Bruchine beetles are highly host-specific seed feeders during the larval stage. Although some specific parasitoid families have been recorded attacking bruchine beetles, most studies have been done at small spatial scales. Therefore, the current knowledge about the diversity and the geographic distribution of parasitoid species parasitizing bruchines is scarce, especially at a wide geographic area that extends over large distances through a latitudinal cline (i.e. large-scale spatial structure). The present study determined the species richness and evenness of parasitoids attacking the bruchine beetle Acanthoscelides macrophthalmus feeding on Leucaena leucocephala seeds, examined their geographic distribution, and characterized the large-scale spatial structure in parasitoid species composition. A total of 1420 parasitoids (all Hymenoptera) belonging to four families, five subfamilies and eight species were collected (genera: Horismenus, Paracrias, Urosigalphus, Stenocorse, Chryseida, Eupelmus). Most parasitoid species showed wide spatial distribution, high evenness in species abundance and the species richness estimators were close to stabilization (approximately eight species). Overall, greater similarity was observed in the species composition of plant populations near to each other than those farther apart, revealing a large-scale spatial structure in parasitoid species composition.

Lecanodiaspis dendrobii Douglas, 1892 (Hemiptera: Coccoidea: Lecanodiaspididae) and the associated parasitoid Cephaleta sp. (Hymenoptera: Pteromalidae) are reported for the first time in Brazil. Specimens of this scale insect were collected on branches and stems of Acacia mangium Willd., Leucaena leucocephala (Lam.) de Wit (Fabaceae), Morus nigra L. (Moraceae), Citrus reticulata Blanco (Rutaceae), Tectona grandis L. f. (Verbenaceae), Anacardium occidentale (Anacardiaceae), Annona squamosa L. and Xylopia aromatica (Lam.) Mart. (Annonaceae), in three municipalities of the Roraima state. All plants here mentioned are recorded for the first time as a host for L. dendrobii. Morphological characters of L. dendrobii and symptoms presented by the host plants infested by this pest are included in this work.

Resumo Lecanodiaspis dendrobii Douglas, 1892 (Hemiptera: Coccoidea: Lecanodiaspididae) e o parasitoide associado Cephaleta sp. (Hymenoptera: Pteromalidae) são registrados pela primeira vez no Brasil. Exemplares dessa cochonilha foram coletados sobre ramos e fustes de Acacia mangium Willd., Leucaena leucocephala (Lam.) de Wit (Fabaceae), Morus nigra L. (Moraceae), Citrus reticulata Blanco (Rutaceae), Tectona grandis L.f. (Verbenaceae), Anacardium occidentale (Anacardiaceae), Annona squamosa L. e Xylopia aromatica (Lam.) Mart. (Annonaceae), em três municípios do Estado de Roraima. Todas as plantas aqui mencionadas são registradas pela primeira vez como hospedeiras de L. dendrobii. Caracteres morfológicos da cochonilha e sintomas das plantas hospedeiras infestadas por esta praga são incluídos neste trabalho.

Current knowledge on coccinellids is primarily focused on their role as natural enemies of soft-bodied insects. However, there is a great diversity of coccinellid parasitoid species that are less studied. Here, we describe new records of coccinellid parasitoids with emphasis on new host-parasitoid interactions in 11 sample sites in Brazil. We collected 122 coccinellid individuals parasitized by six species of parasitoids in the Cerrado and in the Atlantic Rainforest biomes. New records of coccinellid parasitoids and host associations, expansion of habitat ranges and interactions are discussed focusing on the lack of basic information on these interactions in Brazil.

Collections of natural enemies of Bemisia tabaci biotype B (Genn.) (Hemiptera: Aleyrodidae) were made in Lavras, state of Minas Gerais, Brazil. In the greenhouse, 6,495 predators and 16,628 parasitoids belonging to three families were collected. In the field, 267 predators and 344 parasitoids belonging to five families were found. For the first time in Brazil, five species of predators associated with this whitefly were reported. Because of the diversity of natural enemies of B. tabaci biotype B recorded, this study points out the importance of these data for studies on integrated pest management.

The accuracy of two different methods in measuring the ammonia nitrogen (N-NH3) concentration in rumen fluid were evaluated: a catalyzed indophenol colorimetric reaction (CICR) and the Kjeldahl distillation (KD). Five buffered standard solutions containing volatile fatty acids, true protein, and known ammonia concentrations (0, 3, 6, 12, and 24 N-NH3 mg/dL) were used to simulate rumen fluid. Different ratios (10:1, 7.5:1, 5:1, 2.5:1, 1:1, 1:2.5, 1:5, 1:7.5, and 1:10) of a potassium hydroxide solution (KOH, 2 mol/L) to standard solutions were evaluated by the KD method. The accuracy of each method was evaluated by adjusting a simple linear regression model of the estimated N-NH3 concentrations on the N-NH3 concentrations in the standard solutions. When the KD method was used, N-NH3 was observed to be released from the deamination of true protein (P<0.05), and an incomplete recovery of N-NH3 was observed (P<0.05), except for 7.5:1 and 5:1 ratios of KOH solution to standard solutions (P>0.05). The estimates of the N-NH3 concentration obtained by the CICR method were found to be accurate (P>0.05). After the accuracy evaluation, ninety-three samples of rumen fluid were evaluated by the CICR and KD methods (using the 5:1 ratio of KOH solution to rumen fluid sample), assuming that the CICR estimates would be accurate. The N-NH3 concentrations obtained by the two methods were observed to be different (P<0.05) but strongly correlated (r = 0.9701). Thus, it was concluded that the estimates obtained by the Kjeldahl distillation using a 5:1 ratio of KOH solution to rumen fluid sample can be adjusted to avoid biases. Furthermore, a model to adjust the N-NH3 concentration is suggested.
Avaliou-se a exatidão de dois métodos para mensuração da concentração de nitrogênio amoniacal (N-NH3) em fluido ruminal: reação colorimétrica catalisada por indofenol (RCCI) e destilação de Kjeldahl (DK). Cinco soluções-tampão contendo ácidos graxos voláteis, proteína verdadeira e concentrações conhecidas de amônia (0, 3, 6, 12 e 24 N-NH3mg/dL) foram utilizadas para simular o fluido ruminal. Diferentes relações (10:1; 7,5:1; 5:1; 2,5:1; 1:1; 1:2,5; 1:5; 1:7,5 e 1:10) entre a solução de hidróxido de potássio (KOH, 2 mol/L) e as soluções-padrão foram avaliadas no método DK. A exatidão de cada método foi avaliada por intermédio do ajustamento de um modelo de regressão linear simples das concentrações estimadas de N-NH3sobre as concentrações reais de N-NH3nas soluções-padrão. Com a utilização da DK observou-se que N-NH3foi liberada a partir da deaminação de proteína verdadeira (P<0,05). Adicionalmente, a utilização da DK promoveu recuperação incompleta da N-NH3(P<0,05), com exceção das razões entre a solução de KOH e as soluções-padrão de de 7,5:1 e de 5:1 (P>0,05). As estimativas de concentração de amônia obtidas por intermédio do método de RCCI foram exatas (P>0,05). Após a avaliação da exatidão, noventa e três amostras de fluido ruminal foram avaliadas por intermédio dos métodos de RCCI e de DK (neste caso usando a razão entre solução de KOH e fluido ruminal de 5:1). Nesta avaliação assumiu-se que as estimativas obtidas com a RCCI seriam exatas. Observou-se que as concentrações de N-NH3obtidas pelos dois métodos foram diferentes (P<0,05), mas fortemente correlacionadas (r = 0,9701). Concluiu-se que as estimativas obtidas com a DK utilizando-se razão entre a solução de KOH e a amostra de fluido ruminal de 5:1 poderiam ser ajustadas para correções de vieses Um modelo para realização desse ajustamento foi sugerido.

Some parasitoids build a cocoon mass that hangs in the host body until the adults emergence, which is an advantage against attack by predators who troll the vegetation in search of prey. However, such behaviour is not effective against the hyperparasitoid attacks. This study reports the interaction between the caterpillar Manduca sexta Linnaeus, 1763 (Lepidoptera, Sphingidae) parasitised by Meteorus sp. (Hymenoptera, Braconidae) larvae and its hyperparasitoid Toxeumella albipes (Hymenoptera, Pteromalidae). This is the first description of the attack and oviposition of T. albipes.

Leafminers (Diptera: Agromyzidae) are pests of various crops, mainly in greenhouses, and have Diglyphus spp. as important leafminer larval parasitoids. Until recently, only Diglyphus insularis (Gahan) had been reported in Brazil. In here we report the first records of Diglyphus begini (Ashmead), D. intermedius (Girault) and D. isaea (Walker) in Brazil. These parasitoids were found parasitizing leafminer larvae on cultivated and spontaneous plants in some areas of Minas Gerais state, Brazil.

It was reported the occurrence of Spalangia endius Walker, 1839 (Hymenoptera, Pteromalidae) as a parasitoid of pupae of Musca domestica Linnaeus, 1758 (Diptera, Muscidae) and Stomoxys calcitrans Linnaeus, 1758 (Diptera, Muscidae) in the extreme Southern of Brazil. The collection of pupae was performed in January and February, 2008. The pupae of M. domestica and S. calcitrans were collected from bovine feces using the flotation method. The pupae were individualized in glass tubes and maintained in acclimatized chamber at 27±2ºC with relative air humidity > 70 percent until the emergence of the flies or the parasitoids. The referred occurrence consists in the first report to Rio Grande do Sul.

This study reports the first occurrence of Muscidifurax raptorellus (Hymenoptera: Pteromalidae) in Brazil. Chicken manure samples, collected at two-week interval, were taken to the laboratory and the pupae were extracted by the method of flotation. Each pupa was placed in capsules of colorless gelatin until the emergence of dipterous or their parasitoids. In the study, ten pupae of Fannia pusio (Wiedemann, 1830) (Diptera: Fanniidae) were obtained, two of which yielded the parasitoid M. raptorellus . The percentage of parasitism was 5.0 percent.

The properties of recombinant staphylokinase (SakSTAR) expressed in Pichia pastoris cells have been determined. The single consensus N-linked oligosaccharide linkage site in SakSTAR (at Asn28 of the mature protein) was occupied in approximately 50% of the expressed protein with high-mannose-type oligosaccharides. The majority of these glycans ranged in polymerization state from Man8GlcNAc2 to Man14GlcNAc2, with the predominant species being Man10GlcNAc2 and Man11GlcNAc2. Glycosylated SakSTAR (SakSTARg) did not differ from its aglycosyl form in its aggregation state in solution, its thermal denaturation properties, its ability to form a complex with human plasmin (hPm), the amidolytic properties of the respective SakSTAR-hPm complexes, or its ability to liberate the amino-terminal decapeptide required for formation of a functional SakSTAR-hPm plasminogen activator complex. However, this latter complex with SakSTARg showed a greatly reduced ability to activate human plasminogen (hPg) as compared with the same complex with the aglycosyl form of SakSTAR. We conclude that glycosylation at Asn28 does not affect the structural properties of SakSTAR or its ability to participate in the formation of an active enzymatic complex with hPm, but it is detrimental to the ability of the SakSTAR-hPm complex to serve as a hPg activator. This is likely due to restricted access of hPg to the active site of the SakSTARg-hPm complex.
