RESUMO

The objective of this study was to characterize and typify dairy farms and to evaluate society's perception of dairy production in rural settlements in the Brazilian semi-arid region. A questionnaire containing questions related to socioeconomic, livestock, physical, herd management, and production indicators was applied to 17 milk farmers for typological analysis. A second questionnaire was subsequently applied to the same 17 farmers for the holistic perception of dairy farming in rural settlements, in addition to 47 technicians in the area of animal science and 140 laypeople. Milk samples were collected in the settlement's community cooling tank, while the water used for milking management was collected in the milking parlor at the 17 farms. Multivariate analysis techniques and thematic analysis were performed. Three dairy production systems were identified: low production group (0-25 L), medium production group (25-50 L), and high production group (50-100 L). The physicochemical properties (composition, density, and cryoscopy) of milk attended the Brazilian regulation values. The microbiological parameters of milk and water used for milking are at odds with regulations. Regarding the typology, the indicators, livestock, and productivity showed discriminatory power (P < 0.05) between the productive systems. The responses from the segments of society showed different views on dairy production in rural settlements. It is concluded that the dairy farmers have low education and the cleaning product and water used in milking are out of regulation. The livestock and production indicators typified mainly dairy farms, while the holistic perception showed a lack of efficiency in rural extension, ignorance of the potential of the productive system. Finally, the creation of public policies focusing on the development of the milk production system in settlements is recommended.

This work was aimed at the production and characterization of a new nanocarrier based on a Sterculia striata polysaccharide (SSP) modified via acylation reaction with propionic anhydride. Nanocapsules of propionated SSP (PSSP) were produced via spontaneous nanoemulsification process and tested as a potential amphotericin B (AMB) nanocarrier. Stable nanoparticles with a very low polydispersity index (0.08-0.29) and high zeta potential (ζ -42.7 to -53.8 mV) were obtained. Particle size was dependent on the degree of substitution and ranged from 205 to 286 nm. A nanocapsule with a degree of substitution (DS) of 2.53 (NCP 2.53) was selected for encapsulation, biocompatibility, and antifungal evaluation against Candida albicans strains. A maximum of 98.3% AMB encapsulation was achieved. Encapsulated AMB was in its monomeric form and showed good biocompatibility and antifungal activity against four C. albicans strains. Data indicate that PSSP has potential as a nanocarrier system for AMB.

In this study we report the synthesis of silver tungstate microcrystals (α-Ag2WO4) by sonochemistry method (SC) at 65 °C and sonochemistry followed by conventional hydrothermal (SC + HC) for 1, 6 and 12 h, at 140 °C. The structural characterization by XRD confirms the alpha phase of the orthorhombic structure and the space group Pn2n, for all synthesized microcrystals. All the actives modes identified at the Raman spectroscopy were characteristic of alpha phase. The optical band gap by UV-Vis spectroscopy using the diffuse reflectance were 2.98, 3.0, 2.99 and 2.96 eV, for the microcrystals SC, SC + HC-1 h, SC + HC-6 h and SC + HC-12 h, respectively. FE-SEM images show the rod-like microcrystals, however, exhibiting the plane surface (1 0 1) only for the synthesized microcrystals with the assistance of the hydrothermal method (SC + HC-1 h, SC + HC- 6 h and SC + HC-12 h). The antimicrobial potential was confirmed for all α-Ag2WO4 microcrystals synthesized. However, the SC + HC-12 h microcrystals were more susceptible in the bacterial and fungal inhibition, with MIC values for microorganisms C. albicans, T. rubrum, MRSA e EHEC, 0.2-0.5, 4-9, 250 and 31.25 µg mL-1, respectively.

Stable oil nanocapsules based on acetylated Sterculia striata polysaccharide (ASSP) were produced without the use of a surfactant, and derivatives of ASSP with four different degrees of substitution (DS) were synthesised. The data revealed that only derivatives with high DS were able to produce nanocapsules (NC), which exhibited monomodal size distribution profiles with a Z-average particle size, ζ-potential, and polydispersity index (PDI) that were dependent on ASSP DS and concentration. Nanocapsules were loaded with amphotericin B (AMB) with encapsulation efficiencies (EE%) that were dependent on drug and ASSP concentrations and DS. A maximum EE% value of 99.2% was achieved, and the loaded AMB was found to be in a monomeric form, even with a concentration one hundredfold higher than that usually observed for commercial AMB aqueous solutions. Loaded nanocapsules show an in vitro controlled release of AMB. As the monomeric AMB state decreased drug toxicity, ASSP nanocapsules loaded with AMB (NC1.68) have potential for use as a drug delivery system. AMB loaded NC 1.68 keeps its activity against 5 strains of Candida albicans tested.

The aims of this study were to test the antifungal activity, toxicity and chemical composition of essential oil from C. sativum L. fruits. The essential oil, obtained by hydro-distillation, was analyzed by gas chromatography/mass spectroscopy. Linalool was the main constituent (58.22%). The oil was considered bioactive, showing an LC50 value of 23 µg/mL in the Artemia salina lethality test. The antifungal activity was evaluated against Microsporum canis and Candida spp. by the agar-well diffusion method and the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) were established by the broth microdilution method. The essential oil induced growth inhibition zones of 28 ± 5.42 and 9.25 ± 0.5 for M. canis and Candida spp. respectively. The MICs and MFCs for M. canis strains ranged from 78 to 620 and 150 to 1,250 µg/mL, and the MICs and MFCs for Candida spp strains ranged from 310 to 620 and 620 to 1,250 µg/mL, respectively. C. sativum essential oil is active in vitro against M. canis and Candida spp. demonstrating good antifungal activity.
