Background: Resistance to anti-tuberculous drugs is a major challenge in the treatment of tuberculosis (TB). We aimed to evaluate the clinical availability of nanopore-based targeted next-generation sequencing (NanoTNGS) for the diagnosis of drug-resistant tuberculosis (DR-TB). Methods: This study enrolled 253 patients with suspected DR-TB from six hospitals. The diagnostic efficacy of NanoTNGS for detecting Mycobacterium tuberculosis and its susceptibility or resistance to first- and second-line anti-tuberculosis drugs was assessed by comparing conventional phenotypic drug susceptibility testing (pDST) and Xpert MTB/RIF assays. NanoTNGS can be performed within 12 hours from DNA extraction to the result delivery. Results: NanoTNGS showed a remarkable concordance rate of 99.44% (179/180) with the culture assay for identifying the Mycobacterium tuberculosis complex. The sensitivity of NanoTNGS for detecting drug resistance was 93.53% for rifampicin, 89.72% for isoniazid, 85.45% for ethambutol, 74.00% for streptomycin, and 88.89% for fluoroquinolones. Specificities ranged from 83.33% to 100% for all drugs tested. Sensitivity for rifampicin-resistant tuberculosis using NanoTNGS increased by 9.73% compared to Xpert MTB/RIF. The most common mutations were S531L (codon in E. coli) in the rpoB gene, S315T in the katG gene, and M306V in the embB gene, conferring resistance to rifampicin, isoniazid, and ethambutol, respectively. In addition, mutations in the pncA gene, potentially contributing to pyrazinamide resistance, were detected in 32 patients. Other prevalent variants, including D94G in the gyrA gene and K43R in the rpsL gene, conferred resistance to fluoroquinolones and streptomycin, respectively. Furthermore, the rv0678 R94Q mutation was detected in one sample, indicating potential resistance to bedaquiline. Conclusion: NanoTNGS rapidly and accurately identifies resistance or susceptibility to anti-TB drugs, outperforming traditional methods. Clinical implementation of the technique can recognize DR-TB in time and provide guidance for choosing appropriate antituberculosis agents.

Cu(II)-catalyzed peracetic acid (PAA) processes have shown significant potential to remove contaminants in water treatment. Nevertheless, the role of coexistent H2O2 in the transformation from Cu(II) to Cu(I) remained contentious. Herein, with the Cu(II)/PAA process as an example, the respective roles of PAA and H2O2 on the Cu(II)/Cu(I) cycling were comprehensively investigated over the pH range of 7.0-10.5. Contrary to previous studies, it was surprisingly found that the coexistent deprotonated H2O2 (HO2-), instead of PAA, was crucial for accelerating the transformation from Cu(II) to Cu(I) (kHO2-/Cu(II) = (0.17-1) × 106 M-1 s-1, kPAA/Cu(II) < 2.33 ± 0.3 M-1 s-1). Subsequently, the formed Cu(I) preferentially reacted with PAA (kPAA/Cu(I) = (5.84 ± 0.17) × 102 M-1 s-1), rather than H2O2 (kH2O2/Cu(I) = (5.00 ± 0.2) × 101 M-1 s-1), generating reactive species to oxidize organic contaminants. With naproxen as the target pollutant, the proposed synergistic role of H2O2 and PAA was found to be highly dependent on the solution pH with weakly alkaline conditions being more conducive to naproxen degradation. Overall, this study systematically investigated the overlooked but crucial role of coexistent H2O2 in the Cu(II)/PAA process, which might provide valuable insights for better understanding the underlying mechanism in Cu-catalyzed PAA processes.

Sulfonamides are widely used in the clinical and animal husbandry industry because of their antibacterial properties and low cost. However, Sulfonamides cannot be fully absorbed by human bodies or animals, 50 %-90 % will be discharged from the bodies, and enter waters and soils through a variety of ways, causing environmental harm. Phytoremediation as a green in situ repair technology has been proven effective in sulfonamides removal, but the underlying mechanisms are still a question that needs to be further studied. In order to explore the relationship between SAs removal and plants (S. validus), root exudates secreted from plants, and microorganisms, the study conducted a series of experiments and used the structural equation model to quantify the pathways of sulfonamides removal in wetland plants. The removal rate of sulfonamides in the plant treatment group (77.6-92 %) was significantly higher than that in the root exudate treatment group (25.7-36.3 %) and water treatment group (16.3-19.6 %). Plant uptake (λ1 = 0.72-0.77) and microbial degradation (λ2 = 0.31-0.38) were the most important pathways for sulfonamides removal. Sulfonamides could be directly removed through the accumulation, adsorption and metabolism of plants. Meanwhile, plants could indirectly remove sulfonamides by promoting microbial degradation. These results will facilitate our understanding of the underlying mechanism and the improvement of sulfonamides removal efficiency in phytoremediation.

A biological aluminum-based P-inactivation agent (BA-PIA) has been developed and demonstrated to effectively remove nitrogen and phosphorus; however, whether it can control the release of nitrogen and phosphorus in sediment still needs study. This study aimed to examine the effect of BA-PIA on controlling sediment nitrogen and phosphorus release. BA-PIA was prepared by artificial aeration. The use of BA-PIA in controlling nitrogen and phosphorus release was studied using water and sediment from a landscape lake in static simulation experiments. The sediment microbial community was analyzed using high-throughput sequencing. Static simulation showed that the reduction rates of total nitrogen (TN) and total phosphorus (TP) by BA-PIA were 66.8 ± 1.46% and 96.0 ± 0.98%, respectively. In addition, capping of BA-PIA promotes the conversion of easily released nitrogen (free nitrogen) in the sediment to stable nitrogen (acid-hydrolyzable nitrogen). The content of weakly adsorbed phosphorus and iron-adsorbed phosphorus in the sediment was reduced. The relative abundance of nitrifying bacteria, denitrifying bacteria, and microorganisms carrying phosphatase genes (such as Actinobacteria) in the sediment increased by 109.78%. The capping of BA-PIA not only effectively removed the nitrogen and phosphorus in water but greatly reduced the risk of nitrogen and phosphorus release from sediment. BA-PIA was able to make up for the deficiency of the aluminum-based phosphorus-locking agent (Al-PIA) that only removes phosphorus, giving it improved application prospects.

Grain for Green Project (GGP) is one of China's important ecological restoration projects. The key rationale of this Program is to decrease soil erosion and develop ecological conditions. The agricultural sector is putting efforts to promote green innovation and production among farmers to achieve the targets of ecological restoration projects. However, farmers' green values could play a constructive role in building green innovative intention and green technology adoption behaviors. Based on the unified theory of acceptance and use of technology (UTAUT), the present study investigates the association between farmers' green values and green technology adoption behavior. For empirical investigation, the current study assumes that farmers' green values positively correlate with green innovative intention and green technology adoption behavior, respectively. Further, this study proposes that green innovative intention positively impacts green technology adoption behavior. The present study also aims to determine the mediating role of green innovative intention in the relationship between farmer green values and green technology adoption behavior. This study also attempts to check the moderating role of health consciousness in the relationship between farmer green values and green innovative intention and the relationship between farmer green values and green technology adoption behavior, respectively. For empirical analyses, the present study gathered data from303 farmers in China through a structured questionnaire method using a convenient sampling technique. The present study applied partial least square structural equation modeling for empirically examining hypotheses using Smart PLS software. The findings confirmed that farmers' green values have a positive association with green innovative intention and green technology adoption behavior, respectively. The results further verified that green innovative intention positively correlates with green technology adoption behavior. The finding also authenticated that green innovative intention positively mediates the relationship between farmers' green values and green technology adoption behavior. The moderating role of health consciousness in the relationship between farmers' green values and green innovative intention is also confirmed by the results of this study. However, the findings revealed that health consciousness does not moderate the relationship between farmers' green values and green technology adoption behavior. This study's findings serve the literature by providing empirical insight on the importance of farmers' green values for green innovative intention and green technology adoption behavior. Moreover, the findings also have important theoretical and practical implications.

This study focuses on the studies of the main pressure-dependent reaction types of iso-octane (iso-C8H18) pyrolysis, including initial C-C bond fission of iso-octane, isomerization, and ß-scission reactions of the alkyl radicals produced by the C-C bond fission of iso-octane. For the C-C bond fission of iso-octane, the minimum energy potentials are calculated at the CASPT2(2e,2o)/6-31+G(d,p)//CAS(2e,2o)/6-31+G(d,p) level of theory. For the isomerization and the ß-scission reactions of the alkyl radicals, the optimization of the geometries and the vibrational frequencies of the reactants, transition states, and products are performed at the B3LYP/CBSB7 level, and their single point energies are calculated by using the composite CBS-QB3 method. Variable reaction coordinate transition state theory (VRC-TST) is used for the high-pressure limit rate constant calculation and Rice-Ramsperger-Kassel-Marcus/master equation (RRKM/ME) is used to calculate the pressure-dependent rate constants of these channels with pressure varying from 0.01-100 atm. The rate constants obtained in this work are in good agreement with those available from literatures. We have updated the rate constants and thermodynamic parameters for species involved in these reactions into a current chemical kinetic mechanism and also have improved the concentration profiles of main products such as C3H6 and C4H6 in the shock tube pyrolysis of iso-octane. The results of this study provide insight into the pyrolysis of iso-octane and will be helpful in the future development of branched paraffin kinetic mechanisms.

Experimental studies provide a basis for understanding the mechanisms of rill erosion and can provide estimates for parameter values in physical models simulating the erosion process. In this study, we investigated sediment delivery during rill erosion in purple soil. We used the volume-replacement method to measure the volume of eroded soil and hence estimate the mass of eroded soil. A 12 m artificial rill was divided into the following sections: 0-0.5 m, 0.5-1 m, 1-2 m, 2-3 m, 3-4 m, 4-5 m, 5-6 m, 6-7 m, 7-8 m, 8-10 m, and 10-12 m. Erosion trials were conducted with three flow rates (2 L/min, 4 L/min, and 8 L/min) and five slope gradients (5°, 10°, 15°, 20°, and 25°). The eroded rill sections were refilled with water to measure the eroded volume in each section and subsequently calculate the eroded sediment mass. The cumulative sediment mass was used to compute the sediment concentration along the length of the rill. The results show that purple soil sediment concentration increases with rill length before eventually reaching a maximal value; that is, the rate of increase in sediment concentration is greatest at the rill inlet and then gradually slows. Steeper slopes and higher flow rates result in sediment concentration increasing more rapidly along the rill length and the maximum sediment concentration being reached at an earlier location in the rill. Slope gradient and flow rate both result in an increase in maximal sediment concentration and accumulated eroded amount. However, slope gradient has a greater influence on rill erosion than flow rate. The results and experimental method in this study may provide a reference for future rill-erosion experiments.

Studies revealing conflicting results on the role of NAT1 or NAT2 phenotypes on prostate cancer risk led us to perform a meta-analysis to investigate the association of these polymorphisms and prostate cancer risk. The meta-analysis included six studies with NAT1 genotyping (610 prostate cancer cases and 713 controls), and 10 studies with NAT2 genotyping (1,253 cases and 1,722 controls). The fixed effects odds ratio was 0.96 [95% confidence interval (95% CI): 0.75, 1.21; I(2) = 32.9%, P for heterogeneity = 0.189] for the NAT1 genotype, and the random effects odds ratio was 1.10 (95% CI: 0.87, 1.39; I(2) = 49.1%, P for heterogeneity = 0.039) for the NAT2 genotype. For NAT2 polymorphism, a statistically significant association between NAT2 polymorphism and prostate cancer appeared in Asians, but not in Caucasians. In conclusion, the NAT1 or NAT2 phenotypes detoxify carcinogens and their reactive intermediates are unlikely to be the cause of PCa development.

Various kinds of pathogenic bacteria derived from the intestinal tract of animals exist in compost material like cow dung. In order to sterilize the pathogenic bacteria completely in compost material, the cow dung was put into a heat treatment machine in pilot plan, and harmless condition in short time was examined. The results indicated, pathogenic indicator bacteria such as coliform bacteria, fecal coliform, Escherichia coli and salmonella were all 106 cfu/g dw at the beginning, died rapidly when cow dung temperature rose to above 50 degrees C, and not detected at 54-68 degrees C for 6-24 h heat treatment. Coliform bacteria and salmonella in heated cow dung were not detected by re-growth culture and enrichment culture examination. Moreover, it was hardly influenced on the fermentation ability of composting microbe, organic decomposition bacteria. During heat treatment, the mesophile decreased rapidly and the thermophile stabilized or increased, and the most of composting microbe were bacillus in cow dung by fluorescence microscope, this indicated that bacillus was dominator and composting microbe in composting process.

Application of compost in agricultural practice could potentially cause contamination of foodstuffs with pathogenic bacteria such as Escherichia coli O157:H7 (E. Coli O157). We investigated pathogenic bacteria in compost collected from the compost facilities, and evaluated the survival of E. coli K12 and O157 in laboratory experiments. Out of 19 compost product samples, coliform bacteria and salmonella were detected in 7 and 3 samples respectively. The number of coliform bacteria was 1.8 x 10(2) to 2.5 x 10(6) CFU/g dw and that of salmonella was 4.2 x 10(1) to 6.0 x 10(3) CFU/g dw. Moreover, coliform bacteria, fecal coliform, E. coli and salmonella were detected during composting at 54 degrees C to 67 degrees C. The results indicated that moisture content was a very important factor to the heat sensitivity of pathogenic bacteria in compost, E. coil in compost of high moisture content was more sensitive than that in compost of low moisture content, cells harvested in logarithmic phase was more sensitive than these in stationary phase, and E. coli K12 was more sensitive than E. coli O157. Based on the D values, the lethal time of E. coli K12 and O157 from l0(8) to 10(0) CFU/g dw were 16.3 and 28.8 min, respectively, at 60 degrees C in compost with 40% moisture content. However, some E. coil cells survived in composting process at 54 degrees C to 67 degrees C. Water potential (low moisture content) and physiological aspects of bacteria (stationary phase) could explain only in part of the prolonged survival of E. coil in compost, and there should be some other factors that are conducive to bacterial survival in compost.
