RESUMO

Dual-color cross-correlation spectroscopy allows the detection and quantification of labeled biomolecules at ultra-low concentrations, whereby the sensitivity of the assay correlates with the measurement time. We now describe a parallel multifocal dual-color spectroscopic configuration employing multiple avalanche photodiodes and hardware correlators. Cross-correlation curves are obtained from several dual-color excitation foci simultaneously. Multifocal dual-color excitation is achieved by splitting each of two laser beams (488 and 633 nm) into four sub-beams with the help of two 2x2 fan-out diffractive optical elements (DOEs), and subsequent superposition of the two sets of four foci. The fluorescence emission from double-labeled biomolecules is detected by two 2x2 fiber arrays.

RESUMO

The design, construction, and evaluation of a laser beam steerer that uses two binary ferroelectric liquid-crystal (FLC) spatial light modulators (SLMs) operated in conjunction are presented. The system is characterized by having few components and is in principle lossless. Experimentally, a throughput of approximately 20% was achieved. The simple system design was achieved because of the high tilt angle FLC material used in the SLMs, which were specifically designed and manufactured for this study. By coherently imaging the first SLM onto the second SLM, pixel by pixel, we obtained an effective four-level phase structure with a phase step of 90 degrees. An appropriate alignment procedure is presented. The beam steering performance of the system is reported and analyzed.

A gas cell filed with argon gas under pressure is placed in a tightly focused laser beam to provide a limiter for laser pulses above a certain peak power, corresponding to the optical breakdown threshold for the creation of a laser-induced plasma. Measurements of the threshold intensity as a function of argon gas pressure are given for a laser wavelength of 1.064 microm (Nd:YAG) and a pulse length of 6.4 ns. Threshold intensities for optical breakdown in fused silica were measured with the same optical system, enabling a relative comparison of breakdown thresholds, of interest for protecting fused-silica optical components in fiber-optic delivery systems for laser material processing applications. The threshold intensity was measured to 220 GW/cm2 in Ar at 1.0 x 10(5) N/m2 (1 atm), 80 GW/cm2 in Ar at 8.0 x 10(5) N/m2 (7.9 atm), and 55 GW/cm2 in fused silica. Even though the threshold in argon is higher than that in fused silica, the limiter will protect the optical components if the laser beam is focused to a tighter spot in the gas cell than at the input end of the fiber.

RESUMO

A sensitive interferometric sensor scheme that is based on coherent imaging of a first phase grating onto a second phase grating, their periods accurately matched, is suggested. Experimental data, obtained with a setup based on the suggested scheme, are presented. The sensor was found capable of measuring an angular tilt of a mirror less than 0.5 microrad. Compared with a previously suggested measuring scheme, the novelty of the one presented here is the inclusion of a second set of gratings, which eliminates measurement ambiguity. Some characteristics of the sensor scheme are discussed.

RESUMO

Dynamic behavior of an analog-to-digital converter (ADC) based on diffractive optical element(s) (DOE)(s) was studied and found to be in agreement with predictions. The analog signal was translated to an angular deflection of a laser beam by means of an acousto-optic (AO) cell. The number of bits in this experimental demonstration was three, using an eight-element DOE array. The maximum sample rate was found to be 2.5 MS/s, the limiting factor being the transit time for the acoustic wave across the width of the laser beam in the AO cell. The study is intended as a first dynamic demonstration of a proposed ADC scheme previously demonstrated in a quasi-static version. The full potential of the ADC scheme will require the use of a fast tunable diode laser to replace the AO deflection scheme used here.

We have developed a multifocal optical fluorescence correlation spectroscopy system for parallel flow analyses. Multifocal excitation was made possible through a 4 x 1 diffractive optical fan-out element, which produces uniform intensity in all four foci. Autocorrelation flow analyses inside a 20 microm x 20 microm square microchannel, with the 4 x 1 fan-out foci perpendicular to the flow direction, made it possible to monitor different flows in all four foci simultaneously. We were able to perform cross-correlation flow analyses by turning the microstructure, thereby having all four foci parallel to the direction of flow. Transport effects of the diffusion as a function of flow and distance could then also be studied.

RESUMO

Active alignment of a 1 x 8 free-space optical switch was studied experimentally. Optical signals, carried on single-mode fibers, were switched by a ferroelectric liquid-crystal-on-silicon spatial light modulator. Continuous measurement of the in-coupled power to the fibers provided feedback for the switch control. The switch automatically located and locked to the output fibers. An advantage with adaptive switches of a similar kind is relaxed geometrical tolerances in the switch assembly. Further, such switches can adapt to possible geometrical changes and light wavelength drift during operation.

We have developed a multifocal diffractive-optical fluorescence correlation spectroscopy system for parallel excitation and detection of single tetramethylrhodamine biomolecules in microarrays. Multifocal excitation was made possible through the use of a 2 x 2 fan-out diffractive-optical element with uniform intensity in all foci. Characterization of the 2 x 2 fan-out diffractive-optical element shows formation of almost perfect Gaussian foci of submicrometer lateral diameter, as analyzed by thermal motion of tetramethylrhodamine dye molecules in solution. Results of parallel excitation and detection in a high-density microarray of circular wells show single-biomolecule sensitivity in all four foci simultaneously.
