk. The application of anticoagulant rodenticides for rodent control has showed a decrease in effectiveness through time because of the development of resistant populations and the development of aversion behaviour. The goal of the present study was to test the susceptibility to bromadiolone and the existence of anticoagulant resistance in Mus musculus L. (house mouse) in Argentina. We conducted a feeding test with wild animals captured in poultry farms and a laboratory strain that were fed with bromadiolone bait. RESULTS: Three animals of the field experimental group survived the 21 days study period, while for laboratory animals mortality was 100%. Control field animals which were fed without anticoagulant showed 100% survival. CONCLUSION: We found evidence of the presence of anticoagulant resistant M. musculus L. in the study area. Feeding behaviour may have contributed to increasing the time of survival, and may be a mechanism that allows metabolic clearance of the bromadiolone. Under field conditions control with anticoagulants would be less effective because animals have alternative food.

Human neural progenitor cells (hNPCs) are a promising source to treat various neurodegenerative diseases. Potential applications are to use such cells for reprogramming to induce pluripotent stem cells or for secretion of proteins into the brain. These applications usually involve expression of heterologously expressed genes which is difficult to achieve in hNPCs. We tested several protocols for non-viral gene transfer and different promoters. Nucleofection and the cytomegalovirus enhancer/chicken beta-actin promoter allowed expression of foreign genes in hNPCs for up to 6 months. Treatment with the antibiotic G418 enabled us to select stably transfected cells which were subcloned and continued to express the NPC marker nestin. Differentiation of stably nucleofected hNPCs revealed that multipotency was maintained following long-term expansion of subcloned hNPCs. After differentiation for 3 weeks in vitro or in vivo following striatal transplantations transfected hNPCs expressed voltage-gated sodium channels suggesting the development of functional properties during neuronal maturation. In conclusion, stably nucleofected hNPCs can be isolated, subcloned, and expanded for up to 6 months without loss of their differentiation potential. These data provide a basis for future studies using hNPCs to investigate the neuronal differentiation in vivo after transplantation, the feasibility as a vector for gene (protein) therapy, and the induction of pluripotent stem cells.

A prevalência de Trichinella spiralis, Leptospira spp. e Salmonella spp. foi estudada em ratos e gambás que habitam granjas avícolas da região de Exaltación de la Cruz, Buenos Aires, Argentina, com o objetivo de determinar o potencial risco sanitário para pessoas que ficam em contato com esses animais. O estudo foi realizado entre a primavera de 1999 e o inverno de 2001 em 48 granjas avícolas. O estudo em gambás iniciou-se no inverno de 2000. Foram capturados 152 Rattus norvegicus, 3 Rattus rattus, 16 Didelphis albiventris e 1 Lutreolina crassicaudata. Registrou-se a presença de ratos e de gambás em 70 per center e 27 per center das granjas estudadas, respectivamente. A percentagem de granjas com ratos foi independente da presença ou da ausência de suínos. Não foi detectada a presença de Leptospira spp. e de Trichinella spiralis em nenhum animal avaliado. Foi detectada, porém, a presença de Salmonella Enteritidis em um Rattus norvegicus e em um Didelphis albiventris. Nas condições em que esse estudo foi realizado, os nossos resultados indicam que ratos e gambás que habitam granjas avícolas não constituem um fator de risco na transmissão de Trichinella e de Leptospira. A detecção de Salmonella Enteritidis tanto em ratos quanto em gambás, porém, sugere a necessidade de aplicar medidas profiláticas em granjas avícolas para maior controle desse agente.

We studied hantavirus seroprevalence and virus variability in rodent populations in Diego Gaynor, northwest of Buenos Aires province, Argentina. Rodent samplings were conducted in railroads and cropfield borders in March and July 1999, September and December 2000, and March 2001. Antibody detection was performed by an enzyme link immunosorbent assay (ELISA), using the recombinant nucleoprotein of Andes (AND) virus as antigen. Tissue samples were taken from positive antibody individuals in order to confirm the presence of hantavirus genomic material and to identify virus genotypes. Akodon azarae was the most abundant species, followed by Oligoryzomys flavescens, while Calomys laucha and C. musculinus were rarely caught. We found a rate of seroprevalence of 9.3% for a total sample of 291 A. azarae and 13.5% for 37 O. flavescens. After molecular analyses of hantavirus, we confirmed the presence of hantavirus genomic material in 16 individuals with ELISA (+) results and two individuals with ELISA (-). Four amplimers for each species were sequenced and compared to the corresponding sequences of representative hantaviruses. We identified the AND Cent Lec from three O. flavescens, and the Pergamino virus from four A. azarae and from one O. flavescens. A. azarae males had higher seroprevalence than females, and heavier individuals showed higher seroprevalence than lighter ones. We did not find seroprevalence differences according to sex in O. flavescens, although this result may have been produced by the low sample size. The lowest seroprevalence was found in a period of high rodent density, when juveniles prevailed in the population. We found higher seroprevalences than those detected in previous studies for other localities of central Argentina where cases of hantavirus pulmonary syndrome (HPS) have been reported. The presence of AND Cent Lec virus in rodent populations of the study area, which is responsible of HPS cases in central Argentina, suggests that human populations are at risk of HPS disease, although there were not reported cases of this disease until today.

We studied hantavirus seroprevalence and virus variability in rodent populations in Diego Gaynor, northwest of Buenos Aires province, Argentina. Rodent samplings were conducted in railroads and cropfield borders in March and July 1999, September and December 2000, and March 2001. Antibody detection was performed by an enzyme link immunosorbent assay (ELISA), using the recombinant nucleoprotein of Andes (AND) virus as antigen. Tissue samples were taken from positive antibody individuals in order to confirm the presence of hantavirus genomic material and to identify virus genotypes. Akodon azarae was the most abundant species, followed by Oligoryzomys flavescens, while Calomys laucha and C. musculinus were rarely caught. We found a rate of seroprevalence of 9.3 percent for a total sample of 291 A. azarae and 13.5 percent for 37 O. flavescens. After molecular analyses of hantavirus, we confirmed the presence of hantavirus genomic material in 16 individuals with ELISA (+) results and two individuals with ELISA (-).
