RESUMO

Ultimate resolution in scanning transmission electron microscopy (STEM) with state-of-the-art aberration correctors requires careful tuning of the experimental parameters. The optimum aperture semi-angle depends on the chosen high tension, the chromatic aberration and the energy width of the source as well as on potentially limiting intrinsic residual aberrations. In this paper we derive simple expressions and criteria for choosing the aperture semi-angle and for counterbalancing the intrinsic sixth-order three-lobe aberration of two-hexapole aberration correctors by means of the fourth-order three-lobe aberration. It is noteworthy that for such an optimally adjusted electron probe the so-called flat area of the Ronchigram is explicitly not maximized. The above considerations are validated by experiments with a CEOS ASCOR in a C-FEG-equipped JEOL NEOARM operated at 60 kV. Sub-Angstrom resolution is demonstrated for a Si[112] single crystal as well as for a single-layered MoS2 crystalline film. Lattice reflections of 73 pm for silicon and 93 pm for molybdenum disulfide are visible in the Fourier transform of the images, respectively. Moreover, single sulfur vacancies can be clearly identified in the MoS2.

RESUMO

The chemical synthesis of monoatomic metallic copper is unfavorable and requires inert or reductive conditions and the use of toxic reagents. Here, we report the environmental extraction and conversion of CuSO4 ions into single-atom zero-valent copper (Cu0) by a copper-resistant bacterium isolated from a copper mine in Brazil. Furthermore, the biosynthetic mechanism of Cu0 production is proposed via proteomics analysis. This microbial conversion is carried out naturally under aerobic conditions eliminating toxic solvents. One of the most advanced commercially available transmission electron microscopy systems on the market (NeoArm) was used to demonstrate the abundant intracellular synthesis of single-atom zero-valent copper by this bacterium. This finding shows that microbes in acid mine drainages can naturally extract metal ions, such as copper, and transform them into a valuable commodity.

RESUMO

The modification of galactose with α1,2-fucose is involved in symbiosis with intestinal bacteria and elimination of pathogenic bacteria. It is postulated that α1,2-fucosylated mucin secreted from goblet cells is involved in defending an organism against infections, but the detailed molecular mechanisms are yet to be elucidated. It was previously reported that Paneth cells of the small intestine were positive for UEA-1 lectin staining. However, glycoproteins in Paneth cells carrying α1,2-fucose have not yet been identified. Glycoproteomic analysis of ileal lysates identified 3212 O-linked and 2962 N-linked glycopeptides. In particular, cryptdin-related sequence 1 (CRS1) expressed in Paneth cells was found to be α1,2-fucosylated. Unlike other antimicrobial α-defensin proteins, CRS1 contains unique Thr residues, which are modified with O-glycans, with 3HexNAc2Hex1Fuc1NeuAc being the main glycoform. Identification of α1,2-fucose on the O-glycans of CRS1 expressed in Paneth cells will pave the way for a mechanistic understanding of α1,2-fucose-dependent symbiosis with intestinal bacteria and elimination of pathogenic bacteria in the intestine.

RESUMO

Ionic liquids show characteristic properties derived from them being composed of only molecular ions, and have recently been used as solvents for chemical reactions and as electrolytes for electrochemical devices. The liquid structures, i.e., ionic distributions, form when solutes are dissolved in ionic liquids and fundamentally affect the reactions and transfer efficiency in such solutions. In this study, we directly observe the liquid structure in a solution of the long-chain ionic liquid 1-octyl-3-methylimidazolium bromide (C8mim Br) and barium stearate (Ba(C17H35COO)2) using the annular dark-field method of scanning transmission electron microscopy (ADF-STEM). The ADF image shows a 10 nm-scale heterogeneity in the image intensity, which reflects the heterogeneous ionic distribution in the solution. The number density distributions of all the component ions (C8mim+, Br-, Ba2+, and C17H35COO-) were estimated from the ADF image intensity and then visualized. These ionic distribution maps depicted the spatial relationships between the ions at the sub-nanometer scale and revealed that the heterogeneity is largely derived from the large differences in size, charge distributions, and van der Waals interactions.

RESUMO

Estimating the Jacobian matrix of a nonlinear dynamical system through observed time-series data is one of the important steps in predicting future states of the time series. The Jacobian matrix is estimated using local information about divergences of nearby trajectories. Although the basic algorithm for estimating the Jacobian matrix generally works well, it often fails for short or noisy data series. In this paper, we proposed a scheme to effectively use near-neighbor information for more accurate estimation of the Jacobian matrix using the bootstrap resampling method. Then, to confirm the validity of the proposed method, we applied it to a mathematical model and several real time series. As a result, we confirmed that the proposed method greatly improves nonlinear predictability, not only for noise-corrupted mathematical models but also for real time series.
