RESUMO

To implement an animal breeding program, it is important to define the production circumstances of the animals of interest to determine which traits of economic interest will be selected for the breeding goal. The present study defined breeding goals and proposed selection indices for milk production and quality traits of Gir dairy cattle. First, a bioeconomic model was developed to calculate economic values. The genetic and phenotypic parameters were estimated based on records from 22,468 first-lactation Gir dairy cows and their crosses for which calving occurred between 1970 and 2011. Statistical analyses were carried out for the animal model, with multitrait analyses using the restricted maximum likelihood method. Two situations were created in the present study to define the breeding goals: 1) including only milk yield in the breeding goal (HGL1) and 2) including fat and protein in addition to the milk yield (HGL2). The heritability estimates for milk, protein, and fat production were 0.33 ± 0.02, 0.26 ± 0.02, and 0.24 ± 0.02, respectively. All phenotypic and genetic correlations were highly positive. The economic values for milk, fat, and protein were US$0.18, US$0.27, and US$7.04, respectively. The expected economic responses for HGL2 and for HGL1 were US$126.30 and US$79.82, respectively. These results indicate that milk component traits should be included in a selection index to rank animals evaluated in the National Gir Dairy Breeding Program developed in Brazil.

We analyzed 75,555 test-day milk flow records from 2175 primiparous Holstein cows that calved between 1997 and 2005. Milk flow was obtained by dividing the mean milk yield (kg) of the 3 daily milking by the total milking time (min) and was expressed as kg/min. Milk flow was grouped into 43 weekly classes. The analyses were performed using a single-trait Random Regression Models that included direct additive genetic, permanent environmental, and residual random effects. In addition, the contemporary group and linear and quadratic effects of cow age at calving were included as fixed effects. Fourth-order orthogonal Legendre polynomial of days in milk was used to model the mean trend in milk flow. The additive genetic and permanent environmental covariance functions were estimated using random regression Legendre polynomials and B-spline functions of days in milk. The model using a third-order Legendre polynomial for additive genetic effects and a sixth-order polynomial for permanent environmental effects, which contained 7 residual classes, proved to be the most adequate to describe variations in milk flow, and was also the most parsimonious. The heritability in milk flow estimated by the most parsimonious model was of moderate to high magnitude.

Nowadays, hydrogen produced globally has been synthesized from fossil fuel with limited source. Therefore, research has been developed in order to explore biological H2 production by dark fermentation. The purpose of this work was to evaluate the effect of initial pH and ferrous sulfate and ammonium sulfate concentrations on the production of biohydrogen by dark fermentation. The process was carried out in batch mode under anaerobic conditions, in the absence of light, and at standard room temperature and pressure. A microbial consortium provided by the effluent treatment plant of a local dairy company was inoculated into a synthetic medium supplemented with cheese whey permeate (20 g/L of lactose) as a carbon source. The influence of three variables was analyzed by a central composite design 2((3)), and the optimum results of hydrogen yield (4.13 mol H2/mol lactose) and productivity (86.31 mmol H2/L/day) were achieved at initial pH 7.0 and FeSO4 and (NH4)2SO4 concentrations of 0.6 and 1.5 g/L, respectively. Under these conditions, the kinetic parameters of fermentation were investigated by analyzing the profile of H2 yield and productivity, metabolite concentrations, pH, and concentration of dissolved iron. In the kinetic analysis, the modified Gompertz equation described adequately the fermentative hydrogen production from cheese whey permeate (R (2) = 0.98). The profile of ethanol and volatile organic acids showed that lactic acid and butyric acid were the main metabolites produced, and the sum of both by-products corresponded to about 58 % of the total metabolites.

Buffaloes are generally raised in Brazil without milk-recording programs, and thus without genetic evaluations of any of their traits. This study evaluated the economic impacts of three different selection strategies on buffalo populations and the evolution of genetic trends, genetic variances and inbreeding coefficients resulting from each of them. The selection strategies used were as follows: (i) random selection; (ii) phenotypic selection; and (iii) progeny testing (PT). As the numbers of herds enrolled in milk-recording programs increased, phenotypic selection and PT strategies increased both monetary benefits and genetic trends. The extra costs of implementing milk recording (MR) and PT procedures were exceeded by the income resulting from better buffalo performance. Progeny testing is known to result in beneficial genetic trends and the use of artificial insemination promoted better distributions of genetic material into herds that were not enrolled in milk-recording programs. Phenotypic selection and PT increased mean milk production--a key factor in profitability. Inbreeding levels remained stable with phenotypic selection, even as the numbers of MR herds increased. Increases in the numbers of sires that were evaluated reduced the mean inbreeding coefficient in PT. Increasing the number of herds enrolled in milk-recording programs resulted in increased numbers of sires needed for PT, but this did not increase the inbreeding coefficient. In summary, phenotypic selection and PT strategies appear to be economically viable for buffalo husbandry in south-eastern Brazil under current (2007-2008) economic conditions and should be encouraged.

This study evaluated the use of natural coagulants (Moringa oleifera and chitosan) under different conditions with a mixed culture (C1 mixed culture). This culture was used for the biodegradation of hydrocarbons present in the effluent from fuel distribution terminals contaminated with diesel oil and gasoline. The biodegradation was evaluated by two central composite design (CCD) experiments: the first with varying concentrations of Moringa oleifera (MO), drying temperatures (TE) and seed drying times (TI); the second with varying concentrations of chitosan and the hydrochloric acid in which chitosan had been solubilized. The responses monitored in the CCD experiments included the sludge volume index (SVI), the turbidity removal (TR) and the specific rate of oxygen uptake (SOUR). Subsequently, the biodegradation was monitored in a sequencing batch reactor (SBR) under the optimal conditions obtained for each CCD experiment. The results indicated that the best coagulant was chitosan solubilized in 0.25 N HCl at a concentration of 50mg/L. Within five cycles with chitosan as a coagulant, the total organic carbon (TOC) removal increased from 77±1.0% to 82±0.5%, the volatile suspended solids (VSS) increased from 1.4±0.3 to 2.25±0.3 g/L and the total petroleum hydrocarbon (TPH) removal increased from 75±1.0% to 81±0.5%.

A total of 46,089 individual monthly test-day (TD) milk yields (10 test-days), from 7,331 complete first lactations of Holstein cattle were analyzed. A standard multivariate analysis (MV), reduced rank analyses fitting the first 2, 3, and 4 genetic principal components (PC2, PC3, PC4), and analyses that fitted a factor analytic structure considering 2, 3, and 4 factors (FAS2, FAS3, FAS4), were carried out. The models included the random animal genetic effect and fixed effects of the contemporary groups (herd-year-month of test-day), age of cow (linear and quadratic effects), and days in milk (linear effect). The residual covariance matrix was assumed to have full rank. Moreover, 2 random regression models were applied. Variance components were estimated by restricted maximum likelihood method. The heritability estimates ranged from 0.11 to 0.24. The genetic correlation estimates between TD obtained with the PC2 model were higher than those obtained with the MV model, especially on adjacent test-days at the end of lactation close to unity. The results indicate that for the data considered in this study, only 2 principal components are required to summarize the bulk of genetic variation among the 10 traits.

Genetic parameters for test-day milk flow (TDMF) of 2175 first lactations of Holstein cows were estimated using multiple-trait and repeatability models. The models included the direct additive genetic effect as a random effect and contemporary group (defined as the year and month of test) and age of cow at calving (linear and quadratic effect) as fixed effects. For the repeatability model, in addition to the effects cited, the permanent environmental effect of the animal was also included as a random effect. Variance components were estimated using the restricted maximum likelihood method in single- and multiple-trait and repeatability analyses. The heritability estimates for TDMF ranged from 0.23 (TDMF 6) to 0.32 (TDMF 2 and TDMF 4) in single-trait analysis and from 0.28 (TDMF 7 and TDMF 10) to 0.37 (TDMF 4) in multiple-trait analysis. In general, higher heritabilities were observed at the beginning of lactation until the fourth month. Heritability estimated with the repeatability model was 0.27 and the coefficient of repeatability for first lactation TDMF was 0.66. The genetic correlations were positive and ranged from 0.72 (TDMF 1 and 10) to 0.97 (TDMF 4 and 5). The results indicate that milk flow should respond satisfactorily to selection, promoting rapid genetic gains because the estimated heritabilities were moderate to high. Higher genetic gains might be obtained if selection was performed in the TDMF 4. Both the repeatability model and the multiple-trait model are adequate for the genetic evaluation of animals in terms of milk flow, but the latter provides more accurate estimates of breeding values.

We analyzed 152,145 test-day records from 7317 first lactations of Holstein cows recorded from 1995 to 2003. Our objective was to model variations in test-day milk yield during the first lactation of Holstein cows by random regression model (RRM), using various functions in order to obtain adequate and parsimonious models for the estimation of genetic parameters. Test-day milk yields were grouped into weekly classes of days in milk, ranging from 1 to 44 weeks. The contemporary groups were defined as herd-test-day. The analyses were performed using a single-trait RRM, including the direct additive, permanent environmental and residual random effects. In addition, contemporary group and linear and quadratic effects of the age of cow at calving were included as fixed effects. The mean trend of milk yield was modeled with a fourth-order orthogonal Legendre polynomial. The additive genetic and permanent environmental covariance functions were estimated by random regression on two parametric functions, Ali and Schaeffer and Wilmink, and on B-spline functions of days in milk. The covariance components and the genetic parameters were estimated by the restricted maximum likelihood method. Results from RRM parametric and B-spline functions were compared to RRM on Legendre polynomials and with a multi-trait analysis, using the same data set. Heritability estimates presented similar trends during mid-lactation (13 to 31 weeks) and between week 37 and the end of lactation, for all RRM. Heritabilities obtained by multi-trait analysis were of a lower magnitude than those estimated by RRM. The RRMs with a higher number of parameters were more useful to describe the genetic variation of test-day milk yield throughout the lactation. RRM using B-spline and Legendre polynomials as base functions appears to be the most adequate to describe the covariance structure of the data.

This study evaluated the use of coagulants (ferric chloride and aluminium sulphate) and an anionic polyelectrolyte (polyacrylamide) in the settling of a mixed culture (C(1)), which was used for the biodegradation of hydrocarbons present in effluent of fuel distribution terminals contaminated with diesel oil and gasoline. In preliminary investigations conducted in jar tests, the optimal concentrations of coagulant were obtained for further studies. After preliminary tests, biodegradation was evaluated in a central composite design (CCD) with varying concentrations of ferric chloride and polyelectrolyte. Ferric chloride and polyelectrolyte concentrations ranged from 77.9 to 422.12 mg/L and 0.0 to 3.2mg/L, respectively. The responses monitored in CCD experiments included the sludge volume index (SVI), turbidity, and specific rate of oxygen uptake (SOUR), where values of 100mL/g, 840 nephelometric turbidity unit (NTU) and 58 mg O(2)/gh, respectively, were obtained. Subsequently, biodegradation was monitored in a sequencing batch reactor (SBR). The results indicated that within five cycles, total petroleum hydrocarbon (TPH) removal increased from 75 ± 1.0% to 79 ± 0.5%, while the volatile suspended solids (VSS) increased from 1300 to 2500 mg/L.

The objective of the present study was to estimate milk yield genetic parameters applying random regression models and parametric correlation functions combined with a variance function to model animal permanent environmental effects. A total of 152,145 test-day milk yields from 7,317 first lactations of Holstein cows belonging to herds located in the southeastern region of Brazil were analyzed. Test-day milk yields were divided into 44 weekly classes of days in milk. Contemporary groups were defined by herd-test-day comprising a total of 2,539 classes. The model included direct additive genetic, permanent environmental, and residual random effects. The following fixed effects were considered: contemporary group, age of cow at calving (linear and quadratic regressions), and the population average lactation curve modeled by fourth-order orthogonal Legendre polynomial. Additive genetic effects were modeled by random regression on orthogonal Legendre polynomials of days in milk, whereas permanent environmental effects were estimated using a stationary or nonstationary parametric correlation function combined with a variance function of different orders. The structure of residual variances was modeled using a step function containing 6 variance classes. The genetic parameter estimates obtained with the model using a stationary correlation function associated with a variance function to model permanent environmental effects were similar to those obtained with models employing orthogonal Legendre polynomials for the same effect. A model using a sixth-order polynomial for additive effects and a stationary parametric correlation function associated with a seventh-order variance function to model permanent environmental effects would be sufficient for data fitting.

We examined the effects of aeration interval and agitation speed in an effort to optimize the biodegradation of effluent contaminated with diesel oil and gasoline. The biodegradation process employed a C(1) mixed culture and intermittent aeration. Optimization was performed using central composite design (CCD). The independent variables were aeration interval (X(1)) and agitation speed (X(2)) and the dependent variable was the degree of removal of total petroleum hydrocarbons (TPH). The optimum aeration interval (33h) and agitation speed (110rpm) resulted in TPH removal of 75.9% after 3 days. Five hydrocarbons classes were identified using gas chromatography: paraffins, isoparaffins, olefins, naphthenics, and aromatics. The following reductions were observed: 91.8% for the paraffins, 83.3% for the isoparaffins, 80.9% for the olefins, 39.3% for the naphthenics, and 80.9% for the aromatics. In comparative studies performed under constant aeration (CA), without aeration (WA), and intermittent aeration (IA) conditions, intermittent aeration resulted in the highest TPH removals, with 90% reduction after 22 days. Under the IA condition, the degradation percentages were 99.6% for the paraffins, 94% for the isoparaffins, 95.4% for the olefins, 70.8 for the naphthenics, and 83.4% for the aromatics.

Xanthan gum production was studied using sugarcane broth as the raw material and batch fermentation by Xanthomonas campestris pv. campestris NRRL B-1459. The purpose of this study was to optimize the variables of sucrose, yeast extract, and ammonium nitrate concentrations and to determine the kinetic parameters of this bioreaction under optimized conditions. The effects of yeast extract and ammonium nitrate concentrations for a given sucrose concentration (12.1-37.8 g L(-1)) were evaluated by central composite design to maximize the conversion efficiency. In a bioreactor, the maximum conversion efficiency was achieved using 27.0 g L(-1) sucrose, 2.7 g L(-1) yeast extract, and 0.9 g L(-1) NH(4)NO(3). This point was assayed in a shaker and in a bioreactor to compare bioreaction parameters. These parameters were estimated by the unstructured kinetic model of Weiss and Ollis (Biotechnol Bioeng 22:859-873, 1980) to determinate the yields (Y (P/S)), the maximum growth specific rate (mu (max)), and the saturation cellular concentration (X*). The parameters of the model (mu (max), X*, m, lambda, alpha, and beta) were obtained by nonlinear regression. For production of xanthan gum in a shaker, the values of mu (max) and Y (P/S) obtained were 0.119 h(-1) and 0.34 g g(-1), respectively, while in a bioreactor, they were 0.411 h(-1) and 0.63 g g(-1), respectively.

Pseudomonas aeruginosa PACL strain, isolated from oil-contaminated soil taken from a lagoon, was used to investigate the efficiency and magnitude of biosurfactant production, using different waste frying soybean oils, by submerged fermentation in stirred tank reactors of 6 and 10 l capacities. A complete factorial experimental design was used, with the goal of optimizing the aeration rate (0.5, 1.0, and 1.5 vvm) and agitation speed (300, 550, and 800 rpm). Aeration was identified as the primary variable affecting the process, with a maximum rhamnose concentration occurring at an aeration rate of 0.5 vvm. At optimum levels, a maximum rhamnose concentration of 3.3 g/l, an emulsification index of 100%, and a minimum surface tension of 26.0 dynes/cm were achieved. Under these conditions, the biosurfactant production derived from using a mixture of waste frying soybean oil (WFSO) as a carbon source was compared to production when non-used soybean oil (NUSO), or waste soybean oils used to fry specific foods, were used. NUSO produced the highest level of rhamnolipids, although the waste soybean oils also resulted in biosurfactant production of 75-90% of the maximum value. Under ideal conditions, the kinetic behavior and the modeling of the rhamnose production, nutrient consumption, and cellular growth were established. The resulting model predicted data points that corresponded well to the empirical information.

The imprinted gene insulin-like growth factor 2 (IGF2) carries out important functions in the development of placental mammals, during the embryonic and fetal stages and is located on bovine chromosome 29. The main aim of this study was to verify the occurrence of a polymorphism (C/T transversion) in exon 6 of the IGF2 gene in Gir and Nelore cattle, Zebu breeds of great economic importance in Brazil for dairy and beef production, respectively. A 193 bp fragment of bovine IGF2 exon 6 was amplified by PCR with specific primers for this region and digested with Mboll enzyme to analyze the polymorphic segment. In a total of 39 Gir animals (29 females and 10 males), the estimated frequencies of the C and T alleles were 0.42 and 0.58, respectively. The genotypic frequencies were 0.10 for CC, 0.26 for TT and 0.64 for CT. For 28 Nelore animals (17 females and 11 males), the estimated frequencies of the C and T alleles were 0.43 and 0.57, respectively. The genotypic frequencies were 0.18 for CC, 0.32 for TT, and 0.50 for CT. This is the first report of the occurrence of this polymorphism in these breeds. The polymorphic site can be a regulatory motif with functional significance to gene regulation of the IGF2 gene. This polymorphism could be used to investigate the allele-specific expression of the IGF2 gene, its epigenetic status, and its role in developmental, growth and reproductive traits.

In the present work, the production of rhamnolipid from residual soybean oil (RSO) from food frying facilities was studied using a strain of Pseudomonas aeruginosa of contaminated lagoon, isolated from a hydrocarbon contaminated soil. The optimization of RSO, ammonium nitrate, and brewery residual yeast concentrations was accomplished by a central composite experimental design and surface response analysis. The experiments were performed in 500-mL Erlenmeyer flasks containing 50 mL of mineral medium, at 170 rpm and 30 +/- 1 degrees C, for a 48-h fermentation period. Rhamnolipid production has been monitored by measurements of surface tension, rhamnose concentration, and emulsifying activity. The best-planned results, located on the central point, have corresponded to 22 g/L of RSO, 5.625 g/L of NH(4)NO(3), and 11.5 g/L of brewery yeast. At the maximum point the values for rhamnose and emulsifying index were 2.2 g/L and 100%, respectively.

RESUMO

A Bacillus subtilis strain isolated from contaminated soil from a refinery has been screened for biosurfactant production in crystal sugar (sucrose) with different nitrogen sources (NaNO(3), (NH(4))(2)SO(4), urea, and residual brewery yeast). The highest reduction in surface tension was achieved with a 48-h fermentation of crystal sugar and ammonium nitrate. Optimization of carbon/nitrogen ratio (3, 9, and 15) and agitation rate (50, 150, and 250 rpm) for biosurfactant production was carried out using complete factorial design and response surface analysis. The condition of C/N 3 and 250 rpm allowed the maximum increase in surface activity of biosurfactant. A suitable model has been developed, having presented great accordance experimental data. Preliminary characterization of the bioproduct suggested it to be a lipopeptide with some isomers differing from those of a commercial surfactin.

We studied the effects of fuel concentration (diesel and gasoline), nitrogen concentration and culture type on the biodegradation of synthetic effluent similar to what was found at inland fuel distribution terminals. An experimental design with two levels and three variables (2(3)) was used. The mixed cultures used in this study were obtained from lake with a history of petroleum contamination and were named culture C(1) (collected from surface sediment) and C(2) (collected from a depth of approximately 30cm). Of the parameters studied, the ones that had the greatest influence on the removal of total petroleum hydrocarbons (TPH) were a nitrogen concentration of 550mg/L and a fuel concentration of 4% (v/v) in the presence of culture C(1). The biodegradability study showed a TPH removal of 90+/-2% over a process period of 49 days. Analysis using gas chromatography identified 16 hydrocarbons. The aromatic compounds did not degrade as readily as the other hydrocarbons that were identified.

The aim of the present study was to estimate the index and individual responses to selection for milk (MY), fat (FY) and protein (PY) yields for different breeding goals for two commercial buffalo milk production systems in São Paulo State characterized by: 1) all milk produced is sold to the industry (MILK) and 2) all milk produced is used in the mozzarella cheese-making process at the farm (MOZZARELLA). The current payment policy is based exclusively on milk volume. The mozzarella price refers to the wholesale selling price. Index responses to selection (IR) were calculated for three different breeding goals (BG): 1) MY exclusively (BG(1)); 2) FY + PY (BG(2)) and 3) MY + FY + PY (BG(3)). IR for the MILK system were 41.79 US dollars (BG(1)), 5.91 US dollars (BG(2)) and 38.22 US dollars (BG(3)). For the MOZZARELLA system, IR were 179.50 US dollars (BG(1)), 262.85 US dollars (BG(2)) and 402.41 US dollars (BG(3)). The results suggest that for the present circumstances, selection for milk components is not advantageous when milk is produced for sale to the industry. However, when mozzarella making is added to the system, the selection for components and milk volume is the most economically beneficial.

Compararam-se os custos de seis esquemas de controle leiteiro (CL), estabelecidos com base em diferentes freqüências de controles (com possibilidade de controle trimestral supervisionado e aproveitamento do CL da fazenda), no número de ordenhas e na realização de análises qualitativas (composição do leite e/ou contagem de células somáticas). Computaram-se os percentuais que os custos totais do CL representariam na receita bruta mensal do leite (CL/RB) e na receita líquida mensal (CL/RL). O esquema tradicional (freqüência mensal) apresentou o mais elevado custo mensal e os maiores CL/RB e CL/RL. Esquemas com intervalo maior entre controles e amostragem em ordenhas alternadas resultaram na redução dos custos com diárias e quilometragem. O custo mensal do CL variou de 0,68% a 1,8% sobre a receita bruta do leite e de 6,6% a 17,0% sobre a receita líquida.
