The presence of drugs in aquatic environments has been considered a global challenge and several remediation technologies have been proposed, including adsorption. In this study, new diclofenac adsorbents were obtained from the reaction of sodium magadiite (Na-Mag) with surfactants dodecylpyridinium chloride hydrate (C12pyCl) and hexadecylpyridinium chloride monohydrate (C16pyCl)), 1-hexadecyltrimethylammonium bromide (C16Br), and dodecyltrimethylammonium bromide (C12Br). The synthesis was carried out in the microwave at 50 °C for 5 min using surfactant amounts of 100% and 200% in relation to the cation exchange capacity of Na-Mag. The elemental analysis indicated that surfactants with a longer organic chain were more incorporated into Na-Mag, whose values were 1.42 and 1.32 mmol g-1 for C16pyMag200% and C16Mag200%, respectively. X-ray diffraction results suggested formation of intercalated products with basal space in the range of 2.81-4.00 nm. Diclofenac was quickly adsorbed on all organophilic magadiites, at an equilibrium time of 1 min. Drug capacity adsorption was influenced by the arrangement and packing density of organic cations, the basal distance, and the organic contents of the samples at high drug concentrations. Alkylpyridinium magadiites exhibited maximum adsorption capacities higher than alkylammonium magadiites, of 96.4, 100.7, 131.7, and 166.1 mg g-1 for C12pyMag100%, C12pyMag200%, C16pyMag100%, and C16pyMag200%, respectively, at pH 6.0 and 30 °C. Diclofenac removal by samples was not affected by the presence of ibuprofen, which was also removed from binary system by organophilic magadiites reaching removal of 76.5% and 86.9% by C16pyMag100% and C16pyMag200%, respectively. Regeneration studies demonstrated a drug removal percentage of 83-92% for C16pyMag and C16Mag after three cycles of adsorption.

Neurodegenerative diseases represent an increasingly burdensome challenge of the past decade, primarily driven by the global aging of the population. Ongoing efforts focus on implementing diverse strategies to mitigate the adverse effects of neurodegeneration, with the goal of decelerating the pathology progression. Notably, in recent years, it has emerged that the use of nanoparticles (NPs), particularly those obtained through green chemical processes, could constitute a promising therapeutic approach. Green NPs, exclusively sourced from phytochemicals, are deemed safer compared to NPs synthetized through conventional chemical route. In this study, the effects of green chemistry-derived silver NPs (AgNPs) were assessed in neuroblastoma cells, SHSY-5Y, which are considered a pivotal model for investigating neurodegenerative diseases. Specifically, we used two different concentrations (0.5 and 1 µM) of AgNPs and two time points (24 and 48 h) to evaluate the impact on neuroblastoma cells by observing viability reduction and intracellular calcium production, especially using 1 µM at 48 h. Furthermore, investigation using atomic force microscopy (AFM) unveiled an alteration in Young's modulus due to the reorganization of cortical actin following exposure to green AgNPs. This evidence was further corroborated by confocal microscopy acquisitions as well as coherency and density analyses on actin fibers. Our in vitro findings suggest the potential efficacy of green AgNPs against neurodegeneration; therefore, further in vivo studies are imperative to optimize possible therapeutic protocols.

Astrocytes are glial cells that play key roles in neuroinflammation, which is a common feature in diabetic encephalopathy and aging process. Metformin is an antidiabetic compound that shows neuroprotective properties, including in inflammatory models, but astroglial signaling pathways involved are still poorly known. Interferons α/ß are cytokines that participate in antiviral responses and the lack of their signaling increases susceptible to viral infections. Here, we investigated the effects of metformin on astrocytes from hypothalamus, a crucial brain region related to inflammatory processes. Astrocyte cultures were derived from interferon α/ß receptor knockout (IFNα/ßR-/-) and wild-type (WT) mice. Metformin did not change the expression of glial fibrillary acidic protein but caused an anti-inflammatory effect by decreasing pro-inflammatory cytokines (tumor necrosis factor-α and interleukin-1ß), as well as increasing gene expression of anti-inflammatory proteins interleukin-10 and Nrf2 (nuclear factor erythroid derived 2 like 2). However, nuclear factor κB p65 and cyclooxygenase 2 were downregulated in WT astrocytes and upregulated in IFNα/ßR-/- astrocytes. AMP-activated protein kinase (AMPK), a molecular target of metformin, was upregulated only in WT astrocytes, while sirtuin 1 increased in both mice models. The expression of inducible nitric oxide synthase was decreased in WT astrocytes and heme oxygenase 1 was increased in IFNα/ßR-/- astrocytes. Although loss of IFNα/ßR-mediated signaling affects some effects of metformin, our results support beneficial roles of this drug in hypothalamic astrocytes. Moreover, paradoxical response of metformin may involve AMPK. Thus, metformin can mediate glioprotection due its effects on age-related disorders in non-diabetic and diabetic encephalopathy individuals.

Astrocytes have key regulatory roles in central nervous system (CNS), integrating metabolic, inflammatory and synaptic responses. In this regard, type I interferon (IFN) receptor signaling in astrocytes can regulate synaptic plasticity. Simvastatin is a cholesterol-lowering drug that has shown anti-inflammatory properties, but its effects on astrocytes, a main source of cholesterol for neurons, remain to be elucidated. Herein, we investigated the effects of simvastatin in inflammatory and functional parameters of primary cortical and hypothalamic astrocyte cultures obtained from IFNα/ß receptor knockout (IFNα/ßR-/-) mice. Overall, simvastatin decreased extracellular levels of tumor necrosis factor-α (TNF-α) and interleukin-1ß (IL-1ß), which were related to a downregulation in gene expression in hypothalamic, but not in cortical astrocytes. Moreover, there was an increase in anti-inflammatory interleukin-10 (IL-10) in both structures. Effects of simvastatin in inflammatory signaling also involved a downregulation of cyclooxygenase 2 (COX-2) gene expression as well as an upregulation of nuclear factor κB subunit p65 (NFκB p65). The expression of cytoprotective genes sirtuin 1 (SIRT1) and nuclear factor erythroid derived 2 like 2 (Nrf2) was also increased by simvastatin. In addition, simvastatin increased glutamine synthetase (GS) activity and glutathione (GSH) levels only in cortical astrocytes. Our findings provide evidence that astrocytes from different regions are important cellular targets of simvastatin in the CNS, even in the absence of IFNα/ßR, which was showed by the modulation of cytokine production and release, as well as the expression of cytoprotective genes and functional parameters.

Road transport significantly contributes to climate change and air pollution. Efforts to reduce transport sector emissions include deploying battery electric vehicles and designing their powertrains for improved performance. The European H2020 funded Functionally Integrated E-axle Ready for Mass Market Third GENeration Electric Vehicles (FITGEN) developed a novel functionally integrated e-axle (the FITGEN e-axle) for electric vehicles. This paper presents the environmental performance of the FITGEN e-axle. Using the Life Cycle Assessment (LCA) methodology, the study compares the FITGEN e-axle to the 2018 State-of-the-Art (SotA) e-drive, besides diesel and petrol-fuelled powertrains. The FITGEN powertrain reduces climate impacts by 10 % and energy consumption by 17 %, compared with the 2018 SotA e-drive due to the efficiency improvements and components integration. It also outperforms the 2018 SotA e-drive in several other impact categories, such as human toxicity (4-10 %), land use (19 %), and mineral depletion (8 %). However, the FITGEN powertrain only outperforms diesel and petrol powertrains in climate change and fossil resource scarcity impact categories. These findings imply that more efforts are required to improve the environmental profile of electric powertrains. Metal mining and production, especially for copper and aluminium, are critical for toxicity impacts. The sensitivity analysis demonstrates the robustness of the results, with no significant shift in their ranking order. The following aspects should be considered to improve the performance of electric powertrains from a life cycle perspective: improvement of components efficiency, reduced use of electronics and component integration, and use of low-carbon energy mix from their metal mining sites to production and use.

Introdução: O tumor desmoide ou fibromatose agressiva é um tumor de incidência rara (0,03% dos tumores, cerca de dois a cinco casos por milhão de pessoas ao ano) formado por reação inflamatória com proliferação de fibroblastos. Objetivo: O objetivo deste estudo foi identificar as características sociodemográficas e epidemiológicas dos pacientes com tumor desmoide em um grupo de apoio de uma mídia social. Métodos: Trata-se de um estudo transversal, descritivo-exploratório, com abordagem quantitativa, no qual foi aplicado aos pacientes diagnosticados com tumor desmoide de um grupo de apoio de mídia social, um questionário via formulário do Google com questões elaboradas pelas autoras voltadas para a definição do perfil sociodemográfico dos pacientes e para as informações sobre a sua percepção a respeito do diagnóstico, tratamento e acompanhamento do tumor desmoide, após aceite on-line do Termo de Consentimento Livre e Esclarecido e aprovação do Comitê de Ética e Pesquisa. Resultados: Foram entrevistados 173 pacientes com diagnóstico de tumor desmoide de todo o Brasil, com representação de todas as regiões, sendo a maioria dos casos (60%) após algum evento como traumas no local e alterações hormonais como as relacionadas com a gravidez, e 38% com localização na parede abdominal e cerca de 62% foram abordados cirurgicamente no mínimo 01 vez, com recidiva em cerca de 45% dos casos. Conclusão: Este tumor apresenta características desafiadoras para seu diagnóstico, tratamento e acompanhamento. Neste sentido, este estudo pode identificar o desafio quanto à escolha terapêutica no sentido de maior qualidade de vida aos indivíduos, bem como da qualificação das equipes de saúde para uma abordagem mais efetiva clínica e terapeuticamente mais humana


Introduction: Desmoid tumor or aggressive fibromatosis is a tumor of rare incidence (0.03% of tumors, about two to five cases per million people per year) formed by inflammatory reaction with proliferation of fibroblasts. Objective: The aim of this study was to identify the sociodemographic and epidemiological characteristics of patients with desmoid tumor in a social media support group. Methods: This is a cross-sectional, descriptive-exploratory study, with a quantitative approach, in which it was applied to patients diagnosed with desmoid tumor of a social media support group, a questionnaire via Google form with questions prepared by the authors aimed at defining the sociodemographic profile of patients and information about their perception of the diagnosis, treatment and monitoring of the desmoid tumor, after online acceptance of the Informed Consent Form and approval of the Ethics and Research Committee. Results: We interviewed 173 patients diagnosed with desmoid tumor from all over Brazil, representing all regions, with the majority of cases (60%) after some event such as trauma at the site and hormonal changes such as those related to pregnancy, and 38% with location in the abdominal wall and about 62% were surgically approached at least 01 time, with recurrence in about 45% of cases. Conclusion: This tumor presents challenging characteristics for its diagnosis, treatment and follow-up. In this sense, this study can identify the challenge regarding therapeutic choice in the sense of higher quality of life to individuals, as well as the qualification of health teams for a more effective clinical and therapeutically more humane approach

Resultados: Fueron entrevistados 173 pacientes con diagnóstico de tumor demoidal de todo Brasil, representando todas las regiones, con la mayoría de los casos (60%) después de algún evento como trauma local y cambios hormonales como los relacionados con el embarazo, y 38% con localización en el pared abdominal y alrededor del 62% fueron abordados quirúrgicamente al menos 01 vez, con recurrencia en alrededor del 45% de los casos. Conclusión: Este tumor presenta características desafiantes para su diagnóstico, tratamiento y seguimiento. En ese sentido, este estudio puede identificar el desafío en cuanto a la elección terapéutica en el sentido de mayor calidad de vida de los individuos, así como la capacitación de los equipos de salud para un abordaje clínica y terapéuticamente más eficaz y más humano

Decentralized energy systems enable a higher integration of electricity generation by renewable energy sources supported by electric storage and may significantly reduce greenhouse gas emissions for electricity generation. While the environmental impact of single technologies has received great attention in recent years, the environmental impacts of decentralized energy generation and storage technologies remain unaddressed. This study presents a cradle-to-grave life cycle assessment of those technologies in Belgium for 2030 and 2050. The system technologies comprise single-Si photovoltaic installations combined with lithium-ion and second-life batteries. To compile the life cycle inventory (LCI), energy balances are built based on a Belgian impact energy model. The flexibility of the energy system is introduced by different EV charging strategies and distinct modes of stationary battery storage with the Belgium electricity grid, represented by four different scenarios: i) low flexibility, ii) medium flexibility, iii) high flexibility, and iv) high flexibility with high prosumer potential (PPH). The midpoint impact categories climate change, land use, mineral resource scarcity and terrestrial ecotoxicity of ReCiPe life cycle impact assessment method are analyzed. The decentralized energy generation and storage technologies in Belgium in 2050 result in 64.51 gCO2eq/kWh of consumed electricity for the medium flexibility scenario, representing a 72 % decrease compared to 2014. However, these reductions are driven by changes in the national electricity mix. Land use impacts are also reduced, up to 72 % for the high flexibility PPH scenario. In contrast, mineral resource scarcity and terrestrial ecotoxicity rise over time in the high flexibility PPH scenario in 2050 to 46 % and 66 %, respectively. A perturbation analysis is conducted to assess the sensitivity of the results, showing solar irradiation as the most sensitive parameter. One way to further reduce the environmental impacts of decentralized energy systems could be to investigate new strategies for the end-of-life of photovoltaic installations and batteries.

The paper features a computational fluid dynamics study of a flapping NACA0015 hydrofoil moving with a combination of sinusoidal heaving and pitching. Several kinematic configurations are explored, varying sequentially pitch and heave amplitude, Strouhal number and phase angle, in an attempt to determine the influence of each parameter on the propulsive performance. To optimize efficiency the angle of attack should assume the highest value that also avoids the arise of the leading edge vortex generated in the dynamic stall state. At low Strouhal number optimum is reached at high heave amplitudes, which correspond to the configurations minimizing the hysteresis in the (Cy,Cx) plane. The same outcome in terms of hysteresis minimization has been verified to occur when optimal phase shift was considered. Differently, when the Strouhal number and the angle of attack become higher, to exploit efficiently the lift increment owed to dynamic stall it emerged the necessity of adopting low heave amplitude to improve separation resistance, avoiding the occurrence of deep stall.

Over the last decade, education has been evolving to equip students with the fundamental skills required to cope with the challenges of sustainability and inclusivity, such as quality education, access to clean water, cultural heritage preservation and protection of marine life. Technology supports the learning process by providing useful tools that enrich the learning environment, encourage active participation, improve collaboration and prepare students for their future life. Educational Robotics is one of the most popular innovative methodologies that supports the development of many skills by assembling and programming robots in a meaningful way. In this paper, the authors aim at advancing their previous work in the field of Educational Robotics applied to the marine environment by proposing a novel bioinspired educational toolkit whose design and features support activities concerning sustainability, ocean literacy, as well as STEM subjects in kindergarten through to grade twelve education. Exploiting the established educational theories and methodologies underpinning Educational Robotics, the toolkit allows for marine-themed activities, as well promoting activities concerning STEM subjects. To explain the relevance of the toolkit, the authors present the robot design, the workshops that every teacher or student can explore as an Open Educational Resource (OERs), and the results of a case study. Interestingly, the latter shows that the use of the toolkit seems to have complemented the students' initial keen interest in technology itself, with awareness about urgent issues related to the climate and the environment.

INTRODUCTION: The Brazilian Air Force (BAF) personnel must be prepared to perform their professional activities under the worst conditions. This preparation goes beyond habits of practicing physical activity, since it is necessary to perform specific physical tasks, referred to as "combat tasks" (CTs). This study aimed to investigate a combination of specific physical tests (SPTs) for predicting physical performance on simulated tasks (STs) that mimicked the performance of CTs. MATERIALS AND METHODS: Thirty infantry cadets from the BAF took part in anthropometric assessments, 11 SPTs, and 3 STs, during 7 testing days. Bivariate Pearson's correlation was used to determine linear relationships between SPT and ST results, and multiple linear regression models were used to identify test batteries that significantly predicted performance on STs. The level of significance was set at 5%. The study was conducted according to the guidelines of the Declaration of Helsinki and approved by the Ethics Committee of the BAF (protocol code 15796819.4.0000.5250, date of approval September 25, 2019). RESULTS: The greatest predictive power was obtained by the test battery that consisted of sprint-drag-carry, leg tucks, and handgrip strength (R2 = 0.56, P < .01). Conversely, the test battery comprised of push-ups, sit-ups, and 12-minute run (which represents the conventional physical test of the BAF), which presented the lowest predictive power (R2 = 0.14, P < .05). CONCLUSIONS: In conclusion, this study identified a test battery for predicting performance on the following STs: foot march, casualty drag, and move under direct fire. This finding represents the first step to improve the reliability of the BAF physical assessments, focusing on combat readiness levels.

Worldwide, forests are susceptible to fire. Forests with fire and selective logging interactions require monitoring and evaluation. This study evaluated the phytosociology and dynamics of tree vegetation in a disturbed forest (DF) and an undisturbed forest (UF) in selective logging areas affected by fire, in the Brazilian East Amazon. All trees with DBH ≥ 5 cm were measured and identified botanically in 93 plots (5 X 50 m) in the DF area and 58 plots (5 X 50 m) in the UF area, in 2010 (before logging), 2011, 2015 and 2017 (two years after the fire). Analysis of species and tree composition, diversity, similarity, mortality and recruitment were carried out. The fire affected the DF and UF areas in a similar proportion in terms of trees loss and basal area, intensifying the mortality rate. In the short term (2 years), the fire did not cause a significant reduction in species diversity, but there was a tendency towards a similarity loss in species composition in the area disturbed by logging. Subsequent assessments are necessary to understand the forest's recovery mechanisms.

The environmental performance of battery electric vehicles (BEVs) is influenced by their battery size and charging electricity source. Therefore, assessing their environmental performance should consider changes in the electricity sector and refurbishment of their batteries. This study conducts a scenario-based Life Cycle Assessment (LCA) of three different scenarios combining four key parameters: future changes in the charging electricity mix, battery efficiency fade, battery refurbishment, and recycling for their collective importance on the life-cycle environmental performance of a BEV. The system boundary covers all the life-cycle stages of the BEV and includes battery refurbishment, except for its second use stage. The refurbished battery was modelled considering refurbished components and a 50% cell conversation rate for the second life of 5 years. The results found a 9.4% reduction in climate impacts when future changes (i.e., increase in the share of renewable energy) in the charging electricity are considered. Recycling reduced the BEV climate impacts by approximately 8.3%, and a reduction smaller than 1% was observed for battery refurbishment. However, the battery efficiency fade increases the BEV energy consumption, which results in a 7.4 to 8.1% rise in use-stage climate impacts. Therefore, it is vital to include battery efficiency fade and changes to the electricity sector when estimating the use-stage impacts of BEVs; without this, LCA results could be unreliable. The sensitivity analysis showed the possibility of a higher reduction in the BEV climate impacts for longer second lifespans (>5 years) and higher cell conversation rates (>50%). BEV and battery production are the most critical stages for all the other impact categories assessed, specifically contributing more than 90% to mineral resource scarcity. However, recycling and battery refurbishment can reduce the burden of the different impact categories considered. Therefore, manufacturers should design BEV battery packs while considering recycling and refurbishment.

OBJECTIVES: The World Health Organization has recommended social isolation to prevent the transmission of COVID-19. Thus, feelings of loneliness, sedentary behavior, and predisposition to falls have been reported more often due to the adoption of social isolation, especially for older adults. The objective of this study was to compare older adults' loneliness, sedentary behavior, and occurrence of falls before and during social isolation due to the pandemic as well as to analyze the association of loneliness with sedentary lifestyle and falls in older adults. METHOD: Retrospective analytical study conducted through an online survey with older adults from Brazilian states in social isolation, approved by the Research Ethics Committee (number 32168920.0.0000.0068). RESULTS: There was a significant increase in loneliness and sedentary behavior during social isolation (p-value < 0.05 for both), but no increase was observed for falls (p-value = 0.615). There was no correlation between the outcomes, nor was there a correlation between the outcomes and the number of days in social isolation. CONCLUSION: The results of this research show that adoption of social isolation due to the COVID-19 pandemic brought an increase in sedentary behavior and loneliness for older adults but had no effect on the number of falls.

BACKGROUND: Frailty marks a process of increasing dysregulation of physiological systems which increases the risk of adverse health outcomes. This study examines the hypothesis that the association between multiple cardiovascular risk factors (CVRF) and cardiovascular diseases (CVD) becomes stronger with increasing frailty severity. METHODS: Cross-sectional analysis of 339 older adults (55.2% women; aged 75.2 ±â€¯9.1 years) from an outpatient geriatric clinic from a middle-income country. The frailty index (FI) was calculated as the proportion of 30 possible health deficits. We assessed hypertension, diabetes, obesity, dyslipidemia, sedentarism and smoking as CVRF (determinants) and myocardial infarction, stroke, heart failure as CVD. Poisson regression models adjusted for age, sex, and education was applied to estimate the association between frailty as well as CVRF (independent variables) with CVD (dependent variable). RESULTS: Of the 339 patients, 18,3% were frail (FI ≥ 0.25) and 32.7% had at least one CVD. Both frailty and CVRF were significantly associated with CVD (PR = 1.03, 95% CI 1.01 to 1.05; p = 0.001, and PR = 1.46, 95% 1.24 to 1.71; p < 0.001, respectively) adjusted for covariates. The strength of the association between CVRF and CVD decreased with increasing frailty levels, as indicated by a significant interaction term of frailty and CVRF (p < 0.001). CONCLUSION: Frailty and CVRF are both associated with CVD, but the impact of CVRF decreases in the presence of frailty. When confirmed in longitudinal studies, randomized controlled trials or causal inference methods like Mendelian randomization should be applied to assess whether a shift from traditional CVRF to frailty would improve cardiovascular outcome in the oldest old.

BACKGROUND: To investigate whether an exercise intervention using the VIVIFRAIL© protocol has benefits for inflammatory and functional parameters in different frailty status. METHODS/DESIGN: This is a randomized clinical trial in an outpatient geriatrics clinic including older adults ≥60 years. For each frailty state (frail, pre-frail and robust), forty-four volunteers will be randomly allocated to the control group (n = 22) and the intervention group (n = 22) for 12 weeks. In the control group, participants will have meetings of health education while those in the intervention group will be part of a multicomponent exercise program (VIVIFRAIL©) performed five times a week (two times supervised and 3 times of home-based exercises). The primary outcome is a change in the inflammatory profile (a reduction in inflammatory interleukins [IL-6, TNF- α, IL1beta, IL-17, IL-22, CXCL-8, and IL-27] or an increase in anti-inflammatory mediators [IL-10, IL1RA, IL-4]). Secondary outcomes are change in physical performance using the Short Physical Performance Battery, handgrip strength, fatigue, gait speed, dual-task gait speed, depressive symptoms, FRAIL-BR and SARC-F scores, and quality of life at the 12-week period of intervention and after 3 months of follow-up. DISCUSSION: We expect a reduction in inflammatory interleukins or an increase in anti-inflammatory mediators in those who performed the VIVIFRAIL© protocol. The results of the study will imply in a better knowledge about the effect of a low-cost intervention that could be easily replicated in outpatient care for the prevention and treatment of frailty, especially regarding the inflammatory and anti-inflammatory pathways involved in its pathophysiology. OBJECTIVES: This study aimed to investigate whether frailty could be an explanatory factor of the association between depression and the number of geriatric syndromes. METHODS: Cross-sectional baseline data from a cohort study (MiMiCS-FRAIL) were analyzed in a sample of 315 older adults. Depression was measured according to DSM-5 criteria and a self-report questionnaire (PHQ-9). Frailty was assessed according to the FRAIL questionnaire and a 30-item Frailty Index (FI). We considered six geriatric syndromes. Multiple linear regression analyses were performed and adjusted for potential confounders. RESULTS: Multiple linear regression analyses yielded significant associations between depression and geriatric syndromes. These associations decreased substantially in strength when frailty was added to the models. Findings were consistent for different definitions of depression and frailty. CONCLUSIONS: Among depressed patients, frailty may be hypothesized as a causal pathway toward adverse health outcomes associated with depression. Longitudinal studies should explore the causality of this association. CLINICAL IMPLICATIONS: Frailty should be treated or prevented in order to minimize the impact of other geriatric syndromes among depressed older adults. Screening for frailty would be of utmost importance in mental health care, as frailty is neglected especially in this field. Integrated care models are crucial for clinical practice in mental illness care.

BACKGROUND: To explore the mutual relationship between multimorbidity, mental illness and frailty, we have set-up the Multimorbidity and Mental health Cohort Study in FRAILty and Aging (MiMiCS-FRAIL) cohort. At the population level, multimorbidity, frailty and late-life depression are associated with similar adverse outcomes (i.e. falls, disability, hospitalization, death), share the same risk factors, and partly overlap in their clinical presentation. Moreover, these three variables may share a common underlying pathophysiological mechanism like immune-metabolic dysregulation. The overall objectives of MiMiCS-FRAIL are 1) to explore (determinants of) the cross-sectional and longitudinal relationship between multimorbidity, depression, and frailty among non-demented geriatric outpatients; 2) to evaluate molecular levels of senoinflammation as a broad pathophysiological process underlying these conditions; and 3) to examine adverse outcomes of multimorbidity, frailty and depression and their interconnectedness. METHODS: MiMiCS-FRAIL is an ongoing observational cohort study of geriatric outpatients in Brazil, with an extensive baseline assessment and yearly follow-up assessments. Each assessment includes a comprehensive geriatric assessment to identify multimorbidity and geriatric syndromes, a structured psychiatric diagnostic interview and administration of the PHQ-9 to measure depression, and several frailty measures (FRAIL, Physical Phenotype criteria, 36-item Frailty Index). Fasten blood samples are collected at baseline to assess circulating inflammatory and anti-inflammatory cytokines, leukocytes' subpopulations, and to perform immune-metabolic-paired miRome analyses. The primary outcome is death and secondary outcomes are the number of falls, hospital admissions, functional ability, well-being, and dementia. Assuming a 5-year mortality rate between 25 and 40% and a hazard rate varying between 1.6 and 2.3 for the primary determinants require a sample size between 136 and 711 patients to detect a statistically significant effect with a power of 80% (beta = 0.2), an alpha of 5% (0.05), and an R2 between the predictor (death) and all covariates of 0.20. Local ethical board approved this study. DISCUSSION: Frailty might be hypothesized as a final common pathway by which many clinical conditions like depression and chronic diseases (multimorbidity) culminate in many adverse effects. The MiMiCS-FRAIL cohort will help us to understand the interrelationship between these variables, from a clinical perspective as well as their underlying molecular signature.

Bio-inspired solutions devised for autonomous underwater robots are currently being investigated by researchers worldwide as a way to improve propulsion. Despite efforts to harness the substantial potential payoffs of marine animal locomotion, biological system performance still has far to go. In order to address this very ambitious objective, the authors of this study designed and manufactured a series of ostraciiform swimming robots over the past three years. However, the pursuit of the maximum propulsive efficiency by which to maximize robot autonomy while maintaining acceptable maneuverability ultimately drove us to improve our design and move from ostraciiform to carangiform locomotion. In order to comply with the tail motion required by the aforementioned swimmers, the authors designed a transmission system capable of converting the continuous rotation of a single motor in the travelling wave-shaped undulations of a multijoint serial mechanism. The propulsive performance of the resulting thruster (i.e., the caudal fin), which constitutes the mechanism end effector, was investigated by means of computational fluid dynamics techniques. Finally, in order to compute the resulting motion of the robot, numerical predictions were integrated into a multibody model that also accounted for the mass distribution inside the robotic swimmer and the hydrodynamic forces resulting from the relative motion between its body and the surrounding fluid. Dynamic analysis allowed the performance of the robotic propulsion to be computed while in the cruising condition.

Fungi of the genus Trichoderma are important microorganisms used in biocontrol processes and the promotion of plant development. However, they remain poorly studied in the context of forestry programs, especially those related to native Amazonian species. Thus, it is the aim of this study to evaluate the effects of different Trichoderma isolates on the germination and development of Handroanthus serratifolius seedlings. During in vitro germination tests, seeds were immersed for 24 h in respective fungal suspensions each prepared using one of five Trichoderma isolates. The suspensions were held in plastic trays and kept at a temperature of 24 ± 2 °C. Metrics related to germination and development assessed under laboratory conditions include: germination speed index (GSI), germination percentage, length of the roots, and hypocotyls, as well as fungal perseverance. In the nursery, Trichoderma were used in two different applications: pre-planting treatment and as a monthly, post-planting treatment. Pre-planting treatments consisted of 10 g of colonized rice grains bearing each isolate being placed into experimental bags five days before seeding. The post-planting treatment involved the application of 10 mL of fungal suspension per experimental bag. Each month, the height, stem diameter, and leaf number were measured for each seedling. At the end of the experiment, the length and mass of roots as well as the total dry mass were recorded. In laboratory conditions, seeds treated with Trichoderma asperellum -TAM03 obtained the greatest fractional germination (76.5%) and GSI. In the nursery experiments, isolates TAM01 and TAM03, when applied as a post-planting treatment, increased the height, stem diameter, and number of leaves of treated plants with respect to the control group by 180 days post-treatment. After 365 days, plants which received TAM01 pre-planting treatments were observed to have increased root and aerial part length, as well as root mass and overall dry mass. These results suggest that T. asperellum -TAM01 positively affects H. serratifolius development.

This systematic review and meta-analysis examines how exercise modifies brain and behavior in healthy mice, dementia (D) and Parkinson disease (PD) models. A search was performed on the Medline and Scopus electronic databases (2008-2019). Search terms were "mice", "brain", "treadmill", "exercise", "physical exercise". In the total, 430 were found but only 103 were included. Animals n = 1,172; exercised 4-8 weeks (Range 24 h to 32 weeks), 60 min/day (Range 8-120 min per day), and 10/12 m/min (Range 0.2 m/min to 36 m/min). Hippocampus, cerebral cortex, striatum and whole brain were more frequently investigated. Exercise improved learning and memory. Meta-analysis showed that exercise increased: cerebral BDNF in health (n = 150; z = 5.8, CI 3.43-12.05; p < 0.001 I2 = 94.3 %), D (n = 124; z = 4.18, CI = 2.22-9.12; p < 0.001; I2 = 93.7 %) and PD (n = 16 z = 4.26, CI 5.03-48.73 p < 0.001 I2 = 94.8 %). TrkB improved in health (n = 84 z = 5.49, CI 3.8-17.73 p < 0.001, I2 = 0.000) and PD (n = 22; z = 3.1, CI = 2.58-67.3, p < 0.002 I2 = 93.8 %). Neurogenesis increased in health (n = 68; z = 7.08, CI 5.65-21.25 p < 0.001; I2 17.58) and D model (n = 116; z = 4.18, CI 2.22-9.12 p < 0.001 I2 93.7 %). Exercise augmented amyloid clearance (n = 166; z = 7.51 CI = 4.86-14.85, p < 0.001 I2 = 58.72) and reduced amyloid plaques in D models (n = 49; z = 4.65, CI = 3.94-15.3 p < 0.001 I2 = 0.000). In conclusion, exercise improved brain and behavior, neurogenesis in healthy and dementia models, reduced toxicity and cerebral amyloid. Evidence regarding inflammation, oxidative stress and energy metabolism were scarce. Studies examining acute vs chronic exercise, extreme training and the durability of exercise benefit were rare. Vascular or glucose metabolism changes were seldom reported.
