RESUMO

OBJECTIVE: To investigate the correlation of iron metabolic parameters with platelet counts in blood donors. METHODS: A total of 400 blood donors who met requirements of apheresis platelet donation were collected, and their hematological parameters were analyzed. The donors were divided into low ferritin group and normal group, the differences of hematological parameters between the two groups were compared, and the correlation of iron metabolic parameters and routine hematology parameters with platelet counts were analyzed. RESULTS: Whether male or female, low ferritin group had higher platelet counts than normal group (P < 0.01). Among the iron metabolic parameters, the platelet counts was negatively correlated with serum ferritin (SF), serum iron (SI), and transferrin saturation (TSAT) (r =-0.162, r =-0.153, r =-0.256), and positively correlated with total iron binding capacity (TIBC) and unsaturated iron binding capacity (UIBC) (r =0.219, r =0.294) in female blood donors. Platelet counts was also negatively correlated with SF, SI and TSAT (r =-0.188, r =-0.148, r =-0.224) and positively correlated with UIBC (r =0.220) in male blood donors. Among the routine hematology parameters, platelet counts was negatively correlated with mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), and reticulocyte hemoglobin equivalent (Ret-He) in female blood donors (r =-0.236, r =-0.267, r =-0.213, r =-0.284). Platelet counts was also negatively correlated with MCH, MCHC and Ret-He in male blood donors (r =-0.184, r =-0.221, r =-0.209). CONCLUSION: In blood donors with low C-reactive protein level, the lower the iron store capacity, the lower the iron utilization, and the platelet counts tends to rise.

We have previously reported that flavonoid extract from propolis (FP) can improve cardiac function in rats following myocardial infarction (MI). However, the mechanisms responsible for the cardioprotective effects of FP have not been fully elucidated. In the current study, we explored whether FP can reduce inflammatory cytokines and attenuate sympathetic nerve system activity and antiendoplasmic reticulum (ER) stress and whether the cardioprotective effects are related to peroxisome proliferator-activated receptor gamma (PPAR-γ) activation. Sprague Dawley rats were randomly divided into six groups: Sham group received the surgical procedure but no artery was ligated; MI group received ligation of the left anterior descending (LAD) branch of the coronary artery; MI + FP group received FP (12.5 mg/kg/d, intragastrically) seven days prior to LAD ligation; FP group (Sham group + 12.5 mg/kg/d, intragastrically); MI + FP + GW9662 group received FP prior to LAD ligation with the addition of a specific PPAR-γ inhibitor (GW9662), 1 mg/kg/d, orally); and MI + GW9662 group received the PPAR-γ inhibitor and LAD ligation. The results demonstrated that the following inflammatory markers were significantly elevated following MI as compared with expression in sham animals: IL-1ß, TNF-α, CRP; markers of sympathetic activation: plasma norepinephrine, epinephrine and GAP43, nerve growth factor, thyroid hormone; and ER stress response markers GRP78 and CHOP. Notably, the above changes were attenuated by FP, and GW9662 was able to alleviate the effect of FP. In conclusion, FP induces a cardioprotective effect following myocardial infarction by activating PPAR-γ, leading to less inflammation, cardiac sympathetic activity, and ER stress.

Gambogic acid (GA) is in a phase II clinical trial as an antitumor and antiangiogenesis agent. In this study, 36 GA derivatives were synthesized and screened in a zebrafish model to evaluate their antiangiogenic activity and toxicity. Derivatives 4, 32, 35, 36 effectively suppressed the formation of newly grown blood vessels and showed lower toxicities than GA as evaluated by zebrafish heart rates and mortalities. They also exhibited more potent migration and HUVEC tube formation inhibiting activities than GA. Among them, 36 was the most potent one, suggesting that it may serve as a potential new antiangiogenesis candidate with low toxicity. Additionally, 36 showed comparable antiproliferative activity to HUVECs and five tumor cell lines but low cytotoxicity to LO2 cells.

In the purpose of increasing incorporation efficiency and improving the release kinetics of plasmid DNA (pDNA) from poly(D,L-lactide-co-glycolide) (PLGA) nanoparticles, a facile method for the fabrication of calcium phosphate (CaPi) embedded PLGA nanoparticles (CaPi-pDNA-PLGA-NPs) was developed. The effect of several preparation factors on the particle size, incorporation efficiency, pDNA release and transfection efficiency in vitro was studied by Single Factor Screening Method. These preparation factors included the molecular weight (MW), hydrolysis degree (HD) of polyvinyl alcohol (PVA), sonication power and time, composition of organic phase, initial concentration of calcium phosphate and calcium (Ca) to phosphate ion (P) ratio (Ca/P ratio), etc. The CaPi-pDNA-PLGA-NPs made according to the optimal formulation were spherical in shape observed by transmission electron microscopy (TEM) with a mean particle size of 207±5 nm and an entrapment efficiency of 95.7±0.8%. Differential scanning calorimetry (DSC) suggested that there existed interaction between the DNA-calcium-phosphate (CaPi-pDNA) complexes and the polymeric matrices of PLGA. X-ray diffractometry (XRD) further proved the conclusion and indicated that the CaPi-pDNA was in weak crystallization form inside the nanoparticles. The Brunauer-Emmett-Teller (BET) surface area measurement demonstrated that the CaPi-pDNA-PLGA-NPs are mesoporous with specific surface area of 57.5m(2)/g and an average pore size of 96.5 Å. The transfection efficiency of the CaPi-pDNA-PLGA-NPs on human embryonic kidney 293 (HEK 293) cells in vitro was 22.4±1.2%, which was much higher than those of both the pDNA loaded PLGA nanoparticles (pDNA-PLGA-NPs) and the CaPi-pDNA embedded PLGA microparticles (CaPi-pDNA-PLGA-MPs). The CaPi-pDNA-PLGA-NPs are promising vectors for gene delivery.

RESUMO

There are two independent mol-ecules in the asymmetric unit of the title compound, C(20)H(17)ClO(3), each having an E configuration about the -C=C- bond. The dihedral angles between the two benzene rings in the two mol-ecules are 7.17 (11) and 9.82 (11)°. In both mol-ecules, the hy-droxy group is involved in an intra-molecular O-H⋯O hydrogen bond.

Human metapneumovirus (hMPV) has recently been recognized as an important respiratory pathogen, especially in children. At present, our understanding of the characteristics of hMPV from China is very limited. Nasopharyngeal aspirates were taken from 310 hospitalized pediatric patients. Twenty (6.5%) of them were infected with hMPV, and they all developed pneumonia. Sixty five percent (13/20) of the cases were under 12 months. Phylogenetic analysis of F gene fragments indicated that three sub-genotypes of hMPV(A2a/A2b, B1,B2) circulated in Tianjin and A2b was the predominant subtype. The Vero-E6 cell line was better than LLC-MK2 for hMPV isolation. Three hMPV strains were successfully isolated using the Vero-E6 cell line.

Using litter incubation experiment in laboratory, decomposition discrepancies of four typical litters from Zijin Mountain were analyzed. The results show that organic carbon mineralization rates of soil with litters all involve fast and slow decomposition stages, and the differences are that the former has shorter duration,more daily decomposition quantity while the latter is opposite. Organic carbon mineralization rates of soil with litters rapidly reached maximum in the early days of incubation, and the order is soil with Cynodon dactylon litter (CK + BMD) (23.88 +/- 0.62) mg x d(-1), soil with Pinus massoniana litter (CK+ PML) (17.93 +/- 0.99) mg x d(-1), soil with Quercus acutissima litter (CK+ QAC) (15.39 +/- 0.16) mg x d(-1) and soil with Cyclobalanopsis glauca litter (CK + CGO) (7.26 +/- 0.34) mg x d(-1), and with significant difference between each other (p < 0.05). This order has not significant correlation to litter initial chemical elements. The amount of organic carbon mineralized accumulation within three months incubation is (CK + BMD) (338.21 +/- 6.99) mg, (CK + QAC) (323.48 +/- 13.68) mg, (CK + PML) (278.34 +/- 13.91) mg and (CK + CGO) (245.21 +/- 4.58) mg. 198.17-297.18 mg CO2-C are released during litter incubation, which occupies 20.29%-31.70% of the total litter organic carbon amounts. Power curve model can describe the trends of organic carbon mineralization rate and mineralized accumulation amount,which has a good correlation with their change.

OBJECTIVE: To investigate the differences between the sequences of papA and papG of UPEC4030 strain and the related genes, to better understand the genetic variation of UPEC4030 papA and its combination forms with papG so as to identify if it was a new genotype. METHODS: Cloning and sequencing methods were used to analyze the sequences of papG and papA of UPEC4030 strain and to compare their related sequences. RESULTS: Through sequence analysis of papA, it was revealed that there was a 722 bp gene, encoding 192 amino acid polypeptide. The overall homology of the papA genes between UPEC4030 and the standard strains of ten F types were 36.11% - 77.95% and 22.20% - 78.34% at nucleotide and deduced amino acid levels. Homology between the sequences of reverse primers and the corresponding sequence of UPEC4030 papA was 10.00% - 66.67%. The results confirmed that UPEC4030 strain contained a novel papA variant. Through sequence analysis of UPEC4030 papG, we revealed a 1100 bp gene, encoding 337 amino acid polypeptide. The homology of the papG genes between UPEC4030 and UPEC IA2, the standard strain, was 99.00% at nucleotide level and 99.11% at deduced amino acid and UPEC4030 strain carried class II genotype of papG. CONCLUSION: UPEC4030 strain contained an unknown papA variant or the new genotype and carried class II genotype of papG. The pathogenic mechanism and epidemiology call for further study.

Klebsiella pneumoniae was cultured followed by the preparation and immunoactivity elucidating of its polysaccharide (CPS). The lysis of cell is the first key step in the preparation, under the co-action of trypsin, lysozyme and NP-40, the cell lysed within 2h, then the lysate was concentrated by ultrafiltration which serves as concentrating and partial purifying action simultaneously. Crude CPS was got by ethanol precipitation, then purified through the Ion-exchange and gel filtration, the purity of CPS was judged by the gel filtration and agarose gel electrophoresis. The effect of CPS on the cell immunoactivity was studied in detail, the results show that CPS possesses bidirectional immunoregulation on the spleen cells of mice, that is, low concentration of CPS can stimulate the immune response while the high concentration manifests the inhibition significantly. The investigation results will benefit on the exploitation of the CPS.

OBJECTIVE: To analyse the pathogen of child patients with influenza in Tianjin area. METHODS: The influenza virus isolation was performed by MDCK cells and embryonated eggs. The identification of the isolates was carried out with hemagglutination (HA) and hemagglutination inhibition (HI) tests. RESULTS: Two hundred and thirty-eight throat swab specimens from children with influenza-like illness were collected in Tianjin area from Oct. 2001 to Mar. 2002 and 64 strains (26.9%) of influenza virus were isolated. Data showed that there were 42 strains (65.6%) of A (H3N2) subtype, 13 strains (20.3%) of A (H1N1) subtype and 9 strains (14.1%) of B type in these positive isolates. All the isolated viruses grew very well in MDCK cells and hemagglutinated with human "O" red blood cells, and most (62/64 strains) of them were able to multiply in embryonated chick eggs. However, there were only 3 isolates with HA positive in inoculating embryonated eggs with the specimens. Meanwhile, it was revealed that out of 55 strains of A type viruses, 53 strains (96.4%) were from O to D phase, 2 strain of A (H3N2) were D phase characters and all B type isolated viruses being D phase properties. CONCLUSION: There were three endemic types of influenza viruses-A (H3N2), A (H1N1) and B type in Tianjin area, with A, the main type.
