RESUMO

Diabetic wounds are characterized by chronic trauma, with long-term non-healing attributed to persistent inflammation and recurrent bacterial infections. Exacerbation of the inflammatory response is largely due to increased levels of reactive oxygen species (ROS). In this study, catalase (CAT) was used as a biological template to synthesize nanozyme-supported natural enzymes (CAT-Mn(SH)x) using a biomimetic mineralization method. Subsequently, polymyxin B (CAT-Mn(SH)x@PMB) was immobilized on its surface through electrostatic assembly. CAT-Mn(SH)x@PMB demonstrates the ability for slow and sustained release of hydrogen sulfide (H2S). Finally, CAT-Mn(SH)x@PMB loaded microneedles (MNs) substrate were synthesized using polyvinyl alcohol (PVA) and hydroxyethyl methacrylate (HEMA), and named CAT-(MnSH)x@PMB-MNs. It exhibited enhanced enzyme and antioxidant activities, along with effective antibacterial properties. Validation findings indicate that it can up-regulate the level of M2 macrophages and reduce the level of pro-inflammatory cytokine tumor necrosis factor-α (TNF-α). Additionally, it promotes angiogenesis and rapid nerve regeneration, thereby facilitating wound healing through its dual anti-inflammatory and antibacterial effects. Hence,this study introduces a time-space tissue-penetrating and soluble microneedle patch with dual anti-inflammatory and antibacterial effects for the treatment of diabetic wounds.

Objective: The objective is to evaluate the clinical efficacy of cross electro-nape-acupuncture (CENA) in the treatment of pseudobulbar palsy in patients with tracheotomy intubation for severe cerebral haemorrhage and to provide an innovative acupuncture method for the treatment of such patients. Methods: A total of 126 patients from six trial centres who met the inclusion criteria were randomly divided into three groups according to the random number table method in the ratio of 1 : 1 : 1, with 42 patients in each group, and the three groups were divided into CENA group, electro-acupuncture group, and acupuncture group. Each group's acupuncture treatment lasted for 30 minutes, and the needles were removed at the end of the treatment. Acupuncture was performed once a week on Sunday only and twice a day from Monday to Saturday, a total of 4 weeks of treatment. The SWT, FDA, ChSWAL-QOL, and TCRGS scores of the three groups of patients before and after treatment were compared to evaluate the effect of CENA on remodelling the function of swallowing reflex and cough reflex and promoting the recovery of dysarthria and swallowing quality of life in pseudobulbar palsy in patients with tracheotomy intubation for severe cerebral haemorrhage. Results: After treatment, the WST and TCRGS grade scores decreased and the FDA and ChSWAL-QOL scores increased significantly in all three groups compared with the pretreatment scores and were statistically significant. There was a significant difference between the three groups for these four indicators after treatment; the comparison between groups showed significant differences in the CENA group compared to the electro-acupuncture and acupuncture groups. The efficiency of the CENA group was significantly better than that of the electro-acupuncture and acupuncture groups. Conclusion: Compared with the acupuncture and electro-acupuncture groups, the CENA could better promote the remodelling of swallowing function and cough reflex function, promote the recovery of dysarthria, and better improve the quality of life of patients with pseudobulbar palsy from tracheotomy intubation in severe cerebral haemorrhage.

The aim of this study was to develop and validate a prediction model to evaluate the risk of poor sleep quality. We performed a cross-sectional study and enrolled 1,928 college students from five universities between September and November 2021. The quality of sleep was evaluated using the Chinese version of the Pittsburgh Sleep Quality Index (PSQI). Participants were divided into a training (n = 1,555) group and a validation (n = 373) group. The training group was used to establish the model, and the validation group was used to validate the predictive effectiveness of the model. The risk classification of all participants was performed based on the optimal threshold of the model. Of all enrolled participants, 45.07% (869/1,928) had poor sleep quality (PSQI score ≧ 6 points). Multivariate analysis showed that factors such as older age, a higher grade, previous smoking, drinking, midday rest, chronic disease, anxiety, and stress were significantly associated with a higher rate of poor sleep quality, while preference for vegetables was significantly associated with better sleep quality, and all these variables were included to develop the prediction model. The area under the curve (AUC) was 0.765 [95% confidence interval (CI): 0.742-0.789] in the training group and 0.715 (95% CI: 0.664-0.766) in the validation group. Corresponding discrimination slopes were 0.207 and 0.167, respectively, and Brier scores were 0.195 and 0.221, respectively. Calibration curves showed favorable matched consistency between the predicted and actual probability of poor sleep quality in both groups. Based on the optimal threshold, the actual probability of poor sleep quality was 29.03% (317/1,092) in the low-risk group and 66.03% (552/836) in the high-risk group (P < 0.001). A nomogram was presented to calculate the probability of poor sleep quality to promote the applicationof the model. The prediction model can be a helpful tool to stratify sleep quality, especially among university students. Some intervention measures or preventive strategies to quit smoking and drinking, eat more vegetables, avoid midday rest, treat chronic disease, and alleviate anxiety and stress may be considerably beneficial in improving sleep quality.

Camera calibration is the most important aspect of computer vision research. To address the issue of insufficient precision, therefore, a high precision calibration algorithm for binocular stereo vision camera using deep reinforcement learning is proposed. Firstly, a binocular stereo camera model is established. Camera calibration is mainly divided into internal and external parameter calibration. Secondly, the internal parameter calibration is completed by solving the antihidden point of the camera light center and the camera distortion value of the camera plane. The deep learning fitting value function is used based on the internal parameters. The target network is established to adjust the parameters of the value function, and the convergence of the value function is calculated to optimize reinforcement learning. The deep reinforcement learning fitting structure is built, the camera data is entered, and the external parameter calibration is finished by continuous updating and convergence. Finally, the high precision calibration of the binocular stereo vision camera is completed. The results show that the calibration error of the proposed algorithm under different sizes of checkerboard calibration board test is only 0.36% and 0.35%, respectively, the calibration accuracy is high, the value function converges quickly, and the parameter calculation accuracy is high, the overall time consumption of the proposed algorithm is short, and the calibration results have strong stability.

BACKGROUND: Interleukin-1ß (IL-1)-treated mesenchymal stem cells (MSCs) and IL-1-MSCs-conditioned medium (CM) exert anti-inflammatory roles. Astrocytes are essential for the modulation of synaptic activity and neuronal homeostasis in the brain. Exosomes are the critical mediators in intercellular communication. However, the mechanism underlying the anti-inflammatory effect of IL-1-treated MSCs remains unknown. METHODS: In this study, exosomes (IL-1-Exo) were isolated from IL-1-treated MSCs. In addition, lipopolysaccharide (LPS)-treated hippocampal astrocytes and status epilepticus (SE) mice were treated with IL-1-Exo. Inflammatory activity, astrogliosis, and cognitive performance were measured to determine the effect of IL-1-Exo on inflammation. RESULTS: The results revealed that IL-1-Exo significantly inhibited LPS-induced astrogliosis and inflammatory responses of astrocytes. Also, IL-1-Exo reversed the LPS-induced effect on calcium signaling. The Nrf2 signaling pathway was associated with the effect of IL-1-Exo in LPS-treated astrocytes. Furthermore, IL-1-Exo reduced the inflammatory response and improved the cognitive performance of SE mice. CONCLUSION: The results suggest that IL-1-Exo inhibited LPS-induced inflammatory responses in astrocytes and SE mice and that the effect of IL-1-Exo was primarily mediated through the Nrf-2 signaling pathway. This study provides a new understanding of the molecular mechanism of inflammation-associated brain diseases and an avenue to develop nanotherapeutic agents for the treatment of inflammatory conditions in the brain.

Microglia are the resident immune cells in the central nervous system and play an essential role in brain homeostasis and neuroprotection in brain diseases. Exosomes are crucial in intercellular communication by transporting bioactive miRNAs. Thus, this study aimed to investigate the function of microglial exosome in the presence of ischemic injury and related mechanism. Oxygen-glucose deprivation (OGD)-treated neurons and transient middle cerebral artery occlusion (TMCAO)-treated mice were applied in this study. Western blotting, RT-PCR, RNA-seq, luciferase reporter assay, transmission electron microscope, nanoparticle tracking analysis, immunohistochemistry, TUNEL and LDH assays, and behavioral assay were applied in mechanistic and functional studies. The results demonstrated that exosomes derived from microglia in M2 phenotype (BV2-Exo) were internalized by neurons and attenuated neuronal apoptosis in response to ischemic injury in vitro and in vivo. BV2-Exo also decreased infarct volume and behavioral deficits in ischemic mice. Exosomal miRNA-137 was upregulated in BV2-Exo and participated in the partial neuroprotective effect of BV2-Exo. Furthermore, Notch1 was a directly targeting gene of exosomal miRNA-137. In conclusion, these results suggest that BV2-Exo alleviates ischemia-reperfusion brain injury through transporting exosomal miRNA-137. This study provides novel insight into microglial exosomes-based therapies for the treatment of ischemic brain injury.

BACKGROUND: Cerebral infarction ranks as the second leading cause of disability and death globally, and inflammatory response of glial cells is the main cause of brain damage during cerebral infarction. METHODS: Studies have shown that mesenchymal stem cells (MSCs) can secrete exosomes and contribute to cerebral disease. Here, we would explore the function of MSC-derived exosome in cerebral infarction. RESULTS: Microarray indicated a decrease of miR-542-3p and an increase of Toll-Like Receptor 4 (TLR4) in middle cerebral artery occlusion (MCAO) mice comparing with sham mice. And luciferase and RIP analysis indicated a binding of miR-542-3p and TLR4. Then, we injected AAV9-miR-542-3p into paracele of sham or MCAO mice. Functional analysis showed that AAV9-miR-542-3p inhibited infarction area and the number of degenerating neurons and suppressed inflammatory factors' expression and inflammatory cell infiltration. As well, transfection of miR-542-3p mimics into HA1800 cells underwent oxygen and glucose deprivation (OGD). Similarly, overexpression of miR-542-3p alleviated OGD induced cell apoptosis, ROS, and activation of inflammation response. Moreover, miR-542-3p could be packaged into MSCs and secreted into HA1800 cells. The extractive exosome-miR-21-3p treatment relieved MCAO- or OGD-induced cerebral injury and inflammation through targeting TLR4. CONCLUSION: These results confirmed that MSC-derived exosome miR-542-3p prevented ischemia-induced glial cell inflammatory response via inhibiting TLR4. These results suggest possible therapeutic strategies for using exosome delivery of miR-542-3p to cure cerebral ischemic injury.

BACKGROUND: Receptor interacting protein kinase 1 (RIPK1)-mediated cell death, including apoptosis and necroptosis, belongs to programmed cell death. It has been reported that RIPK1-mediated necroptosis exists in lesions of cerebral hemorrhage (CH). Electroacupuncture, a treatment derived from traditional Chinese medicine, could improve neurological impairment in patients with brain injury. AIM: To investigate the protective role of cross electro-nape acupuncture (CENA) in CH, and clarify the potential mechanism. METHODS: CH rat models were established, and CENA was applied to the experimental rats. Neurological functions and encephaledema were then measured. Necrotic cells in the brain of rats with CH were evaluated by propidium iodide staining. Necroptosis was assessed by immunofluorescence. Activation of the necroptosis-related pathway was detected by western blot. Extraction of brain tissue, cerebrospinal fluid and serum samples was conducted to measure the expression and secretion of inflammatory cytokines by quantitative real-time polymerase chain reaction and enzyme-linked immunosorbent assay. RESULTS: The necroptotic marker p-MLKL was detectable in the brains of rats with CH. Next, we found that CENA could ameliorate neurological functions in rat models of CH. Moreover, the upregulation of RIPK1-mediated necroptosis-related molecules in the brains of rats with CH were inhibited by CENA. Further investigation revealed that CENA partially blocked the interaction between RIPK1 and RIPK3. Finally, in vivo assays showed that CENA decreased the expression of the inflammatory cytokines tumor necrosis factor-α, interleukin-6 and interleukin-8 in CH rat models. CONCLUSION: These findings revealed that CENA exerts a protective role in CH models by inhibiting RIPK1-mediated necroptosis.

To attempt to understand certain mechanisms causing the variations between rice cultivars with regard to Cd uptake and accumulation, pot soil experiments were conducted with two rice cultivars at different levels of Cd, i.e., 0 (the control), 10, 50 mg Cd kg(-1 )soil. The two rice cultivars differ significantly with regard to Cd uptake and accumulation. Root secretions of low-molecular-weight organic acids (LMWOA) for each treatment were measured with ion chromatography. The results showed that LMWOA concentrations in the soil planted with Shan you 63 (a high soil Cd accumulator) were all higher than those in the soil planted with Wu yun jing 7 (low soil Cd accumulator) at different soil Cd levels, although the magnitudes of the differences varied for individual LMWOA and depend on soil Cd concentrations. For all six LMWOA, there were significant differences at P < 0.05 or < 0.01 levels for soils treated with 10 and 50 mg kg(-1) Cd. The magnitude of the differences was greater under soil Cd treatments, especially at relatively low levels (for example, 10 mg Cd kg(-1) soil), than in the control. Acetic acid and formic acid constituted more than 96% of the total concentration of the six LMWOA, while citric acid constituted only about 0.1%. The rice cultivar with higher concentrations of LMWOA in soil accumulated more Cd in the plants. The results indicate that LMWOA secretion by rice root, especially in Cd-contaminated soils, is likely to be one of the mechanisms determining the plant Cd uptake properties of rice cultivars.

The variations among six rice cultivars in cadmium (Cd) uptake and translocation were investigated with pot soil experiments. The results showed that only a very small portion (0.73%) of Cd absorbed by rice plant was transferred into grain. With regard to plant total Cd uptake, Cd concentrations and quantity accumulations in roots, stems and leaves, the differences among the cultivars (between the largest one and the smallest one) were less than one time. But for Cd concentrations and Cd quantity accumulations in the grains, the differences were more than five and eight times, respectively. With respect to Cd distribution portions in plant organs, the diversities among the cultivars were also small in roots, stems and leaves, but much larger in grains. Grain Cd concentrations correlated positively and significantly (P<0.01) with Cd quantity accumulations in plant, Cd distribution ratios to aboveground parts, and especially with Cd distribution ratios from aboveground parts to the grain. The results indicated that Cd concentration in rice grain was governed somewhat by plant Cd uptake and the transport of Cd from root to shoot, and in a greater extent, by the transport of Cd from shoot to grain. Cd was not distributed evenly in different products after rice grain processing. The average Cd concentration in cortex (embryo) was five times more than that in chaff and polished rice. With regard to Cd quantity accumulation in the products, near 40% in cortex (embryo), 45% in polished rice and 15% in chaff averagely.
