Trehalose is a reducing disaccharide, acting as a protectant against various environmental stresses in numerous organisms. In plants, trehalose-6-phosphate synthase (TPS) plays a crucial role in trehalose biosynthesis. Anoectochilus roxburghii (Wall.) Lindl. is a prominent species of the Anoectochilus genus, widely utilized as a health food. However, the functional analysis of TPS in this species has been limited. In this study, TPS genes were cloned from A. roxburghii. The ArTPS gene, with an open reading frame spanning 2850 bp, encodes 950 amino acids. Comparative and bioinformatics analysis revealed that the homology was presented between the ArTPS protein and TPSs from other plant species. The ORF sequence was utilized to construct a prokaryotic expression vector, Pet28a-ArTPS, which was then transformed into Escherichia coli. The resulting transformants displayed a significant increase in salt tolerance under the stress conditions of 300 mmol/L NaCl. Quantitative RT-PCR analysis demonstrated that the expression of ArTPS genes responded to NaCl stress. The accumulation of G6P was upregulated, whereas the content of T6P exhibited an opposite expression trend. The glycometabolism products, including trehalose, exhibited notable changes under NaCl stress, although their variations may differ in response to stimulation. The content of kinsenoside, a characteristic product of A. roxburghii, was significantly upregulated under NaCl stress. These results suggest that the ArTPS genes function in response to NaCl stimulation and play a key role in polysaccharide and glycoside metabolism in Anoectochilus. This study provides new insights into the engineering modification of the health food A. roxburghii to enhance the medicinal activity of its ingredients.

The Ecosystem services (ESs), which play an important role in the balance of the natural ecosystem and social-economic development, are suffering from degradation caused by human activities and climate change. However, the manner in which the ESs respond to the land use/cover changes (LUCCs) and the climatic factors respectively remain elusive, especially in the forest-steppe ecotone, which is highly sensitive to climate change and anthroponotic activities. Based on the remote sensing data and in situ meteorological data, we comprehensively modeled and compared 4 key ESs changes caused by 3 LUCC types, land-use change fraction, and climate changes through two simple comparative experiments. Our results showed that: the Grain for the Green Project improved the mean soil conservation, carbon sequestration, and water yield but reduced the sand fixation. The cropland expansion had a positive influence on the water yield and sand fixation, but it induced a decline in soil conservation and carbon sequestration. The urbanization very likely increased the water yield and decreased soil conservation, carbon sequestration, and sand fixation. The unequal change fractions of the same land-use conversion may affect the ESs differently. The ESs changes have different responses to climate change in different landscapes due to the ecological process. The water yield could be well explained by the temperature, precipitation, radiation, and wind speed. Climate change had a stronger effect on the water yield and carbon sequestration than the land use/cover changes but sand fixation and soil conservation were more likely to be affected by LUCCs. The impact of three types of land-use changes and climate change on the ecosystem services should be considered when formulating land-use policies. This paper might aid the decision-makers in achieving ESs sustainable management and develop land-use strategies in the forest-steppe ecotone.

Chromite Ore Processing Residue (COPR) is the byproduct of chromate production process, which contains a large amounts of Cr(VI). The present work developed a new technique to treat COPR, and the process involved mixing the COPR with rice straw followed by pyrolysis. It was found that the gaseous organic fraction generated during pyrolysis of straw was beneficial to Cr(VI) reduction. In the study, process variables, such as the amount of straw added to COPR, heating temperature and particle size, were systematically varied, and their influences on the Cr(VI) reduction in COPR were all investigated. After pyrolysis, Cr(VI) decreased greatly, from 3400 for untreated COPR to less than 30 mg kg(-1) for treated COPR at 600 degrees C.

The effect of pyrolysis process with sewage sludge on the chromium behavior of chromite ore processing residue (COPR) was examined in this study. The behavior of chromium was characterized in term of chromium oxidation test, pH-static leaching tests, column leaching test and sequential extraction test. As a sequence of pyrolysis process, the Cr(VI) in COPR was effectively reduced from 5057 mg kg(-1) for untreated COPR to 8.6 mg kg(-1) for treated COPR at temperature over 600 degrees C, which is far below the New Jersey Department of Environmental Protection regulatory limit of 240 mg kg(-1). As a result, the amount of exchangeable and carbonate-bound Cr fractions, the most mobile for the environment, were largely reduced. At the same time, the amount of the other three Cr fractions which are much less mobile become augmented. pH static test showed that the chromium in the treated COPR at pyrolysis temperature above 400 degrees C was quite stable at pH>7. Column study also indicated that only negligible amount of chromium of the treated COPR at above 600 degrees C can be released by the acid rain.
