BACKGROUND: Biogenic amines (BA) are hazardous components in Huangjiu (HJ). To ensure the quality of Dangshen Huangjiu (DSHJ), an orthogonal experiment L9 (33) was proposed to optimize the process by the main brewing factors (pre-fermentation temperature, pre- and post-fermentation time) that may affect BA and functional factors in DSHJ. DSHJ was produced with low BA content and high functional factors. Gas chromatography-ion mobility spectrometry combined with a multivariate statistical method (GC-IMS-MSM) was used to analyze the volatile components in the brewing process of DSHJ. RESULTS: The optimum brewing process parameters of DSHJ were as follows: pre-fermentation temperature, 28 °C; pre-fermentation time, 9 days; post-fermentation time, 18 days. The average content of BA in DSHJ was 33.12 mg L-1, and the sensory score, total phenol content and DPPH free radical scavenging rate of DSHJ were significantly higher than those of HJ. A total of 14 esters, 7 acids, 7 alcohols, 1 ketone, 5 aldehydes and 1 pyrazine in DSHJ and HJ were identified by GC-IMS. There were no significant differences (P > 0.05) in DSHJ and HJ in the soaking rice and saccharification stage. 11 components, such as ethyl acetate, and 12 components, such as acetic acid, were the different components of HJ and DSHJ in pre-fermentation and post-fermentation stages, respectively. In the post-fermentation stage, the contents of 8 components in DSHJ such as ethyl acetate were higher than in HJ. CONCLUSION: The preparation process parameters of DSHJ optimized by orthogonal experiments can ensure that DSHJ has the advantages of low BA content, high total phenol content and good antioxidant activity. Sensory score and GC-IMS-MSM analysis found that DSHJ prepared using the optimal process had the characteristics of good taste and rich aroma. © 2024 Society of Chemical Industry.

One of the top ten tonic herbs, Dangshen is frequently found in Chinese functional foods. With the inclusion of Dangshen in the list of food and medicine substances in 2020, the Dangshen Huangjiu (DHJ) emerged. In the Bencao, it is written that Huangjiu can "open up the curved veins and thicken the stomach and intestines". Furthermore, increasing investigations have verified the protective effect of Dangshen on the gastric mucosa. Therefore, we propose the hypothesis that the stomach mucosa might be protected by the DHJ. To demonstrate that the effect of solids in Dangshen Huangjiu (DHJG) on damaged human gastric mucosal epithelial cells (GES-1) was reversed, the study used ethanol to induce injury to GES-1 and then used protein immunoblotting (western blotting) to determine the expression levels of p-Akt, p-NF-κB-p65, and NF-κB-p65 proteins in the cells. 0.04 mol L-1 MNNG (5 mL kg-1 body weight) mixed with eating disorders(2 d satiety, l d starvation, 3 d cycle) was used to further establish a chronic non-atrophic gastritis (CNAG) model in Wistar rats, at the same time, the experimental rats were given DHJ and DHJG gavage. Cellular assays confirmed that DHJG (25-100 µg mL-1) dose-dependently increased the viability of ethanol-injured GES-1 and lowered p-Akt and p-NF-κB-p65/NF-κB-p65 protein expression. Animal experiments revealed that 10 mL kg-1 and 20 mL kg-1 DHJ had no significant effect on the basic activity and gastric tissues and related biochemical indices of healthy rats; DHJ (10 mL kg-1, 20 mL kg-1) and DHJG (2.8 g kg-1, 11.4 g kg-1) resulted in some improvement in weight loss and significant improvement in gastric mucosal pathology in CNAG rats with damage. Particularly, DHJ and DHJG significantly decreased the expression of p-Akt, p-NF-κB-p65/NF-κB-p65 and Bcl-2/Bax proteins and Akt, IKKß, IκBα and NF-κB mRNA in the gastric tissues of CNAG rats. These results showed that DHJG ameliorates ethanol-induced GES-1 cell injury; both DHJ and DHJG alleviate CNAG, and the mechanisms by which they do so may be related to DHJ and DHJG increasing the antioxidant capacity (elevating SOD, decreasing MDA), attenuating inflammatory responses (decreasing IL-1ß, IL-6, and TNF-α), reversing apoptosis (reducing the Bcl-2/Bax ratio) and down-regulating gastric tissue p-Akt and p-NF-κB-p65/NF-κB-p65 protein expression as well as Akt, IKKß, IκBα and NF-κB mRNA expression. This study indicates that the interventional effects of DHJ and DHJG in CNAG may act through the Akt/NF-κB signaling pathway.

The quality of traditional Chinese medicines (TCM) has a significant correlation with the source and growth years. However, there is no research on the relationship between the growth period and the quality of Codonopsis Radix (CR). This work aims to evaluate the quality of CR in different growth years (2-5 years). First, the content of 6 efficacy-related and 28 nutrient-related components in different growth years of CR was analyzed. The results showed that with the increase in growth years of CR, the content of some components increased, while some decreased. Then, the AHP-CRITIC method was performed to score the CR in different growth years, the results showed that the comprehensive score of CR increased with the increase of growth years, and the 5-year-CR had the highest score. Finally, in vitro activity assays were designed to verify the evaluation results. The results demonstrated that with the increase of growth years, the spleen lymphocyte proliferation activity and DPPH free radical scavenging activity of CR were enhanced, which proved that the AHP-CRITIC method is reasonable to evaluate the quality of CR in different growth years. The aforementioned findings demonstrated that CR quality improved with longer planting years.

Obesity has become a major health problem worldwide, linked to gut microbiota imbalance and chronic inflammation. This study aims to evaluate whether Codonopsis pilosula oligosaccharides (CPOs) can alleviate obesity and related metabolic complications in high-fat diet (HFD) induced obese mice. Male C57BL/6J mice were fed with a HFD for 16 weeks and treated daily with CPOs (500 mg kg-1). CPO supplementation decreased body weight and fat accumulation and improved glucose tolerance in HFD-fed mice. CPOs also reversed the effects of the HFD on inflammatory markers and improved macrophage infiltration. The results of gut microbiota analysis showed that CPOs could also regulate gut microbiota composition, significantly increasing the abundance of the beneficial bacteria Muribaculaceae spp., Alistipes and Clostridium and decreasing the abundance of the harmful bacteria Rikenella, Enterobacteriaceae spp., Collinsella and Megasphaera in HFD mice. Based on serum non-targeted metabolomics analysis, 20 key metabolites responding to CPO treatment were identified, and their biological functions were mainly related to tryptophan and bile acid metabolism. The results demonstrate that CPO supplementation can ameliorate HFD-induced obesity and obesity-related metabolic disorders. It can be used as a novel gut microbiota modulator to prevent HFD-induced gut dysbiosis.

Although density-dependent benefits to hemipterans from ant tending have been measured many times, few studies have focused on integrated effects such as interactions between ant tending, natural enemy density, and hemipteran density. In this study, we tested whether the invasive mealybug Phenacoccus solenopsis is affected by tending by ghost ants (Tapinoma melanocephalum), the presence of parasitoids, mealybug density, parasitoid density and interactions among these factors. Our results showed that mealybug colony growth rate and percentage parasitism were significantly affected by ant tending, parasitoid presence, and initial mealybug density separately. However, there were no interactions among the independent factors. There were also no significant interactions between ant tending and parasitoid density on either mealybug colony growth rate or percentage parasitism. Mealybug colony growth rate showed a negative linear relationship with initial mealybug density but a positive linear relationship with the level of ant tending. These results suggest that benefits to mealybugs are density-independent and are affected by ant tending level.
