RESUMO

BACKGROUND: The treatment of metabolic system, cardiovascular system, and nervous system diseases remains to be explored. In the internal environment of organisms, the metabolism of substances such as carbohydrates, lipids and proteins (including biohormones and enzymes) exhibit a certain circadian rhythm to maintain the energy supply and material cycle needed for the normal activities of organisms. As a key factor for the health of organisms, the circadian rhythm can be disrupted by pathological conditions, and this disruption accelerates the progression of diseases and results in a vicious cycle. The current treatments targeting the circadian rhythm for the treatment of metabolic system, cardiovascular system, and nervous system diseases have certain limitations, and the identification of safer and more effective circadian rhythm regulators is needed. AIM OF THE REVIEW: To systematically assess the possibility of using the biological clock as a natural product target for disease intervention, this work reviews a range of evidence on the potential effectiveness of natural products targeting the circadian rhythm to protect against diseases of the metabolic system, cardiovascular system, and nervous system. This manuscript focuses on how natural products restore normal function by affecting the amplitude of the expression of circadian factors, sleep/wake cycles and the structure of the gut microbiota. KEY SCIENTIFIC CONCEPTS OF THE REVIEW: This work proposes that the circadian rhythm, which is regulated by the amplitude of the expression of circadian rhythm-related factors and the sleep/wake cycle, is crucial for diseases of the metabolic system, cardiovascular system and nervous system and is a new target for slowing the progression of diseases through the use of natural products. This manuscript provides a reference for the molecular modeling of natural products that target the circadian rhythm and provides a new perspective for the time-targeted action of drugs.

RESUMO

Utilizing electromagnetic hotspots within plasmonic nanogaps is a promising approach to create ultrasensitive surface-enhanced Raman scattering (SERS) substrates. However, it is difficult for many molecules to get positioned in such nanogaps. Metal-organic frameworks (MOFs) are commonly used to absorb and concentrate diverse molecules. Herein, we combine these two strategies by introducing MOFs into plasmon-coupled nanogaps, which has so far remained experimentally challenging. Ultrasensitive SERS substrates are fabricated through the construction of nanoparticle-on-mirror structures, where Au nanocrystals are encapsulated with a zeolitic imidazolate framework-8 (ZIF-8) shell and then coupled to a gold film. The ZIF-8 shell, as a spacer that separates the Au nanocrystal and the Au film, can be adjusted in thickness over a wide range, which allows the electric field enhancement and plasmon resonance wavelength to be varied. By trapping Raman-active molecules within the ZIF-8 shell, we show that our plasmon-coupled structures exhibit a superior SERS detection performance. A range of volatile organic compounds at the concentrations of 10-2 mg m-3 can be detected sensitively and reliably. Our study therefore offers an attractive route for synergistically combining plasmonic electric field enhancement and MOF-enabled molecular enrichment to design and create SERS substrates for ultrasensitive detection.

Metabolic-associated fatty liver disease (MAFLD) is a chronic liver disease characterized by the excessive accumulation of fat in hepatocytes. However, due to the complex pathogenesis of MAFLD, there are no officially approved drugs for treatment. Therefore, there is an urgent need to find safe and effective anti-MAFLD drugs. Recently, the relationship between the gut microbiota and MAFLD has been widely recognized, and treating MAFLD by regulating the gut microbiota may be a new therapeutic strategy. Natural products, especially plant natural products, have attracted much attention in the treatment of MAFLD due to their multiple targets and pathways and few side effects. Moreover, the structure and function of the gut microbiota can be influenced by exposure to plant natural products. However, the effects of plant natural products on MAFLD through targeting of the gut microbiota and the underlying mechanisms are poorly understood. Based on the above information and to address the potential therapeutic role of plant natural products in MAFLD, we systematically summarize the effects and mechanisms of action of plant natural products in the prevention and treatment of MAFLD through targeting of the gut microbiota. This narrative review provides feasible ideas for further exploration of safer and more effective natural drugs for the prevention and treatment of MAFLD.

Obesity is strongly associated with the occurrence and development of many types of cancers. Patients with obesity and cancer present with features of a disordered gut microbiota and metabolism, which may inhibit the physiological immune response to tumors and possibly damage immune cells in the tumor microenvironment. In recent years, bariatric surgery has become increasingly common and is recognized as an effective strategy for long-term weight loss; furthermore, bariatric surgery can induce favorable changes in the gut microbiota. Some studies have found that microbial metabolites, such as short-chain fatty acids (SCFAs), inosine bile acids and spermidine, play an important role in anticancer immunity. In this review, we describe the changes in microbial metabolites initiated by bariatric surgery and discuss the effects of these metabolites on anticancer immunity. This review attempts to clarify the relationship between alterations in microbial metabolites due to bariatric surgery and the effectiveness of cancer treatment. Furthermore, this review seeks to provide strategies for the development of microbial metabolites mimicking the benefits of bariatric surgery with the aim of improving therapeutic outcomes in cancer patients who have not received bariatric surgery.

Locally routing the exciton emissions in two-dimensional (2D) transition-metal dichalcogenides along different directions at the nanophotonic interface is of great interest in exploiting the promising 2D excitonic systems for functional nano-optical components. However, such control has remained elusive. Herein we report on a facile plasmonic approach for electrically controlled spatial modulation of the exciton emissions in a WS2 monolayer. The emission routing is enabled by the resonance coupling between the WS2 excitons and the multipole plasmon modes in individual silver nanorods placed on a WS2 monolayer. Different from prior demonstrations, the routing effect can be modulated by the doping level of the WS2 monolayer, enabling electrical control. Our work takes advantage of the high-quality plasmon modes supported by simple rod-shaped metal nanocrystals for the angularly resolved manipulation of 2D exciton emissions. Active control is achieved, which offers great opportunities for the development of nanoscale light sources and nanophotonic devices.

RESUMO

A novel and efficient strategy for the construction of difluorocarbonyl-oxindole and difluorocarbonyl-isoquinoline-1,3-dione derivatives involving nickel-catalyzed intramolecular Heck-type cyclizations followed by intermolecular cross-couplings has been developed. This approach features high functional group tolerance, broad substrate scope, and operational simplicity under mild conditions, thus providing a new method for the rapid difluorocarbonyl-functionalization of alkenes to construct the structurally diversified five- and six-membered heterocycles.

BACKGROUND: Several patients with erectile dysfunction do not accept or benefit from conventional therapy with phosphodiesterase type 5 inhibitors; thus, alternative and complementary therapies are in need. Traditional Chinese medicine has been treating erectile dysfunction in China, but its clinical value is inconclusive. OBJECTIVE: To systematically evaluate the efficacy and safety of traditional Chinese medicine in treating erectile dysfunction. METHODS: Randomized controlled trials were retrieved from a comprehensive search in the literature published in the past decade from the Web of Science, PubMed, Embase, Cochrane Library, SinoMed, China National Knowledge Internet, WanFang, and VIP. We performed a meta-analysis of the International Index of Erectile Function 5 questionnaire scores, clinical recovery rates, and testosterone levels using Review Manager 5.4 software. The trial sequential analysis was conducted to check the results. RESULTS: A total of 45 trials with 5016 patients were included. Meta-analysis results showed that traditional Chinese medicine effectively improved the International Index of Erectile Function 5 questionnaire scores (weighted mean difference = 3.78, 95% confidence interval: 3.12, 4.44; p < 0.001), clinical recovery rates (risk ratio = 1.57, 95% confidence interval: 1.38, 1.79; p < 0.001), testosterone levels (weighted mean difference = 2.42, 95% confidence interval: 1.59, 3.25; p < 0.001) compared with the controls. The single and add-on applications of traditional Chinese medicine could improve the International Index of Erectile Function 5 questionnaire score (p < 0.001). The trial sequential analysis confirmed the robustness of the analysis of the International Index of Erectile Function 5 questionnaire scores. A significant difference in the incidence of adverse effects between the treatment and control groups was not observed (risk ratio = 0.82, 95% confidence interval: 0.65, 1.05; p = 0.12). CONCLUSION: Traditional Chinese medicine can gain better responses in improving the International Index of Erectile Function 5 questionnaire scores, clinical recovery rates, and testosterone levels as an alternative and complementary treatment, with no increase in side effects. However, more standardized, long-term, traditional Chinese medicine and integrative therapy clinical trials are needed to support the clinical application of traditional Chinese medicine.

OBJECTIVE: To conduct a case series analysis on Professor Wang Qi's prescriptions for oligoasthenospermia and a preliminary discussion on the clinical application of the pathogenesis theory of "kidney deficiency with dampness, heat, stasis, toxin and parasites" so as to provide some reference for the diagnosis and treatment of oligoasthenospermia by traditional Chinese medicine (TCM). METHODS: We selected 13 representative cases of oligoasthenospermia treated by Professor Wang Qi, analyzed the basic conditions of the patients, changes of the disease and prescriptions used, and investigated the treatment of oligoasthenospermia in view of Professor Wang Qi's TCM theory on andrology. RESULTS: Excellent therapeutic effect was achieved in 7, indefinite effect in 5 and no effect in 1 of the 13 cases. Totally 38 prescriptions were employed in the treatment, of which Bajitian was used in 30, Tusizi in 27, Gouqizi in 25, Sangshen in 25, Shuizhi in 24 and Danggui in 22. The drugs used in the treatment of 10 or more patients included Bajitian in 12, Sangshen in 11, Shuizhi in 11, Tusizi in 10 and Gouqizi in 10 cases. CONCLUSIONS: Professor Wang Qi's prescriptions are effective for the treatment of oligoasthenospermia and the pathogenesis theory of "kidney deficiency with dampness, heat, stasis, toxin and parasites" is valuable in guiding the clinical diagnosis and treatment in traditional Chinese medicine.

Objective: The objective of this study was to investigate how knowledge and practice of coronavirus disease 2019 (COVID-19) prevention measures affected concerns about returning to work among supermarket staff. Attitudes about the ability of traditional Chinese medicine (TCM) to prevent COVID-19 were also assessed. Methods: A cross-sectional study was conducted in Huanggang, Hubei Province, China from April 23 to 25, 2020. Participants were invited to fill out an electronic questionnaire on their cell phones. Results: The results showed that from 2,309 valid questionnaires, 61.5% of participants were concerned about resuming work. Major concerns included asymptomatic infection (85.01%) and employees gathering in the workplace (78.96%). Multivariate logistic regression indicated that the female gender, having school-aged children and pregnancy were risk factors for being concerned about resuming work, while good knowledge and practice of preventive measures were protective factors. Knowledge and practice of preventive measures were positively correlated. Among preventive measures, the highest percentage of participants knew about wearing masks and washing hands. Meanwhile, 65.8% of participants expressed confidence in the ability of TCM to prevent COVID-19, where 74 and 51.3% thought there was a need and a strong need, respectively, for preventive TCM-based products. Among them, 71.5% preferred oral granules. Regarding TCM as a COVID-19 preventative, most were interested in information about safety and efficacy. Conclusion: These findings suggested that promoting knowledge and practices regarding COVID-19 prevention can help alleviate concerns about returning to work. Meanwhile, TCM can feasibly be accepted to diversify COVID-19 prevention methods. Clinical Trial Registration:http://www.chictr.org.cn/, identifier: ChiCTR2000031955.

OBJECTIVE: To investigate the role of miR129 in mediating the effect of chloroquine to enhance cisplatin- induced apoptosis in nasopharyngeal carcinoma cells (HNE1). METHODS: MTT assay was used to detect the viability of HNE1 cells treated with different concentrations of cisplatin. Colony formation of HNE1 cells treated with cisplatin and chloroquine, alone or in combination, was observed using crystal violet staining. BALB/C unde mice were inoculated with HNE1 cells and randomly divided into 4 groups with 6 mice in each group. The mice received intraperitoneal injections of cisplatin and chloroquine, alone or in combination once every 3 days for 4 consecutive weeks, and the tumor growth was observed in each group. The expression of miR129 in HNE1 cells treated with chloroquine, cisplatin, or both was detected with qPCR. The effects of miR129 suppression with a miR129 inhibitor on the expressions of autophagy related proteins p62, LC3B, Beclin1 and the drug-resistant related protein P-glycoprotein (P-gp) were examined using Western blotting in HNE1 cells treated with chloroquine, cisplatin, or both; the changes in cell apoptosis were detected Annexin V/PI double staining. RESULTS: Chloroquine combined with cisplatin significantly inhibited HNE1 cell proliferation in vitro and the growth of HNE1 cell-derived tumor in nude mice as compared with cisplatin alone (P < 0.01). In cultured HNE1 cells, inhibition of the expression of miR129 significantly promoted autophagy and up-regulated P-gp expression (P < 0.01); Chloroquine obviously inhibited cisplatin-induced autophagy and up-regulated the expression of miR129 in HNE1 cells (P < 0.01). Transfection of the cells with the miR129 inhibitor abolished the inhibitory effect of chloroquine on cisplatin-induced autophagy, and significantly increased the cell survival rate (P < 0.05) and lower the cell apoptotic rate (P < 0.01) after combined treatment with chloroquine and cisplatin. CONCLUSIONS: Chloroquine enhances the pro-apoptotic effect of cisplatin by up-regulating miR129 to inhibit autophagy and drug resistance in HNE1 cells.

It is of great significance to develop phase change materials (PCMs) with high performance. The reported PCMs usually possess serious defects like low heat capacity and poor thermal stability. Here, core-sheath structured nanofibers with polyvinyl butyral (PVB) as the sheath and octadecane as the core were fabricated by melt coaxial electrospinning. Pure octadecane without any solvents was used as the core solution, thus, the optimal sample possessed very high latent heat up to 118 J g-1. We studied the influence of core feed rate and PVB solution concentration on the encapsulation rate, and the highest encapsulation rate was found when the PVB concentration was 10% and core feed rate was 0.08 mL h-1. And hexagonal cesium tungsten bronze (Cs x WO3, a near infrared absorber) was introduced into the optimal sample partly to improve its conversion efficiency of solar to thermal energy, and partly absorb uncomfortable infrared light; the composite phase change material also possessed high latent heat up to 96.9 J g-1. In addition, 100 thermal cycle test proved that with a minor latent heat decrease, the prepared core-sheath structured smart nanofibers had good thermal stability, which overcomes the leakage problem of pure octadecane. Additionally, the 9 wt% Cs x WO3-loaded sample had an increase in tensile strength and elongation compared with the sample without Cs x WO3, indicating the good compatibility between Cs x WO3 and PVB.
