RESUMO

IMPORTANCE: We present a protocol to efficiently sequence genomes of the MPXV-causing mpox. This enables researchers and public health agencies to acquire high-quality genomic data using a rapid and cost-effective approach. Genomic data can be used to conduct surveillance and investigate mpox outbreaks. We present 91 mpox genomes that show the diversity of the 2022 mpox outbreak in Ontario, Canada.

For the first time, we propose a new modification-free voltammetric platform for simple, fast and reliable quantification of the sport performance-enhancing stimulant synephrine (SYN) based on the usage of a commercial screen-printed boron-doped diamond (BDD) electrochemical sensor. Applying cyclic voltammetry, SYN exhibited one well-shaped, irreversible and diffusion-driven oxidation peak at a peak potential of +1.45 V (vs. the silver pseudoreference electrode) in the presence of 2 M HClO4. Under the selected experimental conditions, the following analytical parameters for determination of SYN were obtained: linear concentration ranges from 19.6 up to 1000 µM and from 9.9 up to 1000 µM for differential pulse (DPV) and square-wave voltammetry (SWV), respectively, detection limits at micromolar concentration levels and intra-day repeatability with a relative standard deviation below 4% for both pulse techniques. The interference study revealed fair selectivity when considering the target dietary supplements. The feasibility of the developed voltammetric platform was verified in the analysis of commercially available dietary supplements and reasonable recovery values were achieved by DPV and SWV, respectively. In addition, the method can be performed directly without higher consumption of chemicals, waste generation, complex sample extraction and higher instrumentation cost. The advanced BDD electrochemical sensor has appeared to be a suitable competitor for efficient applications in food quality control analysis.

Spread of excitation in the cochlea places fundamental limits on the interpretation of audiometric pure-tone hearing loss as a simple map of dysfunction along the cochlear partition, and histologic evidence from human temporal bones will be presented to demonstrate the insensitivity of the audiogram to variations in pathology in the case of low-frequency hearing loss. This article will describe a clinical procedure using simultaneous pure-tone masking to improve upon the localization of cochlear disease, particularly for low-frequency hearing losses, and a model for using the Articulation Index (AI) to develop prognoses for hearing aid performance in these cases, which can then be tested. Fourteen patients with low-frequency hearing loss were divided into two groups based upon threshold shifts caused by a pure-tone masker: those that showed normal low-frequency threshold shifts and those that showed marked shifts at frequencies below the masker, indicating greater loss of function than shown by the unmasked audiogram. Hypothetical audiograms were then generated to model a complete loss of apical function for all patients. Measured speech recognition scores were then compared to AI predictions for the actual and hypothetical audiograms. Best agreement for the patients showing normal masking shifts was between the measured scores and the AI for the actual audiogram, whereas the best agreement for the patients showing marked shifts was with the AI for the hypothetical audiogram. The implications for hearing aid recommendation and fitting in these cases are discussed.
