RESUMO

To ensure food safety and environmental protection, it is crucial to rapidly identify and remove bisphenol A (BPA), a plasticizer commonly used in the inner lining of food containers and beverage packaging. Here, a photocatalytic fuel cell (PFC)-integrated self-powered photoelectrochemical (PEC) sensor is constructed. Unlike conventional single PEC or PFC sensors, this PFC-integrated PEC sensor relies on not only the difference in Fermi energy levels between photoanode and photocathode but also charge accumulation resulted from the oxidation of BPA by photogenerated holes. Consequently, this sensor achieved a remarkable maximum output power (Pmax) of 8.58 µW cm-2, as well as a high sensitivity, wide linear detection range (0.1-200 µM), low detection limit (0.05 µM), great stability, reproducibility, and real sample detection capability. This work integrates PFC and PEC technologies successfully for the rapid identification and efficient removal of BPA.

The treatment of Parkinson's disease is a global medical challenge. α-Synuclein (α-Syn) is the causative protein in Parkinson's disease and is closely linked to its progression. Therefore, inhibiting the pathological aggregation of α-Syn and its neurotoxicity is essential for the treatment of Parkinson's disease. In this study, α-Syn and recombinant human HspB5-ACD structural domain protein (AHspB5) were produced using the BL21(DE3) E. coli prokaryotic expression system, and then the role and mechanism of AHspB5 in inhibiting the pathological aggregation of α-Syn and its neurotoxicity were investigated. As a result, we expressed α-Syn and AHspB5 proteins and characterised the proteins. In vitro experiments showed that AHspB5 could inhibit the formation of α-Syn oligomers and fibrils; in cellular experiments, AHspB5 could prevent α-Syn-induced neuronal cell dysfunction, oxidative stress damage and apoptosis, and its mechanism of action was related to the TH-DA pathway and mitochondria-dependent apoptotic pathway; in animal experiments, AHspB5 could inhibit behavioural abnormalities, oxidative stress damage and loss of dopaminergic neurons. In conclusion, this work is expected to elucidate the mechanism and biological effects of AHspB5 on the pathological aggregation of α-Syn, providing a new pathway for the treatment of Parkinson's disease and laying the foundation for recombinant AHspB5.

Florfenicol (FF) raises significant human health and environmental concerns due to its toxicity to the hematology system and the potential spread of antibiotic-resistant genes. Here, a highly sensitive molecularly imprinted photoelectrochemical (PEC) sensor, featuring an In2O3/Bi2S3 S-scheme heterojunction, is proposed to detect FF without an external voltage supply. Compared with conventional II-type heterojunctions, S-scheme heterojunctions efficiently promote carrier separation and enhance the redox capability of the photocatalytic system. This allows more dissolved O2 and H2O molecules to participate in the redox reaction, resulting in an amplified and stabilized photocurrent response. The electron transfer in the S-scheme heterojunction is confirmed via electron spin resonance (ESR). With the molecular imprinting technique, this PEC platform exhibits exceptional selectivity, wide linear range (1.0 × 10-4-1.0 × 104 ng mL-1), low detection limit (6.4 × 10-5 ng mL-1), and applicability in real milk and chicken samples. This work not only showcases a PEC platform for accurately and portably detecting drugs but also proposes a viable approach for designing S-scheme heterojunctions in sensing analysis.

Conventional photoelectrochemical (PEC) biosensors suffer from the difficulty of visualizing rapid detection and limited accuracy due to a single-signal output. Here, we develop a PEC, electrochromic (EC), and spectral (ST) triple-mode platform for the sensitive visualized detection of nonylphenol (NP). First, the reasonably stepped Fermi energy level arrangement between the defective TiO2 anode and MoO3 cathode enables a remarkable photocurrent response (Mode 1). Then, MoO3 itself is a widely used EC candidate, which can react with free Li-ions to form a LixMoO3 intermediate, and its color will change from white to blue accordingly (Mode 2). More importantly, MoO3 is also a Li-ion host and the potential of LixMoO3 depends on the inserted Li-ion quantity deduced by spectral analysis on residual Li-ions in the electrolyte (Mode 3). The EC signal endows fast visual detection, and triple-mode cross-validation improves reliability and accuracy. As a result, this PEC-EC-ST triple-mode molecularly imprinted sensor has a wide linear range (1-5000 µg L-1), a low detection limit (0.18 µg L-1), selectivity, stability, reproducibility, and actual sample detection capability. This innovative multimode platform not only improves detection reliability but also broadens applications of electrochromic/energy storage materials in biosensors.

Aim: The purpose of this paper is to investigate the relationship between serum pentosidine levels and cardiac function and vascular disease in diabetic patients, and to provide a new reference indicator for the early detection of diabetic cardiovascular complications. Materials and Methods: This was a cross-sectional study. One hundred and twenty-two patients with type 2 diabetes were grouped by LVEF quartiles to compare the differences between their clinical data and serum pentosidine levels. Also, the correlation between serum pentosidine and clinical indicators was assessed. The effect of serum pentosidine on cardiac function and vascular stiffness was analyzed by multiple stepwise regression. Results: Serum pentosidine levels were higher in patients with LVEF ≤57%. Serum pentosidine levels were positively correlated with waist-to-hip ratio, hemoglobin, AIP, baPWV, LVESD, and ARD, and negatively correlated with LVEF. Low serum pentosidine was associated with increased LVESD; high pentosidine was significantly associated with increased ARD, high AIP and high baPWV. Conclusion: The results suggest that serum pentosidine, a member of AGEs, may reflect cardiac remodeling and dysfunction as well as atherosclerosis.

For large ground-based telescopes, static and dynamic disturbances would greatly degrade the optical performance. This is especially true for wide field survey telescopes with prime focus optics. The estimation of disturbance effects on large telescopes is becoming increasingly important during the design phase. Therefore, a wide field survey telescope with 2.5 m aperture and 3.5 deg field of view is studied in this research. This telescope is under construction now, and its first light is expected at the beginning of 2023. The estimation method for the optical performance under static and dynamic disturbances in the temporal domain and the active compensation method to improve the optical alignment, are investigated, which is a supplement for the simulation in the frequency domain. First, based on the mechanical model, the optical misalignment is established, where the deviation of the primary mirror is obtained from the length gauges and the deviation of the corrector is computed using the fitting method. Second, a method for compensating the static and dynamic disturbances is proposed, improving the optical performance. This method uses the disturbed primary mirror as the reference, and the corrector is actively controlled to align with it. Finally, a series of experimental tests and numerical simulations is conducted. The results show that the mechanical modeling error is within 10% and the maximum optical misalignment is reduced from 12''/0.27 to 0.2''/0.006mm for static disturbance and from 1.3''/0.03 to 0.4''/0.01mm for dynamic disturbance. Through active compensation, the telescope optical property is greatly improved. The modeling method and the simulation process mentioned in this research can also be used in the other relevant fields.

Liver cirrhosis remains major health problem. Despite the progress in diagnosis of asymptomatic early-stage cirrhosis, prognostic biomarkers are needed to identify cirrhotic patients at high risk developing advanced stage disease. Liver cirrhosis is the result of deregulated wound healing and is featured by aberrant extracellular matrix (ECM) remodeling. However, it is not comprehensively understood how ECM is dynamically remodeled in the progressive development of liver cirrhosis. It is yet unknown whether ECM signature is of predictive value in determining prognosis of early-stage liver cirrhosis. In this study, we systematically analyzed proteomics of decellularized hepatic matrix and identified four unique clusters of ECM proteins at tissue damage/inflammation, transitional ECM remodeling or fibrogenesis stage in carbon tetrachloride-induced liver fibrosis. In particular, basement membrane (BM) was heavily deposited at the fibrogenesis stage. BM component minor type IV collagen α5 chain expression was increased in activated hepatic stellate cells. Knockout of minor type IV collagen α5 chain ameliorated liver fibrosis by hampering hepatic stellate cell activation and promoting hepatocyte proliferation. ECM signatures were differentially enriched in the biopsies of good and poor prognosis early-stage liver cirrhosis patients. Clusters of ECM proteins responsible for homeostatic remodeling and tissue fibrogenesis, as well as basement membrane signature were significantly associated with disease progression and patient survival. In particular, a 14-gene signature consisting of basement membrane proteins is potent in predicting disease progression and patient survival. Thus, the ECM signatures are potential prognostic biomarkers to identify cirrhotic patients at high risk developing advanced stage disease.

Odontolabis fallaciosa (Coleoptera: Lucanidae) is a giant and popular stag beetle with striking sexual dimorphism and male trimorphism. However, little is known about their intestinal microbiota, which might play an indispensable role in shaping the health of their hosts. The aim of this study was to investigate the intestinal bacterial community structure between the two sexes and among three male morphs of O. fallaciosa from China using high-throughput sequencing (Illumina MiSeq). The gut bacterial community structure was significantly different between males and females, suggesting that sex appeared to be the crucial factor shaping the intestinal bacterial community. Females had higher bacterial alpha-diversity than males. There was little difference in gut bacterial community structure among the three male morphs. However, compared to medium and small males, large individuals were associated with the higher relative abundance of Firmicutes and Firmicutes/Bacteroides (F/B) ratio, which might contribute to nutritional efficiency. Overall, these results might help to further our understanding of beetle-bacterial interactions of O. fallaciosa between the two sexes, and among the three male morphs.

Many pathogenic fungi depend on the development of specialized infection structures called appressoria to invade their hosts and cause disease. Impairing the function of fungal infection structures therefore provides a potential means by which diseases could be prevented. In spite of this extraordinary potential, however, relatively few anti-penetrant drugs have been developed to control fungal diseases, of either plants or animals. In the present study, we report the identification of compounds that act specifically to prevent fungal infection. We found that the organization of septin GTPases, which are essential for appressorium-mediated infection in the rice blast fungus Magnaporthe oryzae, requires very-long-chain fatty acids (VLCFAs), which act as mediators of septin organization at membrane interfaces. VLCFAs promote septin recruitment to curved plasma membranes and depletion of VLCFAs prevents septin assembly and host penetration by M. oryzae. We observed that VLCFA biosynthesis inhibitors not only prevent rice blast disease, but also show effective, broad-spectrum fungicidal activity against a wide range of fungal pathogens of maize, wheat and locusts, without affecting their respective hosts. Our findings reveal a mechanism underlying septin-mediated infection structure formation in fungi and provide a class of fungicides to control diverse diseases of plants and animals.

The systematics of Dorcus MacLeay has been a long-standing debate. Mitochondrial genomes were widely used to deeply understand the phylogeny of problematic taxa in virtue of their genetic importance and comprehensiveness. To provide more useful genetic data for resolving the systematic disputation of Dorcus stag beetles. The complete mitochondrial genomes of Dorcus hopei and Dorcus seguyi were obtained using the next generation sequencing. Characteristics of the two genomes are explicated through comparing their genome organization and base composition, protein-coding genes and codon usage, intergenic spacers and non-coding region, transfer and ribosomal RNA genes and control region. Phylogenetic relationships were reconstructed using Maximum likelihood and Bayesian inference analyses based on the concatenated nucleotide sequences of 13 PCGs from 9 stag beetles and 3 scarab beetles. The complete mitogenomes of D. hopei and D. seguyi was 16,026 bp/17,955 bp long, respectively. A tandem repeat with the length of 940 bp was presented in the A+T-rich region in D. hopei. An unexpected non-coding region of 332 bp was located between nad2 and trnW in D. seguyi. The phylogenetic analyses robustly supported that D. hopei formed a branch with the generic type of D. parallelipipedus. Whereas D. seguyi was not covered in the branch of (D. hopei + D. parallelipipedus), but was sister to them. The results indicated that D. hopei should be a good member of Dorcus MacLeay. The taxonomic status of D. seguyi remained to be studied furtherly.

RESUMO

The complete mitochondrial genome (mitogenome) of Prosopocoilus gracilis (Coleoptera: Lucanidae) that is endemic to Southern China is determined. The circular genome is 736 bp in length and comprises 13 protein-coding genes, 22 tRNA, 2 rRNA genes and a control region. Gene order is identical to that of the putative ancestral arrangement of insects. The nucleotide composition of heavy strand is A (36.6%), C (22.6%), T (29.5%) and G (11.3%). All protein-coding genes start with a typical ATN codon except for the gene COI that uses AAC as the start codon. tRNA-Ser (AGN) uses the anticodon UCU instead of the commonly used GCU. Both maximum likelihood and Bayesian analyses support the monophyly of Lucanidae and the sister relationship of Nigidionus and the remaining sampled genera. Two species of Prosopocoilus were not recovered as a monophyletic group.
