RESUMO

Extracellular vesicles have emerged as promising tools in regenerative medicine due to their inherent ability to facilitate intercellular communication and modulate cellular functions. These nanosized vesicles transport bioactive molecules, such as proteins, lipids, and nucleic acids, which can affect the behavior of recipient cells and promote tissue regeneration. However, the therapeutic application of these vesicles is frequently constrained by their rapid clearance from the body and inability to maintain a sustained presence at the injury site. In order to overcome these obstacles, hydrogels have been used as extracellular vesicle delivery vehicles, providing a localized and controlled release system that improves their therapeutic efficacy. This Review will examine the role of extracellular vesicle-loaded hydrogels in tissue regeneration, discussing potential applications, current challenges, and future directions. We will investigate the origins, composition, and characterization techniques of extracellular vesicles, focusing on recent advances in exosome profiling and the role of machine learning in this field. In addition, we will investigate the properties of hydrogels that make them ideal extracellular vesicle carriers. Recent studies utilizing this combination for tissue regeneration will be highlighted, providing a comprehensive overview of the current research landscape and potential future directions.

BACKGROUND: Metabolic syndrome (MetS) is one of the leading public health issues in the world with a reported prevalence of nearly 25% in the past decades in Iran. The present research aimed to identify the association between the Healthy Eating Index (HEI) and MetS components among female teachers. METHODS: In this cross-sectional study, 97 female teachers aged 31-57 years were enrolled from 2018 to 2019 in Qom, Iran. Usual dietary intakes were assessed using a validated 168-item Food Frequency Questionnaire (FFQ). HEI-2015 was calculated according to the consumption of whole fruits, vegetables, protein foods, beans, seafood, plant proteins, total and refined grain, dairy, fatty acid ratio, saturated fats, added sugars, and sodium. We also measured anthropometric and biochemical parameters. To evaluate the association between HEI-2015 and MetS, multivariate logistic regression was performed. RESULTS: MetS was found in 59.8% of participants. Total HEI-2015 scores were significantly lower in participants with MetS compared to those without MetS (59.69 ± 8.98 vs. 64.21 ± 8.71, respectively; p = 0.02). Daily energy intake, weight, body mass index, waist circumference, systolic and diastolic blood pressure, serum triglyceride, and fasting blood sugar levels were higher in women with MetS (all p < 0.05). Higher HEI-2015 total scores (OR: 0.94; 95% CI: 0.89-0.99; p = 0.02) and scores of total vegetables (OR: 0.62; 95% CI: 0.42-0.91; p = 0.02), dark green vegetables and beans (OR: 0.62; 95% CI: 0.39-0.98; p = 0.04), fatty acid ratio (OR: 0.83; 95% CI: 0.68-0.99; p = 0.04), refined grain (OR: 0.86; 95% CI: 0.75-0.99; p = 0.04), and added sugars (OR: 0.44; 95% CI: 0.26-0.75; p = 0.002) were all associated with lower odds of having MetS. CONCLUSIONS: Higher HEI-2015 scores, particularly in total vegetables, dark green vegetables, beans, and fatty acid ratio, as well as refined grain and added sugars were found to be associated with reduced odds of having MetS among Iranian female teachers. However, further prospective studies are required to confirm this finding.

RESUMO

This is a case report of a 47-year-old male with a history of hypertension and pre-diabetes who presented to the emergency department with dyspnea, progressive unilateral leg swelling and pain. The patient tested positive for coronavirus disease 2019 (COVID-19) infection about a week earlier. The patient was found to have an extensive clot burden of his lower extremity veins, both deep and superficial, which extended to his inferior vena cava (IVC). Based on the patient's clinical exam and ultrasound findings, the patient was diagnosed with impending phlegmasia cerulea dolens. Due to his renal failure, the patient was taken for a ventilation/perfusion (V/Q) scan which found widespread V/Q mismatch highly suggestive of pulmonary embolism. Interventional radiology took the patient for lower extremity venogram, catheter-directed alteplase administration, and IVC filter placement. The patient was admitted to the intensive care unit (ICU) for further management and had a stable recovery.

BACKGROUND: Ineffective integration has been recognized as one of the major causes of early orthopedic failure of titanium-based implants. One strategy to address this problem is to develop modified titanium surfaces that promote osteoblast differentiation. This study explored titanium surfaces modified with TiO2 nanotubes (TiO2 NTs) capable of localized drug delivery into bone and enhanced osteoblast cell differentiation. MATERIALS AND METHODS: Briefly, TiO2 NTs were subjected to anodic oxidation and loaded with Metformin, a widely used diabetes drug. To create surfaces with sustainable drug-eluting characteristics, TiO2 NTs were spin coated with a thin layer of chitosan. The surfaces were characterized via scanning electron microscopy, atomic force microscopy, and contact angle measurements. The surfaces were then exposed to mesenchymal bone marrow stem cells (MSCs) to evaluate cell adhesion, growth, differentiation, and morphology on the modified surfaces. RESULTS: A noticeable increase in drug release time (3 days vs 20 days) and a decrease in burst release characteristics (85% to 7%) was observed in coated samples as compared to uncoated samples, respectively. Chitosan-coated TiO2 NTs exhibited a considerable enhancement in cell adhesion, proliferation, and genetic expression of type I collagen, and alkaline phosphatase activity as compared to uncoated TiO2 NTs. CONCLUSION: TiO2 NT surfaces with a chitosan coating are capable of delivering Metformin to a bone site over a sustained period of time with the potential to enhance MSCs cell attachment, proliferation, and differentiation.

Because of exposure to a wide range of chemical contaminants such as nitrate via potable water resources, the use of the approaches to set standards for drinking water quality and also to do a risk assessment is necessary for maintaining the public health. High levels of nitrate in drinking water can have adverse health effects; primarily for infants and pregnant women. So, the present study aimed to the assessment of nitrate health risk in drinking water resources of the Iranshahr city, Sistan and Baluchistan province and also, evaluation of the uncertainty of nitrate and the probability of contamination occurrence by Monte-Carlo Simulation (MCS) technique. Besides, the geographic information system (Arc GIS, Ver 10.3) was applied to mapping the nitrate concentration in groundwater resources of the studied area. For these aims, the numbers of 66 samples were collected from rural groundwater resources, and nitrate concentration was measured using a Spectrophotometer in wavelength of 220 nm. According to the results, the nitrate concentration was in the range of 6.49 mg/L, and its average level was 6.15 mg/L. Also, the simulation results with 90% confidence showed that the hazard equitant (HQ) in the infant groups, children-teenagers and adults was less than 0.331, 0.311, 0.312, and 0.3, respectively.

Drinking water sources may be polluted by various pollutants depending on geological conditions and agricultural, industrial, and other human activities. Ensuring the safety of drinking water is, therefore, of a great importance. The purpose of this study was to assess the quality of drinking groundwater in Bardaskan villages and to determine the water quality index. Water samples were taken from 30 villages and eighteen parameters including calcium hardness (CaH), total hardness (TH), turbidity, pH, temperature, total dissolved solids (TDS), electrical conductivity (EC), alkalinity (ALK), magnesium (Mg2+), calcium (Ca2+), potassium (K+), sodium (Na+), sulphate (SO4 2-), bicarbonate (HCO3 -), fluoride (F-), nitrate (NO3 -), nitrite (NO2 -) and chloride (Cl-) were analyzed for the purpose for this study. The water quality index of groundwater has been estimated by using the ANFIS. The spatial locations are shown using GPS. The results of this study showed that water hardness, electrical conductivity, sodium and sulfate in 66, 13, 45 and 12.5% of the studied villages were higher than the Iranian drinking water standards, respectively. Based on the Drinking Water Quality Index (DWQI), water quality in 3.3, 60, 23.3 and 13.3% of villages was excellent, good, poor and very poor, respectively. •Groundwater is one of the sources of drinking water in arid and semi-arid regions such as Bardaskan villages, which monitor the quality of these resources in planning for improving the quality of water resources.•The DWQI can clearly provide information associated with the status of water quality resources in Bardaskan villages.•The results of this study clearly indicated that with appropriate selection of input variables, ANFIS as a soft computing approach can estimate water quality indices properly and reliably.•Some parameters were in the undesirable level is some villages. Therefore, the government should try to improve the chemical and physical quality of drinking water in these areas with the necessary strategies.

Early and effective integration of titanium-based materials into bone tissue is of vital importance for long-term stability of implants. Surface modification is commonly used to enhance cell-substrate interactions for improving cell adhesion, proliferation, and activity. Here, the surface of titanium substrates and commercial implants were coated with blood (TiB), fetal bovine serum (TiF), and phosphate-buffered saline (TiP) solution using a spin coating process. Surface roughness and wettability of samples were measured using contact angle measurements and atomic force microscopy. The samples were then exposed to human osteoblast-like MG63 cells in order to evaluate adhesion, growth, differentiation, and morphology on the surface of modified samples. Untreated titanium disks were used as controls. The lowest roughness and wettability values were found in unmodified titanium samples followed by TiP, TiF, and TiB. The percentage of cellular attachment and proliferation for each sample was measured using an MTT (3-[4,5-dimethylthiazol-2yl] 2,5diphenyl-2H-tetrazoliumbromide) assay. Cell adhesion and proliferation were most improved on TiB followed closely by TiF. The results of this study revealed an increased expression of the osteogenic marker protein alkaline phosphatase on TiB and the coated commercial titanium implants. These results suggested that precoating titanium samples with blood may improve cellular response by successfully mimicking a physiological environment that could be beneficial for clinical implant procedures.

Understanding the gas evolution in batteries, caused by decomposition of the electrolyte, is of fundamental importance for improving the long-time performances and cycle life of the battery systems. In general, this phenomenon causes simultaneously an irreversible energy and charge loss, as well as an increase of the internal resistance. Here, we introduce a new cell design capable of performing electrochemical impedance spectroscopy (EIS) and differential electrochemical mass spectroscopy (DEMS) with high resolution. Detailed aspects of the cell fabrication and the different components of the cell are extensively explained. Impedance measurements were validated by using symmetric electrodes. The possibility of performing long-term DEMS measurements was tested on graphite electrodes in Ethylene Carbonate/Dimethyl Carbonate (1:1), 1 M LiPF6 as an electrolyte. Finally, the cell was used to detect hydrogen evolution on the zinc negative electrode of a zinc-ion battery based on copper hexacyanoferrate.

It is now well accepted that the addition of nanoparticles (NPs) can strongly affect the thermomechanical properties of the polymers into which they are incorporated. In the solid (glassy) state, previous work has implied that optimal mechanical properties are achieved when the NPs are well dispersed in the matrix and when there is strong interfacial binding between the grafted NPs and the polymer matrix. Here we provide strong evidence supporting the importance of intermolecular interactions through the use of NPs grafted with polymers that can hydrogen bond with the matrix, yielding to significant improvements in the measured mechanical properties. Our finding thus supports the previously implied central role of strong interfacial binding in optimizing the mechanical properties of polymer nanocomposites.

BACKGROUND: Bilateral neck exploration is the gold standard for parathyroid adenoma localization in primary hyperparathyroidism. But surgeons do not have adequate experience for accurate surgical exploration and new methods are developed for surgery like unilateral exploration and minimally invasive surgery, thus, preoperative localization could reduces time and stress in surgical performance. METHOD: 80 patients with documented primary hyperparathyroidism and with raised serum calcium and parathyroid hormone (PTH) were selected. The results of ultrasonographic localization for each patient were compared with findings of surgery and 99m technetium sestamibi scintigraphy. Also variables such as preoperative serum calcium, PTH level and adenoma weight were compared between patients who had localized and non-localized adenoma with ultrasonography or Sestamibi scan. The data was compared with student's t-test. RESULTS: In a prospective diagnostic tests accuracy study, 80 patients with primary hyperparathyroidism were enrolled. Ultrasonography images detected enlarged parathyroid glands in 61 of 80 patients (76.3%) with sensitivity of 83.5% and positive predictive value (PPV) of 89.7%. Sestamibi scintigraphy detected adenoma in 63 patients (78.8%) with sensitivity of 85% and PPV of 91.3%. There was no significant deference between ultrasonography and scintigraphy in localization of adenomas. Both ultrasonography and scintigraphy used for determining localization, and they located 73 adenomas (91.3%) with sensitivity of 97.3% and PPV of 93.5%. CONCLUSION: Ultrasonography as an accurate method for localization of enlarged parathyroid glands in primary hyperparathyroidism, is comparable in overall utility with sestamibi scintigraphy. This study suggests a strategy for initial testing with one method, followed by the alternate imaging test if the first test happens to be negative.
