RESUMO

Metal-based nanozymes with exceptional physicochemical property and intrinsic enzymatic properties have been widely used in industrial, medical, and diagnostic fields. However, low substrate affinity results in unsatisfying catalytic kinetic and instability in complicated conditions, which significantly decreases their sensitivity and reliability. Herein, an amorphous hollow manganese silicate nanosphere (defined as AHMS) has been successfully synthesized via a facile one-step hydrothermal method and utilized in the archetype for colorimetric detection of biothiols with high sensitivity and high reliability. The experimental data demonstrates that ultrafast affinity of the substrate contributes to enhanced sensitivity with outstanding catalytic kinetic features (Km = 27.1 µM) and low limit of detection (LODGSH = 20 nM). The designed sensor demonstrates a reliable applicability for analysis of biological liquids (fetal calf serum and Staphylococcus aureus) and design of visual logic gates. Therefore, AHMS provides a promising strategy for ultrasensitive and high-reliable biosensing.

Hyperaccumulator Amaranthus hypochondriacus L. has huge potential in the remediation of cadmium (Cd)-contaminated soils and is necessary to understand the mechanism of Cd uptake by the roots. In this study, the mechanism of Cd uptake into the root of A. hypochondriacus was investigated using the non-invasive micro-test technology (NMT) by analyzing the rate of Cd2+ fluxes at different regions of the root tip; also we assessed the impact of different channel blockers and inhibitors on the Cd accumulation in the roots, the real-time Cd2+ fluxes, and the distribution of Cd along the roots. The results showed that the Cd2+ influx was greater near the root tip (within 100 µm of the tip). All the inhibitors, ion-channel blockers, and metal cations had different degrees of inhibition on the absorption of Cd in the roots of A. hypochondriacus. The net Cd2+ flux in the roots was significantly decreased by the Ca2+ channel blockers lanthanum chloride (LaCl3) by up to 96% and verapamil by up to 93%; as for the K+ channel blocker tetraethylammonium (TEA), it also caused a 68%-reduction on the net Cd2+ flux in the roots. Therefore, we infer that the uptake by A. hypochondriacus roots is mainly through the Ca2+ channels. The Cd absorption mechanism appears to be related to the synthesis of plasma membrane P-type ATPase and phytochelatin (PC), which is reflected by the inhibition of Ca2+ upon addition of inorganic metal cations. In conclusion, access of Cd ions into the roots of A. hypochondriacus is achieved through various ion channels, with the most important being the Ca2+ channel. This study will further enhance the literature regarding Cd uptake and pathways of membrane transport in roots of Cd hyperaccumulators.

Automatic respiration monitoring of dairy cows in modern farming not only helps to reduce manual labor but also increases the automation of health assessment. It is common for cows to congregate on farms, which poses a challenge for manual observation of cow status because they physically occlude each other. In this study, we propose a method that can monitor the respiratory behavior of multiple cows. Initially, 4,000 manually labeled images were used to fine-tune the YOLACT (You Only Look At CoefficienTs) model for recognition and segmentation of multiple cows. Respiratory behavior in the resting state could better reflect their health status. Then, the specific resting states (lying resting, standing resting) of different cows were identified by fusing the convolutional neural network and bidirectional long and short-term memory algorithms. Finally, the corresponding detection algorithms (lying and standing resting) were used for respiratory behavior monitoring. The test results of 60 videos containing different interference factors indicated that the accuracy of respiratory behavior monitoring of multiple cows in 54 videos was >90.00%, and that of 4 videos was 100.00%. The average accuracy of the proposed method was 93.56%, and the mean absolute error and root mean square error were 3.42 and 3.74, respectively. Furthermore, the effectiveness of the method was analyzed for simultaneous monitoring of respiratory behavior of multiple cows under movement, occlusion disturbance, and behavioral changes. It was feasible to monitor the respiratory behavior of multiple cows based on the proposed algorithm. This study could provide an a priori technical basis for respiratory behavior monitoring and automatic diagnosis of respiratory-related diseases of multiple dairy cows based on biomedical engineering technology. In addition, it may stimulate researchers to develop robots with health-sensing functions that are oriented toward precision livestock farming.

Nano-catalytic bacterial killing provides new opportunities to address ever-increasing antibiotic resistance. However, the intrinsic catalytic activity usually depends on a much lower pH conditions (pH = 2-5) than that in the weakly acidic bacterial microenvironments (pH = 6-7) for reactive oxygen species production by Fenton reactions. Herein, a MnSiO3 -based pH-ultrasensitive "in situ structure transformation" is first reported to significantly promote the adhesion between material and bacteria, and shorten the diffusion distance (<20 nm) to compensate ultra-short life (<200 ns) of ·OH generated by Mn2+ -mediated Fenton-like reaction, finally enhancing its nano-catalytic antibacterial performance in weakly acidic conditions. A separated spray bottle is further designed to achieve in situ gelation at the wound site, which demonstrates excellent shape adaptability to complicated and rough surfaces of wounds, allowing for long-term nano-catalyst release. As a result, bacterial-infected wound healing is efficiently promoted. Herein, the in situ sprayed nano-catalytic antibacterial gel presents a promising paradigm for bacterial infection treatment.

RESUMO

Non-invasive Micro-test Technology (NMT) is a selective microelectrode technique which can detect the flux rates and three-dimensional motion directions of ions or molecules into and out of living organisms in situ without damaging the sample. It has the advantages of maintaining sample integrity, high temporal and spatial resolution, and being able to measure multiple sites simultaneously. In this paper we provide a comprehensive review on the development of NMT in recent years. Its principles, characteristics, and the differences with other microelectrode techniques are introduced. We discuss the applications of NMT in the field of phytoremediation, plant resistance, water quality monitoring, and toxicity mechanisms of heavy metals on organisms. Furthermore, the challenges and future prospects of NMT in the environmental field are presented.
