RESUMO

Coxsackievirus B3 (CVB3) is the pathogen causing hand, foot and mouth disease (HFMD), which manifests across a spectrum of clinical severity from mild to severe. However, CVB3-infected mouse models mainly demonstrate viral myocarditis and pancreatitis, failing to replicate human HFMD symptoms. Although several enteroviruses have been evaluated in Syrian hamsters and rhesus monkeys, there is no comprehensive data on CVB3. In this study, we have first tested the susceptibility of Syrian hamsters to CVB3 infection via different routes. The results showed that Syrian hamsters were successfully infected with CVB3 by intraperitoneal injection or nasal drip, leading to nasopharyngeal colonization, acute severe pathological injury, and typical HFMD symptoms. Notably, the nasal drip group exhibited a longer viral excretion cycle and more severe pathological damage. In the subsequent study, rhesus monkeys infected with CVB3 through nasal drips also presented signs of HFMD symptoms, viral excretion, serum antibody conversion, viral nucleic acids and antigens, and the specific organ damages, particularly in the heart. Surprisingly, there were no significant differences in myocardial enzyme levels, and the clinical symptoms resembled those often associated with common, mild infections. In summary, the study successfully developed severe Syrian hamsters and mild rhesus monkey models for CVB3-induced HFMD. These models could serve as a basis for understanding the disease pathogenesis, conducting pre-trial prevention and evaluation, and implementing post-exposure intervention.

Background: Rhesus macaques and humans are closely related genetically and share similar physiological and pathological characteristics. Exploring the impact of diet on the early establishment of gut microbiota in non-human primates can provide relevant clinical models for healthy infant growth and development. At present, few writers have focused on the composition and changes of the intestinal microbes of infant rhesus macaques throughout their progression from birth to formula feeding after weaning. In this study, we used 16S rRNA sequencing technology to explore the composition of the intestinal flora of rhesus macaques at different ages and analyzed the trends in the microbial changes. Results: The results showed that the relative abundance of Bifidobacterium and Lactobacillus in the intestinal flora of infant rhesus macaques significantly decreased, and Prevotella increased with age. Bifidobacterium longum and Bifidobacterium breve are effective biomarkers to predict grouping. The metabolic pathways enriched in early life mainly concentrated in glycosphingolipid biosynthesis (lacto and neolacto series) and the degradation and metabolism of alcohols and esters. Conclusions: We found that age was an important factor that affected the changes in the intestinal flora. This study revealed the change trend of flora in breastfed and formula-fed infant rhesus monkeys in different growth months, and found that the dominant flora changed greatly. This research provides a medically relevant theoretical basis for understanding the healthy development of infants.

To explore the relationship between the changes in the physiological period and the fecal microbial population of female rhesus monkeys by measuring microbial composition of fecal samples and the serum hormones. Blood and fecal samples were collected from six female adult rhesus monkeys during the menstrual period (MP), ovulation period (OP), and Luteal period (LP). Serum estradiol (E2) and progesterone (P) levels were determined by the chemiluminescence method and the stool samples were subjected to high-throughput 16S rRNA sequencing. The highest level of E2 and P secretions were during the MP, and LP, respectively. Stool samples produced valid sequences and the number of operational taxonomic unit/OTU was: 810056/3756 (MP), 845242/4159 (OP), 881560/3970 (LP). At the phylum level, the three groups of Firmicutes and Bacteroides accounted for > 95%. The dominant flora at the LP was Bacteroides (53.85%), the dominant flora at the MP and OP was Firmicutes, 64.08 and 56.53%, respectively. At the genus level, the dominant genus at the LP was Prevotella, the dominant genera at the MP were Prevotella, Oncococcus, Streptococcus, and Kurtella. The dominant genera at OP were Prevotella and Nocococcus. At the phylum level, P levels were negatively correlated to Firmicutes, Actinomycetes Actinobacteria, and Fibrobacteres, but positively correlated to Bacteroidetes. Likewise, E2 was positively correlated to Proteobacteria but negatively correlated to Euryarchaeota. At the genus level, P hormone showed a significant correlation with 16 bacterial species, and E2 was significantly correlated to seven bacterial species. Function prediction analysis revealed a high similarity between the MP and OP with six differentially functional genes (DFGs) between them and 11 DFGs between OP and LP (P < 0.05). Fecal microbiota types of female rhesus monkeys varied with different stages of the menstrual cycle, possibly related to changes in hormone levels.

Coxsackievirus A10 (CV-A10) is one of the etiological agents associated with hand, foot and mouth disease (HFMD) and also causes a variety of illnesses in humans, including pneumonia, and myocarditis. Different people, particularly young children, may have different immunological responses to infection. Current CV-A10 infection animal models provide only a rudimentary understanding of the pathogenesis and effects of this virus. The characteristics of CV-A10 infection, replication, and shedding in humans remain unknown. In this study, rhesus macaques were infected by CV-A10 via respiratory or digestive route to mimic the HFMD in humans. The clinical symptoms, viral shedding, inflammatory response and pathologic changes were investigated in acute infection (1-11 day post infection) and recovery period (12-180 day post infection). All infected rhesus macaques during acute infection showed obvious viremia and clinical symptoms which were comparable to those observed in humans. Substantial inflammatory pathological damages were observed in multi-organs, including the lung, heart, liver, and kidney. During the acute period, all rhesus macaques displayed clinical signs, viral shedding, normalization of serum cytokines, and increased serum neutralizing antibodies, whereas inflammatory factors caused some animals to develop severe hyperglycemia during the recovery period. In addition, there were no significant differences between respiratory and digestive tract infected animals. Overall, all data presented suggest that the rhesus macaques provide the first non-human primate animal model for investigating CV-A10 pathophysiology and assessing the development of potential human therapies.

RESUMO

This paper presents a precise, stable, and invertible reflectance model for photometric stereo. This microfacet-based model is applicable to all types of isotropic surface reflectance, covering cases from diffusion to specular reflections. We introduce a single variable to physically quantify the surface smoothness, and by monotonically sliding this variable between 0 and 1, our model enables a versatile representation that can smoothly transform between an ellipsoid of revolution and the equation for Lambertian reflectance. In the inverse domain, this model offers a compact and physically interpretable formulation, for which we introduce a fast and lightweight solver that allows accurate estimations for both surface smoothness and surface shape. Finally, extensive experiments on the appearances of synthesized and real objects evidence that this model is state-of-the-art in our off-the-shelf solution.

Rhesus macaque is an important animal model in biomedical research, especially human disease, developmental, translational, and pre-clinical research. Blood physiological and biochemical parameters are important markers for physiology, pathology, and toxicology research. However, these parameters have not been systematically reported for Chinese rhesus macaques. To characterize the reference for these parameters, this study collected 1805 Chinese rhesus macaques living in Southwestern China. A total of 24 blood physiological indexes and 27 biochemical parameters were determined. Sex and age were found to affect these parameters. In conclusion, a comprehensive and systematic reference of hematological and biochemical parameters for Chinese rhesus macaque was established in this work on the basis of a large cohort. Such reference will benefit biomedical research employing rhesus macaques as animal models.

Calcitonin gene-related peptide (CGRP) has been implicated in multiple functions across many bioprocesses; however, whether CGRP is associated with severe traumatic brain injury (TBI) remains poorly understood. In this study, 96 adult patients with TBI (enrolled from September 2015 to December 2016) were divided into a mild/moderate TBI group (36 males and 25 females, aged 38 ± 13 years) and severe TBI group (22 males and 13 females, aged 38 ± 11 years) according to Glasgow Coma Scale scores. In addition, 25 healthy individuals were selected as controls (15 males and 10 females, aged 39 ± 13 years). Radioimmunoassay was used to detect serum levels of CGRP and endothelin-1 at admission and at 12, 24, 48, 72 hours, and 7 days after admission. CGRP levels were remarkably lower, but endothelin-1 levels were obviously higher in the severe TBI group compared with mild/moderate TBI and control groups. Levels of CGRP were remarkably lower, but endothelin-1 levels were obviously higher in deceased patients compared with patients who survived. Survival analysis and logistic regression showed that both CGRP and endothelin-1 levels were associated with patient mortality, with each serving as an independent risk factor for 6-month mortality of severe TBI patients. Moreover, TBI patients with lower serum CGRP levels had a higher risk of death. Thus, our retrospective analysis demonstrates the potential utility of CGRP as a new biomarker, monitoring method, and therapeutic target for TBI.

To build an ideal animal model for studying the mechanism of occurrence, developing and treating of diabetes become a more important issue, facing with the fact that the big threat of diabetes to human health has been worsen. First, we used the normal control diets or the high-fat/high-sucrose diets to feed the adult rhesus monkeys and the macaques induced by the high-fat/high-sucrose diets in the high-fat/high-sucrose group and the type 2 diabetes mellitus (T2DM) group developed the hyperglycemia, hyperinsulinemia at 6 months in accordance with the precious researches that reported that minipigs, rats and mice could develop hyperglycemia, hyperinsulinemia, hyperlipidemia and obesity after being induced with high-fat/high-carbohydrate diets. Second, the rhesus monkeys in T2DM group were injected STZ at a low dosage of 35 mg/kg BW to induce glucose persistent elevation which maintained pretty well after 12 months. Third, we took the assay of glucose tolerance test and insulin resistance index, assessed the changing tendency of serum resistin and analysed the pathological characteristics of the tissues like pancreas and liver by staining in different ways. The results indicate the rhesus monkeys in T2DM group have lots of clinical features of T2DM. The experimental non-genetic T2DM rhesus monkeys model not only contribute to simulating of clinical manifestations and pathological features of human T2DM, but also may be a good kind of model for research on the treatment of T2DM and for new drugs evaluation.

The present study was conducted to investigate the effects of dietary soybean isoflavones (SI) supplementation on growth performance, innate immune responses, hepatic antioxidant abilities, heat shock protein 70 (HSP70) gene expression and resistance to the pathogen Vibrio harveyi in Trachinotus ovatus. A basal diet was supplemented with SI at 0, 10, 20, 40, 60, 80 mg kg(-1) feed for 8 weeks. Significantly maximum weight gain (WG) and specific growth rate (SGR) were observed in treatment with 40 mg kg(-1) SI supplement (P < 0.05). Feed conversion ratio (FCR), feeding rate (FR) and survival rate were not significantly different among treatments. Fish fed a diet with 40 mg kg(-1) SI showed significant increase in plasma total protein content, complement 3 content, lysozyme activity as well as respiratory burst activity, but decrease in alanine aminotransferase and aspartate aminotransferase activities (P < 0.05). Increased plasma alkaline phosphatase activity, hepatic total antioxidative capacity, catalase activity and superoxide dismutase activity were also noticed in fish fed SI at 40 or 60 mg kg(-1) (P < 0.05). On the contrary, the lowest hepatic malondialdehyde (MDA) content was observed in fish fed SI at 40 mg kg(-1) (P < 0.05). Compared with the control, the relative level of HSP70 mRNA in fish fed SI at 40-80 mg kg(-1) were significantly increased, respectively (P < 0.05). After challenge with V. harveyi, significant higher post-challenge survival was observed in fish fed diets with 40-80 mg kg(-1) SI supplement than that in control group (P < 0.05). These results indicated that dietary intake containing SI could enhance the immune ability of fish and improve its resistance to infection by V. harveyi. Especially supplementation with 40 mg kg(-1) SI to the fish for 8 weeks showed remarkable improvement in the growth, non-specific immune responses, hepatic antioxidant abilities and HSP70 gene expression.

The spectral reflectance and water qualities of 27 stations were acquired in the lake Taihu during the months from May to August, 2008 when blue algal bloomed. Based on the fluorescence characteristics analysis of different chlorophyll a concentration, it was obvious that the position and height of fluorescence peak both have a positive correlation with chlorophyll a concentration, and the correlation coefficients between chlorophyll a concentration and position and half width of fluorescence peak are larger than those of the reference baseline and the normalized height of fluorescence. Estimating of chlorophyll a concentration in case 2 water using fluorescence characteristics is obviously better than the empirical algorithm based on blue to green ratio. Moreover, the common algorithm based on near infrared band to red band ratio is essentially consistent with the normalized height method.

OBJECTIVE: To determine the parameters of refining process of Flos Chrysanthemi extract with macroporous adsorption resin. METHODS: According to the chlorogenic acid and total flavonoids, to investigate the kind of macoroporous resin, the maximum quantity of physic liquor, and the type and quantity of elution solvent. Then combining with the retention rate of luteolin, to verify the parameters of the process. RESULTS: The maximum adsorption quantity of chlorogenic acid was 10.8 mg/g dry resin, and the elution solvent was 5-fold column volume of 80% alcohol; the purity of total flavonoids, chlorogenic acid and luteolin were 589.2%, 605.4%, 650.1%, respectively. CONCLUSION: The resin F can enrich the active components of Flos Chrysanthemi effectively.
