Butter is among the most popular and commercially valuable dairy products. Its high commercial value makes it a major target for adulteration, which aims to reduce production costs by using lower-quality fats and oils from other sources. The annual global market is around USD 30 billion (2023), expected to reach USD 36 billion in 2028, which also justifies the enormous interest in adulteration. In this work, a confirmed case of butter adulteration was studied by Nuclear Magnetic Resonance (NMR) and Stable Carbon Isotopic Ratio Analysis (SCIRA) techniques, employed to detect the inclusion in butter production of vegetable oils, such as soybean and palm oils. A total of 21 samples seized by the Brazilian Federal Police were analysed by NMR and SCIR, and compared to original butter obtained from commercial sources. The composition of all the seized samples was a mixture of butter (dairy fat of animal origin) with fat of vegetable origin (soybean and palm oil) and did not contain milk as a major component. While NMR was an unequivocal choice to discriminate the chemical composition of food samples, identifying the short-chain saturated fatty acids present in milk fat, including the butyryl alkyl chain, SCIRA was able to discriminate the origin of fat present in the butter samples as C3 sources, such as palm vegetable oils.

In terrestrial forested ecosystems, fungi may interact with trees in at least three distinct ways: (i) associated with roots as symbionts; (ii) as pathogens in roots, trunks, leaves, flowers, and fruits; or (iii) decomposing dead tree tissues on soil or even on dead tissues in living trees. Distinguishing the latter two nutrition modes is rather difficult in Hymenochaetaceae (Basidiomycota) species. Herein, we have used an integrative approach of comparative genomics, stable isotopes, host tree association, and bioclimatic data to investigate the lifestyle ecology of the scarcely known neotropical genus Phellinotus, focusing on the unique species Phellinotus piptadeniae. This species is strongly associated with living Piptadenia gonoacantha (Fabaceae) trees in the Atlantic Forest domain on a relatively high precipitation gradient. Phylogenomics resolved P. piptadeniae in a clade that also includes both plant pathogens and typical wood saprotrophs. Furthermore, both genome-predicted Carbohydrate-Active Enzymes (CAZy) and stable isotopes (δ13C and δ15N) revealed a rather flexible lifestyle for the species. Altogether, our findings suggest that P. piptadeniae has been undergoing a pathotrophic specialization in a particular tree species while maintaining all the metabolic repertoire of a wood saprothroph. IMPORTANCE: This is the first genomic description for Phellinotus piptadeniae. This basidiomycete is found across a broad range of climates and ecosystems in South America, including regions threatened by extensive agriculture. This fungus is also relevant considering its pathotrophic-saprotrophic association with Piptadenia goanocantha, which we began to understand with these new results that locate this species among biotrophic and necrotrophic fungi.

Patients with polycythaemia vera (PV) are at increased risk of thrombosis and haemorrhages. Although hydroxyurea (HU) has been the frontline therapy for patients at high risk of vascular complications, about 25% of patients develop resistance/intolerance to this therapy. The aim of this non-interventional, multicentre cohort study was to understand the clinical characteristics and HU treatment response of Portuguese PV patients. HU resistance/intolerance was defined according to adjusted European LeukemiaNet (ELN) criteria. In total, 134 PV patients with a mean (SD) disease duration of 4.8 (5.0) years were included and followed up for 2 years. At baseline, most patients were ≥60 years old (83.2%), at high risk for thrombotic events (87.2%), and receiving HU therapy (79.1%). A total of 10 thrombotic events and 8 haemorrhagic events were reported, resulting in a 5-year probability of thrombo-haemorrhagic events of 17.2%. Haematocrit (p = 0.007), haemoglobin (p = 0.012) and MPN10 symptom score (12.0 (11.6) vs. 10.3 (9.1); p = 0.041) decreased significantly at the 24-month visit compared to baseline. Overall, 75.9% of patients met at least one of the adjusted ELN criteria for HU resistance, and 14.4% of patients remained on HU throughout the study. The results from this real-world study may help identify the subset of patients at higher risk for disease sequelae who may benefit from earlier second-line treatment.

Higher Education Institutions' (HEI) workers were highly impacted by the COVID-19 pandemic, which magnified gender differences in terms of management of work and personal life. Most studies published so far have primarily focused on a group of HEI workers' (i.e., teachers and researchers), but not on staff members, despite their crucial role for HEI functioning. Following the Job Demands-Resources theory, we aimed to: (i) characterize work-life conflict (WLC) among men and women workers from an HEI (staff and teachers/researchers) during the COVID-19 pandemic; and (ii) explore the major predictors of WLC for both staff and teachers/researchers. This study includes a sample of 262 workers from one Portuguese HEI (n = 128 staff members; n = 134 teachers/researchers) who answered an online survey. An Independent Samples T-Test showed that the reported current WLC was significantly higher for teachers/researchers compared to staff. Moreover, women teachers/researchers showed higher WLC than men. Additionally, using a Repeated Measures ANOVA, we found that the increase in the reported levels of WLC (before the pandemic and currently) was significantly more prominent among teachers/researchers than in the staff group. Regarding the predictors of WLC for both groups separately, a Multiple Hierarchical Linear Regression showed that role overload, conceptualized as a demand, was a predictor for both staff and teachers/researchers. As for potential resources, work dedication negatively predicted WLC for staff, whereas family-friendly organization perceptions predicted less WLC for teachers/researchers. These results highlight the importance of understanding HEIs holistically, by considering workers' individual characteristics such as gender, but also distinct careers inside the institutions. As most European HEIs are currently making active efforts to promote gender-equal academic workplaces, these findings may help them design tailored and effective measures to address employees' work-life balance issues, not only considering gender, but also the different types of demands associated with each group of workers within HEIs.

O artigo pretende fazer a leitura de textos que marcaram a história da teoria da Gestalt nas suas fases iniciais de desenvolvimento. Assim, procura redescobrir as origens do termo a partir de conceitos como Tongestalt, de Ernst Mach, e Gestaltqualitäten, de Christian von Ehrenfels, para alcançar as raízes filosóficas da psicologia da Gestalt. Em seguida, focando sobre a base da psicologia da Gestalt, a pesquisa se dedica aos fatores da Gestalt descritos por Max Wertheimer como leis gerais da teoria da Gestalt, à introdução da Gestalt theory nos Estados Unidos por Kurt Koffka a partir da apresentação do ponto de vista da Gestalt sobre a teoria da percepção e ao esclarecimento sobre o conceito de insight por Wolfgang Köhler por meio da abordagem exaustiva da perspectiva do leigo. Logo, o texto tenta ser capaz de dar mostras da evolução do conceito de Gestalt ao longo do tempo e ao mesmo tempo conectar uma variedade de definições e facetas da Gestalt.


The article intends to read texts that marked the history of Gestalt theory in its early stages of development. Thus, it seeks to rediscover the origins of the term from concepts such as Ernst Mach's Tongestalt and Christian von Ehrenfels's Gestaltqualitäten in order to reach the philosophical roots of Gestalt psychology. Next, focusing on the basis of Gestalt psychology, the research focuses on the Gestalt factors described by Max Wertheimer as general laws of the Gestalt theory, the introduction of Gestalt theory into the United States by Kurt Koffka from the presentation of the point of view of Gestalt over the theory of perception, and the clarification on the concept of insight by Wolfgang Köhler by the means of an exhaustive approach in layman's terms. So the text tries to be able to show off the evolution of the concept of Gestalt over time and at the same time to connect a variety of definitions and facets of Gestalt.


El artículo pretende leer textos que marcaron la historia de la teoría de la Gestalt en sus primeras etapas de desarrollo. Así, busca redescubrir los orígenes del término a partir de conceptos como Tongestalt de Ernst Mach y Gestaltqualitäten de Christian von Ehrenfels para llegar a las raíces filosóficas de la psicología de la Gestalt. A continuación, centrándose en la psicología de la Gestalt, la investigación se concentra en los factores de la Gestalt descritos por Max Wertheimer como leyes generales de la teoría de la Gestalt, la introducción de la Gestalt-theorie en Estados Unidos por Kurt Koffka a partir de la presentación del punto de vista de la Gestalt sobre la teoría de la percepción y la clarificación acerca del concepto de insight por Wolfgang Köhler por medio de un enfoque exhaustivo en términos legos. Food fraud is still a recurrent practice throughout food supply chains. In the case of seafood, misidentification of species and products repackaging constitute the most common frauds. Therefore, the development of appropriate analytical approaches to be used against food fraud is necessary. The present study goal is to explore for the first time, the possibility to differentiate between Sparus aurata from two different mariculture farms located in Madeira Island (Caniçal and Ribeira Brava), using the mass fingerprint of fish mucus obtained from MALDI-TOF MS and analyzed using Mass-UP software for multivariate statistical analysis and biomarker identification. It was possible to establish, from the mucus protein fraction, a set of potential biomarkers for each location in a total of 35 peaks, being 17 peaks specific to Caniçal located farm and 18 to Ribeira Brava. The proposed analytical approach revealed a useful strategy providing accurate and fast results for fish geographical origin discrimination.

Sugarcane honey (SCH) is a syrup produced on Madeira Island and recognized by its unique aroma, a complex attribute of quality with an important influence on the final consumer's acceptance of the product, and determined by a complex mixture of a large number of volatile organic compounds (VOCs) generated during its traditional making process and storage. Therefore, the purpose of this study was to establish the volatile profile of genuine SCH produced by a regional certified producer for seven years and compare it with syrups from non-certified regional producers and with producers from different geographical regions (Spain, Egypt, Brazil and Australia), as a powerful strategy to define the volatomic fingerprint of SCH. Different volatile profiles were recognized for all samples, with 166 VOCs being identified belonging to different chemical classes, including furans, ketones, carboxylic acids, aldehydes and alcohols. Chemometric analysis allowed (i) the differentiation between all syrups, being more pronounced between SCH and other syrups; and (ii) the identification of 32 VOCs as potential markers for the traceability and authenticity of SCH on the global market.

Sugarcane honey (SCH) is a syrup from Madeira Island recognized by its unique and excellent aroma, associated to volatile organic compounds (VOCs) generated during the well-defined five stages of its traditional making process. The establishment of volatile profile throughout all SCH-making stages during four years, allowed the evaluation of the influence of each stage in the typical characterisitcs of SCH. One hundred eighthy seven VOCs were identified, being associated to several origins and formation pathways. VOCs formed during stage 1 and 2 were originate from raw material, and its oxidation (i.e. enzymatic browning) and thermal degradation (i.e. lipid oxidation, Maillard reactions, Strecker degradation). In stage 3 and 4, the caramelization and melanoidin degradation also occurred, while in stage 5, the thermal degradation continues, followed by microbial activity. Chemometric analysis allowed to identify 35 VOCs as potential markers for processing control by the producers and as guarantee of the typicality and authenticity of SCH. Based on the obtained results, we propose for the first time an innovative schematic diagram explaining the potential reactions and pathways for VOCs formation during the different steps of the SCH production.

Sugarcane (SC) is a perennial grass widely cultivated in tropical and subtropical regions. However, its cultivation in Europe is residual, where Madeira Island, Portugal, is the only region where SC continues to be extensively cultivated. For the first time, the volatile profiles of regional cultivars were established by solid-phase microextraction combined with gas chromatography-mass spectrometry. Different volatile profiles for each cultivar were recognized, identifying 260 volatile organic compounds belonging to 15 chemical classes, such as aldehydes, alcohols, ketones, hydrocarbons, esters, and terpenes. Chemometric analysis procedure, namely, one-way ANOVA with Tukey's test, principal component analysis, partial least-square analysis, linear discriminant analysis, and hierarchical clustering analysis, allowed the differentiation between all regional cultivars. This study represents an important contribution for the maintenance of biodiversity and subsistence of the SC industry in Europe. Furthermore, it is also a valuable contribution to establish the typicality of traditional SC-based products, such as SC honey.

Paciente masculino, de 32 años, acudió a la Clínica de Prótesis Parcial Fija (PPF), de la Facultad de Odontología de Araçatuba-Universidad Estadual Paulista, manifestando como queja principal que sus dientes anteriores estaban "feos y rotos". Después del exa- men clínico, el paciente fue diagnosticado con desorden temporomandibular, bruxismo y presencia de erosiones dentarias (intrínsecas y extrínsecas). Se propuso el acompaña- miento de un médico gastroenterólogo y la rehabilitación con prótesis libres de metal (11, 12, 21 y 22) confeccionadas en silicato de litio reforzado con zirconio por el sistema CAD/CAM. Después de la endodoncia de los incisivos antero-superiores, fueron insta- lados los pernos de fibra de vidrio anatomizados con resina compuesta; la impresión fue realizada con hilo retractor y silicona de adición. Después de la prueba estética y ajustes oclusales, las PPFs de e-max Ceram, fueron preparadas para cementación resinosa con el sistema Variolink II color Light. Después de 1 semana, se tomó la impresión de la arcada superior y posteriormente se le instaló una placa miorrelajante. El paciente y los profesionales involucrados aprobaron el resultado final, comprobando la eficacia estética asociada al uso de prótesis libre de metal, a pesar de que el paciente presenta compromiso gastroesofágico y desorden temporomandibular.


A 32-year-old male patient attended in the Fixed Partial Prosthesis Clinic (PPF), of the Araçatuba Dental School-Paulista State University, stating as the main complaint that his anterior teeth were "ugly and broken". After clinical examination, the patient was diagnosed with temporomandibular disorder, bruxism and the presence of dental ero- sions (intrinsic and extrinsic). It was proposed to be followed up by a gastroenterologist and rehabilitation with metal-free prostheses (11, 12, 21 and 22) made of lithium silicate reinforced with zirconium by the CAD / CAM system were proposed. After endodontics of the anterior-superior incisors, the anatomized fiberglass posts with composite resin were installed; the impression was made with retractor wire and addition silicone. After aesthetic testing and occlusal adjustments, the e-max Ceram PPF ̈s were prepared for resin cementation with the Variolink II color Light system. After 1 week, the impression of the upper arch was taken and subsequently a muscle relaxant plate was installed. The patient and the professionals involved approved the final result, verifying the aesthetic efficacy associated with the use of a metal-free prosthesis, despite the fact that the patient has gastroesophageal involvement and temporomandibular disorder.

The volatile amines trimethylamine (TMA) and dimethylamine (DMA) could be used as important spoilage indices for seafood products, assisting in the determination of the rejection period. In the present study, a systematic analytical duality-by-design (AQbD) approach was used as a powerful strategy to optimize the most important experimental parameters of headspace solid-phase microextraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS) conditions for the quantification of TMA and DMA in Sparus aurata. This optimization enabled the selection of the best points in the method operable design region for HS-SPME extraction (30 min; 35 °C; NaOH 15 M and NaCl 35%, w/v) and GC-MS analysis (80 °C; gradient 50 °C/min; flow rate 1 mL/min and splitless mode). The rejection day, estimated through the TMA concentration (>12 mg/100 g, at days 9-10), was compared with sensory (quality index method: day 7-8), physical (Torrymeter: day 8-9), and microbial (day 9-10) analysis, corroborating the suitability of the proposed approach for estimating the period for which they will retain an acceptable level of eating quality from a safety and sensory perspective.

One of the main goals of the Spanish and Portuguese-Speaking Working Group of the International Society for Forensic Genetics (GHEP-ISFG) is to promote and contribute to the development and dissemination of scientific knowledge in the field of forensic genetics. The GHEP-ISFG supports several Working Commissions which develop different scientific activities. One of them, the Working Commission on "Massively Parallel Sequencing (MPS): Forensic Applications", organized its first collaborative exercise on forensic applications of MPS technology in 2019. The aim of this exercise was to assess the concordance between the MPS results and those obtained with conventional technologies (capillary electrophoresis and Sanger sequencing), as well as to compare the results obtained within the different MPS platforms and/or the different kits/panels and analysis software packages (commercial and open-access) available on the market. The seven participating laboratories analyzed some samples of the annual GHEP-ISFG proficiency test (EIADN No. 27 (2019)), using Ion Torrent™ or MiSeq FGx® platforms. Six of them sent autosomal STR sequence data, five laboratories performed MPS analysis of individual identification SNPs, four laboratories reported MPS data of Y-chromosomal STRs, and X-chromosomal STRs, three laboratories performed MPS analysis of ancestry informative SNPs and phenotype informative SNPs, two labs performed MPS analysis of the mitochondrial DNA control region, and only one lab produced MPS data of lineage informative SNPs. Autosomal STR sequencing results were highly concordant to the consensus obtained by capillary electrophoresis in the EIADN No. 27 (2019) exercise. Furthermore, in general, a high level of concordance was observed between the results of the participating laboratories, regardless of the platform used. The main discordances were due to errors during the analysis process or from sequence data obtained with low depth of coverage. In this paper we highlight some issues that still arise, such as standardization of the nomenclature for STRs analyzed by sequencing with MPS, the universal uptake of a nomenclature framework by the analysis software, and well established validation and accreditation of the new MPS platforms for use in routine forensic case-work.

OBJECTIVE: To evaluate the long-term color stainability, translucency, and contrast ratio (CR) of different resins used to fabricate interim prostheses after immersion in acidic/staining solutions. MATERIALS AND METHODS: A total of 160 specimens were divided into 16 groups (n = 10) according to the material (heat-polymerized acrylic resin [HPAR], auto-polymerized acrylic resin [APR], nanoparticulated bis-acrylic resin [BR], and prefabricated poly(methyl methacrylate) block for CAD/CAM [CADR]) and immersion solutions (artificial saliva, cola beverage, coffee, and red wine). A spectrophotometer was used before and after each immersion period (7, 14, 28, 90, and 180 days). Color differences (CIEDE2000 and CIELab) were calculated. A three-way repeated-measures analysis of variance and Bonferroni test (α = .05) were used. RESULTS: After 180 days, the APR presented the highest value for coffee and the CADR presented the smallest value for the cola (P < .001). For the CR, the highest values were obtained at 180 days for BR in coffee (1.35) and wine (1.18) (P < .001). Higher translucency parameters were obtained in the BR and CADR in the initial, 14, 28, and 90 days (P < .001). CONCLUSIONS: From the greatest to the smallest staining potential, the solutions were classified as: wine > coffee > cola beverage > saliva, while for the materials as: APR > BR > HPAR > CADR. CLINICAL SIGNIFICANCE: The knowledge of the long-term optical behavior of interim prosthetic materials is important for clinicians to decide which material to use to match the dietary intake of their patients and their esthetic demands. Prefabricated blocks for CAD/CAM systems have been shown to maintaining their optical characteristics even after a long period of immersion in acidic/staining solutions.

Fish and fish-based products are easily perishable foods due to different factors, including fragile organization, abundant endo-enzymes, psychrophilic bacteria, and impact of pre-harvest operations, that contribute to reducing its value. Therefore, a timely effective method for fish freshness and shelf-life evaluation is important. In this context, this study aimed to develop a sensory scheme based on the Quality Index Method (QIM) (sensory table and point system) for freshness monitorization and shelf-life prediction for Seriola dumerili from aquaculture in Madeira Island. Evaluation of appearance, texture, eyes, and gills was performed during 20 days of storage on ice (0 ± 1 °C). The shelf-life prediction was supported by the analysis of microorganisms (total viable colonies, TVC, counts), texture (Torrymeter), and production of trimethylamine (TMA), evaluated by HS-SPME-GC-MS and validated according to Association of Official Analytical Chemists AOAC guidelines. The result is a QIM scheme with 25 demerit points, where zero indicates total freshness. From the integration of sensory analysis, microbial growth at the time of rejection (TVC, 108 cfu/cm2 and H2S producers, 107 cfu/cm2), texture (Torrymeter value < 8), and TMA analyses (>12.5 mg/100 g), shelf-life was estimated as 12 days (±0.5 days). The obtained results show the high-throughput potential of the developed method for fish freshness assessment and shelf-life prediction. This QIM scheme is a secure way to measure quality and provide users with a reliable standardized fish freshness measure.

RESUMO

Brazilians form one of the most heterogeneous populations in the world, as the result of five centuries of miscegenation between its native populations with migrants from Europe, Africa and Asia. The present study intended to characterize the frequencies of mtDNA haplotypes in a dataset of 306 individuals from Brasília, Federal District of Brazil. Brasília was built from scratch in the late 1950s and its construction attracted migrants from different regions of Brazil, mostly from Central-West, Northeast and Southeast regions. Due to its formation, its population is admixed. The goal of this study was to collect mtDNA population data and contribute to databases for a better use of mtDNA for forensic purposes. The haplotypes are available at EMPOP website under accession number EMP00695.

Recently, Brazilian Federal Police used forensic chemistry and forensic botany techniques on a case. Two packets containing fragmented plant matter were seized and sent for forensic analysis. Forensic chemistry, the gold standard for evaluating plant material suspected to contain illicit substances, did not find illicit materials. Gas chromatography coupled mass spectrometry (GC-MS) identified thujone in the botanical material. Thujone is a chemical compound naturally found in many plant species, notably Artemisia absinthium. Because doubt remained, we next used plant DNA barcoding methods. Total DNA from plant tissue fragments was extracted and five different DNA regions were amplified, sequenced, and analyzed using plant DNA barcoding methods. Genetic analysis yielded 30 good quality sequences representing five taxa. Most specimens were identified as A. absinthium. Few studies focus on practical forensic applications of plant DNA barcoding methods using a case solved in a forensic laboratory with its difficulties and limitations. To the best of our knowledge, this is the first study to report an effective joint effort of forensic chemistry and botany techniques to assess plant material in Brazil. The availability of a new technical approach for the genetic sequencing of plant species will enhance many forensic investigations and inspire similar initiatives.

Sotolon is widely associated with the quality of fortified aged wines, and has also been linked to premature oxidative aging (premox). Here we developed a single, fast and environmental-friendly microextraction by packed sorbent ultra-high pressure liquid chromatography analysis (MEPS/UHPLC-PDA) for sotolon quantification in different wines. The best extraction conditions (loading three times 250µL samples through the MEPSPEP sorbent and elution with 100µL of 50% MeOH) were combined with a fast UHPLC separation (5min separation using acidified 10% MeOH isocratic flow in a CORTECS C18 column) to allow unparalleled minimum sample and solvents volumes usage. The validated methodology showed good linearity (r(2)>0.993) and precision (<5.6%); high recovery (>81%) and detection limits (0.45-2.51µg/L) far below sotolon odor threshold for any type of wine. The methodology was successfully applied to selected white table and Madeira wines, encompassing therefore a wide range of alcohol and sugar contents. Furthermore, as far we may know, this is the first time a single methodology can be used to assess both wine aging or premox according to the type of wine.
