PURPOSE: To assess the effect of nanoglass (NG) particles and multiwalled carbon nanotubes' (MWCNTs) addition on Vickers hardness (VH), degree of conversion (DC), and abrasion resistance of 3D-printed denture base resin. MATERIALS AND METHODS: 3D-printed denture base resin was reinforced using silanized NG and MWCNTs to obtain four groups: Control, 0.25 wt% NG reinforced resin, 0.25 wt% MWCNTs reinforced resin, and a combination group of 0.25 wt% of both fillers. All specimens (N = 176) were tested before and after thermal aging (600 cycles) for VH (n = 22), DC, and abrasion resistance (n = 22). Abrasion resistance specimens were subjected to 60,000 brushing strokes, and then assessed for surface roughness (Ra) and weight loss. Specimens were then scanned with a benchtop scanner before and after abrasion to produce a color map of topographical changes from superimposed images. Data were analyzed using ANOVA tests followed by Tukey post hoc test. Kruskal-Wallis test was used to compare percent change among groups, followed by Dunn post hoc test (α = 0.05). RESULTS: The interaction between nanofiller content and thermal cycling displayed a significant effect on VH and DC. The 0.25% NG expressed the highest VH before aging but revealed the highest percent decrease after aging. Nanofiller content, thermal aging, and brushing displayed a significant interaction impact on the Ra values. CONCLUSIONS: The addition of nanofillers resulted in an overall improvement in resin microhardness and abrasion resistance. The 0.25% MWCNTs group revealed the lowest Ra with the least percent change in VH and DC, while the combination one displayed the least change in weight.

OBJECTIVE: Evaluation of biaxial flexural strength (BFS) of nanoglass (NG) and multiwalled carbon nanotubes (MWCNTs) reinforced 3D-printed denture base resins and their shear bond strength (SBS) to 3D-printed and acrylic denture teeth. METHODS: Silanized NG and MWCNTs were added to 3D-printed denture base resin to obtain four groups: Control, 0.25 wt% NG, 0.25 wt% MWCNTs, and a combination group with 0.25 wt% of both fillers. All specimens were tested before and after 600 cycles of thermal aging. BFS (n = 88) was tested using disk-shaped specimens (12 ×2 mm) centralized on an O ring in a universal testing machine. Weibull analysis was conducted to assess predictability of failure. SBS (n = 176) was tested for acrylic and 3D-printed denture teeth attached to bar-shaped specimens in a universal testing machine followed by failure mode analysis using stereomicroscope. Two and three-way ANOVA tests followed by Tukey post hoc test were conducted for BFS and SBS. Kruskal-Wallis test compared percent change among groups with subsequent Dunn post hoc test with Bonferroni correction (α = 0.05). RESULTS: BFS was affected significantly by filler content (P < 0.001) and thermal cycling (P < 0.001), with thermal cycling displaying the uppermost effect (È p2 =0.551). A significant interaction between filler content, thermal cycling, and teeth type was displayed by SBS results (P < 0.001, F=10.340, È p2 =0.162). The highest BFS values belonged to 0.25 % MWCNTs while the highest SBS to printed teeth was displayed by the combination. SIGNIFICANCE: The combination group displayed higher BFS and SBS to printed teeth compared to control which allows 3D-printed materials to have a long-term clinical success.

BACKGROUND: Cement spacer is essential for compensating deformation of zirconia restoration after sintering shrinkage, allowing proper seating and better fracture resistance of the restoration. Studies assessing the effect of cement spacer on fit accuracy and fracture strength of zirconia frameworks are missing in the literature. Therefore, the aim of this study was to evaluate the effect of different cement spacer settings on fit accuracy and fracture strength of 3-unit and 4-unit zirconia frameworks. METHODS: Sixty standardized stainless-steel master dies were manufactured with 2 prepared abutments for fabricating 3-unit and 4-unit zirconia frameworks. The frameworks were assigned into 6 groups (n = 10) according to cement spacer setting (30 µm, 50 µm, and 80 µm) as follows: 3-unit frameworks; 3u-30, 3u-50, 3u-80, and 4-unit frameworks; 4u-30, 4u-50, and 4u-80. The frameworks were assessed for fit accuracy with the replica method. The specimens were cemented to their corresponding dies, and the fracture strength was measured in a universal testing machine. The Weibull parameters were calculated for the study groups and fractured specimens were inspected for failure mode. Two-Way ANOVA followed by Tukey test for pairwise comparison between study groups (α = 0.05). RESULTS: The cement spacer had a significant effect on both fit accuracy and fracture strength for 3-unit and 4-unit frameworks. The 50 µm spacer had significantly better fit accuracy followed by 80 µm, and 30 µm spacers. Both 50 µm and 80 µm spacers had similar fracture strength, and both had significantly better strength than 30 µm spacer. CONCLUSIONS: For both 3-unit and 4-unit zirconia frameworks, 50 µm cement spacer can be recommended over 30 µm and 80 µm spacers for significantly better fit accuracy and adequate fracture strength.

RESUMO

The mechanisms underlying adult hippocampal neurogenesis (AHN) are not fully understood. AHN plays instrumental roles in learning and memory. Understanding the signals that regulate AHN has implications for brain function and therapy. Here we show that Caveolin-1 (Cav-1), a protein that is highly enriched in endothelial cells and the principal component of caveolae, autonomously regulates AHN. Conditional deletion of Cav-1 in adult neural progenitor cells (nestin +) led to increased neurogenesis and enhanced performance of mice in contextual discrimination. Proteomic analysis revealed that Cav-1 plays a role in mitochondrial pathways in neural progenitor cells. Importantly, Cav-1 was localized to the mitochondria in neural progenitor cells and modulated mitochondrial fission-fusion, a critical process in neurogenesis. These results suggest that Cav-1 is a novel regulator of AHN and underscore the impact of AHN on cognition.

This study aimed to evaluate carbon nanotubes-hydroxyapatite nanocomposites as bioactive titanium implant coats and to assess the effect of near-infrared radiation on these nanocomposites. Carbon nanotubes were acid-functionalized, and hydroxyapatite was prepared by the wet-chemical precipitation method. Both precursors were used to prepare the carbon nanotubes-hydroxyapatite nanocomposites in two concentrations of hydroxyapatite (0.5 and 1 wt.%). The formed nanocomposites were characterized and used to coat silanized titanium discs and cylinders. Half the specimens of each group were radiated by near-infrared laser, then wettability and shear bond strength were tested for all specimens. Bioactivity was tested by monitoring the formation of calcium phosphate compounds after soaking in simulated body fluid. A significant increase in wettability and bond strength was found in the radiated coats compared to the non-radiated ones with the 1% hydroxyapatite group showing the highest values followed by 0.5% hydroxyapatite then the carbon nanotubes group. The two-way ANOVA test showed that both the difference in material and the laser treatment have had a statistically significant contribution to the increase in wettability and bond strength. The radiated groups also contributed to the formation of more calcium phosphate crystals of larger sizes and higher degrees of crystallinity.

Dietary nitrate and nitrite are sources of gastric NO, which modulates blood flow, mucus production, and microbial flora. However, the intake and importance of these anions in infants is largely unknown. Nitrate and nitrite levels were measured in breast milk of mothers of preterm and term infants, infant formulas, and parenteral nutrition. Nitrite metabolism in breast milk was measured after freeze-thawing, at different temperatures, varying oxygen tensions, and after inhibition of potential nitrite-metabolizing enzymes. Nitrite concentrations averaged 0.07 ± 0.01 µM in milk of mothers of preterm infants, less than that of term infants (0.13 ± 0.02 µM) (P < .01). Nitrate concentrations averaged 13.6 ± 3.7 µM and 12.7 ± 4.9 µM, respectively. Nitrite and nitrate concentrations in infant formulas varied from undetectable to many-fold more than breast milk. Concentrations in parenteral nutrition were equivalent to or lower than those of breast milk. Freeze-thawing decreased nitrite concentration ~64%, falling with a half-life of 32 minutes at 37°C. The disappearance of nitrite was oxygen-dependent and prevented by ferricyanide and 3 inhibitors of lactoperoxidase. Nitrite concentrations in breast milk decrease with storage and freeze-thawing, a decline likely mediated by lactoperoxidase. Compared to adults, infants ingest relatively little nitrite and nitrate, which may be of importance in the modulation of blood flow and the bacterial flora of the infant GI tract, especially given the protective effects of swallowed nitrite.

OBJECTIVE: To measure the circulating concentrations of nitric oxide (NO) adducts with NO bioactivity after inhaled NO (iNO) therapy in infants with pulmonary hypertension. STUDY DESIGN: In this single center study, 5 sequential blood samples were collected from infants with pulmonary hypertension before, during, and after therapy with iNO (n = 17). Samples were collected from a control group of hospitalized infants without pulmonary hypertension (n = 16) and from healthy adults for comparison (n = 12). RESULTS: After beginning iNO (20 ppm) whole blood nitrite levels increased approximately two-fold within 2 hours (P<.01). Whole blood nitrate levels increased to 4-fold higher than baseline during treatment with 20 ppm iNO (P<.01). S-nitrosohemoglobin increased measurably after beginning iNO (P<.01), whereas iron nitrosyl hemoglobin and total hemoglobin-bound NO-species compounds did not change. CONCLUSION: Treatment of pulmonary hypertensive infants with iNO results in increases in levels of nitrite, nitrate, and S-nitrosohemoglobin in circulating blood. We speculate that these compounds may be carriers of NO bioactivity throughout the body and account for peripheral effects of iNO in the brain, heart, and other organs.
