OBJECTIVE: The Manihot esculenta Crantz (Cassava) is a typical South American plant rich in nutrients and energetic compounds. Lately, our group has shown that non-pharmacological interventions with natural antioxidants present different neuroprotective effects on oxidative balance and memory deficits in AD-like animal models. Here, our objective was to evaluate the neuroprotective effects of Cassava leaves' extract (CAS) in an AD-like model induced by amyloid-beta (Aß) 25-35 peptide. METHODS: Male Wistar rats (n = 40; 60 days old) were subjected to 10 days of CAS supplementation; then, we injected 2 µL Aß 25-35 in the hippocampus by stereotaxic surgery. Ten days later, we evaluated object recognition (OR) memory. Cassavas' total polyphenols, flavonoids, and condensed tannins content were measured, as well as hippocampal lipid peroxidation and total antioxidant capacity. RESULTS: CAS protected against Aß-induced OR memory deficits. In addition, Aß promoted antioxidant capacity decrease, while CAS was able to prevent it, in addition to diminishing lipoperoxidation compared to Aß. DISCUSSION: We show that treatment with Cassava leaves' extract before AD induction prevents recognition memory deficits related to Aß hippocampal injection. At least part of these effects can be related to the Cassava leaves' extract supplementation effects on diminishing lipid peroxidation and preventing a decrease in the hippocampal total antioxidant capacity in the hippocampus of AD-like animals without adverse effects. Once cassavais a plant of warm and dry ground that can adapt to growon various soil types and seems to resist several insects, our results enable Cassava to be considered asa potential preventive intervention to avoid or minimizeAD-induced memory deficits worldwide.

COVID-19 Acute Respiratory Distress Syndrome (CARDS) is the most serious complication of COVID-19. The SARS-CoV-2 outbreaks rapidly saturated intensive care unit (ICU), forcing the application of non-invasive respiratory support (NIRS) in respiratory intermediate care unit (RICU). The primary aim of this study is to compare the patients' clinical characteristics and outcomes (Helmet-Continuous Positive Airway Pressure (H-CPAP) success/failure and survival/death). The secondary aim is to evaluate and detect the main predictors of H-CPAP success and survival/death. A total of 515 patients were enrolled in our observational prospective study based on CARDS developed in RICU during the three Italian pandemic waves. All selected patients were treated with H-CPAP. The worst ratio of arterial partial pressure of oxygen (PaO2) and fraction of inspired oxygen (FiO2) PaO2/FiO2 during H-CPAP stratified the subjects into mild, moderate and severe CARDS. H-CPAP success has increased during the three waves (62%, 69% and 77%, respectively) and the mortality rate has decreased (28%, 21% and 13%). H-CPAP success/failure and survival/death were related to the PaO2/FiO2 (worst score) ratio in H-CPAP and to steroids' administration. D-dimer at admission, FiO2 and positive end expiratory pressure (PEEP) were also associated with H-CPAP success. Our study suggests good outcomes with H-CPAP in CARDS in RICU. A widespread use of steroids could play a role.

Metalworkers are exposed to numerous chemicals in their workplace environment, such as solvents, heavy metals, and metalworking fluids, that have a negative impact on their health. Furthermore, there is an increase in the prevalence of chronic diseases among metalworkers; however, the molecular mechanisms involved in this increased predisposition to chronic diseases are unclear. Considering that occupational exposure represents a potential risk for metalworkers, the aim of this study was to measure biomarkers of oxidative stress, inflammation, and cytotoxicity in the peripheral blood of metalworkers from Southern Brazil. The study included 40 metalworkers and 20 individuals who did not perform activities with any recognized exposure to chemical substances, such as those working in administration, commerce, and education, as controls. Cellular and molecular biomarkers as leukocyte viability, intracellular production of reactive species, mitochondrial mass and membrane potential and plasma lipid peroxidation, sulfhydryl groups, total antioxidant capacity, and butyrylcholinesterase activity were evaluated in the blood of metalworkers and controls. Metalworkers were found to have higher rates of apoptosis, increased production of reactive species, and increased mitochondrial potential and mass in leukocytes associated with decreased antioxidant defenses and increased activity of the butyrylcholinesterase enzyme in their plasma. It can be concluded that cytotoxicity, oxidative stress, and inflammation are involved in the multiplicity of health outcomes related to chemical exposure in the metalworking industry.

The health of family farmers is at risk due to occupational exposure to pesticides. The aims of the current study were to investigate the level of farmers' perception of risks associated with pesticide use and to assess their health condition based on biochemical and immunological tests. Family farmers living in a vegetable-producing region in Southern Brazil were selected to participate in the study. More than 70% of the family farmers were often exposed to more than one type of pesticides; 41.2% were intensively using several pesticides for more than one decade and 74.4% were not using personal protective equipment (PPE) at the time of pesticide handling due to low perception of the risks posed by these chemicals. Enzymatic analysis performed in participants' blood samples showed changes in catalase (CAT) and glutathione reductase (GR) activity, in lipid peroxidation (TBARS) and carbonylated protein levels, as well as in chemoattractant (IL-8) and anti-inflammatory (IL-10) interleukin expression. Low perception of health-related risks posed by pesticides can be attributed to factors such as low schooling and lack of information, which put farmers' health at risk, as evidenced by blood biochemical and immunological changes.

BACKGROUND: We evaluated the ability of quercetin, a natural antioxidant formulated in a specific delivery system, to reduce skin inflammation induced by a variety of stimuli, including UV radiation, stimulation with a histamine solution, or contact with chemical irritants. In particular, we tested the soothing and anti-itch effect of Quercevita(®), 1% cream for external use, a formulation characterized by a phospholipids-based delivery system. PATIENTS AND METHODS: The study was a monocentric, single blind trial that enrolled a group of 30 healthy volunteers. The back of each subject was examined to identify four quadrants with no previous skin damage or naevi that were treated in order to induce a controlled and reversible form of skin stress. The areas were treated as follows: no product; Quercevita(®) 1% cream, 2 mg/cm(2); placebo; positive control (a commercially available topical formulation containing 1% dexchlorpheniramine). RESULTS: Only quercetin phospholipids 1% and dexchlorpheniramine 1% achieved a significant reduction in erythema with comparable results: (-10.05% [P=0.00329] for quercetin phospholipids 1% vs -14.05% [P=0.00046] for the positive control). Moreover, quercetin phospholipids 1% and dexchlorpheniramine 1% were both associated with a significant decrease in mean wheal diameter: (-13.25% and -12.23% for dexchlorpheniramine 1%, respectively). Similar findings were reported for the other tested parameters. CONCLUSION: Quercetin has a skin protective effect against damage caused by a variety of insults, including UV radiation, histamine, or contact with toxic chemical compounds. Indeed, quercetin is able to reduce redness, itching, and inflammation of damaged skin; it may also help restore skin barrier function, increasing hydration, and reducing water loss.

Noninvasive ventilation (NIV) was introduced as an alternative to invasive mechanical ventilation for acute respiratory failure caused from exacerbations of chronic obstructive pulmonary disease in the 1980s, and its use gradually rose worldwide. Seventy-eight patients (57 males, mean age 78.3 ± 9.2 years) undergoing NIV were evaluated. Of them, 48 (62.3%) had acute hypercapnic respiratory failure because of a chronic obstructive pulmonary disease exacerbation, and the remaining 30 had acute hypercapnic respiratory failure from other causes, mainly cardiac failure. All patients were treated by NIV using the bi-level positive airway pressure set up at high pressure/high backup rate. NIV was successful in 67 subjects (85.9%) and the patients were discharged, 57 of whom continued NIV at home and ten had spontaneous breathing. NIV was unsuccessful in eleven patients, ten of whom died and one was successfully treated by invasive mechanical ventilation. Significant differences were detected for a higher basal Glasgow Coma Scale score in successfully treated patients (P = 0.007), a higher basal Acute Physiology and Chronic Health Evaluation score in unsuccessfully treated patients (P = 0.004), and a lower pH after 1 hour in unsuccessfully treated patients (P = 0.015). These findings show a very high rate of success of NIV in patients with acute hypercapnic respiratory failure not only from chronic obstructive pulmonary disease but also from cardiac failure. This suggests that the use of invasive mechanical ventilation may be further reduced, with a decrease in its known complications as well.

The aim of this research was to evaluate possible toxic effects of commercial formulation containing fipronil on Cyprinus carpio tissues under rice field conditions. Antioxidant profile (SOD, catalase, glutathione S-transferase), oxidative stress parameters (thiobarbituric acid-reactive substances, protein carbonyl), and growth were investigated in carp exposed to fipronil under rice field conditions for 7, 30, and 90 days. Waterborne insecticide concentrations were measured and the detectable concentration of fipronil was observed up to 45 day after application. Common carp survival and growth was not affected by fipronil. Liver superoxide dismutase activity was enhanced while liver catalase activity was inhibited at 7, 30, and 90 days. Alterations were not observed in the glutathione S-transferase activity in any experimental periods. Protein carbonyl increased only after 30 and 90 days of exposure. The thiobarbituric acid-reactive substances levels were enhanced in all analyzed tissues (liver, muscle, and brain) and periods of exposure. This study demonstrates that fipronil insecticides cause alterations in the biochemical parameters in different tissues of carp without affecting the growth or the survival of the fish.

Field and laboratory experiments were performed to evaluate toxicological responses of Cyprinus carpio exposed to the commercial herbicide clomazone (500 mg l(-1)). Fish were exposed to 0.5 mg l(-1) of the formulated herbicide for 7, 30, and 90 days. Fish were exposed to clomazone in field conditions (7, 30, or 90 days trapped in submersed cages together with rice crops) and in laboratory conditions where the fish were placed in 45-l tanks with tap water only for 7 days. Fish exposed for 7, 30, or 90 days showed no alterations in acetylcholinesterase (AChE) activity under field conditions. Under laboratory conditions, decreased muscle AChE activity was observed only after 7 days of exposure. During the same evaluation period (7 days), oxidative stress parameters changed under both field and laboratory conditions; however, metabolic parameters were altered only under field conditions. Disorders in oxidative stress parameters and metabolism were evident in different tissues up to day 90 after treatment. These overall results show that AChE activity changed only under laboratory conditions. Oxidative stress, along with metabolic parameters, may be good indicators of herbicide contamination in C. carpio under rice-field conditions.

The effects of commercial glyphosate herbicide formulation on the activity of acetylcholinesterase (AChE) enzyme and oxidative stress were studied in Cyprinus carpio exposed for 96 h to 0.0, 0.5, 2.5, 5.0 and 10.0 mg/L and then allowed to equal recovery period in water without herbicide. The activity of AChE was inhibited in the brain and in the muscle after exposure. However, after recovery period brain and muscle AChE activity increased. Brain thiobarbituric acid reactive species (TBARS) were measured as an indicator of oxidative stress. Increased TBARS levels were observed with all concentrations tested of the glyphosate formulation, and remained increased after the recovery period. The results recorded clearly indicate lipid peroxidation and anti-AChE action induced by Roundup(®) exposure.

BACKGROUND: Noninvasive mechanical ventilation (NIMV) is an effective tool in treating patients with acute respiratory failure (ARF), since it reduces both the need for endotracheal intubation and the mortality in comparison with nonventilated patients. A particular issue is represented by the outcome of NIMV in patients referred to the emergency department for ARF and with a do-not-intubate (DNI) status because of advanced age or excessively critical conditions. This study evaluated long-term survival in a group of elderly patients with acute hypercapnic ARF who had a DNI order and who were successfully treated by NIMV. METHODS: The population consisted of 54 patients with a favorable outcome after NIMV for ARF. They were followed up for 3 years by regular control visits, with at least one visit every 4 months, or as needed according to the patient's condition. Of these, 31 continued NIMV at home and 23 were on long-term oxygen therapy (LTOT) alone. RESULTS: A total of 16 of the 52 patients had not survived at the 1-year follow-up, and another eight patients died during the 3-year observation, with an overall mortality rate of 30.8% after 1 year and 46.2% after 3 years. Comparing patients who continued NIMV at home with those who were on LTOT alone, 9 of the 29 patients on home NIMV died (6 after 1 year and 3 after 3 years) and 15 of the 23 patients on LTOT alone died (10 after 1 year and 5 after 3 years). CONCLUSION: These results show that elderly patients with ARF successfully treated by NIMV following a DNI order have a satisfactory long-term survival.

Cyprinus carpio fish were exposed to penoxsulam (Ricer) in field conditions. The experiment in the rice field was carried out for 7, 21 and 72 days. Oxidative stress parameters and antioxidant profile were studied. The acetylcholinesterase (AChE) enzyme activity in the brain was increased after 7 days and reduced after 21 and 72 days of the experiment in the rice field. The AChE activity in muscle was reduced only after 72 days of exposure. Thiobarbituric acid-reactive species were increased in the liver, brain and muscle at 7 days of the trial, reduced at 21 days in the brain and unaltered after 72 days of exposure in muscle. However, an increase in this parameter in the brain and liver was observed. Liver glutathione S-transferase was reduced at 7 days, unchanged at 21 days and increased after 72 days of exposure. Catalase of the liver changed only in the second experimental period, when it was reduced. Liver protein carbonyl was reduced at 7 days and increased at 21 and 72 days of exposure. This study shows long-term effects of rice herbicide at environmentally relevant concentrations on toxicological parameters in different tissues (brain, muscle and liver) of Cyprinus carpio.

The effects of sublethal cadmium concentrations on oxidative stress parameters were evaluated in Rhamdia quelen. The fish were exposed to 0.44, 236, and 414 µg l⁻¹ cadmium for 7 and 14 days, followed by the same time periods for recovery. Enzymes, such as catalase (CAT), superoxide dismutase (SOD), and glutathione S-transferase (GST), and indicators of oxidative stress, such as thiobarbituric acid-reactive species (TBARS) and protein carbonyl, were verified in fish tissues. In addition, the accumulation of cadmium was evaluated in these tissues. Our results indicate that CAT and GST levels decreased in gills after exposure periods associated with increased TBARS levels. In hepatic tissue, CAT, GST, TBARS, and protein carbonyl levels increased after 7 days of exposure, whereas SOD activity decreased after exposure for 14 days. In the kidney, TBARS levels decreased after exposure for 7 days and increased after exposure for 14 days. During the recovery periods, some variations persisted in gills, liver, and kidney. Cadmium accumulation was most significant in liver, followed by kidney and gills. These results indicate that cadmium concentrations studied invoke a stress response in silver catfish.

Antioxidant enzymes and oxidative stress indicators were evaluated in fish exposed to different concentrations of the herbicide Roundup 48% (Monsanto, St. Louis, MO): control (none), 0.45, or 0.95 mg/l. After exposure for 8 days to herbicide, fish were transferred to clean water for a recovery response period (also 8 days). Herbicide increased thiobarbituric acid reactive species in liver and muscle at the higher concentration and in the brain at both concentrations. Protein carbonyl in liver increased after exposure. Catalase (CAT) and superoxide dismutase (SOD) activities and ascorbic acid levels in liver did not change in fish exposed to both concentrations. Glutathione S-transferase (GST) levels decreased at both concentrations. The nonprotein thiol levels decreased at the 0.95 mg/l concentration. During the recovery period, some of the parameters that had altered, such as protein carbonyl content, later recovered. However, some enzymes reacted during this period, e.g., GST increased its activity, possibly indicating a compensatory response against the toxic conditions. In contrast, CAT and SOD activities decreased during the recovery period, indicating herbicide toxicity. Oxidative stress that occurred during the exposure period was likely due to the increased lipid peroxidation and protein carbonyl content. The results concerning oxidative and antioxidant profiles indicate that short-term exposure to herbicide is capable of causing oxidative stress in fish tissues.

Pesticides can have an effect on the biochemical and physiological functions of living organisms. The changes seen in fish and their response to pesticides can be used as an example for vertebrate toxicity. In this study, carp fish (Cyprinus carpio) were exposed to different concentrations of tebuconazol fungicide, by rice field (31.95 µg/L) and laboratory (33.47 and 36.23 µg/L) conditional testing, during a 7 day period. Parameters such thiobarbituric acid-reactive substance levels (TBARS), protein carbonyl, catalase, glutathione S-transferase and acetylcholinesterase activities were studied, using the liver, brain and white muscle of the fish. The field experiment showed that the TBARS levels were increased in all the analyzed tissues. Similarly, the protein carbonyl of the liver and the brain AChE activity increased after 7 days. The laboratory experiment demonstrated that the TBARS levels in the liver were increased in both of the concentration tests. TBARS levels in the muscle increased only by the lowest test concentration. On the other hand, the protein carbonyl was increased only by the highest concentration. The results indicate that the tebuconazol exposure from the field and laboratory conditions directly affected the health of the fish, showing the occurrence of oxidative stress.

Cyprinus carpio were exposed under field conditions to 20.87 microg l(-1) of commercial herbicide bispyribac-sodium (Nominee, SC), during 7, 21 and 72 days. Enzymatic parameters such as catalase (CAT), glutathione S-transferase (GST) and acetylcholinesterase (AChE) activities, as well as thiobarbituric acid-reactive substances (TBARS) and protein carbonyl contents were studied in different tissues. After 7 days of exposure, GST activity decreased. At the same period, brain AChE activity increased, but a reduction of activity was observed in muscle tissue. Brain TBARS levels increased at 7 days. After 21 days of exposure liver CAT levels and muscle AChE activities decreased. In the same period, liver protein carbonyl and muscle TBARS increased. After 72 days of exposure in the field, AChE activity was reduced in both brain and muscle. Protein carbonyl contents in liver and brain TBARS levels increased. Muscle AChE activity, TBARS and protein carbonyl can be used as biomarkers of exposure to the herbicide bispyribac-sodium. This study demonstrates effects of exposure to bispyribac-sodium under rice field conditions on oxidative stress parameters in tissues of Cyprinus carpio.

Tobacco dependence reaches nearly one third of the world population, and is the second leading cause of deth around the world. Cotinine, a major metabolite of nicotine, is conssidered the most appropriate parameter to evaluate tobacco exposure HLPC (High Performance Liquid Chromatography) was choses to analyze cotinine concentrations in urine. Aiming at validating this method of analysis, urine samples with added cotinine were submitted to liquid-liquid extractions using2-phenylimidazole as the internal standard. After evaporation, residue was injected in HLPC with ultraviolet detection and C8 column. The following parameters were stabilishe: linearity, precision, detection and qualification limit, accuracy, ans stability. The calibration curve, determined ian a interval of 10ng/mL and 1000ng/mL, presented linearity (R2 = 0,09979). The method limit of detection was of 5ng/mL, and the limit of quantification was of 10ng/mL. In 50, 500, and 1000ng/mL concentrations, the precision was of 11.2; 12.3, and 8.1 per cent, accuracy found was of 107.0; 101.7, and 97.0 per cent, and loss during the stability test was of 8.1...
