Weaning is a critical period for pigs. Some plant extracts showing antioxidant, anti-inflammatory or antibacterial properties, provided to piglets and/or their dam, may improve piglets' robustness at weaning, thus reducing the need for antobiotics. This study investigated the effects of a maternal and/or a direct supplementation of piglets with a combination of plant extracts on sow and piglet performance and their metabolic, immune, inflammatory, and oxidative status during lactation and around weaning. Sixty-four sows were assigned to the control or treated group. Treated sows were supplemented with a powdered plant extracts supplement daily top-dressed on feed from day of gestation (DG) 106 to day of lactation (DL) 28 and a liquid solution top-dressed on feed on DG109. Within each sow group, litters were divided into two groups: a control piglet group and a treated piglet group. A single dose of a liquid solution was orally given to piglets in the treated piglet group. Piglets were weaned on DL28. Blood samples were collected from sows on DG94, DG112, and DL26 and from 2 piglets per litter on DL3, DL14, DL25, and 5 d postweaning to analyze indicators of metabolic, immune, inflammatory, and oxidative status. Colostrum and milk samples were collected at farrowing, DL6, and 26. Maternal supplementation had no effect on sow metabolic, immune, inflammatory, and oxidative status except for fewer lymphocytes on DG112 (Pâ <â 0.05) and a lower plasma concentration of non-esterified fatty acids on DL26 (Pâ <â 0.05). Maternal supplementation tended to decrease dry matter and gross energy (Pâ <â 0.10) and reduced fat and haptoglobin concentrations (Pâ <â 0.01) in milk on DL26. Maternal supplementation had no effect on piglets' growth performance and blood indicators during lactation and around weaning. On DL25, the direct supplementation of piglets decreased their neutrophils proportion (Pâ <â 0.05), increased the expression of genes encoding pro- and anti-inflammatory cytokines in whole blood culture in response to lipopolysaccharide (Pâ <â 0.05) and tended to decrease the oxidative stress index (Pâ =â 0.06). After weaning, these beneficial effects were no longer observed but the supplementation improved piglets' growth performance during the postweaning period (Pâ <â 0.05). Plant extract supplementation could thus modify the composition of mammary secretions and improve postweaning performance of piglets potentially related to the modification of their immune and oxidative status before weaning.

This study investigated the effects of a maternal and/or a direct supplementation of piglets with a combination of plant extracts on sow and piglet metabolic, immune, inflammatory, and oxidative status during lactation and around weaning. The maternal supplementation corresponded to a powdered supplement top-dressed on sow diet during late gestation and lactation and a liquid solution administered once 1 wk before parturition. The piglet supplementation was a liquid solution administered once on day 3 of age. The most concentrated components of the powder were extracts of fenugreek, Siberian ginseng, and cat's claw. The liquid solutions contained mostly oregano and eucalyptus essential oils. The maternal supplementation had few effects on sow immune, inflammatory, and oxidative status but modified milk composition at the end of lactation. It did not improve growth performance and the immune, inflammatory, and oxidative blood parameters of piglets around weaning. The direct supplementation of piglets modified their immune and oxidative status before weaning and increased their growth performance during the postweaning period, showing the potential of plant extracts as part of preventive strategies dedicated to improve piglets' robustness during the suckling and postweaning periods.

To meet the demands of the beef cattle sector in France, weaned beef calves are transported to sorting facilities and sorted into batches composed of animals of similar body weight (BW) before the beginning of the fattening period. This procedure aims to facilitate animal management. However, it leads to practices that affect animal welfare, health and performance, such as transporting weaned beef calves over long distances and mixing animals originating from different cow/calf farms. In contrast, other potentially beneficial practices, such as pre-weaning vaccination against bovine respiratory diseases (BRD), are seldom taken into consideration when batches are formed. This observational study, based on field data from 15,735 Charolais bulls, aimed to investigate which criteria should be favored for batch constitution by quantifying the effect of batch characteristics on the growth performance of young bulls during the fattening period. Clustering analysis was used to group young bulls exhibiting similar batch characteristics and define batch types. Associations between batch characteristics/batch types and individual growth performance/homogeneity of growth performance (mean and standard deviation (SD) of average daily gain (ADG) and fattening period duration) were studied using linear mixed models. The mean BW and the percentage of animals vaccinated against BRD before weaning were positively associated with ADG (+35 g/d for each additional 50 kg and +28 g/d for a high percentage of vaccinated animals, P < 0.05). In contrast, transportation distance was negatively associated with ADG (-12 g/d for each additional 120 km travelled). Mixing animals and BW homogeneity did not affect growth performance (P > 0.05). Only the mean BW and mixing animals negatively influenced the homogeneity of ADG (P < 0.01). The clustering analysis revealed that batches with the most BW heterogeneity, the least mixing, the shortest transportation distance and a high percentage of pre-weaning animals vaccinated against BRD had better growth performance compared to batches with the opposite characteristics (+61 g/d, P < 0.001). Our results suggest that major improvements of growth performance of fattening young bulls could be obtained by minimizing transportation distance, providing vaccination programs against BRD before weaning, and maintaining groups from the same cow/calf farm instead of constituting groups of animals with similar BW at the beginning of fattening.

Avoidance of novelty, termed neophobia, protects animals from potential dangers but can also impair their adaptation to novel environments or food resources. This behaviour is particularly well described in birds but the neurobiological correlates remain unexplored. Here, we measured neuronal activity in the amygdala and the striatum, two brain regions believed to be involved in novelty detection, by labelling the early gene c-fos following chicks exposure to a novel food (NF), a novel object (NO) or a familiar food (FF). NF and NO chicks showed significantly longer latencies to touch the food, less time eating and emitted more fear-vocalizations than control chicks. Latency to touch the food was also longer for NO than for NF chicks. Significantly higher densities of c-fos positive cells were present in all the nuclei of the arcopallium/amygdala of NF and NO chicks compared to FF chicks. Also, NO chicks showed higher positive cell densities than NF chicks in the posterior amygdaloid, the intermediate and the medial arcopallium. Exposure to novel food or object induced a similar increase in c-fos expression in the nucleus accumbens and the medial striatum. Our data provide evidence activation of the arcopallium/amygdala is specific of the type of novelty. The activation of striatum may be more related to novelty seeking.

Milk is produced in the udder by mammary epithelial cells (MEC). Milk contains MEC, which are gradually exfoliated from the epithelium during lactation. Isolation of MEC from milk using immunomagnetic separation may be a useful non-invasive method to investigate transcriptional regulations in ruminants' udder. This review aims to describe the process of isolating MEC from milk, to provide an overview on the studies that use this method to analyze gene expression by qRT PCR and to evaluate the validity of this method by analyzing and comparing the results between studies. In several goat and cow studies, consistent reductions in alpha-lactalbumin mRNA levels during once-daily milking (ODM) and in SLC2A1 mRNA level during feed restriction are observed. The effect of ODM on alpha-lactalbumin mRNA level was similarly observed in milk isolated MEC and mammary biopsy. Moreover, we and others showed decreasing alpha-lactalbumin and increasing BAX mRNA levels with advanced stages of lactation in dairy cows and buffalo. The relevance of using the milk-isolated MEC method to analyze mammary gene expression is proven, as the transcript variations were also consistent with milk yield and composition variations under the effect of different factors such as prolactin inhibition or photoperiod. However, the RNA from milk-isolated MEC is particularly sensitive to degradation. This could explain the differences obtained between milk-isolated MEC and mammary biopsy in two studies where gene expression was compared using qRT-PCR or RNA Sequencing analyses. As a conclusion, when the RNA quality is conserved, MEC isolated from milk are a valuable, non-invasive source of mammary mRNA to study various factors that impact milk yield and composition (ODM, feeding level, endocrine status, photoperiod modulation, and stage of lactation).
