Objective: Investigating the effect of different parameters of photobiomodulation (PBM) with low-power laser on multi-potent mesenchymal stem cells (MSCs) derived from adipose tissue in terms of proliferation and cell death. Methods: MSCs were submitted to PBM applications with combinations of the following physical parameters: control group (no intervention), wavelengths of 660 and 830 nm; energy of 0.5, 2, and 4 J; and power of 40 and 100 mW. MSC analysis was performed using MetaXpress® software at 24, 48, and 72 h. Results: Irradiation promoted a significant increase in cell proliferation (p < 0.05), with 830 nm laser, 100 mW, with energy of 0.5, 2, and 4 J in relation to the control group at all times. PBM with 660 nm, power of 40 mW, and energy of 0.5, 2, and 4 J produced greater cell death at 24 h compared with the control group. At the time of 72 h, there was no significant difference concerning cell death. Conclusions: According to the results found, we can conclude that both wavelengths were effective; however, the 830 nm laser was more effective in terms of cell proliferation compared with the 660 nm laser. The 660 nm wavelength showed a significant increase in cell death when compared with the 830 nm laser.

Due to its regenerative action, extracorporeal shock wave therapy (ESWT) is applied in treating integumentary and musculoskeletal diseases. However, other potential therapeutic interventions are being investigated. It is essential to fully understand its mitochondrial signaling pathway to achieve this, which plays a fundamental role in elucidating the mechanism of action and possible therapeutic interventions. Thus, this study aimed to analyze the effect of ESWT on mitochondrial pathways through the relationship between lipolysis and adipocyte apoptosis, as well as cellular functionality. This is a non-randomized case-control clinical trial where obese women received ESWT sessions in the abdominal region, after which tissue samples were collected for histological and immunohistochemical analyses of adipose tissue. The data demonstrated positivity in the expression of mitochondrial markers related to cell apoptosis, such as FIS1 (p < 0.0203) and OPA1 (p < 0.0283), in addition to the positivity of anti-MFN1, responsible for regulating mitochondrial cell proliferation (p < 0.0003). In summary, this study demonstrates that ESWT was able to activate specific mitochondrial signaling pathways, which may be associated with its ability to stimulate lipolysis and apoptosis in superficial adipose tissue. However, no significant improvements in cellular functionality were observed.

Low-level laser therapy (LLLT) is known for its ability to induce a photochemical process, primarily targeting mitochondria, a process referred to as photobiomodulation (PBM). Recently, its use has been attributed as an adjunct in obesity treatment, to stimulate lipolysis and apoptosis. However, the pathway of stimulation remains uncertain. Thus, the objective of this study was to understand whether mitochondrial stimulation occurs in adipose tissue cells after PBM therapy, which could lead to the processes of lipolysis and apoptosis. A non-randomized clinical trial was conducted using a split abdomen design in obese women who received red and infrared LED photobiomodulation therapy (PBMT). The patients underwent bariatric surgery, and adipose tissue samples were collected for immunohistochemical analysis with primary mitochondrial antibodies. Adipose tissue samples subjected to LED intervention exhibited positivity in mitochondrial antibodies for cAMP, DRP1, FAS, FIS1, MFN2, and OPA1 (p<0.001) compared to the control group. In conclusion, we observed that PBMT was capable of generating mitochondrial stimulation in adipose tissue cells, as evidenced by the positive antibody signals. This finding suggests that mitochondrial stimulation could be the mechanism and action underlying adipose tissue lipolysis and apoptosis.

OBJECTIVE: This study aimed to compare the effects of therapeutic ultrasound and paraffin with or without vacuum massage on the biomechanical properties of grafted skin after a burn. METHODS: A total of 44 patients with deep second- and third-degree burns, with a mean age of 35.89 (±11.53) years, who visited the Hospital Burn Unity, were included in the study. The therapeutic interventions were randomly defined by drawing lots, with a crossover design (crossover), and a minimum interval of 7 days (washout) between interventions. Skin biomechanical parameters such as distensibility (R0) and viscoelasticity (R6) were noninvasively evaluated by Cutometer before and after 0, 10, 20, and 30 min of intervention with therapeutic ultrasound and paraffin alone, as well as associated with negative pressure therapy of the skin (vacuum therapy). In this study, all groups showed increased distensibility (R0) in the period immediately after the application of the resources and a progressive reduction in the effects in the consecutive tests. Participants with skin grafts showed a decrease in viscoelasticity (R6) in all groups, except therapeutic paraffin and therapeutic ultrasound and vacuum massage. CONCLUSION: The biomechanical properties of grafted skin after a burn are altered after therapeutic intervention with ultrasound alone or associated with vacuum massage, such as intervention with paraffin associated with vacuum massage, for both parameters evaluated, skin distensibility (R0) and skin viscoelasticity (R6). However, the same did not occur for the intervention with isolated paraffin. There was no significant difference between the interventions therapeutic ultrasound and therapeutic paraffin.

Objectives: The objective of this study was to evaluate the immediate effect of manual therapy on ankle joint mobility and static balance in patients with diabetes. Methods: Forty patients, at a mean age of 59.35 ± 7.85, with type 2 diabetes mellitus and neurologic symptoms according to a Neuropathy Symptom Score protocol with amplitude, were included. The patients were divided into 2 groups: sham group and intervention group, which underwent manual manipulation intervention and 7-day follow-up. Joint range-of-motion analysis was performed using digital goniometry and static discharge of weights assessed by computerized baropodometry with open and closed eyes. The Shapiro-Wilk normality test was used to analyze the distribution. The data showed normal distribution, so the analysis of variance tests followed by Tukey's tests were used. SAS statistical software was used and the significance level was 5%. Results: The results of the intervention group showed an increase in the variable ankle goniometry over time compared to the sham group. The dorsiflexion movement on the right side obtained major gains over time; in addition, plantar flexion increased. Conclusion: Based on the participants evaluated in this study, manual therapy increased the ankle joint amplitude and improved the static balance in individuals with diabetes.

Caffeate 3-O-methyltransferase (COMT) catalyzes the methylation of the 3-hydroxyl group of caffeate to produce ferulate, an important precursor of the lignin biosynthesis. As a crucial drawback for biofuel production, lignin limits the enzymatic hydrolysis of polysaccharides to result in fermentable sugars. We hypothesized that a controlled inhibition of maize COMT can be an efficient approach to reduce ferulate and lignin, thus improving the saccharification process. First, we applied in silico techniques to prospect potential inhibitors of ZmaysCOMT, and the nitrocatechol entacapone was selected. Second, in vitro assays confirmed the inhibitory effect of entacapone on maize COMT. Finally, in vivo experiments revealed that entacapone reduced the contents of cell-wall-esterified hydroxycinnamates and increased saccharification of stems (18%) and leaves (70%), without negatively affecting maize growth and lignin biosynthesis. This non-genetically modified approach can be an alternative strategy to facilitate the enzymatic hydrolysis of biomass polysaccharides and increase saccharification for bioethanol production.

Biomimetically incorporated into the lignin structure, rosmarinic acid improves in vitro maize cell wall saccharification; however, no in planta studies have been performed. We hypothesized that rosmarinic acid, itself, could inducer saccharification without disturbing plant growth. Its effects on growth, enzymes of the phenylpropanoid pathway, lignin, monomeric composition, and saccharification of maize were evaluated. In a short-term (24 h) exposure, rosmarinic acid caused deleterious effects on maize roots, inhibiting the first enzymes of the phenylpropanoid pathway, phenylalanine ammonia-lyase and tyrosine ammonia-lyase, altering lignin composition and slightly increasing saccharification. In a long-term (14 d) exposure, rosmarinic acid increased saccharification of maize stems by about 50% without any deleterious effects on plant growth, the phenylpropanoid pathway and lignin formation. This demonstrated that exogenous application of rosmarinic acid on maize plants improved saccharification, and represented an interesting approach in facilitating enzymatic hydrolysis of biomass polysaccharides and increasing bioethanol production.

Grasses producing trans-aconitic acid, a geometric isomer of cis-aconitic acid, are often used in Glycine max rotation systems. However, the effects of trans-aconitic acid on Glycine max are unknown. We conducted a hydroponic experiment to evaluate the effects of 2.5-10 mM trans-aconitic acid on Glycine max growth and photosynthesis. The results revealed that the enhanced H2O2 production in the roots increased the membrane permeability and reduced the water uptake. These effects culminated with a reduced stomatal conductance (gs), which seems to be the main cause for a decreased photosynthetic rate (A). Due to low gs, the limited CO2 assimilation may have overexcited the photosystems, as indicated by the high production of H2O2 in leaves. After 96 h of incubation, and due to H2O2-induced damage to photosystems, a probable non-stomatal limitation for photosynthesis contributed to reducing A. This is corroborated by the significant decrease in the quantum yield of electron flow through photosystem II in vivo (ΦPSII) and the chlorophyll content. Taken together, the damage to the root system and photosynthetic apparatus caused by trans-aconitic acid significantly reduced the Glycine max plant growth.

OBJECTIVE: assess the effects of ischemic compression and kinesiotherapy on the rehabilitation of breast cancer survivors with chronic myofascial pain. METHODS: A randomized, controlled, blinded clinical trial was performed with 20 breast cancer survivors with myofascial trigger point in the upper trapezius muscle. Patients were randomly allocated to ischemic compression + kinesiotherapy (G1, n = 10) and kinesiotherapy (G2, n = 10). Both groups were submitted to 10 sessions of treatment. The variables evaluated were: Numeric Rating Scale, Pain Related Self-Statement Scale, pressure pain threshold, Functional Assessment of Cancer Therapy-Breast and Infrared thermography. RESULTS: A significant reduction (p < 0.05) was observed in pain intensity after 10 sessions in Groups 1 and 2, a significant increase (p < 0.05) in pressure pain threshold in both the operated and non-operated side after 10 sessions for Group 1. CONCLUSION: Ischemic compression associated with kinesiotherapy increases the pressure pain threshold on the myofascial trigger point in the upper trapezius muscle and reduces the intensity of pain in breast cancer survivors with myofascial pain.

The objective of the study was to evaluate the intra- and inter-rater reliability of bioimpedance in determining upper limb volume in women with lymphedema resulting from breast cancer treatment, as well as its correlation with the upper limb volume calculated by circumferential measurement. A blind cross-sectional study was performed in which 27 women (62.59 ± 10.50 years) were evaluated with upper limb lymphedema secondary to breast cancer treatment. Two examiners performed assessments in the same volunteers independently and twice, with an interval of 1 week between assessments. The collections were performed by the direct tetrapolar multifrequency segmental bioimpedance and by the circumference of the upper limb homolateral to the breast cancer. The results of the circumference showed that the homolateral limb had lymphedema. The intra- and inter-rater analysis showed excellent reliability with intraclass correlation coefficient (ICC) values ranging from 0.954 to 0.999 for the amount of liquid and excellent reliability with ICC values ranging from 0.852 to 0.999 for the electrical impedance of the upper limb with lymphedema. The correlation was strong and negative (p < 0.05) between intracellular, extracellular, and total water when associated with electrical impedance and moderate and positive (p < 0.05) when associated with upper limb volume for all frequencies. The correlations between upper limb volume and water quantities were moderate and positive (p < 0.05). The results indicate that bioimpedance is a reliable method for the evaluation of lymphedema, and the volume of the homolateral upper limb is associated with the amount of water in women with lymphedema secondary to the treatment of breast cancer.

We compared the amount of lignin as determined by the three most traditional methods for lignin measurement in three tissues (sugarcane bagasse, soybean roots and soybean seed coat) contrasting for lignin amount and composition. Although all methods presented high reproducibility, major inconsistencies among them were found. The amount of lignin determined by thioglycolic acid method was severely lower than that provided by the other methods (up to 95%) in all tissues analyzed. Klason method was quite similar to acetyl bromide in tissues containing higher amounts of lignin, but presented lower recovery of lignin in the less lignified tissue. To investigate the causes of the inconsistencies observed, we determined the monomer composition of all plant materials, but found no correlation. We found that the low recovery of lignin presented by the thioglycolic acid method were due losses of lignin in the residues disposed throughout the procedures. The production of furfurals by acetyl bromide method does not explain the differences observed. The acetyl bromide method is the simplest and fastest among the methods evaluated presenting similar or best recovery of lignin in all the tissues assessed.

Sä inúmeras as vantagens de audiçäo binaural, uma vez que proporciona melhor localizaçäo da fonte sonora, somaçäo binaural, eliminaçäo do efeito sombra da cabeça e, principalmente, melhora do reconhecimento de fala no ruído. Com base nesses fatos, ressaltamos a importância da adaptaçäo do aparelho de amplificaçäo sonora individual (AASI) nas deficiências auditivas unilaterais. Esse estudo teve como objetivo avaliar o desempenho do AASI nas diferentes situaçöes de vida diária, assim como, as possíveis queixas e satisfaçäo com a utilizaçäo do mesmo. Para issp, foi aplicado um questionário de auto-avaliaçäo em portadores de deficiência audutiva unilateral, de grau leve à profundo, usuários de AASI com tempo de adaptaçäo de seis meses a dois anos, atendidos no Centro de Distúrbios da Audiçäo, Linguagem e Visäo (CEDALVI), do Hospital de Reabilitaçäo de Anomalias Cranofaciais (HRACF-USP), em Bauru-SP. A maioria dos pacientes apontou o AASI como muito útil nas situaçöes diárias de comunicaçäo e encontravam-se satisfeitos com o uso do mesmo.
