RESUMO

The dataset available here comes from an experimental dairy cattle farm located in a mountain region in Central France, where the feeding systems are mostly pasture grazing combined with a period of indoor overwintering during the colder months. The dataset comprises individual productive and functional traits covering over 36 different variables in 185 primiparous Montbéliarde and Holstein cows, followed by data on productive longevity and reason for culling. The data was collected over a 20-year period during which animal husbandry and data collection protocols remained consistent. Potential re-users of the data are private-sector professionals, farmer associations, and researchers interested in developing statistical and mechanistic models and simulations of individual dairy cows under low-input grassland-based systems.

The experiment reported in this research paper aimed to evaluate the effects of high-starch or starch and oil-supplemented diets on rumen and faecal bacteria, and explore links between the structure of bacterial communities and milk fatty acid (FA) profiles. We used four Holstein dairy cows in a 4 × 4 Latin square design. Cows were fed a diet rich in cereals (high-starch diet with 23% starch content on dry matter (DM) basis), a diet supplemented with saturated FA from Ca salts of palm oil + 18% DM starch, a diet with high content of monounsaturated FA (from extruded rapeseeds) + 18% DM starch or a diet rich in polyunsaturated FA (from extruded sunflower seeds) + 17% DM starch. At the end of each experimental period, cows were sampled for rumen and faecal contents, which were used for DNA extraction and amplicon sequencing. Partial least squares (PLS) regression analysis highlighted diet-related changes in both rumen and faecal bacterial structures. Sparse PLS discriminant analysis was further employed to identify biologically relevant operational taxonomical units (OTUs) driving these differences. Our results show that Butyrivibrio discriminated the high-starch diet and linked positively with higher concentrations of milk odd- and branched-chain FA. YS2-related OTUs were key taxa distinguishing diets supplemented with Ca salts of palm oil or sunflower seeds and correlated positively with linoleic acid in milk. Similarly, diets modulated faecal bacterial composition. However, correlations between changes in faecal and rumen bacteria were poor. With this work, we demonstrated that high-starch or lipid-supplemented diets affect rumen and faecal bacterial community structure, and these changes could have a knock-on effect on milk FA profiles.

A previous study identified differences in rind aspects between Cantal-type cheeses manufactured from the same skimmed milk, supplemented with cream derived either from pasture-raised cows (P) or from cows fed with maize silage (M). Using an integrated analysis of multiomic data, the present study aimed at investigating potential correlations between cream origin and metagenomic, lipidomic and volatolomic profiles of these Cantal cheeses. Fungal and bacterial communities of cheese cores and rinds were characterized using DNA metabarcoding at different ripening times. Lipidome and volatolome were obtained from the previous study at the end of ripening. Rind microbial communities, especially fungal communities, were influenced by cream origin. Among bacteria, Brachybacterium were more abundant in P-derived cheeses than in M-derived cheeses after 90 and 150 days of ripening. Sporendonema casei, a yeast added as a ripening starter during Cantal manufacture, which contributes to rind typical aspect, had a lower relative abundance in P-derived cheeses after 150 days of ripening. Relative abundance of this fungus was highly negatively correlated with concentrations of C18 polyunsaturated fatty acids and to concentrations of particular volatile organic compounds, including 1-pentanol and 3-methyl-2-pentanol. Overall, these results evidenced original interactions between milk fat composition and the development of fungal communities in cheeses.

Metabolome profiling in biological fluids is an interesting approach for exploring markers of methane emissions in ruminants. In this study, a multiplatform metabolomics approach was used for investigating changes in milk metabolic profiles related to methanogenesis in dairy cows. For this purpose, 25 primiparous Holstein cows at similar lactation stage were fed the same diet supplemented with (treated, n = 12) or without (control, n = 13) a specific antimethanogenic additive that reduced enteric methane production by 23% with no changes in intake, milk production, and health status. The study lasted 6 wk, with sampling and measures performed in wk 5 and 6. Milk samples were analyzed using 4 complementary analytical methods, including 2 untargeted (nuclear magnetic resonance and liquid chromatography coupled to a quadrupole-time-of-flight mass spectrometer) and 2 targeted (liquid chromatography-tandem mass spectrometry and gas chromatography coupled to a flame ionization detector) approaches. After filtration, variable selection and normalization data from each analytical platform were then analyzed using multivariate orthogonal partial least square discriminant analysis. All 4 analytical methods were able to differentiate cows from treated and control groups. Overall, 38 discriminant metabolites were identified, which affected 10 metabolic pathways including methane metabolism. Some of these metabolites such as dimethylsulfoxide, dimethylsulfone, and citramalic acid, detected by nuclear magnetic resonance or liquid chromatography-mass spectrometry methods, originated from the rumen microbiota or had a microbial-host animal co-metabolism that could be associated with methanogenesis. Also, discriminant milk fatty acids detected by targeted gas chromatography were mostly of ruminal microbial origin. Other metabolites and metabolic pathways significantly affected were associated with AA metabolism. These findings provide new insight on the potential role of milk metabolites as indicators of enteric methane modifications in dairy cows.

The aims of this work were to study on dairy farm conditions: i) the repeatability of long-term enteric CH4 emissions measurement from lactating dairy cows using GreenFeed (GF); ii) the ranking of dairy cows according to their CH4 emissions across diets. Forty-five Holstein lactating dairy cows were randomly assigned to 3 equivalent groups at the beginning of their lactation. The experiment was composed of 3 successive periods: i) pre-experimental period (weeks 1 to 5) in which all cows received a common diet; ii) a dietary treatment transition period (weeks 6 to 10); and iii) an experimental period (weeks 11 to 26) in which each group was fed a different diet. Experimental diets were formulated to generate more or less CH4 production: i) a diet based on ryegrass silage and concentrates, low in starch and lipid, designed to induce high CH4 emissions (CH4+); ii) a diet based on maize silage and concentrates, rich in starch, designed to induce intermediate CH4 emissions (CH4int); iii) a diet based on maize silage and concentrates, rich in starch and lipid, designed to induce low CH4 emissions (CH4-). Gas emissions were individually measured using GF systems. Repeatability of gas emissions, dry matter intake (DMI) and dairy performances measurements was calculated from data averaged over 1, 2, 4, and 8 weeks for each animal. Hierarchical cluster analysis was performed to rank individual animals according to their CH4 emissions. No significant differences were observed for daily CH4 emissions (g/day) among diets, because of lower DMI of CH4+ cows. When CH4 emissions were referred to units of DMI or milk, the differences among diets emerged as significant and persistent over the observed period of lactation. Repeatability values of gas emissions measurements were higher than 0.7 averaged over 8 weeks of measurement, but still higher than 0.6 for CH4 g/day, CO2 g/day, CH4 g/kg milk, and CH4/CO2 even averaging only 2 weeks of measurement. The repeatability of CH4 emissions measurement was systematically lower than those of DMI or dairy performance parameters, like milk and FPCM yield, irrespective of the averaged measurement period. The dairy cow ranking was not stable over time between all individuals or within any of the diets. In our experimental conditions, the GF performance in the long term can be considered reliable in differentiating dairy herds by their CH4 emissions according to diets with different methanogenic potential, but did not allow the ranking of individual dairy cows within a same diet. Our data highlight the importance of phenotyping animals across environment in which they will be expected to perform.

The objective of this work is to compare the ability of three spectroscopy techniques: molecular fluorescence, near-infrared (NIR), and mid-infrared with attenuated total reflectance (MIR-ATR) spectroscopy to predict the concentrations of 8 carotenoids, 6 vitamins and 22 fatty acids (FA) in cow's milk. A dataset was built through the analysis of 242 frozen milk samples from different experiments. The milk compounds were analysed using reference methods and by NIR, MIR-ATR, and fluorescence to establish different predictive models. NIR spectroscopy allowed for better prediction of cis9-ß-carotene, ß-cryptoxanthin and the sum of carotenoids than the other techniques, with a coefficient of cross-validation in calibration (R2CV) > 0.60 and a coefficient of determination in validation (R2V) > 0.50. Their standard errors of prediction (SEP) were equal to 0.01, except for the sum of carotenoids (SEP = 0.15). However, MIR-ATR and fluorescence seem usable for the prediction of lutein and all-trans-ß-carotene, respectively. These three spectroscopy methods did not allow us to predict (R2CV < 0.30) vitamin contents except, for vitamin A (the best R²CV = 0.65 with NIR and SEP = 0.15) and α-tocopherol (the best R²CV = 0.56 with MIR-ATR and SEP = 0.41), but all R²V were <0.30. NIR spectroscopy yielded the best prediction of the selected milk FA.

Food safety crises involving persistent organic pollutants [POPs, e.g. dioxins, polychlorinated biphenyls (PCBs), organochlorine pesticides] lead to systematic slaughter of livestock to prevent their entry into the food chain. Therefore, there is a need to develop strategies to depurate livestock moderately contaminated with POPs in order to reduce such economic and social damages. This study aimed to test a POPs depuration strategy based on undernutrition (37% of energy requirements) combined with mineral oil (10% in total dry matter intake) in nine non-lactating ewes contaminated with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and PCBs 126 and 153. In order to better understand the underlying mechanisms of the depuration process, POPs kinetics and body lipids dynamics were followed concomitantly over 57-day of depuration in POPs storage (adipose tissue, AT), central distribution (blood) and excretion (faeces) compartments. Faecal POPs concentrations in underfed and mineral oil supplemented ewes increased by 2.0 to 2.6-fold, but not proportionally to lipids concentration which increased by 6-fold, compared to the control ewes. Nonetheless, after 57 days of depuration in undernutrition and mineral oil supplementation, AT POPs concentrations were 1.5 to 1.6-fold higher while serum concentrations remained unchanged compared to the control ewes. This was concomitant with a decrease by 2.7-fold of the AT estimated lipids weight along the depuration period. This reduction of the volume of the storage compartment combined with the increase of POPs faecal excretion in underfed and mineral oil supplemented ewes led to a reduction by 1.5-fold of the PCB 126 AT burden, while no changes were observed for TCDD and PCB 153 burdens (vs. no change for PCB 126 and increases for TCDD and PCB 153 AT burdens in control ewes). The original approach of this study combining the fine description at once of POPs kinetic and of body lipids dynamic improved our understanding of POPs fate in the ruminant.

Therefore, there is a need to develop strategies to depurate livestock moderately contaminated with POPs to reduce economic and social damage. This study aimed to test undernutrition (37% of energy requirements) combined with mineral oil (10% in total dry matter intake) in nine non-lactating ewes contaminated with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and polychlorinated biphenyls (PCBs) 126 and 153 as a strategy to enhance the depuration of POPs through faecal excretion. To better understand the underlying mechanisms of the depuration process, lipophilic POPs and lipid fluxes were co-monitored in various body and excretion compartments. Body compartments (adipose tissues, muscle, liver and blood) and the total empty body were analyzed for lipids and POPs concentrations and burdens at slaughter, as well as excretion compartments (faeces and wool) collected during the depuration period. Decreases in empty body total and lipid weights were 6-fold higher in underfed and supplemented ewes compared to control ewes. In addition, over the depuration period undernutrition and supplementation treatment increased faecal TCDD, PCBs 126 and 153 excretions by 1.4- to 2.1-fold but tended to decrease wool PCB 153 excretion by 1.4-fold. This induced 2- to 3-fold higher decreases in the empty body POPs burdens for underfed and supplemented ewes. Nonetheless, when expressed relative to the calculated initial empty body burdens, burdens at slaughter decreased only slightly from 97%, 103% and 98% for control ewes to 92%, 97% and 94% for underfed and supplemented ones, for TCDD, PCBs 126 and 153, respectively. Fine descriptions at once of POPs kinetic (companion paper 1) and mass balance (companion paper 2), and of body lipid dynamics were very useful in improving our understanding of the fate of POPs in the ruminants.

Direct-fed microbials (DFM) are considered as a promising technique to improve animal productivity without affecting animal health or harming the environment. The potential of three bacterial DFM to reduce methane (CH4) emissions, modulate ruminal fermentation, milk production and composition of primiparous dairy cows was examined in this study. As previous reports have shown that DFM respond differently to different diets, two contrasting diets were used in this study. Eight lactating primiparous cows were randomly divided into two groups that were fed a corn silage-based, high-starch diet (HSD) or a grass silage-based, high-fiber diet (HFD). Cows in each dietary group were randomly assigned to four treatments in a 4 × 4 Latin square design. The bacterial DFM used were selected for their proven CH4-reducing effect in vitro. Treatments included control (without DFM) and 3 DFM treatments: Propionibacterium freudenreichii 53-W (2.9 × 1010 colony forming units (CFU)/cow per day), Lactobacillus pentosus D31 (3.6 × 1011 CFU/cow per day) and Lactobacillus bulgaricus D1 (4.6 × 1010 CFU/cow per day). Each experimental period included 4 weeks of treatment and 1 week of wash-out, with measures performed in the fourth week of the treatment period. Enteric CH4 emissions were measured during 3 consecutive days using respiration chambers. Rumen samples were collected for ruminal fermentation parameters and quantitative microbial analyses. Milk samples were collected for composition analysis. Body weight of cows were recorded at the end of each treatment period. Irrespective of diet, no mitigating effect of DFM was observed on CH4 emissions in dairy cows. In contrast, Propionibacterium increased CH4 intensity by 27% (g CH4/kg milk) in cows fed HSD. There was no effect of DFM on other fermentation parameters and on bacterial, archaeal and protozoal numbers. Similarly, the effect of DFM on milk fatty acid composition was negligible. Propionibacterium and L. pentosus DFM tended to increase body weight gain with HSD. We conclude that, contrary to the effect previously observed in vitro, bacterial DFM Propionibacterium freudenreichii 53-W, Lactobacillus pentosus D31 and Lactobacillus bulgaricus D1 did not alter ruminal fermentation and failed to reduce CH4 emissions in lactating primiparous cows fed high-starch or high-fiber diets.

Although the effects of cow diet on cheese sensory properties have been well documented, the putative interactions between the biochemical and microbial milk components and their respective roles in the development of the sensory properties of cheeses have yet to be explored in depth. The aim of this study was to evaluate the specific contribution of milk fat composition to the formation of cheese sensory properties. Two creams with different fat compositions were obtained from cows fed either pasture or maize silage. Cheeses were manufactured from the same skim milk (identical chemical and microbial composition) with either the pasture- or maize silage-origin pasteurized cream added. The gross composition and microbial composition of milks did not vary with cream origin. In milks and cheeses, the fatty acid (FA) profiles were modified by the origin of the cream. The concentrations of C18:0 and unsaturated FA such as cis-9 C18:1, trans-11 C18:1, C18:3n-3, total conjugated linoleic acids, and mono- and polyunsaturated FA were higher in milks and cheeses with the pasture-origin cream than in those with the maize-origin cream. In contrast, the maize milks and cheeses had higher concentrations of short- and medium-chain saturated FA, C16:0, and C18:2n-6. The level of lipolysis was 11% in the cheese rind and only 0.30% in the cheese core. The rind of pasture cheeses had a higher concentration of free C18:0 and C18:3n-3 and a lower concentration of free C14:0 and free C16:0 than the rind of maize cheeses. The levels of major microbial groups were similar in pasture and maize cheeses at different stages of ripening. The pasture cheeses had a more elastic and creamier texture, a yellower color, and a thinner rind than the maize cheeses, but the odor and aroma of cheeses were not affected by the origin of the cream, despite a few modifications in the balance of volatile compounds from FA catabolism. Based on these results, we conclude that milk fat composition modulated by cow diet had a direct role in the texture of the cheese but no effect on flavor. The high degree of lipolysis in cheese rind, along with the higher concentration of long-chain unsaturated free FA in pasture cheeses may be responsible for antimicrobial activity, which could explain differences in the appearance of cheese rind.

n-3 highly unsaturated fatty acids (n-3 HUFA) directly and indirectly regulate lipid metabolism, energy balance and the inflammatory response. We investigated changes to the n-3 HUFA score of healthy adults, induced by different types and amounts of conjugated linoleic acid (CLA)-enriched (ENCH) cheeses consumed for different periods of time, compared to dietary fish oil (FO) pills (500 mg, each containing 100 mg of eicosapentaenoic and docosahexaenoic acids­EPA+DHA) or α-linolenic acid (ALA)-rich linseed oil (4 g, containing 2 g of ALA). A significant increase in the n-3 HUFA score was observed, in a dose-dependent manner, after administration of the FO supplement. In terms of the impact on the n-3 HUFA score, the intake of ENCH cheese (90 g/day) for two or four weeks was equivalent to the administration of one or two FO pills, respectively. Conversely, the linseed oil intake did not significantly impact the n-3 HUFA score. Feeding ENCH cheeses from different sources (bovine, ovine and caprine) for two months improved the n-3 HUFA score by increasing plasma DHA, and the effect was proportional to the CLA content in the cheese. We suggest that the improved n-3 HUFA score resulting from ENCH cheese intake may be attributed to increased peroxisome proliferator-activated receptor alpha (PPAR-α) activity. This study demonstrates that natural ENCH cheese is an alternative nutritional source of n-3 HUFA in humans.

The objectives of this study were to explore bacterial community assembly from cow teat skin to raw milk cheeses and to evaluate the role of farming systems on this assembly using 16S rRNA gene high-throughput sequencing. The two grazing systems studied (extensive vs. semi-extensive) had a greater effect on the microbiota of cow teat skin than on that of raw milks and cheeses. On teat skin, the relative abundance of several taxa at different taxonomic levels (Coriobacteriia, Bifidobacteriales, Corynebacteriales, Lachnospiraceae, Atopobium, and Clostridium) varied depending on the grazing system and the period (early or late summer). In cheese, the abundance of sub-dominant lactic acid bacteria (LAB) varied depending on the grazing system. Overall, 85% of OTUs detected in raw milks and 27% of OTUs detected in ripened cheeses were also found on cow teat skin. Several shared OTUs were assigned to taxa known to be involved in the development of cheese sensory characteristics, such as Micrococcales, Staphylococcaceae, and LAB. Our results highlight the key role of cow teat skin as a reservoir of microbial diversity for raw milk, and for the first time, that cow teat skin serves as a potential source of microorganisms found in raw-milk cheeses.

Consumption of milk and dairy products is important in Western industrialised countries. Fat content is an important constituent contributing to the nutritional quality of milk and dairy products. In order to improve the health of consumers, there is high interest in improving their fatty acid (FA) composition, which depends principally on rumen and mammary metabolism. This paper reviews the lipid metabolism in ruminants, with a particular focus on the production of trans and conjugated linoleic acids (CLA) and conjugated linolenic acids (CLnA) in the rumen. After the lipolysis of dietary lipids, an extensive biohydrogenation of unsaturated FA occurs by rumen bacteria, leading to numerous cis and trans isomers of 18:1, non-conjugated of 18:2, CLA and CLnA. The paper examines the different putative pathways of ruminal biohydrogenation of cis9-18:1, 18:2n-6, 18:3n-3 and long-chain FA and the bacteria implicated. Then mechanisms relative to the de novo mammary synthesis are presented. Ruminant diet is the main factor regulating the content and the composition of milk fat. Effects of nature of forage and lipid supplementation are analysed in cows and small ruminants species. Finally, the paper briefly presents the effects of these FA on animal models and human cell lines. We describe the properties of ruminant trans 18:1, when compared to industrial trans 18:1, CLA and CLnA on human health from meta-analyses of intervention studies and then explore the underlying mechanisms.

Near (NIR) and medium (MIR) infrared reflectance spectroscopy (IR) predictions of fatty acid (FA) composition, expressed as g/kg of milk or g/100g of FA, on fresh and thawed milk were compared. Two-hundred-and-fifty bulk cow milks, collected from 70 farms in northwest Italy, were scanned by MIR in liquid form and by NIR in liquid and oven-dried forms. MIR and NIR FA (g/100g FA) predictions on oven-dried milk were similar for the sum of even chain-saturated FA (ECSFA), odd chain-FA (OCFA), unsaturated FA (UFA), conjugated linoleic acid (CLA), n-3 FA, and C18:1cis9 to C16 ratio. The monounsaturated FA (MUFA), n-6 to n-3 ratio, polyunsaturated FA (PUFA), and n-6 FA were predicted better by NIR on oven-dried milk. The NIR showed worse predictions than MIR for almost all FA, when expressed as g/kg of milk. The NIR predictions on fresh liquid and oven-dried milk were similar, but the reliability decreased for thawed liquid milk. The high performance shown by NIR and MIR allows their use for routine milk FA composition recording.

In intensively reared dairy cows, milk fat secretion is reduced in response to high-concentrate diets and it is often referred to as the "milk fat depression" (MFD) syndrome. Some trans fatty acid (FA) isomers produced in the rumen of the cows, including t10,c12-18:2, are known for their inhibitory effect on mammary lipogenesis. To study whether this effect depends on the basal diet, duodenal infusions of t10,c12-18:2 were performed on cows fed four different diets (a factorial arrangement of forage:concentrate ratio and linseed oil supplementation). The overall response obtained with t10,c12-18:2 infusion was consistent with previous studies: a decrease in milk fat content and yield without significant variations in milk yield. Mean transfer efficiency of infused t10,c12-18:2 was 19.6%. However, the decrease in milk fat and FA yields (both de novo synthesis and preformed long-chain FA) was less pronounced in cows fed high-concentrate diets (-27% of the initial level), compared with cows fed low-concentrate diets (-42% of initial level). This difference was independent of dietary oil supplementation and milk FA yield before infusion. Results pertaining to effects of dietary forage:concentrate ratio were confirmed by statistical meta-analysis of data from previously published t10,c12-18:2 infusion experiments. This study shows that in cows fed MFD diets the mammary gland becomes more resistant to or experiences a lower response potential to further inhibition of lipogenesis and/or delta-9 desaturation of FA.

Based on the potential benefits to long-term human health there is interest in developing sustainable nutritional strategies for reducing saturated and increasing specific unsaturated fatty acids in ruminant milk. The impact of plant oil supplements to diets containing different forages on caprine milk fatty acid composition was examined in two experiments using twenty-seven Alpine goats in replicated 3 x 3 Latin squares with 28 d experimental periods. Treatments comprised of no oil (control) or 130 g/d of sunflower-seed oil (SO) or linseed oil (LO) supplements added to diets based on grass hay (H; experiment 1) or maize silage (M; experiment 2). Milk fat content was enhanced (P<0.01) on HSO, HLO and MLO compared with the corresponding H or M control diets, resulting in 17, 15 and 14% increases in milk fat secretion, respectively. For both experiments, plant oils decreased (P<0.05) milk 10:0-16:0 and odd- and branched-chain fatty acid content and increased 18:0, trans-Delta(6-9,11-14,16)-18:1 (and their corresponding Delta-9 desaturase products), trans-7, trans-9-conjugated linoleic acid (CLA), trans-9, trans-11-CLA and trans-8, cis-10-CLA concentrations. Alterations in the distribution of cis-18:1, trans-18:1, -18:2 and CLA isomers in milk fat were related to plant oil composition and forage in the diet. In conclusion, plant oils represent an effective strategy for altering the fatty acid composition of caprine milk, with evidence that the basal diet is an important determinant of ruminal unsaturated fatty acid metabolism in the goat.

The first objective of the present paper was to assess the potential of both isotopic ( (18)O/ (16)O in milk water) and molecular biomarkers (terpenes, fatty acids, carotenoids, and vitamins) and milk color to discriminate the production zone (lowland or upland areas) from which 49 tanker bulk milks were collected over one year from a total of 204 farms. The milk water (18)O enrichment was higher in lowland (<500 m altitude) than in upland (>700 m altitude), but the delta (18)O values failed to discriminate systematically the production zone at the scale of the year because of its high variability related to the sampling period. In contrast with vitamins A and E, carotenoids, and milk color measurements, terpenes and fatty acids were confirmed to be relevant tracers of the production zone. The milk compounds with the strongest discriminative potential were fatty acids, which were determined by high-resolution gas chromatography. The calculation of fatty acid ratios, which permits the limitation of using fatty acid relative quantity expressed in percentage of total fatty acids to be overcome, was shown to be particularly relevant in discriminating upland from lowland milk ratios. The selection of two pairs of ratios, namely, iso-C17:0/C18:3 n-3 and iso-C15:0/iso-C14:0, enabled the authentication of 100% of the highland versus lowland milks whatever the season. The second objective was to evaluate the relevance of fatty acid composition to discriminate milks according to the proportion of corn silage in the diets of dairy cows. The selection of two fatty acids ratios, namely, trans11 cis15-C18:2/trans11-C18:1 and cis9-C16:1/iso-C16:0, enabled the correct classification of 100% of the milk samples according to the proportion of corn silage in the basic fodder rations (<25% vs >30%). The relationship between the milk production zone and the type of forage fed to the cows is discussed.

The experiment was designed to study the effects of butters differing in conjugated linoleic acid (CLA) and trans 18:1 contents on lipoproteins associated with the risk of atherogenesis. New Zealand White male rabbits (9.6 weeks; 2.1 kg) were assigned for 6 or 12 weeks to three diets (n = 6 per diet) made of conventional pellets with 0.2% cholesterol and with 12% fat provided from a butter poor in trans-10 and trans-11 18:1 and in CLA (standard group), or rich in trans-10 18:1 (trans-10 18:1 group) or rich in trans-11 18:1 and in cis-9,trans-11 CLA (trans-11 18:1/CLA group). Blood samples were collected at the end of dietary treatments. Lipoproteins were separated by gradient-density ultracentrifugation. Lipid classes were determined enzymatically and apolipoproteins A-I and B by radial immunodiffusion. Mainly in the 12-week rabbits, higher plasma triglycerides and apolipoprotein B levels shown in the standard and trans-10 18:1 groups compared with those in the trans-11 18:1/CLA group are associated with higher plasma levels of very low density lipoproteins (VLDL) and low density lipoproteins (LDL) also shown in these two groups. In the 12-week rabbits, a shift towards denser LDL, considered as more atherogenic, was shown only in the trans-10 18:1 group. In these animals, the VLDL + LDL to HDL ratio was 1.7-2.3 times higher in the trans-10 18:1 group than in the other groups (P = 0.076). These results suggest a rather neutral effect of trans-11 18:1/CLA butter towards the risk of atherogenesis, whereas trans-10 18:1 butter would tend to be detrimental.

Experimental butters with a high content of trans-18 : 1 fatty acids and/or cis-9,trans-11-18 : 2 (rumenic acid; RA) were fed to thirty-six New Zealand White rabbits to investigate their effects on adipose tissue (AT) and liver lipogenic activities. Animals received one of three atherogenic (0.2 % cholesterol) diets containing 12 % butter with either a standard fatty acid composition (rich in saturated fatty acids), rich in trans-10-18 : 1 (T10 diet) or in trans-11-18 : 1 plus RA (T11+ RA diet) for 6 or 12 weeks. The ingestion of butters rich in trans fatty acids and/or RA for 6 weeks had little or no effect on liver and AT lipogenesis. The ingestion for 12 weeks of butter rich in T11+ RA decreased perirenal AT weight, lipogenic enzyme and lipoprotein lipase activities, without affecting liver lipid concentration or lipogenic activities except for a decrease in glycerol-3-phosphate dehydrogenase activity. Similar trends, but of a lower magnitude, were observed in rabbits fed the T10 diet for 12 weeks. Ingestion of the T10 or T11+ RA diets for 6 or 12 weeks had no significant effect on plasma metabolites and hormones except for glucose which increased at 6 weeks in the T10 group. Plasma leptin concentration was positively correlated with AT weight but did not differ between the three diets. In conclusion, the supply of butters rich in either T10 or T11+ RA in an atherogenic diet for 12 weeks decreased rabbit AT lipogenesis, with a more marked effect of the T11+RA diet, but had no effect on liver lipogenesis.

Trans fatty acids are suspected to be detrimental to health, particularly to cardiovascular function. Trans fatty acids include a wide range of fatty acids, with isomers of C18:1, conjugated and non-conjugated C18:2 as major components. A vaccenic acid (trans11-C18:1) + rumenic acid (cis9,trans11-CLA)-rich butter has been shown previously to exhibit health beneficial effects, but less is known concerning another trans-C18:1 present in hydrogenated vegetable oil-based products and sometimes in milk fat, the trans10-isomer. The present experiment was conducted to produce butters from milk of variable fatty acid composition for use in biomedical studies with rodents, with the overall aim of evaluating the specific effect of trans10-C18:1 and trans11-C18:1 + cis9,trans11-CLA on cardiovascular function. Milks from lactating dairy cows fed two types of maize-based diets supplemented (5% of dry matter)--or not--with sunflower oil were collected, and used to manufacture butters either rich in trans10-C18:1 (14% of total fatty acids, 64.5% of fat content) or rich in trans11-C18:1 + cis9,trans11-CLA (7.4 and 3.1% of total fatty acids, respectively, 68.5% of fat content), or with standard fatty acid composition (70% of fat content). Additionally, total saturated fatty acid percentage was reduced by more than one third in the enriched butters compared with the standard butter. An understanding of the role of nutrition on milk fatty acid composition in cows allows for the production of dairy products of variable lipid content and composition for use in biomedical studies in animal models and human subjects.
