RESUMO

Cholangiocarcinoma (CCA) is characterized by rapid onset and high chance of metastasis. Therefore, identification of novel therapeutic targets is imperative. E26 transformation-specific homologous factor (EHF), a member of the E26 transformation-specific transcription factor family, plays a pivotal role in epithelial cell differentiation and cancer progression. However, its precise role in CCA remains unclear. In this study, through in vitro and in vivo experiments, we demonstrated that EHF plays a profound role in promoting CCA by transcriptional activation of glioma-associated oncogene homolog 1 (GLI1). Moreover, EHF significantly recruited and activated tumor-associated macrophages (TAMs) through the C-C motif chemokine 2/C-C chemokine receptor type 2 (CCL2/CCR2) axis, thereby remodeling the tumor microenvironment. In human CCA tissues, EHF expression was positively correlated with GLI1 and CCL2 expression, and patients with co-expression of EHF/GLI1 or EHF/CCL2 had the most adverse prognosis. Furthermore, the combination of the GLI1 inhibitor, GANT58, and CCR2 inhibitor, INCB3344, substantially reduced the occurrence of EHF-mediated CCA. In summary, our findings suggest that EHF is a potential prognostic biomarker for patients with CCA, while also advocating the therapeutic approach of combined targeting of GLI1 and CCL2/CCR2-TAMs to inhibit EHF-driven CCA development.

Subsurface karst caves provide unique opportunities to study the deep biosphere, shedding light on microbial contribution to elemental cycling. Although ammonia oxidation driven by both ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) is well explored in soil and marine environments, our understanding in the subsurface biosphere still remained limited to date. To address this gap, weathered rock and sediment samples were collected from the Xincuntun Cave in Guilin City, an alkaline karst cave, and subjected to high-throughput sequencing and quantification of bacterial and archaeal amoA, along with determination of the potential nitrification rates (PNR). Results revealed that AOA dominated in ammonia oxidation, contributing 48-100% to the PNR, and AOA amoA gene copies outnumbered AOB by 2 to 6 orders. Nitrososphaera dominated in AOA communities, while Nitrosopira dominated AOB communities. AOA demonstrated significantly larger niche breadth than AOB. The development of AOA communities was influenced by deterministic processes (50.71%), while AOB communities were predominantly influenced by stochastic processes. TOC, NH4+, and Cl- played crucial roles in shaping the compositions of ammonia oxidizers at the OTU level. Cross-domain co-occurrence networks highlighted the dominance of AOA nodes in the networks and positive associations between AOA and AOB, especially in the inner zone, suggesting collaborative effort to thrive in extreme environments. Their high gene copies, dominance in the interaction with ammonia oxidizing bacteria, expansive niche breadth and substantial contribution to PNR collectively confirmed that AOA better adapted to alkaline, oligotrophic karst caves environments, and thus play a fundamental role in nitrogen cycling in subsurface biosphere.

OBJECTIVE: Our study was to elucidate the role of RNA helicase DEAD-Box Helicase 17 (DDX17) in NAFLD and to explore its underlying mechanisms. METHODS: We created hepatocyte-specific Ddx17-deficient mice aim to investigate the impact of Ddx17 on NAFLD induced by a high-fat diet (HFD) as well as methionine and choline-deficient l-amino acid diet (MCD) in adult male mice. RNA-seq and lipidomic analyses were conducted to depict the metabolic landscape, and CUT&Tag combined with chromatin immunoprecipitation (ChIP) and luciferase reporter assays were conducted. RESULTS: In this work, we observed a notable increase in DDX17 expression in the livers of patients with NASH and in murine models of NASH induced by HFD or MCD. After introducing lentiviruses into hepatocyte L02 for DDX17 knockdown or overexpression, we found that lipid accumulation induced by palmitic acid/oleic acid (PAOA) in L02 cells was noticeably weakened by DDX17 knockdown but augmented by DDX17 overexpression. Furthermore, hepatocyte-specific DDX17 knockout significantly alleviated hepatic steatosis, inflammatory response and fibrosis in mice after the administration of MCD and HFD. Mechanistically, our analysis of RNA-seq and CUT&Tag results combined with ChIP and luciferase reporter assays indicated that DDX17 transcriptionally represses Cyp2c29 gene expression by cooperating with CCCTC binding factor (CTCF) and DEAD-Box Helicase 5 (DDX5). Using absolute quantitative lipidomics analysis, we identified a hepatocyte-specific DDX17 deficiency that decreased lipid accumulation and altered lipid composition in the livers of mice after MCD administration. Based on the RNA-seq analysis, our findings suggest that DDX17 could potentially have an impact on the modulation of lipid metabolism and the activation of M1 macrophages in murine NASH models. CONCLUSION: These results imply that DDX17 is involved in NASH development by promoting lipid accumulation in hepatocytes, inducing the activation of M1 macrophages, subsequent inflammatory responses and fibrosis through the transcriptional repression of Cyp2c29 in mice. Therefore, DDX17 holds promise as a potential drug target for the treatment of NASH.

RNA modifications play a pivotal role in regulating cellular biology by exerting influence over distribution features and molecular functions at the post-transcriptional level. Among these modifications, N7-methylguanosine (m7G) stands out as one of the most prevalent. Over recent years, significant attention has been directed towards understanding the implications of m7G modification. This modification is present in diverse RNA molecules, including transfer RNAs, messenger RNAs, ribosomal RNAs, and other noncoding RNAs. Its regulation occurs through a series of specific methyltransferases and m7G-binding proteins. Notably, m7G modification has been implicated in various diseases, prominently across multiple cancer types. Earlier studies have elucidated the significance of m7G modification in the context of immune biology regulation within the tumor microenvironment. This comprehensive review culminates in a synthesis of findings related to the modulation of immune cells infiltration, encompassing T cells, B cells, and various innate immune cells, all orchestrated by m7G modification. Furthermore, the interplay between m7G modification and its regulatory proteins can profoundly affect the efficacy of diverse adjuvant therapeutics, thereby potentially serving as a pivotal biomarker and therapeutic target for combinatory interventions in diverse cancer types.

In this study, the contents of eight heavy metal(loid)s (As, Pb, Zn, Cd, Cr, Cu, Sb and Tl) in 50 sediment samples from a headwater of Beijiang River were studied to understand their pollution, ecological risk and potential sources. Evaluation indexes including sediment quality guidelines (SDGs), enrichment factor (EF), geo-accumulation index (Igeo), risk assessment code (RAC) and bioavailable metal index (BMI) were used to evaluate the heavy metal(loid)s pollution and ecological risk in the sediments. Pearson's correlation analysis and principal component analysis were used to identify the sources of heavy metal(loid)s. The results showed that the average concentration of heavy metal(loid)s obviously exceeded the background values, except Cr. Metal(loid)s speciation analysis indicated that Cd, Pb, Cu and Zn were dominated by non-residual fractions, which presented higher bioavailability. The S content in sediments could significantly influence the geochemical fractions of heavy metal(loid)s. As was expected, it had the most adverse biological effect to local aquatic organism, followed by Pb. The EF results demonstrated that As was the most enriched, while Cr showed no enrichment in the sediments. The assessment of Igeo suggested that Cd and As were the most serious threats to the river system, while Cr showed almost no contamination in the sediments. Heavy metal(loid)s in sediments in the mining- and smelting-affected area showed higher bioavailability. According to the results of the above research, the mining activities caused heavier heavy metal(loid)s pollution in the river sediment. Three potential sources of heavy metal(loid)s in sediment were distinguished based on the Pearson's correlation analysis and PCA, of which Cd, Pb, As, Zn, Sb and Cu were mainly derived from mining activities, Cr was mainly derived from natural sources, Tl was mainly derived from smelting activities.

Subterranean karst caves are windows into the terrestrial subsurface to deconstruct the dimensions of mycobiome fingerprints. However, impeded by the constraints of remote locations, the inaccessibility of specimens and technical limitations, the mycobiome of subterranean karst caves has remained largely unknown. Weathered rock and sediment samples were collected from Luohandu cave (Guilin, Southern China) and subjected to Illumina Hiseq sequencing of ITS1 genes. A total of 267 known genera and 90 known orders in 15 phyla were revealed in the mycobiomes. Ascomycota dominated all samples, followed by Basidiomycota and Mortierellomycota. The sediments possessed the relatively highest alpha diversity and were significantly different from weathered rocks according to the diversity indices and richness metrics. Fifteen families and eight genera with significant differences were detected in the sediment samples. The Ca/Mg ratio appeared to significantly affect the structure of the mycobiome communities. Ascomycota appeared to exert a controlling influence on the mycobiome co-occurrence network of the sediments, while Ascomycota and Basidiomycota were found to be the main phyla in the mycobiome co-occurrence network of weathered rocks. Our results provide a more comprehensive dimension to the mycobiome fingerprints of Luohandu cave and a new window into the mycobiome communities and the ecology of subterranean karst cave ecosystems.

Water quality is one of the most important environmental issues in the sustainable development of karst areas. To investigate heavy metal pollution and assess health risk in karst water basins around mines, 18 groups of water samples were collected from the river and groundwater in the Sidi River karst basin, and the concentrations of nine types of heavy metals(Cu, Pb, Zn, Cd, Mn, Fe, As, Cr, and Sr) were determined. Sample data were analyzed using principal component analysis, correlation analysis, water quality index, the Nemerow comprehensive pollution index, hazard quotient, and hazard index. The results showed that the Sidi River was slightly alkaline. The farther the river water samples were from the tailings reservoir, the lower were the concentrations of Cu, Pb, Zn, Cd, Mn, Fe, As, and Sr in the river water. Principal component and correlation analyses showed that heavy metals in the Sidi River karst basin mainly came from mine discharge(55.42%), carbonate weathering dissolution(21.41%), and human activities(14.72%). Eighty-two percent of the samples in the river and all the samples in the groundwater were excellent water. The Nemerow comprehensive pollution index in the river was 4.12 with strong pollution. All the hazard indices were below 1, and Pb, Zn, As, Cd, and Cr were potentially threatening metals in the Sidi River karst basin. The concentration of heavy metals changed significantly after entering the karst conduit, indicating that the unique properties of the karst aquifer affected the spatial variation of the heavy metal concentration. The results of this study can provide data reference for water resource prevention and human health protection in the Sidi River karst basin and similar karst basins.

The abnormal enrichment of cadmium (Cd) in soil caused by rock weathering and mining activities is an issue in southern China. Although the soil Cd content in these regions is extremely high, the bioavailability of Cd in the soils differs significantly. The carbonate area (CBA) and tin-mining area (TIA) in Hezhou City were investigated to determine the primary features of soil Cd mobility in these regions and improve environmental management. Lateral and vertical spatial distributions revealed different accumulation and migration mechanisms of soil Cd in the CBA and TIA. Further analyses revealed that mining activities and geological weathering resulted in different soil geochemical parameters, thus yielding significantly lower levels of Cd in rice grains in the CBA than in the TIA. The random forest (RF) model predicted the bioaccumulation factor (BAF) (R2 = 0.69) better than the support vector machine (SVM) model (R2 = 0.68). Subsequently, a novel land management scheme was proposed based on soil Cd and the prediction of Cd in rice to optimize the spatial resources of agricultural land and ensure the safety of rice for consumption. This study provides a novel approach for land management in Cd-contaminated areas.

BACKGROUND: N6-methyladenosine (m6A) is a prevalent modification of mRNA and is known to play important roles in tumorigenesis in many types of cancer. The function of N6-methyladenosine (m6A) RNA methylation depends on a variety of methyltransferases and demethylases. AlkB homolog 5 (ALKBH5) is a demethylase, and its biological function has not been completely explored in HCC. RESULTS: ALKBH5 is downregulated and has antitumor effects in HCC cells. In addition, Progestin and AdipoQ Receptor 4 (PAQR4) was identified as a downstream target of ALKBH5 based on transcriptome sequencing and validation studies. We found that ALKBH5 decreases PAQR4 mRNA and protein expression in an N6-methyladenosine (m6A)-dependent manner. The study also showed that ALKBH5 changes PAQR4 expression via the m6A reader IGF2BP1. In both in vivo and in vitro experiments, PAQR4 showed a strong association with the development of HCC. Finally, we found that PAQR4 interacts with AKT and enhances PI3K/AKT pathway activation. CONCLUSIONS: ALKBH5 inhibits HCC growth by downregulating PAQR4 expression in an m6A-dependent manner, therefore suppressing PI3K/AKT pathway activation.

Karst water quality is one of the most important environmental issues in karst areas. The study's purpose was to investigate dissolved heavy metal pollution and health risk assessment in karst water basins around mines. River water and groundwater samples were analyzed by principal component analysis, correlation analysis, water quality index, hazard quotient, and hazard index. Median concentrations of dissolved heavy metals in the Sidi River were similar to the world average with a slightly alkaline characteristic. The concentrations of most dissolved heavy metals in river water were higher than those in groundwater. The concentrations of Zn, Pb, and Cd around the mine exceeded the limits of drinking water indicators. The poor water quality samples with high water quality index values were distributed around the mine. Lead (Pb), Zn, As, Cd, and Cr were potentially threatening metals in the study area. The pollution level of dissolved heavy metals in the Sidi River was at a medium level compared with other rivers worldwide. Principal component analysis and correlation analysis showed that Cu, Pb, Zn, Cd, Mn, Fe, As, and Sr mainly came from mine drainage; Ca2+, Mg2+, and Cr mainly came from the contribution of carbonate rocks; Na+ and K+ were related to local human agricultural activities. The concentrations of dissolved heavy metals in groundwater were affected by karst aquifers. The results of this study can provide a data reference for water resources prevention and human health protection in the Sidi River's karst basin and similar karst basins.

N6-methyladenosine (m6A) is a widely investigated RNA modification in studies on the "epigenetic regulation" of mRNAs that is ubiquitously present in eukaryotes. Abnormal changes in m6A levels are closely related to the regulation of RNA metabolism, heat shock stress, tumor occurrence, and development. m6A modifications are catalyzed by the m6A writer complex, which contains RNA methyltransferase-like 3 (METTL3), methyltransferase-like 14 (METTL14), Wilms tumor 1-associated protein (WTAP), and other proteins with methyltransferase (MTase) capability, such as RNA-binding motif protein 15 (RBM15), KIAA1429 and zinc finger CCCH-type containing 13 (ZC3H13). Although METTL3 is the main catalytic subunit, WTAP is a regulatory subunit whose function is to recruit the m6A methyltransferase complex to the target mRNA. Specifically, WTAP is required for the accumulation of METTL3 and METTL14 in nuclear speckles. In this paper, we briefly introduce the molecular mechanism of m6A modification. Then, we focus on WTAP, a component of the m6A methyltransferase complex, and introduce its structure, localization, and physiological functions. Finally, we describe its roles and mechanisms in cancer.

The corrected version of Fig. 5 is shown on the next page, featuring the correct data for Fig. 5C and E, including new bar charts showing the quantification of these data. In this paper, the ultra-thin layer (UTL) is defined as the dense framework structure mixture made of asphalt binder, fine aggregate with nominal maximum aggregate size (NMAS) not greater than 13.2 mm and possible additives (mineral or organic), thickness of 2-4 cm. The study aims to investigate comprehensive performance of UTL asphalt mixture. The method of impact freeze thaw split test and the index of impact freeze-thawing damage degree (IFTDD) are proposed to reflect the durability. The indoor tire-rolling-down test system and accelerated abrasion machine are used to simulate the tire-pavement interaction and test road noise and skid resistance, respectively. Though evaluating the influencing factors (pavement thickness, gradation, asphalt binder type, and the content of KS additive) on durability, the optimum parameters with excellent durability are recommended. Combined with the test of noise and skid resistance, the factors affecting the surface function are analyzed. Moreover, the prediction mathematical model of skid resistance and the long-term safety benefit value Eeff are put forward. Results indicate that pavement thickness is the most significant factor effecting on durability, and gradation is the most significant factor affecting noise. Compared with KS additive, gradation has a greater influence on skid resistance index of Texture Depth (TD), whereas, KS additive is the most significant factor affecting British Pendulum Number (BPN). Furthermore, with the addition of asphalt rubber (AR), IFTDD and noise are reduced by 29.17% and 1.6 dB, and BPN and TD increase by 0.7 and 0.03 mm, remarkably. Compared with different asphalt types, the noise of UTL asphalt rubber mixture with 13.2 mm NMAS (UTL13 AR) is the lowest. Additionally, when KS content increases by 0.6%, the noise increases by 3 dB. Furthermore, on the basis of the calculation results of Eeff, UTL13 AR mixture with 0.5% KS has the best long-term benefit of pavement safety and is recommended for field project.

Construction and demolition waste material is of great potential for use in pavement engineering. This paper aims to investigate the feasibility of ceramic waste aggregate (CA) used in cooling asphalt pavement through a series of test methods and simulation techniques. Stone mastic asphalt (SMA) containing 10%, 20%, 30%, 40%, and 50% coarse ceramic waste aggregate (CASMAs) was first designed using the Marshall method. Afterward, the road performance and thermal insulation performance of the five different CASMAs were assessed by a comprehensive lab test, including a wheel rutting test, moisture susceptibility test, bending beam test, fatigue beam test, and indoor thermal insulation test. Finally, a 2D finite-element (FE) model was developed to investigate the transient thermal field and rutting deformation response of the cooling asphalt pavement with CASMAs. Results show that CASMAs experienced degradation of rutting resistance, moisture susceptibility, and anti-cracking performance while still meeting technical requirements with CA content of up to 40%. On the other hand, CASMAs can cool the pavement's temperature by 11.5 °C at the bottom of asphalt layers. The permanent rutting deformation of cooling asphalt pavement was 45.36% smaller than that of conventional asphalt pavement without CASMAs. Based on the test results and numerical simulation results, the optimum content of ceramic waste aggregate in stone mastic asphalt was recommended as 40%.

This study experimentally investigated heavy metal removal and accumulation in the aquatic plant Eichhornia crassipes. Pb, Zn, Cd, and Mn concentrations, plant morphology, and plant functional groups were analyzed. Eichhornia crassipes achieved high removal efficiency of Pb and Mn from karst water (over 79.5%), with high proportion of Pb, Zn, and Cd absorption occurring in the first eight days. The highest removal efficiencies were obtained at initial Pb, Zn, Cd, and Mn concentrations of 1 mg/L, 2 mg/L, 0.02 mg/L, and 0.2 mg/L, respectively. Eichhornia crassipes exhibited a high bioconcentration factor (Mn = 199,567 > Pb = 19,605 > Cd = 3403 > Zn = 1913) and a low translocation factor (<1). The roots accumulated more Pb, Zn, Cd, and Mn than the stolons and leaves due to the stronger tolerance of roots. The voids, stomas, air chambers, and airways promoted this accumulation. Pb, Cd, Zn, and Mn likely exchanged with Mg, Na, and K through the cation exchange. C≡C, C=O, SO42-, O-H, C-H, and C-O played different roles during uptake, which led to different removal and accumulation effects.

The interfacial zone between aggregate particles and asphalt mortar presents a significant effect on the strength of an asphalt mixture. In this paper, basalt, limestone, and diabase were selected, and the influence of these aggregates on the mechanical characteristics and microstructures of the interfacial zone was investigated. Nanoindentation was employed to measure the change law of mechanical behavior in the region of the interfacial zone, and corresponding viscoelastic parameters were deduced; microstructural morphology was observed by scanning electron microscopy, and the effect of the mineralogical components on the interfacial zone was analyzed as well. Results show that the mechanical behavior of the interfacial transition zone is complicated. The modulus and hardness of asphalt mortar decrease with the increases in the aggregate surface distance, and then keep stable after the distance is greater than 40 µm. Both the relaxation time and dissipated energy ratio of the interfacial zone affected by the different aggregate types show a similar change law. These states indicate that aggregate types have little influence on the stress dissipation of asphalt mortar. However, creep compliances that quantify the ability of the deformation resistance show a significant difference; microstructure morphologies of the interfacial zone are affected by aggregates obviously, and micro pores present a different distribution and state in the interfacial zone.

Karst water is rich in calcium ions (Ca2+) and exhibits poor metal availability and low biodegradation efficiency. This study sought to analyze the effects and mechanisms of Ca2+ on lead (Pb) removal and absorption by Eichhornia crassipes (a floating plant common in karst areas). Moreover, the morphology and functional groups of E. crassipes in water were characterized via SEM, and FTIR. The results demonstrated that the removal rate of Pb in karst water (85.31%) was higher than that in non-karst water (77.04%); however, the Pb bioconcentration amount (BCA) in E. crassipes roots in karst water (1763 mg/kg) was lower than that in non-karst water (2143 mg/kg). With increased Ca2+ concentrations (60, 80, and 100 mg/L) in karst water, the Pb removal rate increased (85.31%, 88.87%, and 92.44%), the Pb BCA decreased (1763, 1317, and 1095 mg/kg), and the Ca BCA increased (6801, 6955, and 9368 mg/kg), which was attributed to PbCO3 and PbSO4 precipitation and competitive Ca and Pb absorption. High Ca2+ concentrations increased the strength of cation exchange, alleviated the fracture degree of fibrous roots, reduced the atrophy of vascular bundles, protected the cell wall, promoted C-O combined with Pb, enhanced the strength of O‒H, SO42-, C=O, and reduced the oxidization of alkynyl acetylene bonds.

Most wild fish caught by anglers (WFAs) are likely to be contaminated by toxic metals, particularly the fish collected from the waterways in urban and suburban areas; hence, the determination of health risk caused by WFAs consumption associated with toxic metals is vital. Therefore, Liuzhou, one of the largest industrial cities in China, was considered as an example city in this study. Eight toxic elements were analysed to uncover the pollution status and consumption safety of WFAs. Moreover, the suitable angling waterways were identified in the urban and suburban areas. The obtained results suggested relatively high concentrations of Zn, Cr and Cd, which were also found to be beyond corresponding Maximum Residue Limit. Among all analysed elements, only the mean pollution indices of Cr and Cd were observed to be beyond 0.2, revealing that the observed WFAs were generally contaminated by these metals. However, the potential health risk of WFAs can be predominately attributed to Cr, confirmed by the significantly higher Target hazard quotients (THQ). For adults, all the THQ values were below 1, indicating no significant health risk being associated with WFAs consumption in the case of adults. On the contrary, all the THQ values for Children were beyond 1, suggesting children being susceptible to great health risks due to WFAs consumption. Furthermore, the weekly recommended consumption of WFAs in urban area is remarkably lower than the current rate of fish consumption observed among urban residents; therefore, the waterways in urban areas can be evaded for fish angling.

To investigate the environmental quality and human health risks of different types of groundwater at Xiangshui, Chongzuo, several regular water quality indexes and concentrations of metals in 60 groundwater samples were measured and analyzed. The environmental quality of groundwater was analyzed by means of the Nemerow index. The health risks were assessed by using a human health risk assessment model. The regular water quality indexes and concentrations of metals of the well water, spring water, and underground river water exceeded the standards to different degrees. The environmental quality of groundwater was at a poor grade. The comprehensive evaluation score of underground river water (F=4.26) was the lowest. The well water had the same score as spring water (F=7.10). The high hardness and salinity were conducive to enrichment of Cr, and the reducing environment was of great advantage for the enrichment of As. The environmental geochemistry of Zn, Pb, Cd, and Cu was similar. The sources of Fe, Al, and Mn were similar. The results of the health risk assessment indicated that the health risks of well water, spring water, and underground river water were relatively high. The health risks decreased in the order of well water > underground river water > spring water. The health risks mainly came from the carcinogenic metallic element Cr. Carcinogenic risks were 4-6 orders of magnitude higher than non-carcinogenic risks. Carcinogenic risks were higher than the maximum allowance levels (5.0×10-5 a-1). Non-carcinogenic risks were lower than the allowance levels (10-6 a-1). Children had greater health risks than adults. The health risks of metals through the drinking pathway were 2-3 orders of magnitude higher than the values caused by the dermal contact pathway. For the sake of drinking water safety, the well water, underground river water, and spring water should be properly treated and the concentration of Cr in groundwater should be controlled before drinking.
