OBJECTIVE: Recent studies have determined the crucial role of long noncoding RNAs (lncRNAs) in cancer development. Cervical cancer (CC) is a common type of fatal gynecological cancer worldwide. This study aims to identify the role of lncRNA AB073614 in the progression of CC. PATIENTS AND METHODS: Relative level of AB073614 in 3 CC cell lines and 48 paired CC samples was determined by the quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). The potential regulatory effects of AB073614 on the cellular behaviors of CC cells were explored through apoptosis assay, colony formation assay, and proliferation assay in vitro. The underlying mechanism of AB073614 in mediating the progression of CC was also conducted. RESULTS: The AB073614 expression was remarkably higher in CC tissues than that in adjacent tissues. The knockdown of AB073614 inhibited proliferation but induced apoptosis of CC cells. In addition, RBM5 (RNA binding motif 5) was upregulated in CC cells after knockdown of AB073614 in vitro. Furthermore, a negative correlation was seen between the expressions of RBM5 and AB073614 in CC tissues. CONCLUSIONS: AB073614 could promote proliferation and inhibit apoptosis of CC cells through downregulating RBM5, which may offer a new therapeutic intervention for CC patients.

While machine learning has been making enormous strides in many technical areas, it is still massively underused in transmission electron microscopy. To address this, a convolutional neural network model was developed for reliable classification of crystal structures from small numbers of electron images and diffraction patterns with no preferred orientation. Diffraction data containing 571,340 individual crystals divided among seven families, 32 genera, and 230 space groups were used to train the network. Despite the highly imbalanced dataset, the network narrows down the space groups to the top two with over 70% confidence in the worst case and up to 95% in the common cases. As examples, we benchmarked against alloys to two-dimensional materials to cross-validate our deep-learning model against high-resolution transmission electron images and diffraction patterns. We present this result both as a research tool and deep-learning application for diffraction analysis.

OBJECTIVE: This study aims to investigate the effect of curcumin concentrations on the proliferation and invasion of endometrial cancer cells. MATERIALS AND METHODS: Endometrial cancer cells were cultured with 0, 15, and 30 µmol/l curcumin, and were divided into control group, low concentration group, and high concentration group. The treatment included the silencing and overexpression of MMP-2. The expression of MMP-2 and E-cadherin were detected by Western blot and the expression level changes were recorded after MMP-2 was silenced and overexpressed. Pearson's analysis was used to determine the relationship of curcumin concentration between MMP-2 mRNA. CCK-8 was used to detect the optical density of cancer cells in three groups. Transwell invasion assay was performed to analyze the invasion inhibition rate in the three groups. RESULTS: Western blot and qPCR results: MMP-2 expression levels were lower and E-cadherin was higher in high concentration group than that in the low concentration group (p<0.01). MMP-2 protein and mRNA decreased after ShRNA and increased after overexpression (p<0.01). Pearson analysis revealed that the curcumin concentration was negatively correlated with the MMP-2 mRNA (r=-0.497, p=0.036). Cell optical density was lower with curcumin and the lowest was in high concentration group (p<0.01). After MMP-2 silencing, optical cell density decreased and this value increased after overexpression (p=0.000). Cell invasion results: Curcumin improves the rate of cell invasion (p<0.01). After silencing of MMP-2, cell invasion inhibition rate increased, while the invasion inhibition rate decreased after overexpression (p<0.01). CONCLUSIONS: Curcumin can downregulate MMP-2, inhibit the proliferation and invasion of cancer cells.

RESUMO

The M² factor could be decomposed as amplitude term and phase term. A method to improve the beam quality of laser beams is proposed. In our method, the amplitude and phase of a laser beam are both compensated in order to improve the beam quality completely. In experiment, a laser amplifier is set to modulate the amplitude to Gaussian type, and a deformable mirror is used to compensate the phase aberration. The laser beam is well compensated by our method; the beam quality factor of the laser is improved from 1.7 to 1.1.

We examined the distribution of major allelic variants of CYP2C9 and CYP2C19 in the Mongolian population of China and compared it with that of other populations. The polymorphisms of CYP2C9 (including the CYP2C9*1, CYP2C9*2 and CYP2C9*3 alleles) and CYP2C19 (including the CYP2C19*1, CYP2C19*2 and CYP2C19*3 alleles) were analyzed in 280 healthy unrelated Chinese Mongolian subjects, using a PCR-RFLP assay. The frequencies of CYP2C9*1, *2 and *3 alleles were 0.97, 0.00 and 0.03, respectively. The frequencies of CYP2C19*1, *2 and *3 alleles were 0.72, 0.24 and 0.04, respectively. We did not find any differences in the allelic distribution of these two genes between age groups. However, the genotype frequency of CYP2C9 *1/*3 was significantly higher in males than in females. Compared with other populations, we found that the allele frequencies of the CYP2C9*2 and CYP2C9*3 allelic variants in this Mongolian population of China were similar to those reported for other Asian populations, with significant differences compared to Caucasians and African-Americans.

A novel 2-positional linked carbazole based ß-diketone with methoxyl in the 7-position (EMOCTFBD), and its europium(III) ternary complex Eu(EMOCTFBD)(3)phen were synthesized via a dexterously designed indirect routine, where EMOCTFBD was 1-(9-ethyl-7-methoxyl-9H-carbazol-2-yl)-4,4,4-trifluorobutane-1,3-dione and phen was 1,10-phenanthroline. Eu(EMOCTFBD)(3)phen shows high thermal stability. The introduction of a methoxyl in the 7-position of the carbazole ring remarkably enhanced the excitation band intensity in the blue region, and the complex exhibited intense red emission under blue-light excitation. The lowest triplet state energy was measured and suggests the photoluminescence process as a ligand-sensitized luminescence process (antenna effect). A bright red-emitting diode was fabricated by coating the complex phosphor onto a ∼460 nm-emitting InGaN chip. All the results indicate that Eu(EMOCTFBD)(3)phen is an interesting red-emitting material excited by blue light, and therefore may be applied in many fields without UV radiation.

Two novel carbazole-based beta-diketones with 2- or 2,7-substituted groups in the carbazole ring, 2-(4'4'4'-trifluoro-1'3'-dioxobutyl)-carbazole (2-TFDBC) and 2,7-bis(4'4'4'-trifluoro-1''3'-dioxobutyl)-carbazole (2,7-BTFDBC), and their europium(III) ternary complexes Eu(2-TFDBC)(3)phen and Eu(2)(2,7-BTFDBC)(3)(phen)(2) were synthesized via a dexterously designed routine, where phen is 1,10-phenanthroline. The Eu(III) complexes were characterized by elementary analysis, IR and UV-visible absorption spectroscopy, thermogravimetric analysis (TGA), and photoluminescence (PL) measurements. TGA shows that the decomposition temperatures of the complexes are 361.4 and 367.3 degrees C, respectively. PL measurement results indicated that suitably expanded pi-conjugation in the complex molecules makes the excitation band redshift to the visible region, and both the Eu(III) complexes exhibit intense red emission under blue-light excitation. The triplet state energy levels of 2-TFDBC and 2,7-BTFDBC in the complexes are higher than that of the lowest excited level of Eu(3+) ion, (5)D(0), so the photoluminescence mechanism of the Eu(III) complexes was proposed as a ligand-sensitized luminescence process. All of the results indicate that Eu(2-TFDBC)(3)phen and Eu(2)(2,7-BTFDBC)(3)(phen)(2) are promising candidates as visible-light excitable red phosphor for luminescence applications.

Start-up of a continuously stirred tank reactor for bio-hydrogen production under different initial organic loading rate (OLR) of 3, 7 and 10 kgCOD/m3 d, respectively, was carried out with sewage sludge as inoculum. Molasses wastewater was used as substrate and hydraulic retention time was kept at 6 h. This study aimed to assess OLR on the formation of fermentation types and the structure of microbial communities during the start-up period. It was found that at an initial OLR of 7 kgCOD/m3 d and an initial biomass of 6.24 gVSS/L, an equilibrial microbial community of ethanol-type fermentation could be established within 30 days. The observed average specific hydrogen production rate was 276 mLH2/gVSS d, which was 40% higher than that of the one acclimated with 3 kgCOD/m3 d. Based on denaturing gradient gel electrophoresis profiles, significant microbial population shifts took place at the first 15 days, but a longer period up to 30 days was required to establish a microbial community with stable metabolic activity.

Fluorescence of Ce(III) aqua ion at pH 6.0 is found to be in good linear relationship with its concentration, and the intensity is strong enough to be employed to determine its concentration. TRNA(Phe) evidently quench this fluorescence. Fluorescence titration experiments were performed to Ce(III)-tRNA(Phe) system, the binding number and association constant was estimated with a Scatchard plot. Two classes of binding sites with association constant of 5.2 x 10(7) and 4.4 x 10(6) M, respectively, were found. Addition of spermine slightly decrease binding number and association constant of Ce(III) ion.
