RESUMO

The critical role of RNA, its use and targetability concerning different aspects of human health are gaining more attention because our understanding of the versatility of RNA has dramatically evolved over the last decades. We now appreciate that RNA is far more critical than a messenger molecule and possesses many complicated functions. As a multifunctional molecule with its sequence, flexible structures and enzymatic abilities, RNA is genuinely powerful. Mammalian transcriptomes consist of a dynamically regulated plethora of coding and noncoding RNA types. However, some aspects of RNA metabolism remain to be explored. In this Viewpoint, we focus on the transcriptome's unconventional and possibly overlooked aspects to emphasize the importance of RNA in mammalian systems.

Objective: Reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) is one of the time-saving, accurate, and cost-effective alternative methods to real-time polymerase chain reaction (RT-PCR). This study aimed to identify the robustness of a colorimetric RT-LAMP assay kit that we developed, detecting SARS-COV-2 viral RNA within 30 minutes using a primer set special to the N gene against RT-PCR, the gold standard. Materials and Methods: Both symptomatic and asymptomatic subjects were included from a single university hospital and the status of both RT-PCR and RT-LAMP assay results were compared, and the consistency of these two assays was analyzed. Results: We showed that the RT-LAMP and RT-PCR assay results confirmed 90% consistency. When we consider the epidemiologic, clinical, and radiologic evaluation, the consistency reached 97%. Conclusion: The results revealed that the colorimetric RT-LAMP assay was efficient, robust, and rapid to be used as in vitro diagnostic tool to display competitiveness compared with RT-PCR.

Olive mill wastewater, also known as olive wastewater, contains biologically active components with various beneficial effects on health. The development of novel adsorbent materials for the recovery of these biologically active substances is important area of research. In this study, copolymeric hydrogels based on N-vinylimidazole (VIm), a new material that has never been used as an adsorbent in the separation of phenolic components, were synthesized. The hydrogels synthesized in this study is copolymer structures based on N-vinylimidazole (VIm) containing [2- (methacryloxy) ethyl] dimethylpentylammonium bromide (QDMAC5) in different moles. QDMAC5 was obtained by quaternization of 2- (dimethylamino) ethyl methacrylate (DMA) with 1-bromopentane (C5). The production of copolymer hydrogels was carried out by free radical solution polymerization. The syntheses were carried out only by changing the monomer composition so that the crosslinker ratio remained constant (1.2 mol%). The QDMAC5 content in the copolymers was 5, 10, 20, 30, and 50 mol%. So, the resulting structures were named PVQ-5%, PVQ-10%, PVQ-20%, PVQ-30%, and PVQ-50%, respectively. Functional group characterizations of hydrogels were made by Fourier Transform Infrared Spectrometry (FTIR). The surface of the hydrogels was analyzed by Scanning Electron Microscopy (SEM). Finally, thermogravimetric analyzes (TGA) were performed to investigate the thermal degradation behavior. The recovery of the rutin present in olive mill wastewater has been investigated as a model study. Kinetic data has been represented by the selected models (pseudo-first order, pseudo-second order, and intraparticle diffusion) convincingly (R2 > 0.76), while the equilibrium findings have fitted well to Langmuir, Freundlich, and Temkin equations (R2 > 0.77). Rutin adsorption process on N-vinylimidazole (VIm) based copolymeric hydrogels has been found as exothermic and spontaneous chemisorption process depending on the thermodynamic analysis.

The solid-state reaction was employed as a suitable method to produce orthorhombic Yb3+/Er3+ co-doped La2Ti2O7 phosphors. The pump power and temperature dependencies of spectra of the Yb3+/Er3+ co-doped La2Ti2O7 were studied for color tunability and optical thermometry. The pump power significantly affected the color coordinates of the phosphors. The upconversion green light transformed to the broad white light at higher pump power. A two-photon transition was proposed to explain the upconversion energy transfer process. The fluorescence intensity ratio technique was used to calculate the optical temperature sensor sensitivities. The ΔE energy difference obtained from the intensity ratios of the thermally coupled 2H11/2 and 4I15/2 energy levels of Er3+ are 763.64 ± 76.62 cm-1, 777.77 ± 44.62 cm-1, 640.71 ± 137.39 cm-1 for La1.95Yb0.04Er0.01Ti2O7, La1.94Yb0.04Er0.02Ti2O7, and La1.93Yb0.04Er0.03Ti2O7, respectively. Absolute temperature sensitivities of the phosphors decreased with increasing Er3+ concentration. The maximum absolute sensitivity was determined to be 0.51x10-2 K-1 at 553 K for La1.95Yb0.04Er0.01Ti2O7, 0.64x10-2 K-1 at 571 K for La1.94Yb0.04Er0.02Ti2O7, and 0.38x10-2 K-1 at 457 K for La1.93Yb0.04Er0.03Ti2O7.

Roles of HNRNPA1 are beginning to emerge in cancers; however, mechanisms causing deregulation of HNRNPA1 function remain elusive. Here, we describe an isoform switch between the 3'-UTR isoforms of HNRNPA1 in breast cancers. We show that the dominantly expressed isoform in mammary tissue has a short half-life. In breast cancers, this isoform is downregulated in favor of a stable isoform. The stable isoform is expressed more in breast cancers, and more HNRNPA1 protein is synthesized from this isoform. High HNRNPA1 protein levels correlate with poor survival in patients. In support of this, silencing of HNRNPA1 causes a reversal in neoplastic phenotypes, including proliferation, clonogenic potential, migration, and invasion. In addition, silencing of HNRNPA1 results in the downregulation of microRNAs that map to intragenic regions. Among these miRNAs, miR-21 is known for its transcriptional upregulation in breast and numerous other cancers. Altogether, the cancer-specific isoform switch we describe here for HNRNPA1 emphasizes the need to study gene expression at the isoform level in cancers to identify novel cases of oncogene activation.

Novel 4,4'-(((2,4,8,10-tetraoxaspiro[5.5]undecane-3,9-diyl)bis(2-methylpropane-2,1-diyl))bis(oxy)) (SUBO) bridged ball-type metallophthalocyanines were synthesized starting from 4,4'-(((2,4,8,10-tetraoxaspiro[5.5]undecane-3,9-diyl)bis(2-methylpropane-2,1-diyl))bis(oxy))diphthalonitrile with convenient metal salts in 2-N,N-dimethylaminoethanol. A new bisphthalonitrile compound was obtained from 2,2'-(2,4,8,10-tetraoxaspiro[5.5]undecane-3,9-diyl)bis(2-methylpropan-1-ol) and 4-nitrophthalonitrile in acetonitrile at reflux temperature in the presence of potassium carbonate as a catalyst. The structural characterization of the compounds was performed by elemental analysis, and infrared, ultraviolet-visible and matrix-assisted laser desorption/ionization time-of-flight mass spectroscopic methods. Nonlinear absorptions of the phthalocyanine complexes were measured using the Z-scan technique with 7 ns pulse duration at a 532 nm wavelength. It is obvious that ball-type copperphthalocyanine has a high nonlinear absorption coefficient and imaginary component of the third-order susceptibility compared to other complexes. Therefore, ball-type copperphthalocyanine can be regarded as a very good candidate for optical limiting applications. Density functional theory was used for geometry optimizations and time-dependent density functional theory calculations of electronic transitions in order to compare with the experimental results. Molecular orbital and nonlinear optical analyses were also performed with density functional theory at the CAM-B3LYP/6-31G(d,p)/LANL2DZ level. The nonlinear optical analyses show that ball-type copperphthalocyanine has significantly better nonlinear optical properties in comparison to a common reference compound, urea.

The ER-bound kinase/endoribonuclease (RNase), inositol-requiring enzyme-1 (IRE1), regulates the phylogenetically most conserved arm of the unfolded protein response (UPR). However, the complex biology and pathology regulated by mammalian IRE1 cannot be fully explained by IRE1's one known, specific RNA target, X box-binding protein-1 (XBP1) or the RNA substrates of IRE1-dependent RNA degradation (RIDD) activity. Investigating other specific substrates of IRE1 kinase and RNase activities may illuminate how it performs these diverse functions in mammalian cells. We report that macrophage IRE1 plays an unprecedented role in regulating phosphatidylinositide-derived signaling lipid metabolites and has profound impact on the downstream signaling mediated by the mammalian target of rapamycin (mTOR). This cross-talk between UPR and mTOR pathways occurs through the unconventional maturation of microRNA (miR) 2137 by IRE1's RNase activity. Furthermore, phosphatidylinositol (3,4,5) phosphate (PI(3,4,5)P3 ) 5-phosphatase-2 (INPPL1) is a direct target of miR-2137, which controls PI(3,4,5)P3 levels in macrophages. The modulation of cellular PI(3,4,5)P3 /PIP2 ratio and anabolic mTOR signaling by the IRE1-induced miR-2137 demonstrates how the ER can provide a critical input into cell growth decisions.

Yb3+,Er3+,Tm3+:Y2Si2O7 nanopowders were fabricated by a sol-gel method and then annealed at two different temperatures of 1050 °C and 1480 °C for 12 h to investigate the phase effect on the structural and optical properties. X-ray diffraction patterns (XRD) of all powders revealed that most of the peaks were in good harmony with the standard JCPDS cards of Y2Si2O7 (PDF #38-0223 and PDF #38-0440). SEM images of the powders were taken to examine the morphology and chemical composition of the powders. Diffuse reflection spectra of Y2Si2O7 exhibited varying number of transitions depending on the dopant ion in all powders in the range of 400-1050 nm. The spectral properties of the powders were investigated to identify the possible up-conversion (UC) emission mechanism in the range between 400 and 900 nm under 975 nm excitation. A broadening white light emission was observed with increasing laser pumping power at room temperature as well. Spectral profiles of the transitions observed in both reflectance and up-conversion luminescence spectra were found to be crystalline phase dependent.

OBJECTIVE: Bipolar disorder (BD) is a chronic mood disorder characterized by recurrent episodes that has a lifetime prevalence of 0.4- 5.5%. The neurochemical mechanism of BD is not fully understood. Oxidative stress in neurons causes lipid peroxidation in proteins associated with neuronal membranes and intracellular enzymes and it may lead to dysfunction in neurotransmitter reuptake and enzyme activities. These pathological processes are thought to occur in brain regions associated with affective functions and emotions in BD. The relationship between the number of manic episodes and total oxidant-antioxidant capacity was investigated in this study. METHODS: Eighty-two BD patients hospitalized due to manic symptoms and with no episodes of depression were enrolled in the study. Thirty of the 82 patients had had their first episode of mania, and the other 52 patients had had two or more manic episodes. The control group included 45 socio-demographically matched healthy individuals. Serum total antioxidant capacity (TAC) and total oxidant capacity (TOC) measurements of the participants were performed. The oxidative stress index (OSI) was calculated by TOC/TAC. RESULTS: There were no significant differences in OSI scores between BD patients with first-episode mania and BD patients with more than one manic episode. However, OSI scores in both groups were significantly higher than in the control group. TOC levels of BD patients with first-episode mania were found to be significantly higher than TOC levels of BD patients with more than one manic episode and healthy controls. There were no significant differences in TAC levels between BD patients with first-episode mania and BD patients with more than one manic episode. TAC levels in both groups were significantly higher than in the control group. CONCLUSION: Significant changes in oxidative stress indicators were observed in this study, confirming previous studies. Increased levels of oxidants were shown with increased disease severity rather than with the number of manic episodes. Systematic studies, including of each period of the disorder, are needed for using the findings indicating deterioration of oxidative parameters.

OBJECTIVE: In obstructive sleep apnea syndrome (OSAS), any of the activated neural, vascular, hemodynamic, metabolic, inflammatory, and thrombotic mechanisms may be related to increased cerebrovascular disease and risk of death; however, the possible pathophysiological process between obstructive sleep apnea syndrome and stroke has not been clearly explained. We hypothesize that alterations in vasomotor reactivity in patients may be responsible for their altered cerebral blood flow, and may contribute to the increased risk of ischemic stroke. METHODS: A total of 30 untreated patients with severe obstructive sleep apnea and 26 control subjects were included in the study. The mean blood flow velocity and breath holding index were measured in middle cerebral artery bilaterally in both patient and control groups by using transcranial Doppler ultrasound. We compared the values between two groups. RESULTS: The mean blood flow velocity and breath holding indexes were significantly decreased in the patient group when compared with the control group. There were no correlations between cerebral hemodynamic parameters and polysomnographic findings in patients. CONCLUSION: Our findings suggest that there was a deteriorated vasodilator response to hypercapnia in patients with OSAS. This deterioration may stem from chemoreceptors or endothelial damages that lead to vascular relaxation and vasodilatation in cerebrovascular circulation. This impaired cerebral vascular regulation may contribute to the increased risk of stroke in patients with OSAS.

OBJECTIVE: Aggressive behaviour is associated with reduced serotonin metabolism in the brain, but there is not enough knowledge on potential changes of the serotonin precursor levels among violent offenders. In this study, we aimed to evaluate the relationships among the tendency of psychopathy, anger and the basic amino acids. METHODS: Fifty-two young adult male patients with antisocial personality disorder (APD) and 30 healthy men included the study. Serum amino acid levels were measured by HPLC method. Aggression questionnaire and Hare Psychopathology Scale were used for all participants. RESULTS: Blood levels of phosphoserine, aspartic acid, glutamic acid, aminoadipic acid and 1-methylhistidine in group of patients with APD were significantly higher than the control group. Blood levels of TRP, asparagine, citrulline, cystine, isoleucine, tyrosine, histidine, hydroxylysine, lysine, ethanolamine and arginine in the group of patients were found lower than the control group. A significant positive correlation between anger scores and histidine, methionine and GABA was found. GABA and methionine showed a significant correlation with the indirect aggression score. CONCLUSION: Our study showed a relationship between serum amino acid levels and the scores of aggression and psychopathy. We think that this is a productive research area for understanding the relationship among biochemical factors, aggression and psychopathy.

Pure, Eu(3+) - doped and Eu(3+),B(3+) co-doped CdNb2O6 powders have been prepared by a molten salt synthesis method using Li2SO4/Na2SO4 salt mixture as a flux at a relatively low temperatures as compared to solid state reaction. X-ray diffraction patterns of pure CdNb2O6 samples indicated orthorhombic single phase. Photoluminescence investigations of CdNb2O6 samples showed a strong blue emission band centered at 460 nm. For Eu-doped CdNb2O6 samples, the luminescence of Eu(3+) was observed with the host red emission varying with the Eu-doping concentrations. This PL characteristic of the doped samples may be attributed to the energy transfer between Eu(3+) and niobate groups (NbO6). Boron incorporation has remarkably increased the luminescence of Eu(3+)-doped CdNb2O6.

BACKGROUND: Studies that have focused on the mitochondrial electron transport chain indicate that bipolar disorder (BD) is associated with pathology in mitochondrial function. These pathological processes occur in the brain circuits that regulate affective functions, emotions, and motor behaviors. The present study aimed to determine the relationship between mitochondrial complex dysfunction and BD. METHODS: The BD group included 32 male patients diagnosed with first-episode manic BD. The control group included 35 sociodemographically matched healthy males. Messenger ribonucleic acid (mRNA) was isolated from peripheral blood samples obtained from the patients and control group, and the mRNA levels of the NDUFV1, NDUFV2, and NDUFS1 genes of mitochondrial complex I and the UQCR10 gene of mitochondrial complex III were investigated. RESULTS: Significant differences were identified in complex I gene mRNA levels between the BD group (n = 32) and the control group (n = 35) for the following genes: NDUFV1 (P = 0.01), NDUFV2 (P < 0.01), and NDUFS1 (P = 0.02). The UQCR10 gene (complex III) mRNA level did not differ between the groups (P = 0.1). The mRNA levels of the four genes studied were lower at the 3-month follow-up; however, these differences were not significant (P > 0.05). LIMITATIONS: All of the BD patients were in manic episodes; thus, we were unable to separately compare these levels with those during depressive and euthymic episodes. CONCLUSIONS: The mRNA levels of all of the genes representing the subunits of mitochondrial complex I (NDUFV1, NDUFV2, and NDUFS1) were significantly higher in the present study's BD patients during manic episodes than in the controls. With the data obtained from further research, biomarkers that could be used for the diagnosis and follow-up of neuropsychiatric disorders may be discovered.

This is a study of in-plane and out-of-plane distribution of rotational torque (ROT-T) and effective electric field (EEF) on electrorotation (ER) devices with 3D electrodes using finite element modeling (FEM) and experimental method. The objective of this study is to investigate electrical characteristics of the ER devices with five different electrode geometries and obtain an optimum structure for ER experiments. Further, it provides a comparison between characteristics of the 3D electrodes and traditionally used 2D electrodes. 3D distributions of EEF were studied by the time-variant FEM. FEM results were verified experimentally by studying the rotation of biological cells. The results show that the variations of ROT-T and EEF over the measurement area of the devices are considerably large. This can potentially lead to misinterpretation of recorded data. Therefore, it is essential to specify the boundaries of the measurement area with minimum deviation from the central EEF. For this purpose, FE analyses were utilized to specify the optimal region. Thereby, with confining the measurements to these regions, the dependency of ROT-T on the spatial position of the particles can be eliminated. Comparisons have been made on the sustainability of the EEF and ROT-T distributions for each device, to find an optimum design. Analyses of the devices prove that utilization of the 3D electrodes eliminate irregularities of EEF and ROT-T along the z-axis. The Results show that triangular electrodes provide the highest sustainability for the in-plane ROT-T and EEF distribution, while the oblate elliptical and circular electrodes have the lowest variances along the z-axis.

Dielectrophoresis (DEP), a technique used to separate particles based on different sizes and/or dielectric properties under nonuniform electric field, is a promising method to be applied in label-free, rapid, and effective cell manipulation and separation. In this study, a microelectromechanical systems-based, isolated 3D-electrode DEP device has been designed and implemented for the label-free detection of multidrug resistance in K562 leukemia cells, based on the differences in their cytoplasmic conductivities. Cells were hydrodynamically focused to the 3D-electrode arrays, placed on the side walls of the microchannel, through V-shaped parylene-C obstacles. 3D-electrodes extruded along the z-direction provide uniformly distributed DEP force through channel depth. Cell suspension containing resistant and sensitive cancer cells with 1:100 ratio was continuously flown through the channel at a rate of 10 µL/min. Detection was realized at 48.64 MHz, the cross-over frequency of sensitive K562 cells, at which sensitive cells flow with the fluid, while the resistant ones are trapped by positive DEP force. Device can be operated at considerably low voltages (<9 Vpp ). This is achieved by means of a very thin (0.5 µm) parylene coating on electrodes, providing the advantages offered by the isolation of electrodes from the sample, while the working voltage can still be kept low. Results prove that the presented DEP device can provide an efficient platform for the detection of multidrug resistance in leukemia, in a label-free manner.

Pure Nd(3+)- or Eu(3+)-doped CoNb2O6 powders have been prepared by a molten salt synthesis method using a Li2SO4-Na2SO4 or NaCl-KCl salt mixture as a flux at relatively low temperatures as compared to the solid state reaction method. X-ray diffraction patterns of pure CoNb2O6 samples indicated an orthorhombic single phase. For Eu(3+)-doped CoNb2O6 samples, the luminescence of Eu(3+) was observed at 615 nm as red emission while the Nd(3+) doped sample showed a typical emission at 1064 nm varying with the Eu(3+) or Nd(3+) doping concentrations. These luminescence characteristics of the doped samples may be attributed to the energy transfer between rare earth ions and CoO6 octahedral groups in the columbite structure.

BACKGROUND: Military personnel deployed abroad could be exposed to more risk factors that adversely affect quality of life. In this study, we examined psychiatric symptoms and quality of life in Turkish Armed Forces deployed to Afghanistan. METHOD: A total of 289 Turkish military personnel working in Afghanistan enrolled in this study. They completed two surveys containing questions about socio-demographic characteristics. Data were collected and analyzed from 258 of the participants. RESULTS: The general symptom scores (GSI) were above 1 in 20.8 Percent (n=54) of the participants. The lowest SF-36 scores by the sub-groups were mental health (59.14 ± 18.56) and vitality (59.25 ± 21.17). The highest score was in the physical function subscale (84.42 ± 19.53). All Quality of Life Questionnaire Short Form (SF-36)subscale scores were lower in the GSI above 1 group than the GSI below 1 group. In the GSI above 1 group: education level and depression affected SF-36 physical functioning; paranoid ideation and somatization affected SF-36 role limitations due to physical health; age and somatization affected SF-36 pain; age affected SF-36 general health; phobic anxiety affected SF-36 vitality; age, tenure of occupation, tenure abroad; and phobic anxiety affected SF-36 mental health. CONCLUSIONS: The negative effects of psychiatric symptoms on the quality of life were similar to those in the general population and in specific disease groups. These results should be considered when evaluating the mental health of military personnel deployed abroad.

INTRODUCTION: Hyperarousal and alertness play an important role in the clinical presentation of Post-traumatic stress disorder (PTSD). Strenuous effort has been made to shed light on the mechanisms that cause these symptoms of patients. Based on the claim that there is a relationship between some subtypes of hyperarousal symptoms and aggression in patients with PTSD, we aimed to examine the relationship between electrophysiological measurements that was measured through transcranial magnetic stimulation (TMS) and aggression scale scores of PTSD patients in this study. METHODS: The study included 37 patients with a diagnosis of PTSD according to DSM-IV criteria and 25 healthy volunteers. Electrophysiological measurements of participants were made with TMS. The Buss Perry Aggression Questionnaires was administered to patients and control group. RESULTS: In the patient group, a positive correlation was found between scores of aggression and arousal symptoms. Motor excitability threshold, one of TMS measurements, which is a sign of cortical excitability, was significantly lower in the patient group than the control group. There was a negative correlation between aggression scale scores and the parameters of motor excitability threshold and cortical silent period which both shows cortical excitability of the patients. CONCLUSION: We concluded that there was an increase in cortical excitability in PTSD patients and we suggest that this increase might be associated with hyperarousal symptoms and aggressive behavior.

Nitric oxide (NO) is thought to be involved in the pathogenesis of schizophrenia as well as many neuropsychiatric disease. Asymmetric dimethylarginine (ADMA) reduces the level of NO by inhibiting nitric oxide synthase (NOS) enzyme. In this study it is aimed to be investigated ADMA in patients with first-episode schizophrenia. In this study, according to DSM-IV diagnostic criteria for schizophrenia-like psychotic disorder, 49 male first-episode schizophrenia patients-whose mean age was 23.4±3.5 year-and age and education matched 30 healthy male subjects were included for comparison. ADMA levels of the patients were measured before and after 2 months of therapy. In order to rule out the conditions that may affect the levels of ADMA, people whose physical examination and laboratory findings were within normal range were included in the study. In this study plasma ADMA levels of first-episode schizophrenia patients and control group were 3.6±1.5 µmol/L and 1.02±1.02 respectively. After 2 months of antipsychotic treatment plasma ADMA levels of the schizophrenia patients decreased compared to baseline. There was no relationship between the ADMA levels and the clinical severity of the disease. It is considered to be the role of ADMA in the etiopathogenesis of schizophrenia.

Crimean-Congo haemorrhagic fever (CCHF) is a tick-borne disease endemic in Turkey since 2002. Çorum is one of the leading five cities in Turkey in which CCHF disease is seen most. We studied characteristics of the patients with tick bites in our emergency department (ED) and determined the fatality rate of the disease in city of Çorum for the first time. We retrospectively analysed the characteristics of the patients admitted to our ED from the medical files of 21,680 patients in a 5-year period. The number of patients with definite diagnosis and who have died was determined. Our results demonstrated that the fatality rate of CCHF in Çorum is 6.78%. Among 21,680 patients, blood samples of 970 patients were sent to an advanced centre in Ankara for polymerase chain reaction (PCR) testing. Results of 560 patients were reported to be PCR (+) and 38 of them have died.
