RESUMO

Circular aptamers are promising candidates for analytical and therapeutic applications due to their enhanced biological and structural stability. However, the process of circular aptamer selection remains a great challenge, as it requires multiple rounds of binding-separation-amplification that involves issues with nonspecific binding and amplification bias. Here, we develop a highly practical solution for reliable selection of circular aptamers in a single round based on magnetosome-like magnetic chain cross-linked graphene oxide (separation efficiency ≈ 105). High-affinity aptamer candidates can be rapidly selected from a preenriched circular DNA library, while low-affinity candidates are effectively adsorbed and separated by magnetosome-like magnetic chain cross-linked graphene oxide. With lipopolysaccharide as a representative model, the single-round selected lipopolysaccharide circular aptamer has been identified to have a high binding affinity with a Kd value of low to nanomolar range. Using this method, circular aptamers for protein and small-molecule targets were also successfully generated. We envision that this approach will accelerate the discovery of various new circular aptamers and open up a new avenue for analytical and therapeutic studies.

RESUMO

Behcet's disease (BD) is a chronic multi-systemic disease characterized by relapsing-remitting oral ulcers, genital ulcers, ocular inflammatory involvements, and numerous other systemic features. Ocular involvements are quite common in BD and may cause severe tissue damage and potentially blindness. Even though the pathogenesis of BD remains ambiguous, growing evidences have shown that genetic factors, environmental triggers and immunological abnormalities play significant roles in its development and progression. Novel biotherapies targeting IFN-γ, TNF-α and interleukins have been used in recent years. In this review, we mainly pay attention to the ocular involvement of BD, and discuss the current understanding of mechanisms and advances in therapeutic approaches, especially novel biologics. Finally, we discuss the management in patients with pregnancy.

Probiotic-based therapy is a promising approach, which can positively modulate bacterial composition and maintain homeostasis. However, exogenous probiotics are easily destroyed by harsh conditions in vivo; thus, their application prospects have been severely limited. Specifically, oxytetracycline (OTC), a broad-spectrum antibiotic widely used in aquaculture, results in adverse intestinal environments, such as dysbacteriosis, oxidative damage, and gut dysmotility. Here, we describe a facile method to apply glycol chitosan/alginate as armor on the surface of probiotics to effectively protect them from the changed enteric environments induced by OTC. The results demonstrated that the coated Lacticaseibacillus rhamnosus GG (LGG) for only 2 h administration could significantly improve the colonization rate of LGG, and the relative abundance of Lacticaseibacillus can reach 80% in OTC-treated larvae intestines. We also explored the specific mechanisms of the coated LGG to diminish reactive oxygen species (ROS) generation and rescue gut dysmotility for OTC treatment, including enhancing the activity of antioxidative enzymes (CAT, SOD and GPx) and increasing 5-HT synthesis. The mitigation effect of the coated LGG for 2 h administration was comparable to that of uncoated LGG for 24 h administration. Encapsulation of LGG with polysaccharides provides a unique application example for generating useful bacterial therapeutics in harsh intestinal environments.

OBJECTIVES: Our aim was to evaluate the renal response rate of patients with lupus nephritis (LN) undergoing standard treatment during a 2-year follow-up and to investigate its predictive factors. METHODS: A prospective cohort study that enrolled 56 clinically diagnosed LN patients with urinary protein positivity was carried out. All patients underwent standard treatment. All patients were followed up at 6-month intervals for 2 years. Data on renal response and clinical characteristics were collected and analyzed. RESULTS: Among 56 patients, 27 (48.2%) and 13 (23.2%) patients achieved complete renal response (CR) and partial renal response (PR) at 6 months after induction therapy, respectively, and 42 (75.0%) and 4 (7.1%) patients developed CR and PR at 2 years. Among patients who achieved PR at 6 months, 90.0% achieved CR at 24 months, while only 37.5% of the patients who were unresponsive at 6 months achieved CR. In the multivariable Cox proportional-hazards model, female (OR 6.51, 95% CI 1.23-34.52, p = 0.028), disease duration (OR 0.84, 95% CI 0.73-0.98, p = 0.021), achieving PR within 6 months (OR 8.09, 95% CI 2.06-31.73, p = 0.003), and urine protein/creatinine ratio (UPCR) (OR 0.998, 95% CI 0.996-1.000, p = 0.025) were found to be predictive factors of CR. CONCLUSION: A total of 48.2% of patients achieved CR at 6 months of induction therapy, and the response rates gradually increased to 60.7%, 64.3%, and 75.0% at 12, 18, and 24 months. Besides, female, disease duration, partial response within 6 months, and UPCR were predictive factors for a complete renal response. Key Points • We evaluate the renal response rates in Chinese patients with lupus nephritis in the real world for 2 years. • A total of 48.2% and 75.0% of patients achieved a complete response after standard treatment for 6 months and 2 years. • Female, disease duration, partial response within 6 months, and UPCR are predictors of complete renal response.

AIM: Genetic factors are believed to be implicated in the pathogenesis of relapsing polychondritis (RP). However, the molecular genetic determinants remain to be elucidated. This study aimed to detect the susceptibility genes of RP with whole-exome sequencing (WES) in a Chinese family and deepen our understanding of the pathogenesis of RP thereafter. METHOD: A 32-year-old Chinese female proband with RP and her family including her mother with RP were enrolled in the study. The genomic DNA of 6 human subjects was extracted from peripheral blood and then gene allele mutations were identified using WES. Candidate variants with low frequency (<0.1%) in the general population and predicted deleterious effects on gene function were identified. Sanger sequencing was applied subsequently to confirm the analyzed gene variants in 12 human blood samples. RESULTS: Nine single nucleotide polymorphism variants from different genes were identified to associate with RP by WES and further confirmed by Sanger sequencing, including Ring finger protein 207 (RNF207), collagen type XXII alpha 1 chain (COL22A1) rs200464636, glycosylphosphatidylinositol anchor attachment 1 (GPAA1) rs201424010, recQ like helicase 4 (RECQL4) rs757703895, folliculin (FLCN) NM_144606: c.G838A: p.E280K, DNA ligase 3 (LIG3) rs761808558, NM_207396: c.T425C:p.I142T, myosin heavy chain 15 (MYH15) NM_014981: c.G4462A: p.A1488T, purkinje cell protein 2 (PCP2) rs144974437 and coiled-coil domain containing 61 (CCDC61) rs777816675. CONCLUSIONS: This study suggests that coinheritance of multigene mutation may contribute to RP predisposition. The candidate genes mutated which we discovered are potential targets for in-depth functional studies.

The recommender system is widely used in the field of e-commerce and plays an important role in guiding customers to make smart decisions. Although many algorithms are available in the recommender system, collaborative filtering is still one of the most used and successful recommendation technologies. In collaborative filtering, similarity calculation is the main issue. In order to improve the accuracy and quality of recommendations, we proposed an improved similarity model, which takes three impact factors of similarity into account to minimize the deviation of similarity calculation. Compared with the traditional similarity measure, the advantages of our proposed model are that it makes full use of rating data and solves the problem of co-rated items. To validate the efficiency of the proposed algorithm, experiments were performed on four datasets. Results show that the proposed method can effectively improve the preferences of the recommender system and it is suitable for the sparsity data.

Inflammatory back pain (IBP) is an important clinical feature for axial spondyloarthritis (SpA). Yet, little is known about their prevalences in China. We conducted an epidemiological study in a university to detect the prevalences of IBP and axial SpA according to the Assessment of SpondyloArthritis International Society (ASAS) criteria. We investigated 3770 participants from South China Agricultural University by face-to-face questionnaires and evaluated the prevalences of chronic low back pain (CLBP) and IBP. In addition, 333 students including all IBP patients volunteered to do HLA-B27 test, and we performed X-ray examination on students with suspect axial SpA. Axial SpA was confirmed by rheumatologists according to ASAS criteria. The mean (± SD) age of screened population was 19.48 (± 2.80) years, while female to male ratio was 1.45:1 (2229/1541). Seven hundred thirty-one (19.39%) of all participants had CLBP and 111 (2.94%) had IBP. Among the 333 students receiving HLA-B27 test, 13 (0.34%, 13/3770) fulfilled ASAS criteria for axial SpA. Nine students had sacroiliitis on imaging plus at least one SpA feature (IBP and positive HLA-B27 results). Four students had positive HLA-B27 plus at least two other SpA features (arthritis/enthesitis and good response to NSAIDs). For CLBP, female/male was 485/246. For axial SpA, female/male was 4/9(P = 0.014). In southern China, the prevalences of CLBP and IBP were respectively 19.39 and 2.94% in university, and the prevalence of axial SpA was 0.34%. Although more female students had CLBP, males were more likely to suffer from axial SpA.
