RESUMO

The only genus of the Francisellaceae family known to contain species pathogenic to mammals is Francisella, for which reported cases in the Southern Hemisphere have been limited to Australia. We describe severe necrotizing and inflammatory lesions and intralesional immunohistochemical identification of Francisella sp. lipopolysaccharide among aborted ovine fetuses in Uruguay.

Rotavirus A (RVA) is amongst the most widespread causes of neonatal calf diarrhea. Because subclinical infections are common, the diagnosis of RVA-induced diarrhea cannot rely solely on molecular viral detection. However, RT-qPCR allows for quantification of RVA shedding in feces, which can be correlated with clinical disease. Here, we determine an optimal cutoff of rotaviral load quantified by RT-qPCR to predict RVA causality in diarrheic neonate calves, using RVA antigen-capture ELISA as reference test. Feces from 328 diarrheic (n = 175) and non-diarrheic (n = 153), <30-day-old dairy calves that had been tested by ELISA and tested positive by RT-qPCR were included. Of 82/328 (25.0%) ELISA-positive calves, 53/175 (30.3%) were diarrheic, whereas 124/153 (81.0%) non-diarrheic calves tested negative by ELISA. The median log10 viral load was significantly higher in diarrheic vs. non-diarrheic and ELISA-positive vs. -negative calves, indicating a higher viral load in diarrheic and ELISA-positive calves. A receiver operating characteristic (ROC) analysis was conducted using the viral loads of the 175 diarrheic calves that had tested either positive (n = 53, cases) or negative (n = 122, controls) by ELISA. The optimal log10 viral load cutoff that predicted RVA causality in diarrheic calves was 9.171. A bootstrapping procedure was performed to assess the out-of-bag performance of this cutoff point, resulting in sensitivity = 0.812, specificity = 0.886, area under the curve = 0.922, and positive and negative diagnostic likelihood ratios of 11.184 and 0.142, respectively. The diagnostic accuracy of the cutoff was excellent to outstanding. This information will help in the interpretation of RVA RT-qPCR results in feces of diarrheic calves submitted for laboratory testing.

The aim of this work was to identify causes of abortion through laboratory investigations in sheep flocks in Uruguay. One hundred cases of abortion, comprising 58 fetuses, 36 fetuses with their placentas, and 6 placentas were investigated in 2015-2021. Cases were subjected to gross and microscopic pathologic examinations, and microbiological and serological testing for the identification of causes of abortion, including protozoal, bacterial, and viral pathogens. An etiologic diagnosis was determined in 46 (46%) cases, including 33 (33%) cases caused by infectious pathogens, as determined by the detection of a pathogen along with the identification of fetoplacental lesions attributable to the detected pathogen. Twenty-seven cases (27%) were caused by Toxoplasma gondii, 5 (5%) by Campylobacter fetus subspecies fetus, and 1 (1%) by an unidentified species of Campylobacter. Fourteen cases (14%) had inflammatory and/or necrotizing fetoplacental lesions compatible with an infectious etiology. Although the cause for these lesions was not clearly identified, T. gondii was detected in 4 of these cases, opportunistic bacteria (Bacillus licheniformis, Streptococcus sp.) were isolated in 2 cases, and bovine viral diarrhea virus 1 subtype i (BVDV-1i) was detected in another. Campylobacter jejuni was identified in 1 (1%) severely autolyzed, mummified fetus. BVDV-2b was identified incidentally in one fetus with an etiologic diagnosis of toxoplasmosis. Microscopic agglutination test revealed antibodies against ≥1 Leptospira serovars in 15/63 (23.8%) fetuses; however, Leptospira was not identified by a combination of qPCR, culture, fluorescent antibody testing nor immunohistochemistry. Neospora caninum, Chlamydia abortus, Chlamydia pecorum, Coxiella burnetii and border disease virus were not detected in any of the analyzed cases. Death was attributed to dystocia in 13 (13%) fetuses delivered by 8 sheep, mostly from one highly prolific flock. Congenital malformations including inferior prognathism, a focal hepatic cyst, and enterohepatic agenesis were identified in one fetus each, the latter being the only one considered incompatible with postnatal life. Toxoplasmosis, campylobacteriosis and dystocia were the main identified causes of fetal losses. Despite the relatively low overall success rate in establishing an etiologic diagnosis, a systematic laboratory workup in cases of abortion is of value to identify their causes and enables zoonotic pathogens surveillance.

Abstract Campylobacter fetus fetus (Cff) is a major infectious cause of abortion in sheep worldwide, and an opportunistic human pathogen. Information on Cff as an ovine abortifacient in South America is limited. We describe a case of abortion caused by a multidrug resistant strain of Cff in a sheep in Uruguay. In August 2017, 3/57 pregnant ewes (5.3%) aborted whithin one week. Histopathologic examination of the placenta of an aborted ewe revealed severe neutrophilic and fibrinonecrotizing placentitis with vasculitis and thrombosis of the chorionic arterioles. Cff was isolated on microaerobic culture in Skirrow agar, and further confirmed by 16S rDNA PCR amplification and sequencing, and endpoint and real time PCR assays. Antimicrobial sensitivity testing revealed resistance to tetracyclines, nalidixic acid, telithromycin and clindamycin. Other abortifacients were not detected. Further studies are necessary to determine the geographic distribution, ecology, epidemiology, economic impact, and antimicrobial resistance of Cff in sheep flocks in Uruguay.


Resumen Campylobacter fetus fetus (Cff) es una importante causa de abortos en ovinos y un patógeno oportunista en humanos. La información sobre Cff como abortifaciente en ovinos en Sudamérica es limitada. Describimos un caso de aborto causado por una cepa de Cff mul tirresistente a antibióticos en una oveja en Uruguay. En agosto de 2017, 3/57 ovejas prenadas (5,3%) abortaron en una semana. El examen histopatológico de la placenta de una de ellas reveló placentitis neutrofílica fibrinonecrosante severa, vasculitis y trombosis. Cff fue aislado en microaerobiosis en agar Skirrow, y confirmado mediante amplificación del ADNr 16S por PCR seguida de secuenciación, y por PCR punto final y qPCR. Las pruebas de sensibilidad antimicrobiana revelaron resistencia a tetraciclinas, ácido nalidíxico, telitromicina y clindamicina. No se detectaron otros abortifacientes. Son necesarios más estudios para determinar la distribución geográfica, ecología, epidemiología, el impacto económico y la resistencia antimicrobiana de Cff en majadas ovinas de Uruguay.

Escherichia coli ETEC, EPEC, NTEC and STEC/EHEC pathotypes are often isolated from bovine feces. The objective of this study was to detect 21 E. coli virulence genes in feces from 252 dairy calves in Uruguay (149 with neonatal diarrhea - NCD - and 103 asymptomatic). Genes iucD, f17A, afa8E, papC, clpG and f17G(II) were the most prevalent (81.3%; 48.4%; 37.3%; 35.7%; 34.1%; 31.3%, respectively). Genes eae, stx1and stx2 were poorly represented; 13/252 animals harbored one or a combination of these genes. The prevalence of the cnf gene was 4.4%, while that of cdt-IV and cdt-III genes was 24.2% and 12.7% respectively. This study reports updated data about the virulence profiles of E. coli in dairy calves in Uruguay. A large number of adhesins and toxin genes were detected. Our results demonstrate that E. coli from bovine feces has diarrheagenic and extraintestinal profiles although other NCD risks factors may contribute to the disease outcome.

Hypomagnesemia is a major cause of death in grazing beef cows in countries of the Southern Hemisphere such as Argentina, Australia and New Zealand. Here we review the literature on hypomagnesemia in beef cattle and describe an outbreak in Uruguay. The disease occurred in late autumn, affecting 6 to 11-year-old Aberdeen Angus and Hereford x Aberdeen Angus lactating multiparous cows in good body condition, grazing on natural grasslands that had been improved by fertilization and sowing of seeds of ryegrass and oat. Approximately 40 out of 225 cows were affected and 24 (10.7%) died. A presumptive diagnosis of hypomagnesemia was established based on sudden death, acute neuromuscular clinical signs, epidemiological data, and the response to Mg administration. The diagnosis was confirmed by detecting low Mg concentrations in serum (0.47-0.57mmol/L), vitreous humor (0.47-0.80mmol/L), aqueous humor (0.66mmol/L) and cerebrospinal fluid (0.59mmol/L). The largest component of the diet corresponding to fast-growing and exuberant forages of ryegrass and oat had high concentrations of K (3.48%), N (4.38%) and P (0.94%), suggesting secondary hypomagnesemia. In addition, the K/(Ca+Mg) ratio was 2.38 in forages of ryegrass and oat (reference value: 2.2), and 0.15 in the soil (reference value: 0.09), which represent risk factors for hypomagnesemia. In conclusion, hypomagnesemia is an important cause of mortality in beef cattle grazing improved natural grasslands in Uruguay and it can be easily prevented by correct seasonal Mg supplementation.(AU)

Aqui, revisamos a literatura sobre hipomagnesemia em bovinos de corte e descrevemos um surto no Uruguai. A doença ocorreu no final do outono, afetando vacas Aberdeen Angus e Aberdeen Angus x Hereford de 6-11 anos de idade em boas condições corporais, lactantes, multíparas e pastando em campos naturais que foram melhorados com fertilização e plantio de azevém e aveia. Aproximadamente 40 de 225 vacas foram afetadas e 24 (10,7%) morreram. O diagnóstico presuntivo de hipomagnesemia foi estabelecido com base na morte súbita, sinais clínicos neuromusculares agudos, dados epidemiológicos e a resposta à administração de Mg. O diagnóstico foi confirmado pela detecção de baixos níveis de Mg no soro (0,47-0,57mmol/L), humor vítreo (0,47-0,80mmol/L), humor aquoso (0,66mmol/L) e líquido cefalorraquidiano (0,59mmol/L). O componente mais importante da dieta correspondia a pastagens de azevém e aveia de rápido crescimento, as quais apresentaram altas concentrações de K (3,48%), N (4,38%) e P (0,94%), que sugerem hipomagnesemia secundária. Além disso, a relação K/(Ca+Mg) foi de 2,38 em forragens de azevém e aveia (valor de referência: 2,2) e 0,15 no solo (valor de referência: 0,09), que representam fatores de risco para hipomagnesemia. Despite the importance of dairy farming in Uruguay, little information on dairy systems in this country is available in the scientific literature, and management practices that influence calf welfare at the herd level have not been explored. The aims of this study were to (1) describe the prepartum and calf-rearing systems, as well as the management practices that may influence calf welfare in pastured dairy herds in Uruguay, (2) estimate the annual calf mortality risk from birth to weaning, and (3) identify the primary clinical disease syndromes shown by the calves before death. A survey comprising a farm visit and a questionnaire was conducted on 225 randomly selected dairies with >30 milking cows, in 3 strata (31-99, 100-299, and ≥300 milking cows) of 6 Uruguayan departments where dairies are concentrated. Retrospective information from July 2013 to June 2014 was collected. A descriptive analysis was performed and results were inferred into the national dairy cattle population. Several management practices that could contribute to poor calf welfare were identified in a large proportion of farms. The annual calf mortality risk (calves that died between birth and weaning/calves born death or alive × 100, n = 149 farms) was 15.2%. Age at weaning averaged 75 d. Farmers reported that the most common clinical syndromes were diarrhea and respiratory disease in 85.2% and 47.5% of the farms, respectively. There was no continuous veterinary advice in 61.3% of the farms, 20.0% lacked data records, 38.5% had poor drainage in the prepartum area with waterlogging after rainfall, 52.1% monitored the prepartum area ≤2 times per day during the calving season, 65.1% did not perform navel antisepsis on newborns, 62.3% separated the calves from their dams at >24 h postpartum, 95.2% did not have a colostrum management program, 72.4% did not rotate the calf-rearing areas, 59.0% did not disinfect the calf feeders, 85.7% did not have staff dedicated exclusively to calf rearing, and 39.8% did not separate sick from healthy calves. The average volume of milk or milk replacer offered per calf was 4.5 L/day. Several of the identified management practices that affect calf welfare in the prepartum and calf-rearing periods could explain the high mortality risk. An effort should be made to conduct extension work focusing on the dissemination of good management practices to improve calf welfare and reduce calf mortality in Uruguayan dairy farms.

We describe and illustrate lesions in an outbreak of lead arsenate poisoning in beef cattle that ingested pesticide residues stored in an abandoned building of a former orange orchard. Of 70 exposed cattle, 14 had diarrhea, paresis, ataxia, recumbency, and/or seizures. Ten of the affected animals died after a clinical course of 12-18 h. Pathologic findings in 3 steers included extensive necrohemorrhagic, ulcerative rumenitis, omasitis, and abomasitis; lymphocytolysis in lymphoid organs; and nephrosis. Hepatic arsenic and lead levels in cases 1-3 were 20, 24, and 31 ppm, and 8.3, 25, and 9.4 ppm, respectively. Lesions in the forestomachs and lymphoid tissues have been rarely reported in cases of lead arsenate poisoning. In southern South America, these lesions are indistinguishable from those produced by Baccharis coridifolia, a toxic plant that contains macrocyclic trichothecenes, thus these conditions should be considered in the differential diagnosis of necrotizing lesions in alimentary and lymphoid organs.

Bovine actinobacillosis is typically characterized by pyogranulomatous glossitis (wooden tongue). The involvement of other tissues, generally the skin or lymph nodes, has been regarded as atypical or cutaneous. We describe herein 2 outbreaks of actinobacillosis affecting primarily the lymph nodes of the head and neck. The disease affected 40 of 540 lactating cows in a dairy herd, and 5 of 335 two-y-old steers in a beef herd. Multiple or single, occasionally ulcerated nodules were observed in the region of the mandible, neck, and shoulder, including the parotid, submandibular, retropharyngeal, and prescapular lymph nodes. The histologic lesions were multifocal pyogranulomatous lymphadenitis, dermatitis, and cellulitis with Splendore-Hoeppli material. One steer had an exophytic pyogranuloma in the gingiva and another died because of ruminal tympany secondary to oropharyngeal and esophageal obstruction by a pyogranulomatous mass. Actinobacillus lignieresii was isolated from the lesions and identified by amplification, sequencing, and analysis of the 16S ribosomal (r)DNA gene. Seven of 8 cows recovered after treatment with sodium iodide. Lymphatic actinobacillosis is a frequent disease in Uruguay, southern Brazil, and Argentina. Morbidity is 1-50%; mortality is <1%. A. lignieresii apparently penetrates the intact oral and pharyngeal mucosa, infecting primarily the regional lymph nodes. Later, lesions may extend to the subcutaneous tissue and the skin, causing ulceration. Affected cattle with draining pyogranulomas contaminate the environment, favoring disease transmission, and should be treated with sodium iodide or antibiotics and isolated from the herd in order to control the disease.

Solanum glaucophyllum, a toxic plant known for its calcinogenic effects, causes enzootic calcinosis in ruminant and monogastric animals. We describe an outbreak of enzootic calcinosis that occurred in a herd of 110 horses grazing pastureland heavily contaminated with S. glaucophyllum in Buenos Aires province, Argentina. Ten horses developed clinical signs, and 6 horses died. Clinical signs included abnormal gait (stiff-legged action, short strides), stiffness, thoracolumbar kyphosis, reluctance to move, wide stance, chronic weight loss, weakness, recumbency, and difficulty standing. Autopsy of 2 horses revealed severe mineralization of the aorta, pulmonary arteries, heart, and lungs, consistent with enzootic calcinosis. Although horses usually have very selective grazing behavior, under food restriction conditions, they can ingest the toxic plants and can develop the disease. Enzootic calcinosis should be considered as a differential diagnosis in horses grazing S. glaucophyllum-invaded pasturelands with compatible clinical signs and lesions.
