RESUMO

The present study aimed to quantify and compare in situ the primary and secondary physiological stress responses, related to the changing operations of floating net cages, in both subadult (523 days post hatching [dph]) and adult (916 dph) European sea bass Dicentrarchus labrax under intensive farming conditions in the Moroccan M'diq Bay. The blood levels of cortisol, glucose, total cholesterol, total protein, and lactate, as well as the percentage of haematocrit, were measured before and after this operation. The results showed significantly elevated levels of cortisol and blood glucose in both age groups, whereas total cholesterol and protein levels were unaffected. In fact, blood lactate significantly decreased in subadults, whereas in adults this parameter was not affected by the operation. However, the haematocrit percentages measured after the operation were significantly higher than those found before the operation in both groups of fish, which is attributed to the increased rate of oxygen renewal in the new net cages and the lower water temperature inside the cages. With regard to the age-specific response during this essential operation plasma cortisol, blood glucose, and lactate concentrations, as well as plasma total protein levels, were significantly higher in subadults than in adults, in both pre- and post- stress measurement, with the presence of individual-specific response. It is concluded that aquaculture practices such as changing the aquaculture net cage could have repercussions in terms of the classic physiological responses to stress in D. labrax.

RESUMO

Globally, aquaculture contributes to sustainable food and nutritional stability. However, stress conditions constitute a major threat affecting farmed-fish welfare and fish farms' performances. In this regard, the present study was aimed at measuring and comparing in-situ (offshore) the physiological stress responses caused by recurrent sampling operations for length−weight measurement. Studied fish were European sea bass Dicentrarchus labrax sub-adults and adults reared in intensive farming conditions in M'diq Bay, on the Moroccan Mediterranean coast. The physiological stress response was evaluated by measuring blood biomarkers, including cortisol, glucose, lactate, total cholesterol and total proteins levels, and hematocrit percentage. The hypercortisolemia, hyperglycemia, hyperlactatemia and hypoproteinemia detected in the post-sampling state in both age groups of D. labrax indicated stress tendencies and a high sensitivity to aquaculture practice-related stress factors, with major and minor responses detected in the same age and same stress conditions. It is also interesting to note that the fish age and the time-course of the sampling operation had a statistically significant effect in terms of the physiological response (with p < 0.01 and p < 0.001, respectively). In conclusion, the present study showed that sea bass subjected to various stressful situations in intensive farming conditions displayed a physiological stress response specific to its age, to the individual status, as well as to the duration of stressor.

RESUMO

Isolation and identification of novel microalgae strains with high lipid productivity is one of the most important research topics to have emerged recently. However, practical production processes will probably require the use of local strains adapted to commanding climatic conditions. The present manuscript describes the isolation of 96 microalgae strains from seawater located in Bay M'diq, Morocco. Four strains were identified using the 18S rDNA and morphological identification through microscopic examination. The biomass and lipid productivity were compared and showed good results for Nannochloris sp., (15.93 mg/L/day). The lipid content in the four species, namely Nannochloropsis gaditana, Nannochloris sp., Phaedactylum tricornutum and Tetraselmis suecica, was carried out by high performance liquid chromatography coupled to mass spectrometry (HPLC-MS )highlighting the identification of up to 77 compounds.

RESUMO

The U2 snRNA multigene family has been analyzed in four species of the Engraulidae family--Engraulis encrasicolus, Engraulis mordax, Engraulis ringens, and Engraulis japonicas--with the object of understanding more about the structure of this multigene family in these pelagic species and studying their phylogenetic relationships. The results showed that the cluster of this gene family in the Engraulis genus is formed by the U2-U5 snRNA with highly conserved sequences of mini- and micro-satellites, such as (CTGT)n, embedded downstream of the transcription unit; findings indicate that this gene family evolved following the concerted model. The phylogenetic analysis of the non-transcribed spacer of cluster U2-U5 snDNA in the 4 species showed that the sequences of the species E. encrasicolus and E. japonicus are closely related; these two are genetically close to E. mordax and slightly more distant from E. ringens. The data obtained by molecular analysis of U2-U5 snDNA and their secondary structure, with the presence of the micro-satellite (CTGT)n and mini-satellites, show clearly that the species E. encrasicolus and E. japonicus are closely related and would be older than E. mordax and E. ringens.

RESUMO

The four species of the Engraulidae family: European anchovy (Engraulis encrasicolus), Californian anchovy (Engraulis mordax), Peruvian anchoveta (Engraulis ringens), and Japanese anchovy (Engraulis japonicus) studied in this work are very similar morphologically, and it is very difficult to distinguish between them, especially when frozen or processed. We have used the 5S rDNA as a molecular marker to discriminate these four species and used specific primers designed for each species in the nontranscribed spacers (NTS) of these genes. Multiplex PCR was performed with three pairs of primers, and three different sizes were obtained: 597 bp E. encrasicolus, 598 bp E. japonicus, 380 bp E. mordax, and 250 bp E. ringens. For the species E. encrasicolus and E. japonicus, PCR-RFLP was used as an additional technique to distinguish between them because their NTS sequences showed considerable similarity.

RESUMO

By analyzing three multigene families, two closely related and commercially important species, Dicentrarchus labrax and Dicentrarchus punctatus, were characterized by cytogenetic and molecular methods. The interspecies hybrid Dicentrarchus labrax (♀) × Dicentrarchus punctatus (♂) was also analyzed. The multigene families studied were the 5S rDNA, 45S rDNA and the U2 snRNA. A microsatellite GTT motif was found within the non transcribed spacers (NTS) of the 5S rDNA from the two species. However, hexanucleotide duplication next to this microsatellite was observed in the D. labrax and hybrid clones, but not in D. punctatus. The U2 snRNA appeared to be linked to the U5 gene and showed two variant sequences, in both D. labrax and D. punctatus. They differed in one insertion/deletion of 7 nucleotides. The first internal transcribed spacer (ITS-1) region showed higher nucleotide variability in D. punctatus than in D. labrax. Nucleotide polymorphism within species and also nucleotide divergence between species were determined in the different gene regions. In a FISH analysis we obtained three chromosomal markers, because the 5S rDNA, 18S rDNA and U2 snRNA probes hybridized each in three different chromosome pairs. Hence none of them was co-localized. The 5S rDNA cluster and U2 snRNA were localized in acrocentric chromosome pairs, while the 18S rRNA gene probe hybridized in a subtelocentric pair. Finally, the usefulness of the results in developing tools for phylogenetic analysis and species identification are discussed in relation to other fish species.
