Background: Obesity and central precocious puberty (CPP) are associated with increased anxiety, depression, and anger in girls. The contribution of exercise as an efficacious component in decreasing anxiety, depression, and anger has been increasingly recognized. Objectives: This study aims to evaluate the effects of combined training on cortisol, anxiety, depression, and anger in overweight and obese girls with CPP. Methods: The study involved 30 girls aged 7-9 years diagnosed with CPP (undergoing triptorelin treatment) and dealing with obesity. In addition, these girls scored higher than the cut-off line for anxiety, depression, and anger. The participants were divided into two groups, with 15 individuals in each group. The exercise group engaged in 60 min of combined aerobic and resistance training three times per week for a duration of 12 weeks. On the other hand, the control group did not receive any training. Throughout the study, the serum cortisol levels were measured in both groups. Anxiety, anger, and depression questionnaires were also completed at three different stages, namely, baseline, 12 weeks, and 16 weeks (after a 4-week period of detraining). Results: In the exercise group, there was a significant decrease (P < 0.05) in cortisol serum levels and anxiety, depression, and anger scores. These changes were observed consistently during detraining (P > 0.05). However, in the control group, only the depression score significantly decreased (P < 0.05). Conclusions: Based on the results, it can be concluded that combined training is a method to improve the mental health of CPP girls. BACKGROUND: The present study analyzes the influence of resistance training (RT) and high-intensity interval training (HIIT) on metabolic indices and serum levels of Sirtuin1 (SIRT1) in postmenopausal women who suffer from the metabolic syndrome (MetS). METHODS: 45 postmenopausal women aged 45-65 years with MetS were divided into two intervention groups (RT and HIIT) and one control group, each consisting of 15 people. The RT group performed resistance training for both the upper and lower body, while the HIIT group completed 3 min(min) of high-intensity training at 80-90% of their maximum heart rate (HRmax), followed by moderate walking for 3 min at 55-65% of HRmax. These sessions were conducted for a duration of eight weeks and three times a week, with the samples being collected at the baseline and at the end of the treatment, i.e., week 8. RESULTS: The results showed that weight, waist circumference, body mass index, fat mass, low-density lipoprotein, triglyceride, cholesterol, fasting blood sugar (FBS), hemoglobin A1c (HbA1C), systolic and diastolic blood pressure decreased, and SIRT1 increased significantly in both training groups. Systolic blood pressure, cholesterol, HbA1C, and FBS decreased more in the HIIT group. Skeletal muscle mass and 1-repetition maximum (1-RM) increased more in the RT group. CONCLUSIONS: RT and HIIT serve as one of the most effective strategies for therapeutically treating patients with metabolic syndrome. TRIAL REGISTRATION: IRCT, IRCT20221120056548N1. This study was an attempt to examine the changes in serum levels of ghrelin and leptin after 12-weeks of aerobic training and gonadotropin releasing hormone agonist (GnRH) treatment in girls with central precocious puberty. Thirty girls (6-8 years old) with precocious puberty who had received Triptorelin were randomly divided in two groups (medication and medication + training). Fifteen age-matched healthy girls (without precocious puberty) were also included as the control group. The medication + training group submitted an aerobic training program for 3 days/week with 20-75 min per day and 45-75% of maximum heart rate for 12-weeks. Serum levels of leptin, ghrelin, cholesterol, triglycerides and body mass index (BMI) were determined at baseline and 48 h after the last training session. The results indicated that leptin significantly decreased (p = 0.001) and ghrelin significantly increased (p = 0.001) in the medication + training group but no significant difference was observed in the ghrelin (p = 1) and leptin (p = 0.78) in the medication group. Leptin to ghrelin ratio indicated a decrease in medicine + training group (p = 0.028). Ghrelin were negatively correlated with leptin and BMI. The data indicated that aerobic training increased ghrelin and reduced leptin and leptin to ghrelin ratio but GnRH agonist treatment had no effect on plasma leptin and ghrelin levels.

Aims: This study was designed to investigate the effects of resistance training (RT) and hawthorn extract (Ha) on Glypican-4 (GPC-4) and Insulin-regulated glycosylphosphatidylinositol-specific phospholipase D (GPLD1) serum levels in T2DM and to examine the relationship of these variables with glycemic indexes. Method: 40 male Wistar rats were divided into five equal groups: Healthy Control (H-C), Diabetes Control (D-C), Diabetes Resistance training (D-RT), Diabetes Hawthorn (D-Ha), and Diabetes Resistance training Hawthorn (D-RT-Ha). T2DM was induced with a 4-week high-fat diet (HFD) and one dose of STZ intraperitoneal injection (35 mg/kg). 1-week after the injection, RT (with a range of 50%-100%1RM/3 day/week) and gavage of Ha extract (100 mg/kg/day) was performed for 12 weeks. Results: The glycemic indices improvement (reducing blood glucose and increasing serum insulin level) caused by RT and/or Ha increased GPC-4 and decreased GPLD1 in the T2DM rats, but these positive changes were more effective in the combination of RT + Ha. A strong correlation was also observed between GPC-4 and GPLD1 with blood glucose and insulin. Conclusion: The increase in serum GPC-4 levels was probably due to the direct effect of RT + Ha, and the improvement of glycemic indexes after RT and Ha. The double effect of RT + Ha can be a regulatory mechanism for GPC-4 and its related factors in controlling T2DM complications.

PURPOSE: We examined the effects of aerobic exercise and detraining on adiponectin, resistin, tumor necrosis factor alpha (TNF-α), white blood cell, and pubertal signs (uterine length, ovarian volume, luteinizing hormone [LH], and follicle-stimulating hormone [FSH]) in girls with central precocious puberty (CPP). METHODS: Thirty girls with CPP (7-9 yr old) were randomly divided into CPP and CPP + Exercise (EX) groups. Fifteen healthy age-matched girls without precocious puberty participated for comparison with CPP. Subjects in the CPP + EX group attended 12 wk of aerobic exercise protocol three sessions a week. In the baseline, after completing the exercise protocol and after 4 wk of detraining, all the parameters were measured. In the CPP and healthy groups, all the parameters were measured in all three stages. To measure adiponectin, resistin, and TNF-α, the ELISA method was used. LH and FSH were measured by electrochemiluminescence immunoassay. White blood cell counts were analyzed by an automated blood cell counter. Repeated-measures ANOVA was used to analyze the results. RESULTS: CPP girls have lower adiponectin ( P = 0.01) and higher TNF-α levels ( P = 0.001) than healthy girls. In the CPP + EX group, after 12 wk, body weight and fat mass decreased, and adiponectin increased significantly ( P = 0.02). Resistin ( P = 0.02), TNF-α ( P = 0.02), neutrophils ( P = 0.01), and signs of puberty significantly ( P < 0.05) decreased. After detraining, no significant change was observed except TNF-α, which increased significantly ( P = 0.03). In the CPP group, no significant change was observed in any of the parameters; only LH ( P = 0.0001) and uterine length and ovarian volume ( P = 0.003, P = 0.001) decreased after 12 and 16 wk, respectively. CONCLUSIONS: Aerobic exercise can have a positive effect on the state of inflammation and pubertal signs. Positive effects remain after 4 wk of detraining.

Background: Omentin-1 is a circulating adipokine that can serve as a biomarker for assessment of metabolic risk factors. We investigated the effect of eight weeks of aerobic exercise training on serum omentin-1, insulin resistance and lipid profile in nonsmokers and smokers. Methods: Nineteen male nonsmokers (aged 27.88 ± 2.47 years, and with BMI of 22.69 ± 1.77 kg.m-2) and twenty male smokers (aged 30.11 ± 1.96 years, and with BMI of 23.12 ± 1.91 kg.m-2) were randomly assigned into nonsmokers control group (C), nonsmokers exercise group (E), control smoker group (CS), and exercise smoker group (ES). Exercise groups participated in an eight-week aerobic exercise training program (three times a week, 20-35 min per session at 55%-70% of maximum heart rate). Serum omentin-1 and insulin values were determined by ELISA. The Homeostatic Model Assessment for Insulin Resistance (HOMA-IR), glucose level and lipid profile were measured before and after the intervention. Pearson correlation test, Eta test, paired samples t-test, one and two-way analysis of variance (ANOVA) and post-hoc Tukey test were applied for data analysis (p < .05). Results: Aerobic exercise improved both serum omentin-1 and high lipoprotein cholesterol (HDL-C) in the exercise groups (P < .05). Also, exercise training reduced insulin, glucose, HOMA-IR, triglyceride (TG), total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) levels (p < .05). Omentin-1 was significantly correlated with glucose, insulin, HOMA-IR, TG, TC, LDL-C and HDL-C in both nonsmokers and smokers. Conclusions: The findings suggest that aerobic exercise-induced changes in omentin-1 in the exercise-trained groups may be associated with the beneficial effects of exercise on reduced insulin resistance and lipid profile.

BACKGROUND: We investigated the effects of high-fat diet (HFD) consumption combined with diabetes induction, resistance exercise training (RET) and a de-training period on circulating irisin levels and selective metabolic parameters. MATERIAL AND METHODS: Rats were assigned to four groups (n = 8): healthy non-diabetic rats (NDC), non-diabetic rats that performed RET (NDR), sedentary HFD-fed/STZ-treated rats (HFD/STZ) and HFD-fed/STZ-treated rats that performed RET (HFD/STZ + RE). RESULTS: HFD consumption reduced irisin level and Quicki (p < .01). After the 12-week period, levels of TC, TG, HOMA1-IR, HOMA2-IR and irisin were also lower in the HFD/STZ + RE group compared to the HFD/STZ group. Body weight and HOMA1-IR showed a positive (r = 0.558 and r = 0.538) whereas TC and LDL-C had a negative correlation (r = -0.461 and r = -0.630) with irisin level (p < .05). CONCLUSIONS: Irisin level increased along with the progress of obesity and T2DM. It seems that RET can attenuate the increase of irisin in those conditions by improvement of glucose/lipid metabolic disorders.

BACKGROUND: The prevalence of precocious puberty is increasing. Obesity has been demonstrated to be associated with changes in the adipokine profile and incidence of early puberty in girls. This study assessed the pubertal signs, the levels of adiponectin, resistin, and tumor necrosis factor-alpha (TNF-α) after 12 weeks of combined exercise and 4 weeks of detraining in overweight and obese girls with precocious puberty. METHODS: Thirty overweight and obese girls (aged 7-9) with precocious puberty, who had received Triptorelin, were randomly divided into two groups (15 exercise and 15 control). Initially, serum levels of adiponectin, resistin, TNF-α, luteinising hormone (LH), and follicle-stimulating hormone (FSH) and the signs of puberty progression (bone age, uterine length, and ovarian volume) were measured. The exercise group performed 60 min of combined (aerobic and resistance) exercise three times/week for 12 weeks. The control group did not receive any exercise. 48 h after the last training session and after 4 weeks of detraining, all research variables were measured (also in the control group). The statistical method used for data analysis was repeated measures ANOVA. RESULTS: In the exercise group, adiponectin significantly increased and resistin significantly decreased after 12 weeks. After 4 weeks of detraining, adiponectin significantly decreased, but resistin significantly increased. TNF-α levels did not change significantly during the study. There was no significant difference in all of the factors in the control group. Throughout the 16-week study period, the rate of puberty and LH significantly decreased in both exercise and control groups, but FSH, LH/FSH and ovarian volume significantly decreased in the exercise group alone (P<0.05). CONCLUSIONS: Combined exercise increased adiponectin and decreased resistin and the rate of puberty. However, after 4 weeks of detraining, these effects diminished but did not disappear. BACKGROUND: Irisin and betatrophin are involved in insulin resistance. We investigated the effects of aerobic and resistance exercise (AE/RE) and de-training (cessation of the training after the AE/RE) on betatrophin, irisin and some metabolic factors in rats. METHODS: Wistar rats were assigned into six groups: non-diabetic rats (C), non-diabetic rats that performed AE/RE, diabetic rats (Dia), and diabetic rats that performed AE/RE (Dia+AE and Dia+RE). Diabetes was induced by high-fat diet/streptozotocin model. The rats de-trained for four weeks after the 12-week exercise training. Blood samples were analyzed by enzyme-linked immunosorbent assay (ELISA) method and statistical analyses were performed using repeated measures and 1-way analysis of variance (ANOVA). RESULTS: The 12-week ET improved homeostasis model assessment of IR (HOMA-IR), high-density lipoprotein cholesterol (HDL-C), total cholesterol (TC) and triglycerides (TG) in the trained diabetic groups (p<0.05). ET reduced betatrophin level in the Dia+RE but not in the Dia+AE group. Positive correlations between betatrophin and body weight (r=0.547; p<0.01), and HOMA-IR (r=0.461; p<0.05) but a negative correlation with LDL-C and TC (r=-0.684, r=-0.669; both p<0.01) were observed, whereas no significant correlation was found between betatrophin and HDL-C and TG (r=-0.225, r=-0.360; both p>0.05). Betatrophin was correlated with irisin in the healthy rats but not the diabetic rats (p<0.01). CONCLUSIONS: It seems that RE has greater efficiency than AE in reducing betatrophin level and irisin resistance. However, de-training caused most of the improvements resulting from RE to be lost, but not the improvements resulting from AE.

BACKGROUND: Adipocytokines, which are secreted by the adipose tissue, contribute to the pathogenesis of obesity-related complications. To evaluate this assumption, we investigated the effects of aerobic exercise training (AET), resistance exercise training (RET), and 4 weeks of de-training on serum leptin and TNF-α levels in diabetic rats. METHOD: 36 Wistar rats were divided into normal diet (ND) (control, RET, AET) and high-fat diet (HFD) + STZ (control, RET, AET) groups. Serum insulin, leptin, and TNF-α levels were assessed by commercial ELISA kits. Also fasting blood glucose (FBG), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG) levels were measured by the colorimetric kits. RESULTS: Diabetes induction increased body weight (BW) and FBG, and decreased insulin compared to the ND rats' groups (p < 0.001). 12-weeks of AET and RET programs in the trained diabetic rats led to a decrease in TG, LDL-C, leptin, TNF-α, and FBG, and an increase in insulin compared to the HFD + STZ-C group (p < 0.001). Besides, there was no difference between AET and RET in improving the variables studied (p > 0.05). Also, de-training led to increased BW, TG, leptin, and TNF-α compared to the end of the exercise training (p < 0.05). The correlation between the variables studied was established at different stages of the study (p < 0.05), and only BW was not correlated with insulin during exercise training and de-training (p > 0.05). CONCLUSION: These findings indicate that both AET and RET are useful in reducing levels of serum adipocytokines (TNF-α, leptin) in diabetic and non-diabetic rats. At the same time, 4 weeks of de-training was sufficient to lose the metabolic adaptations.

Diabetes mellitus is mainly associated with degeneration of the central nervous system, which eventually leads to cognitive deficit. Although some studies suggest that exercise can improve the cognitive decline associated with diabetes, the potential effects of endurance training (ET) accompanied by Matricaria chamomilla (M.ch) flowers extract on cognitive impairment in type 2 diabetes has been poorly understood. Forty male Wistar rats were randomized into 5 equal groups of 8: healthy-sedentary (H-sed), diabetes-sedentary (D-sed), diabetes-endurance training (D-ET), diabetes-Matricaria chamomilla. (D-M.ch), and diabetes-endurance training-Matricaria chamomilla. (D-ET-M.ch). Nicotinamide (110 mg/kg, i.p.) and Streptozotocin (65 mg/kg, i.p.) were utilized to initiate type 2 diabetes. Then, ET (5 days/week) and M.ch (200 mg/kg body weight/daily) were administered for 12 weeks. After 12 weeks of the experiment, cognitive functions were assessed using the Morris Water Maze (MWM) test and a passive avoidance paradigm using a shuttle box device. Subsequently, using crystal violet staining, neuron necrosis was examined in the CA3 area of the hippocampus. Diabetic rats showed cognitive impairment following an increase in the number of necrotic cells in region CA3 of the hippocampal tissue. Also, diabetes increased serum levels of lipid peroxidation and decreased total antioxidant capacity in serum and hippocampal tissue. ET + M.ch treatment prevented the necrosis of neurons in the hippocampal tissue. Following positive changes in hippocampal tissue and serum antioxidant enzyme levels, an improvement was observed in the cognitive impairment of the diabetic rats receiving ET + M.ch. Therefore the results showed that treatment with ET + M.ch could ameliorate memory and inactive avoidance in diabetic rats. Hence, the use of ET + M.ch interventions is proposed as a new therapeutic perspective on the death of hippocampal neurons and cognitive deficit caused by diabetes.

Cisplatin, a common chemotherapeutic drug, can induce testicular toxicity. Methylene blue, a potent antioxidant, can inhibit the generation of free radicals. This research aimed to study the protective effect of methylene blue against the cisplatin-induced toxicity of the reproductive system in rats. 35 male Wistar rats were divided into five groups: the control group, the cisplatin group (a single dose of 5 mg/kg cisplatin), the low-dose and high-dose methylene blue + cisplatin (2 and 4 mg/kg of methylene blue, respectively, for 7 days) and the methylene blue group (4 mg/kg of methylene blue, for 7 days). The treatments were applied through intraperitoneal injection. Cisplatin treatment reduced the sperm parameters and serum testosterone levels significantly. Methylene blue treatment increased the sperm count (p < .001), viability (p < .001) and motility (p < .001) compared to the cisplatin group. The methylene blue group showed a significant increase in the levels of testosterone compared to the cisplatin group (p < .001) and reverted histopathological changes in cisplatin-treated groups. Immunohistochemical evaluation of the caspase-3 protein revealed that the treatment with methylene blue has significant anti-apoptotic effects on testicular tissue damage. In conclusion, methylene blue can attenuate the cisplatin-induced histological damages and improve the sperm parameters.

BACKGROUND: Glycosylphosphatidylinositol-specific phospholipase D (GPLD1) is responsible for cleaving membrane-associated glycosylphosphatidylinositol (GPI) molecules, which is affected by diabetes. We aimed to examine the effect of 14 weeks treadmill running on serum GPLD1 levels and its association with glycemic indexes and serum glypican-4 (GPC-4), a novel GPI-anchored adipokine, in streptozotocin-nicotinamide-induced diabetic rats. METHODS: Thirty-six male Wister rats were randomly divided into three groups of twelve animals each, involving sedentary control (SC), sedentary diabetic (SD), and trained diabetic (TD) groups. The diabetes was induced through intraperitoneal injection of 120 mg/kg nicotinamide 15 min prior to intraperitoneal injection of 65 mg/kg streptozotocin in SD and TD groups. The TD group was exercised on a treadmill for 60 min/days, 5 days/wk at 26 m/min, and zero grade for 14 weeks. Following the experiment period, blood samples were taken from all animals and analyzed for experimental indexes via sandwich ELISA. RESULTS: Exercise training caused a significant decrease in the elevated blood glucose levels and a significant increase in the lowered blood insulin levels in TD rats (both p < 0.001). Glucose tolerance of TD rats significantly improved following experimental protocol, as indicated by OGTT (p < 0.001). Experimental diabetes significantly increased serum GPLD1 levels (p < 0.001), while exercise training significantly decreased its levels (p < 0.001). Serum GPLD1 levels correlated directly with glycemic indexes involving FBS, 2hOGTT, and AUC of glucose (r = 0.80, r = 0.79, r = 0.79, respectively, all p < 0.001) and inversely with serum insulin levels (r = - 0.83, p < 0.001). There were no significant differences in serum GPC-4 levels among groups, and no association with GPLD1 alteration. CONCLUSIONS: Sedentary diabetic rats have higher circulating GPLD1 compared to controls, which can be reversed by exercise training and is associated with modifying in glycemic and insulin profile.

Background The aim of this study was to investigate the effect of 12 weeks of aerobic training, 4 weeks of detraining and use gonadotropin-releasing hormone agonist (GnRHa) on serum C-reactive protein (CRP) and cortisol levels in girls with central precocious puberty (CPP). Methods Forty-five girls (aged 6-8 years) with precocious puberty were randomly divided into three groups (medication, training and medicine + training groups). Fifteen healthy girls (without precocious puberty) were also included as the control group. Serum CRP and cortisol levels were measured at baseline by the enzyme-linked immunosorbent assay (ELISA) technique. Then, the experimental groups performed an aerobic training program for 3 days/week 20-75 min per day at 45-75% maximum heart rate for 12 weeks. The medication groups also received GnRHa during the study, once a month (1 mL every 4 weeks) by intramuscular injection. Serum CRP and cortisol levels were measured again 48 h after the last training session and also after 4 weeks of detraining. Results Analysis of variance (ANOVA) with repeated measures showed a significant decrease in CRP (p = 0.02) and cortisol levels (p = 0.01) in the training group and the medicine + training group. Detraining led to return of CRP and cortisol levels to the pre-training levels (p = 0.001). No significant difference in serum CRP (p = 0.43) and cortisol levels (p = 0.06) was observed in the medication group. Further, no significant difference was observed between groups in CRP and cortisol. Conclusions Long-term regular moderate training decreases inflammation indices, and detraining eliminates the benefits of training in girls with precocious puberty.

PURPOSE: Diabetes mellitus type 2 with damaging effects on reproductive hormones and sperm quality parameters can often cause infertility in men. The aim of this study was to evaluate the effects of endurance, resistance and concurrent training on reproductive hormones, sperm parameter in the diabetic type 2 male rats. METHODS: In this experimental study 60 Wistar rats (200 ± 50 g) were randomly assigned into 5 groups: control; diabetic; diabetic endurance training; diabetic resistance training and diabetic concurrent training. For inducing diabetes, after 12 hours of food starvation nicotinamide (120 mg/kg) and STZ (65 mg/kg) were intraperitoneally injected. Twenty-four hours after the last training session, left epididymis of the rats was examined for studying sperm parameters and blood serum samples were examined for evaluating reproductive hormones. Data were analyzed using one-way ANOVA and Turkey's Post Hoc test. RESULTS: Ten weeks of endurance and concurrent training induced significant decrease in the blood glucose in comparison to the diabetic group (P < 0.05). In addition, endurance, resistance and concurrent training induced significant increases in serum testosterone and LH levels in the comparison to the diabetic group (P < 0.005). In addition, sperm parameters revealed significant improvements in compared to the diabetic group (P = 0.002). CONCLUSION: Endurance, resistance and combined training might improve sperm parameters, including viability and motility of sperms through increasing the serum testosterone and LH levels in rat model of diabetes mellitus type 2.

AIMS: Glypican-4 (GPC-4) is a novel adipomyokine that enhances insulin signaling. Glycosylphosphatidylinositol-specific phospholipase D (GPLD1) is thought to release GPC-4 and is itself an insulin-regulated enzyme. Beneficial effects of exercise training and chamomile flowers extract (CFE) are shown through activation of PPARγ, which is a promising drug target in diabetes and associated with GPC-4 synthesis. This study investigated the effects of 14-week treadmill running and CFE on serum GPC-4, GPLD1, and insulin levels in streptozotocin-nicotinamide (STZ-NA)-induced diabetic rats. METHODS: Thirty-two STZ-NA-induced diabetic male Wistar rats were randomly assigned to four groups: control (C), training (T), CFE treatment (CFE), and training plus CFE treatment (TCFE) groups. The training groups were exercised on treadmill 5 days/week and the treating groups were fed with 200 mg/kg/day CFE in drinking water for 14 weeks. Finally, serum GPC-4, GPLD1, and insulin levels were analyzed via sandwich ELISA. RESULTS: Compared to the control group, serum insulin levels were significantly higher in the T, CFE, and TCFE groups (p < 0.05, p < 0.05, p < 0.01, respectively), while OGTT and serum GPLD1 levels were significantly lower in the T, CFE, and TCFE groups (all p < 0.001). Changes in serum GPC-4 levels were not significant. Serum GPLD1 levels were negatively correlated with insulin levels and HOMA-IS (both p < 0.001). CONCLUSIONS: This study suggests that endurance training and CFE may downregulate serum GPLD1 levels in STZ-NA-induced diabetic rats, which associate with the serum insulin profile. However, the results show that endurance training and CFE may not cause serum GPC-4 adaptation in STZ-NA-induced diabetic rats.

BACKGROUND: Drug addiction triggers the infliction of a variety of diseases. Various subjects have indicated that during the withdrawal syndrome period, the immune system is weakened. OBJECTIVES: This study aimed to investigate the changes in serum levels of interferon-gamma (IFN-γ) and interleukin-17 (IL-17) during the morphine withdrawal syndrome induced by 8 weeks of moderate exercise and their effects on the immune system function. MATERIALS AND METHODS: Twenty-four male Wistar rats (220 ± 10 g) were divided into four groups (n = 6): healthy control (HC), addicted control (AC), healthy trained (HT), and addicted trained (AT) groups. AC and AT groups were made addicted to morphine sulfate (0.4 mg/mL) in 21 days. To ensure their dependence on morphine, naloxone (3 mg/kg, i.p.) was injected into the body of a number of the rats. HT and AT groups were made to run on a treadmill 5 days per week for 8 weeks while time and speed gradually increased. Both prior to the exercises and 24 hours after the last training session, blood samples were collected from all the animals, and serum IFN-γ and IL-17 serum levels were measured using the ELISA method. This research was performed at the Bu-Ali Sina University, Hamadan, Iran. RESULTS: After 8 weeks of exercise, a significant increase was observed in the serum IFN-γ level in the HT group (251.17 ± 13.045) in comparison with the HC group (234 ± 12.884) (P = 0.045). Furthermore, the serum IFN-γ level in the AT group (218.33 ± 5.164) in comparison to the AC group (190.67 ± 8.477) showed a significant increase (P = 0.000). In addition, the serum level of IFN-γ in the HT group showed a significant increase compared to the AT group (P = 0.000). After 8 weeks of exercise, there was a significant decrease in the serum IL-17 level in the HT group (22.67 ± 4.46) compared with the HC group (38.17 ± 7.68) (P = 0.005). In addition, a significant decrease was observed in serum IL-17 in the AT group (42.17 ± 7.41) in comparison with the AC group (57.17 ± 7.83) (P = 0.007). In addition, the serum IL-17 level in the HT group significantly decreased in comparison with the AT group (P = 0.001). CONCLUSIONS: Moderate exercise for 8 weeks increased the IFN-γ and decreased the IL-17 serum levels in the morphine-dependent rats. For this reason, this type of exercise can improve the function of the immune system during morphine withdrawal syndrome.

AIMS: Continuous morphine consumption contributes to the development of cognitive disorders. This work investigates the impacts of different types of exercise on learning and memory in morphine-dependent rats. MAIN METHODS: Forty morphine-dependent rats were randomly divided into five groups: sedentary-dependent (Sed-D), endurance exercise-dependent (En-D), strength exercise-dependent (St-D), and combined (concurrent) exercise-dependent (Co-D). Healthy rats were used as controls (Con). After 10weeks of regular exercise (endurance, strength, and concurrent; each five days per week), spatial and aversive learning and memory were assessed using the Morris water maze and shuttle box tests. KEY FINDINGS: The results showed that morphine addiction contributes to deficits in spatial learning and memory. Furthermore, each form of exercise training restored spatial learning and memory performance in morphine-dependent rats to levels similar to those of healthy controls. Aversive learning and memory during the acquisition phase were not affected by morphine addiction or exercise, but were significantly decreased by morphine dependence. Only concurrent training returned the time spent in the dark compartment in the shuttle box test to control levels. SIGNIFICANCE: These findings show that different types of exercise exert similar effects on spatial learning and memory, but show distinct effects on aversive learning and memory. Further, morphine dependence-induced deficits in cognitive function were blocked by exercise. Therefore, different exercise regimens may represent practical treatment methods for cognitive and behavioral impairments associated with morphine-related disease.

BACKGROUND: Diabetes mellitus (types 1 and 2) leads to secondary complications such as neuropathy, which reduce a patient's quality of life. Apelin and its receptor, APJ, have been shown to have antinociceptive effects and to decrease blood glucose levels. OBJECTIVES: The present experimental study was conducted in Iran and investigated the role of apelin, which is used to manage type 1 diabetes mellitus, during exercise training. MATERIALS AND METHODS: Male Wistar rats (n = 36) were assigned by simple random allocation to six groups (n = 6): non-diabetic (ND), diabetic (D), sedentary non-diabetic (SND), sedentary diabetic (SD), exercise non-diabetic (END), and exercise diabetic (ED). Diabetes was induced by a single subcutaneous injection of streptozotocin (50 mg/kg). Exercise training consisted of treadmill running 60 minutes/day × 5 days/week for 10 weeks. The tail-flick test was used to assess the thermal pain threshold, and an apelin enzyme immunoassay kit was utilized to assess plasma apelin levels. RESULTS: Plasma apelin level was higher (0.3 vs. 0.1, P < 0.0001) and the tail-flick latency was lower (2.2 vs. 3.8, P < 0.0001) in the D group than in the ND group. After the training program, plasma apelin levels decreased in the exercise groups, and the tail-flick latency increased in the ED group. No correlation was found between apelin blood concentrations and tail-flick latency following the training program in the ED group. CONCLUSIONS: These findings suggest that apelin does not play any significant role in regulating the pain threshold in type 1 diabetes mellitus during exercise training.

Microvascular disease is a major feature of type1 diabetes and results from long-standing structural and functional changes especially in the skin microvasculature. Magnesium (Mg) deficiency has recently been proposed as a novel factor implicated in the pathogenesis of diabetes complications such as vascular disturbance, but its mechanism of action is not completely elucidated. The present study was designed to determine whether chronic magnesium sulfate administration could control streptozocin-induced diabetes and improve endothelium-dependent and endothelium-independent dilatation, and identify its probable mechanism in the skin microvasculature of diabetic rats. Fifty male Wistar rats (220 ± 10 g) were divided into two diabetic and one control groups. One subgroup of diabetic received magnesium sulfate (10 g/l) in their drinking water, while two other groups had only tap water. Laser Doppler flow meter with iontophoresis was used to measure the relative changes in skin blood flow. We used acetylcholine (Ach), sodium nitroprusside (SNP), and N (w)-nitro-L-arginine (LNNA; NO synthase inhibitor) with magnesium sulfate (0.1 M) in control and experimental animal by microsyringe pump microinjection. SNP- and Ach-induced cutaneous perfusion increased significantly by Mg treatment in the diabetic groups, and local microinjection of magnesium sulfate (0.1 M) increased cutaneous blood flow in all groups (p < 0.01). However, the administration of LNNA prior to magnesium sulfate attenuated (p < 0.05) but not abolished the increase in cutaneous blood flow in diabetic and normal rats. From the results of this study, it may be concluded that Mg could improve skin microvasculature of diabetic rats with potentiation of nitric oxide pathway.
