RESUMO

AIM: The aim of this study was to describe the clinical and pathological findings of acute bovine pulmonary edema and emphysema (ABPEE) and left endocardial fibroelastosis in an adult dairy cow. In addition, a review of recent literature of these two conditions is provided. MATERIALS AND METHODS: Necropsy and histopathological examination were performed using conventional techniques. A review of the literature was carried out using internet search engines such as PubMed and Google Scholar. Only published papers in scientific and refereed journals were reviewed. RESULTS: Concurrent pathologies of ABPEE and left endocardial fibroelastosis were described in an adult Holstein Friesian cow. A review of recent literature concerning ABPEE and endocardial fibroelastosis revealed seven and two scientific reports of these conditions in cattle, respectively. CONCLUSION: Although rare, combined pathologies involving multiple organs such as the lungs and heart can be diagnosed in animals on careful clinical and histopathological examinations.

The effects of thermal manipulation (TM) at 38.5°C and 40°C for 6 h at embryonic day (ED) 16, 9 h at ED 17, and 12 h at ED 18 on body weight (BW) and cloacal body temperature (Tb) during the first wk and later at post-hatch d 10, 14, 21, 28, and 42 were evaluated. Furthermore, chicks' ability to cope with a thermal challenge (TC; 41°C for 6 h) at post-hatch d 14 and 42 was also evaluated. A chick's response to TC was measured by determining the cloacal body temperature; the plasma thyroid hormones (thyroxin (T4) and triiodothyronine (T3)); the packed cell volume (PCV); the heterophil (H), lymphocyte (L), monocyte, basophil, and eosinophil percentages; and the heterophil-to-lymphocyte ratios (H/L). Thermal manipulation did not affect the hatchability. However, the body weight of TM chicken was higher compared with controls at marketing age (post-hatch d 42). At post-hatch d 14 and 42, no significant changes in Tb were observed among the different treatment groups. However, during TC at d 14 and 42, the Tb of TM chicks was lower compared with the controls. During TC, a significant increase in plasma T4 and a significant decrease in plasma T3 of TM chicks compared with controls were reported. Furthermore, during TC, a significant increase in the PCV and heterophil, monocyte, and H/L ratios, and a reduction in the lymphocyte percentages also were observed in TM chicks compared with the controls. Results of this study showed that chicks subjected to heat manipulation during late embryogenesis respond better to heat stress later in the growth and development period.

Lumpy skin disease (LSD) is an emerging disease in the Middle East region and has been recently reported in Jordan. The aim of this study was to investigate the adverse reactions that were reported after vaccine administration. Geographical areas enrolled in the study were free of the disease and away from the outbreak governorate. Sixty-three dairy cattle farms, with a total of 19,539 animals, were included in the study. Of those, 56 farms reported adverse clinical signs after vaccine administration. The duration between vaccine administration and appearance of adverse clinical signs ranged from 1 to 20 days (Mean = 10.3, SD ± 3.9). Clinical signs were similar to those observed with natural cases of lumpy skin disease. These were mainly fever, decreased feed intake, decreased milk production and variable sized cutaneous nodules (a few millimetres to around 2 cm in diameter) that could be seen anywhere on the body (head, neck, trunk, perineum), udder, and/or teats. Nodules were raised and firm initially and then formed dry scabs that could be peeled off the skin. The characteristic deep 'sit fast' appearance was rarely seen and most lesions were superficial. Some cattle had swollen lymph nodes, while a few pregnant animals aborted. The percentage of affected cattle ranged from 0.3 to 25% (Mean = 8, SD ± 5.1). Fever, decreased feed intake, and decreased milk production were seen in 83.9, 85.7, and 94.6% in cattle on the affected farms, respectively. All affected cattle displayed skin nodules over their entire bodies, while 33.9 and 7.1% of the affected farms reported nodular lesions present on the udders and teats, respectively. No mortalities were reported due to vaccine adverse reactions. Duration (course) of clinical signs ranged from 3 to 20 days (Mean = 13.7, SD ± 4.1). Two types of LSD vaccines were used by the farmers in this study. The first one was a sheep pox virus (SPPV) vaccine derived from the RM65 isolate [Jovivac, manufactured by Jordan Bioindustries Centre (JOVAC)] and the other an unlabelled one, which was later identified using PCR as a strain of lumpy skin disease virus (LSDV). Blood and skin samples collected from cattle vaccinated with the LSDV vaccine were positive for LSDV using both general and species-specific PCR primers, whereas those from cattle vaccinated with the Jovivac vaccine were negative. Adverse reactions observed in cattle after administration of the LSDV vaccine were reported to be more severe than those seen after Jovivac vaccine administration and were comparable with clinical signs observed in natural infections.

An outbreak of suspected malignant catarrhal fever (MCF) was investigated by molecular and histopathological assays. Of the 70 Holstein beef calf herds, 14 were affected by multiple clinical signs suggestive of MCF infection. These beef calves were housed next to sheep flocks. In the complete blood count, the 14 affected calves had severe anaemia with leucopaenia, lymphopaenia and neutropaenia. Upon PCR amplification using a hemi-nested PCR assay for the detection of the Ovine herpesvirus 2 (OvHV-2), bovine tissue samples from the mesenteric lymph nodes and spleen and ovine blood samples were shown to be positive with the expected PCR bands amplified. Direct sequencing of the hemi-nested PCR product confirmed the identity of the causative virus as OvHV-2. The histopathological findings confirmed the clinical and laboratory diagnosis of MCF. Collective clinical, PCR and histopathological data confirmed the identity of this outbreak to be a sheep-associated malignant catarrhal fever (SA-MCF).

A total of 29 brain tissue samples (BTS) were examined for rabies infection by different diagnostic techniques. None of the examined brain tissues were presented as a whole intact brain. Twenty-seven brain tissue samples from various animal species - dog (13 cases), cat (one case), fox (one case), pig (one case), cow (three cases), sheep (two cases), goat (one case), camel (one case), horse (one case) and donkey (three cases) - were provided by the Vaccine and Sera Department/Al-Bashir Central Hospital in Amman/Jordan from July 2009 up to May 2010. All these samples were frozen at -20°C, for a period of time and then fixed in 10% formalin after being tested for rabies virus by fluorescence antibody test (FAT). The results showed that 21 (77.77%) of 27 BTS were positive for rabies by FAT. Seventeen samples (58.62%) of 29 were positive by histopathology, 2 (6.90%) were positive by histopathology, immunohistochemistry (IHC) and of those which were fixed for 24h only, and 21 (72.42%) were positive using RT-PCR assay. Five of 29 BTS had no pathological lesions, 17 had Negri bodies and the remaining had non-suppurative encephalitis and necrosis. Thirteen BTS that were diagnosed positive by FAT were also positive by RT-PCR and histopathology, but negative by IHC. Four BTS that were positive by FAT were negative by histopathology, IHC and RT-PCR. Also, 3 BTS (cases 19, 22, and 25) that were negative by FAT were positive by RT-PCR and negative by IHC. One of these was negative, while two were positive by histopathology. Therefore, definitive diagnosis of rabies under these conditions in Jordan needs one or more other diagnostic tests in addition to FAT. Also, freezing and prolonged formalin fixation of BTS is not suitable for the detection of rabies virus antigen using IHC.
