RESUMO

It has been shown that endogenous opioid dermorphin induces effective analgesia after intranasal injection to fish (0.02-0.20 mg/kg) and rats (0.005 mg/kg). Maintenance of dermorphin analgesia after olfactory and trigeminal denervation indicates that the analgetic effect is not realized via the olfactory receptors or free terminals of the trigeminal nerve.

RESUMO

The effects of dermorphin and DSIP were studied in male Wistar rats chronically treated with ethanol. Both peptides (200 mg/kg, i. p., daily during 10 days) significantly reduced volitional alcohol intake for 3-4 weeks. These effects were associated with a marked decrease in serotonin and its metabolite 5-HIAA levels in the hypothalamus. The findings suggest that antialcoholic effects of both peptides are actualized through mechanisms involving serotoninergic system of the brain.

RESUMO

The aim of the study was to determine whether opioid peptides could modulate the intracardiac ganglionic excitation. It has been shown that postganglionic impulse activity recorded in the frog intracardiac nerve was inhibited by endogenous amphibian opioid peptide-dermorphin (10(-12)-10(-6) M). The inhibition was blocked by naloxone (10(-5) M). Dermorphin (10(-6) M) had no influence on transitory impulse activity in the intracardiac postganglionic sympathetic fibers. It is suggested that opioid peptides take part in the mediatory process in the intracardiac ganglia.

RESUMO

The analgetic activity of dermorphin and its analogue A-2 was assessed by the tail-flick test. Following intraperitoneal administration at doses 5 mg/kg and above the peptides showed significant effects. After intranasal application of the peptides a significant effect was observed in the range of low doses 0.001-0.1 mg/kg. After intranasal application of high dermorphin doses (1 or 5 mg/kg) the analgetic activity decreased. The effect of analogue A-2 lasted longer after intranasal, than after intraperitoneal administration. It is assumed that the neurophysiological mechanisms of the analgetic activity of dermorphins depend on the route of their administration.
