RESUMO

The Jishishan Ms 6.2 earthquake occurred at 23:59 on December 18, 2023 in Gansu Province, China. We conducted a field survey to assess the hazards and damages caused by the earthquake and its associated geo-activities. Subsequently, we organized a seminar to discuss the possible causes of the destruction of a prehistoric site-Lajia Settlement-dated back to four thousand years B.P. and located only several kilometers away from the epicenter of the Jishishan earthquake. The Jishishan earthquake was unique for its hazard and disaster process, which featured ground shaking and a series of complex geological and geomorphological activities: sediment and soil spray piles, liquefaction, collapse, landslide, and mudflow along water channels. We define this phenomenon as the Jishishan earthquake ripple hazard (JERH). The most recent evidence from the JERH suggests that a prehistoric earthquake similar to the JERH, instead of riverine floods or earthquake-induced landslide dam outburst flood, as previously hypothesized, destroyed the Lajia Settlement.

RESUMO

Osteonecrosis of the femoral head (ONFH) is a elaborate hip disease characterized by collapse of femoral head and osteoarthritis. RNA N6-methyladenosine (m6A) plays a crucial role in a lot of biological processes within eukaryotic cells. However, the role of m6A in the regulation of ONFH remains unclear. In this study, we identified the m6A regulators in ONFH and performed subtype classification. We identified 7 significantly differentially expressed m6A regulators through the analysis of differences between ONFH and normal samples in the Gene Expression Omnibus (GEO) database. A random forest algorithm was employed to monitor these regulators to assess the risk of developing ONFH. We constructed a nomogram based on these 7 regulators. The decision curve analysis suggested that patients can benefit from the nomogram model. We classified the ONFH samples into two m6A models according to these 7 regulators through consensus clustering algorithm. After that, we evaluated those two m6A patterns using principal component analysis. We assessed the scores of those two m6A patterns and their relationship with immune infiltration. We observed a higher m6A score of type A than that of type B. Finally, we performed a cross-validation of crucial m6A regulatory factors in ONFH using external datasets and femoral head bone samples. In conclusion, we believed that the m6A pattern could provide a novel diagnostic strategy and offer new insights for molecularly targeted therapy of ONFH.

RESUMO

Osteoporosis (OP) is a systemic disorder characterized by decreased bone mass as well as deteriorated microarchitecture. Although OP in men is common, it has received much less attention than that in women. Ginseng, a famous traditional herb in Asia, is used to strengthen and repair bones by invigorating vital bioenergy and maintaining body homeostasis in dietary intake and clinical applications. However, there is currently no study investigating the impact of ginseng and its active compounds on male osteoporosis. In this study, RNA sequencing and bioinformatic analysis were conducted to reveal the influence of Ginsenoside-Rb2 on RAW264.7 cells and its underlying signaling pathways. The potential anti-osteoporosis effects of Rb2 as well as its molecular mechanisms were elucidated in RAW264.7 cells and BMMs by TRAP staining, F-actin belt staining, qRT-PCR and WB. Moreover, orchiectomy (ORX) was utilized to demonstrate the influence of Rb2 on bone mass loss in vivo by micro-CT scanning, and H&E, TRAP, and IHC staining. The results suggested that Rb2 suppressed osteoclastogenesis and mitigated bone loss in orchiectomy mice through NF-κB/MAPK signaling pathways. These findings indicate that ginseng as well as its active component Rb2 have potential therapeutic value in the management of osteoporosis in men.

RESUMO

The association between sarcopenia and OA still presents many uncertainties. We aimed to assess whether sarcopenia is associated with occurrence of OA in US adults. We conducted a cross-sectional study consisting of 11,456 participants from National Health and Nutrition Examination Survey 1999-2006. Sarcopenia was defined by a low muscle mass. The skeletal muscle index (SMI) was calculated as the appendicular skeletal muscle mass divided by body mass indexes (BMI) or body weight. OA status was assessed by using self-reported questionnaire. We evaluated the association between sarcopenia and OA using multivariate regression models. In addition, subgroup and interaction analysis were performed. Sarcopenia was associated with OA when it was defined by the BMI-adjusted SMI (OR = 1.23 [95% CI, 1.01, 1.51]; P = 0.038) and defined by the weight-adjusted SMI (OR = 1.30 [95% CI, 1.10, 1.55]; P = 0.003). Subgroup and interaction analysis found that the strongest positive association mainly exists in smoker (OR = 1.54 [95% CI, 1.21, 1.95], Pint = 0.006), and this association is not significant in other groups. In conclusion, we found that sarcopenia was associated with occurrence of OA. Subgroup analysis revealed that the association between sarcopenia and OA was more pronounced in smoker. Further well-designed prospective cohort studies are needed to assess our results.

RESUMO

BACKGROUND: Oxidative stress was closely related to the occurrence and development of Steroid-induced osteonecrosis of the femoral head (SIONFH). 8-hydroxy-2'-deoxyguanosine (8-OHdG) is a important index of oxidative stress. The aim of this study is to investigate the role of 8-OHdG in the development of SIONFH. METHODS: From May 2021 and November 2021, 33 patients diagnosed with SIONFH and 26 healthy controls were recruited in this study. Assessment included the radiography and pathology evaluation of clinical bone tissue, expression position and level of 8-OHdG, level of plasma 8-OHdG, as well as the receiver operating characteristic (ROC) curve. RESULTS: We observed that expression levels of 8-OHdG in bone samples decreased with Association Research Circulation Osseous (ARCO) stages. Plasma 8-OHdG levels were significantly increased in the SIONFH group compared to the healthy control group. Plasma 8-OHdG level of pre-collapse patients was higher than that of post-collapse patients, the decreased plasma 8-OHdG level was related to higher ARCO stages. CONCLUSION: Plasma 8-OHdG may represent potential biomarkers during SIONFH at different stages. Higher plasma 8-OHdG levels indicated early stage of SIONFH. The current study provided new clues for early diagnosis and treatment for SIONFH.

RESUMO

Surgical hip dislocation (SHD) is a powerful and safe approach used to address pathologic lesions around the hip joint, and therefore, many studies have been conducted in this field. However, no bibliometric studies regarding the global research trend concerning SHD have been studied yet. This study aims to determine the research status in the field of SHD research between 2001 and 2021. The publications related to SHD from 2001 to 2021 were retrieved from the Web of Science Core Collection. Three bibliometric tools were used for this study. The main analyses include publication counts, contributions of countries, institutions, authors, journals and funding agencies, as well as analyses on clustering of references and keywords. In total, 498 articles were identified. The annual publication counts of SHD showed an ascending tendency as a whole. The United States has the most prominent contributions, with the most number of publications and the highest value of H-index. The University of Bern was the organization that produced the most literature. Professors Ganz R, Siebenrock KA, Tannast M, Steppacher SD and Leunig M were the core authors in this field. The most productive journal was Clinical Orthopaedics and Related Research. Burst keyword detection suggested that the following research directions, including 'surgical hip dislocation', 'outcome', 'fixation' and 'pain', are considered the research hotspots and deserve more attention. In conclusion, this is the first bibliometric analysis that provides a comprehensive overview of SHD research, which may assist investigators in exploring new directions for this technique.

RESUMO

BACKGROUND: The association between serum ferritin and bone mineral density (BMD) is still controversial. This study aims to investigate the association of serum ferritin level with BMD in US adults. METHODS: We conducted a cross-sectional study consisting of 8445 participants from National Health and Nutrition Examination Survey. Serum ferritin and lumbar spine BMD were used as independent variables and dependent variables, respectively. We evaluated the association between serum ferritin and lumbar spine BMD through a weighted multivariable linear regression model. Subgroup and interaction analysis was also performed in this study. RESULTS: After adjusting for other confounding factors, serum ferritin was negatively correlated with lumbar spine BMD [ß = - 0.090, 95% CI (- 0.135, - 0.045)]. Further subgroup analysis found that the strongest negative association mainly exists in females aged over 45 years [ß = - 0.169, 95% CI (- 0.259, - 0.079)], and this association is not significant in other groups. CONCLUSIONS: The results found that the association between serum ferritin and lumber spine BMD differed by gender and age. Increased level of serum ferritin may indicate a higher risk of osteoporosis or osteopenia in females aged over 45 years.

RESUMO

Background: The association between serum total cholesterol (TC) and bone mineral density (BMD) is still controversial. We aimed to evaluate the association of serum TC with BMD in general US adult women. Methods: A cross-sectional study consisting of 7,092 (age range 20-85) participants from the National Health and Nutrition Examination Survey (NHANES) database was conducted. Weighted multivariate linear regression analyses were performed to evaluate association between serum TC and lumbar spine BMD. In addition, subgroup and interaction analysis were used in this study. Results: The serum TC was negatively correlated with lumbar spine BMD after adjusting for confounders. Subgroup analysis found that the strongest negative association mainly exists in women aged over 45 years with body mass index (BMI) < 24.9 kg/m2, and this association is not significant in other groups. Conclusions: This study found that serum TC exhibit an inverse association with lumbar spine BMD in Us women aged over 45 years. The measurement of serum TC may provide information for predicting poor bone health outcomes in these women.

RESUMO

Objectives: Low-density lipoprotein cholesterol (LDL-C) plays an essential part in bone metabolism. However, the correlation between LDL-C levels and bone mineral density (BMD) is still controversial. This study aimed to explore the relationship between LDL-C levels and lumbar BMD in young- and middle-aged people. Methods: We conducted a cross-sectional study comprising 4,441 participants aged 20-59 from the National Health and Nutrition Examination Survey (NHANES). LDL-C levels and lumbar BMD were used as independent and dependent variables, respectively. We evaluated the correlation between LDL-C levels and lumbar BMD through a weighted multivariate linear regression model. We performed a subgroup analysis of the relationship between LDL-C levels and lumbar BMD based on age, gender, and body mass index (BMI). Results: After adjusting for confounding factors, LDL-C levels were negatively correlated with lumbar BMD. In subgroup analyses stratified by gender, this negative association was statistically significant in men and women. In the subgroup analysis stratified by age, a negative connection existed in people aged 30-49 years. In the subgroup analysis divided by BMI, there was an inverse correlation in overweight people (25 ≤ BMI < 30). Conclusions: Our research observed an inverse association between LDL-C levels and lumbar BMD in young- and middle-aged people, especially in people aged 30-49 years and who are overweight. Close monitoring of BMD and early intervention may be required for these people.

RESUMO

Background: Femoral neck fracture (FNF) is a commonly encountered injury in orthopedic practice, and many studies have been conducted in this field. However, no bibliometric studies regarding the global research trend concerning FNF have been performed. This study aims to analyze the knowledge framework, research hotspots, and theme trends in the field of FNF research. Methods: The scientific outputs related to FNF from 1994 to 2021 were retrieved from the Web of Science Core Collection. Three bibliometric tools were used for this study. The main analyses include publication and citation counts, contributions of countries, institutions, authors, funding agencies and journals, and clustering of keywords. Results: In total, 3,553 articles were identified. The annual publication counts of FNF showed an ascending tendency as a whole. The United States has the most prominent contributions, with the most number of publications and the highest H-index. Karolinska Institutet devoted the most in this domain. Professors Bhandari M, Schemitsch EH, Frihagen F, Parker MJ, and Rogmark C were the core authors in this field. The most productive journal was Injury International Journal of the Care of the Injured. Keywords were divided into four clusters: epidemiology and mortality, fracture prevention, internal-fixation and risk factors, and hip replacement. A trend of balanced and diversified development existed in these clusters. Keywords with the ongoing bursts, including "outcome," "reoperation," "complication," "revision," "displaced intracapsular," "fracture," and "adult," are considered as the research hotspots in the future and deserve more attention. Conclusions: The management of FNF in young patients is drawing more attention from orthopedic surgeons, and it is expected that these research topics may continue to be the research hotspots and focus in the near future.

RESUMO

Calcium oxalate (CaOx) crystals are the main component of kidney stones. Macrophages have the function of eliminating these crystals, and the underlying mechanism remains unclear. Here, we attempted to determine the role of macrophage-derived exosomes exposed to CaOx crystals in regulating apoptosis of human proximal tubular cells (HK-2). Exosomes (CaOx-Exo) were isolated from CaOx-treated macrophages and then incubated with HK-2 cells. CaOx-Exo treatment reduced cell viability and promoted apoptosis of HK-2 cells. The expression of Caspase-3 and Bax was increased, and Bcl-2 expression was decreased in HK-2 cells following CaOx-Exo treatment. Moreover, CaOx-Exo treatment caused an increase of LC3-II/LC3-I ratio and Beclin-1 expression and a downregulation of p62 in HK-2 cells. GFP-LC3 puncta were increased in HK-2 cells following CaOx-Exo treatment. Additionally, CaOx-Exo-treated HK-2 cells were treated with 3-methyladenine (3-MA) to inhibit autophagy activity. 3-MA treatment weakened the impact of CaOx-Exo on cell viability and apoptosis of HK-2 cells. 3-MA treatment also reduced the LC3-II/LC3-I ratio and Beclin-1 expression and enhanced p62 expression in CaOx-Exo-treated HK-2 cells. In conclusion, these data demonstrated that exosomes derived from CaOx-treated macrophages promote apoptosis of HK-2 cells by promoting autophagy. Thus, this work suggests that macrophage-derived exosomes may play a vital role in CaOx-induced human proximal tubular cell damage.

RESUMO

Solar thermal fuels offer a closed cycle and a renewable energy storage strategy by harvesting photon energy within the chemical conformations of molecules and retrieving energy by an induced release of heat. However, the majority of reports are limited to the ultraviolet light storage, which potentially interferes with the surrounding environment and reduces the material lifetime. Here, we present a novel arylazopyrazole (AAP)-containing dendrimer that not only addresses the hindrance of visible light storage for solar thermal fuels but also exhibits outstanding performances of abundant energy conversion and stable storage, which are attributed to the substantial absorbance in visible wavelengths of para-thiomethyl-substituted AAP groups and the stability of cis isomers, respectively. The energy density of the dendrimer fuel after efficiently harvesting blue light (405 nm) is as high as 0.14 MJ kg-1 (67 kJ mol-1), and the storage half-life of the fabricated dendrimer film can reach up to 12.9 days. Moreover, the heat release of the dendrimer film can be triggered by different stimuli (light and heat). The dendrimer film displays a 6.5 °C temperature difference between trans isomers and cis isomers during green light irradiation. Our work provides a fascinating avenue to fabricate visible light storage solar thermal fuels and unlocks the possibility of developing natural sunlight storage in the future.

RESUMO

Dendrimers are well-defined, highly branched macromolecules that have been widely applied in the fields of catalysis, sensing, and biomedicine. Here, we present a novel multifunctional photochromic dendrimer fabricated through grafting azobenzene units onto dendrimers, which not only enables controlled switching of adhesives and effective repair of coating scratches but also realizes high-performance solar energy storage and on-demand heat release. The switchable adhesives and healable coatings of azobenzene-containing dendrimers are attributed to the reversible solid-to-liquid transitions because trans-isomers and cis-isomers have different glass transition temperatures. The adhesion strengths increase significantly with the increase in dendrimer generations, wherein the adhesion strength of fifth-generation photochromic dendrimers (G5-Azo) can reach up to 1.62 MPa, five times higher than that of pristine azobenzenes. The solar energy storage and heat release of dendrimer solar thermal fuels, the isomers of which possess different chemical energies, can be also enhanced remarkably with the amplification of azobenzene groups on dendrimers. The storage energy density of G5-Azo can reach 59 W h kg-1, which is much higher than that of pristine azobenzenes (36 W h kg-1). The G5-Azo fuels exhibit a 5.2 °C temperature difference between cis-isomers and trans-isomers. These findings provide a new perspective and tremendously attractive avenue for the fabrication of photoswitchable adhesives and coatings and solar thermal fuels with dendrimer structures.

RESUMO

BACKGROUND: Bladder cancer is a common malignancy characterized by a high recurrence rate and the development of drug resistance. Frequent mutations and gene expression alterations in the PI3K/AKT and mitogen-activated protein kinase-ERK pathways lead to deregulated cell growth and the acquisition of invasive properties, which facilitates tumour progression and confers resistance to chemotherapy. Therefore, identification of the underlying mechanisms that trigger the activation of these signalling pathways and control the invasive phenotype of tumour cells is of urgent need. METHODS: We utilized publicly available gene expression databases (GEO and TCGA) and bioinformatics analysis to identify key gene expression changes in human bladder cancer . The key gene expression was detected using BC tissue microarrays. Cell proliferation, apoptosis, migration, invasion and related signalling pathways were analysed flowing transfection with key gene overexpression plasmids. RESULTS: The analysis revealed that inhibited expression of the alpha-crystallin B chain was a common feature in all analysed datasets. The decrease in alpha-crystallin B expression was further confirmed at the protein level using BC tissue microarrays. Overexpression of alpha-crystallin B in T24 and J82 BC cell lines resulted in significant inhibition of tumour cell migration and invasion, which was associated with a decrease in PI3K, AKT and ERK activation. Moreover, alpha-crystallin B overexpression increased the expression of E-cadherin, while reducing the expression of N-cadherin, which indicated suppression of the epithelial-mesenchymal transition. CONCLUSIONS: Overall, the results of our study suggested that alpha-crystallin B may function as a tumour-suppressive factor in bladder cancer.

RESUMO

The aim of this study was to investigate the regulatory mechanism of cancer-associated fibroblasts (CAFs) exosome in bladder cancer (BC) cell proliferation and invasion. CAFs and normal fibroblasts (NFs) were isolated from tumor tissues and adjacent normal tissues of BC patients, and examined by immunocytochemistry for the expression of fibroblast activation protein alpha (FAP) and α-smooth muscle actin (α-SMA). Exosomes were extracted from CAFs and NFs and observed under a transmission electron microscope, and expression of the exosome markers CD9 and CD63 was confirmed by western blotting. The distribution and intensity of fluorescence were observed by confocal laser microscopy to analyze exosomes uptake by BC cell lines T24 or 5367. BC cell proliferation and invasion were detected by MTT and Transwell assays, respectively. LINC00355 levels in CAFs, NFs, CAFs exosome, NFs exosome, and BC cells were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Results showed that CAFs exosome significantly promoted BC cell proliferation and invasion relative to NFs exosome. LINC00355 expression was significantly elevated in CAFs exosome when compared with that in NFs-exosome. Up-regulated LINC00355 expression was observed both in T24 and 5367 cells co-incubated with CAFs exosome. Exosomes derived from LINC00355 siRNA-transfected CAFs observably repressed BC cell proliferation and invasion when compared with control siRNA-CAFs exosome. In conclusion, CAFs exosome-mediated transfer of LINC00355 regulates BC cell proliferation and invasion. Significance of the study. In this study, our data suggest that the exosomes released from CAFs promote BC cell proliferation and invasion. The mechanism of this effect is, at least in part, related to the increased LINC00355. Regulation of LINC00355 expression in exosomes released from CAFs might be a putative therapeutic strategy against the pathogenesis of BC.

RESUMO

The Ministry of Industry and Information Technology (MIIT) universities are important bases for science and technology research and play a critical role in China's National Innovation System. Based on the Web of Science (WoS), this article analyzes the statistics of paper published by MIIT universities and universities from across China including MIIT universities. The results are as follows: (1) Both the MIIT universities and universities nationwide in China have increased their international academic publications, and MIIT have shown a greater increase for the past decade. (2) In terms of U-I-G interaction, for UG relations, the Tug value of MIIT universities has remained stable, while that of universities in China has become declined. For UI relations, the Tui value of both MIIT universities and universities in China has shown steady growth. For UIG relations, MIIT universities have a greater synergistic effect of Triple Helix relationship than universities in China. (3) For more details in seven MIIT universities, universities elected into "Project 985", including HIT, BUAA, BIT and NPU, have published more papers, and been more synergistic with government and industry (UIG relations) than other three universities, including NUAA, NUST and HEU. Based on the empirical results, we discuss our findings, and make certain suggestions regarding policy incentives, reasonable administrative system and U-I-G interaction mode, which is significant not only for Chinese universities but also for universities in other developing countries.

RESUMO

Here, a novel triple-responsive graphene oxide hybrid supramolecular hydrogel based on the electrostatic self-assembly between graphene oxide and a quaternized polymer and the host-guest inclusion between α-cyclodextrins and poly(ethylene glycol) monomethyl ether (mPEG) was constructed. The quaternized polymer was synthesized by quaternization between pH-sensitive poly( N, N-dimethylaminoethyl methacrylate) and bromine end-capped poly(ethylene glycol) monomethyl ether. The supramolecular hydrogels prepared from the host-guest inclusion of poly(ethylene glycol) monomethyl ether and α-cyclodextrins would turn into a mobile sol phase when the temperature was increased above a certain temperature (Tgel-sol). Graphene oxide sheets not only acted as a core material to provide additional cross-linking but also absorbed NIR light and converted NIR light into heat to trigger the gel-sol transition. The constructed graphene oxide hybrid cyclodextrin-based supramolecular hydrogels could respond to NIR light, temperature, and pH, which could be beneficial for controlled release of cargoes and would hold great promise in the field of delivery systems.

RESUMO

Premature ejaculation (PE) is one of the most common sexual dysfunctions, which were associated with prostatitis-like symptoms (PLS). We intended to explore the prevalence of prostatitis-like symptoms and outcomes of National Institutes of Health-Chronic Prostatitis Symptom Index (NIH-CPSI) scores in outpatients with lifelong (LPE) and acquired premature ejaculation (APE). From December 2013 to December 2015, a total of 498 consecutive heterosexual men with PE and 322 male healthy subjects without PE were enrolled. Each of them completed a detailed questionnaire on demographics information, sexual and medical histories, and the NIH-CPSI. Assessment of NIH-CPSI and definition of PLS and PE were used to measure the PLS and NIH-CPSI scores and ejaculatory function for all subjects. Finally, a total of 820 subjects (including 498 men in PE group and 322 men in control group) were enrolled in our study. The mean ages were significantly different between PE and no PE groups. Men with PE reported worse PLS and higher NIH-CPSI scores (P < 0.001 for all). Similar findings were also observed between men with LPE and APE. Men with APE also reported higher rates of PLS and scores of NIH-CPSI (P < 0.001 for all). Multivariate analysis showed that PLS and NIH-CPSI scores were significantly associated with PE.

RESUMO

Fifteen cases of chyluria were diagnosed by Sudan staining test and the sides of chylous reflux were determined by cystoscopy. All patients underwent the modified laparoscopic technique. The operations had been successfully without unexpected injury. The postoperative urine chyle tests were negative in all patients. Recurrence developed in 1 patient during the follow-up. The modified technique does not require complete disconnection of perirenal fat tissue and fasciectomy diminishing necessity of nephropexy and preventing renal torsion. It is a feasible and effective surgical procedure for chyluria with a short operation time, minimal invasion and few complications.
